1.Establishment of modified gas chromatography-mass spectrometry for determination of plasma sufentanil concentrations

Objective To modify gas chromatography-mass spectrometry (GC-MS) for determination of plasma sufentanil concentrations.Methods Fentanyl was used as the internal standard.The plasma samples were extracted with ethyl acetate and petroleum ether.An HP-5MS capillary column was used.The initial temperature of the column was set at 130 ℃,and the final temperature was 320 ℃.The injector port temperature was set at 290 ℃,and the interface temperature was 300 ℃.The carrier gas was high purity helium (purity 99.999%) with a constant flow rate of 1 ml/min.The injection volume was 1 μl with splitless injection.The MS conditions were as follows:EI source,ion source temperature 230 ℃,four-pole temperature 150 ℃,electron bombardment energy 70 eV,multiply voltage 2112 V,selective ion monitoring mode,solvent delay 7 min.The characteristic ions of sufentanil and fentanyl were obtained in 8.20-8.40 min and 7.60-7.80 min,respectively,according to the characteristic ion curve of sufentanil and fentanyl.Results The standard curve of sufentanil had good linear relationship in the range of 0.02-10.00 ng/ml and the equation was Y =0.1625X + 0.0316,R2 =0.9987.The extraction recovery was more than 80%,intra-day and inter-day determinations were less than 8%,and the limit of detection was 0.02 ng/ml.Conclusion When modified GC-MS is applied to determine the concentrations of sufentanil in plasma,not only the determination is sensitive and accurate,but also the procedures are simplified,and it is more suitable for the pharmacological research of sufentanil.
2.Effect of extracorporeal circuit on the concentration of sufentanil the priming solution

Objective To investigate the effect of extracorporeal cjrcuit on the concentration of sufentanil the priming solution.Methods The extracorporeal circuit (ECC) of Xi-jing-90 type (group A) was used in the study,while incontrol group (group B) a glass container was used.The ECC and glass container were filled with priming solution (succinylated Gelatin 1000 ml + lactated Ringer's solution 1000 ml).Sufentanil 15 μg was then added to the priming solution (the final concentration was 7.5 ng/ml).The priming solution was circulated in the closed ECC or stirred in the glass container.The concentration of sufentanil in the priming solution was determined at 3,5,10,20,30,40,50,60,70,80 and 90 min after addition of sufentanil by gas chromatography.Results The sufentanil concentration in.the priming solution decreased in group A at the different time points respectively as compared with group B (P < 0.05).Conclusion Sufentanil can significantly be absorbed by the extracorporeal circuit.

Objective To evaluate the effects of SHENG JI YU HONG herbs loading on modified gelatin sponges on the wound healing of mechanical trauma.Methods The round size of 1.8cm diameter of full thickness back skin was cut mechanically and covered with petrolatum gauzes,modified gelatin sponges,SHENG JI YU HONG plasters and SHENG JI YU HONG herbs loading on modified gelatin sponges respectively.On day3,day7 and day14 after model mading,the wound surface healing rate,the amount of hydroxyproline,hemoglobin,the number of capillaries and the cells with positive rhVEGF165 by immunochemical method inside granulation of the wound were detected individually.Results On day3,the wound surface healing rate and the number of capillaries and the cells with positive rhVEGF165 of SHENG JI YU HONG herbs loading on modified gelatin sponges were significant enhanced than those in the other groups.On day14,all the experimental indicators of SHENG JI YU HONG herbs loading on modified gelatin sponges were better than those in the other groups with P
4.Analysis of urinary ostomy bag to reduce urinary tract infections in ICU patients with diarrhea

Objective To analyze the effect of urinary ostomy bag on reduction of urinary tract infection in ICU patients with diarrhea.Methods A total of 445 diarrhea patients with indwelling catheter were admitted from June 2013 to May 2014,patients were divided into two groups according to the occurrence of diarrhea after admission,219 cases were in the observation group,226 cases were in the control group.The observation group used urinary tract ostomy bag connected with Kangwei anti-inverse drainage device,the control group used traditional methods of perianal care.Then,the incidence of urinary tract infection between two groups and the results of urine culture between patients of two groups with urinary tract infection were compared.Results The incidence of urinary tract infection in the observation group was 5.5% (12/219),significantly lower than that of the control group,10.6% (24/226),x2=3.952,P<0.05.The urine culture results showed that 4 cases were intestinal strains in the observation group,and 17 cases in the control group,the difference was significant,x2=4.629,P<0.05.Conclusions Urinary tract ostomy bag connected with Kangwei anti-inverse drainage device can effectively reduce the incidence of urinary tract infection in patients with diarrhea,it is simple to operate,and can reduce the workload of nurses and increase the comfort degree of patients,which is worthy of clinical application.
5.Preparation and anti-carcinoma effect of magnetic polybutylcyanoacrylate nanosphere loaded with aclacinomycin A

Objective To prepare magnetic polybutylcyanoacrylate (PBCA) nanosphere loaded with aclacinomycin A (ACM) and investigate its anti carcinoma effect and toxicity. Methods Magnetic PBCA nanospheres loaded with ACM were prepared by interfacial polymerization method. Female BABL/c nude mice were injected with MKN 45 gastric carcinoma cell line mass subcutaneously near right forefoot to establish human gastric carcinoma model. The mice were divided into 5 groups with 6 mice in each group: ACM (8 mg/kg), nanosphere with low dosage of ACM(1.6 mg/kg), nanosphere with high dosage of ACM (8 mg/kg ), magnetic nanosphere without ACM and normal saline (NS). Magnets (2500 Gs) were embedded in the tumor mass of all the mice one day before the date of therapy. The agents were administered again the same as the first time after 5 days. Tumor weight was measured and assay of colony forming unit granulocyte and macrophage (CFU GM) was performed on semi solid culture. Results The drug content of the agent was 12.0% and the mean particle size was 210 nm. The tumor inhibition rates of ACM, nanosphere with low dosage of ACM, nanosphere with high dosage of ACM and carrier without ACM on human gastric carcinoma in nude mice were 22.63%, 30.66%, 52.55% and 10.22% respectively. Objective To explore the difference of the oxytocin levels between social phobia patients and normal controls,and the relationship of symptoms severity of social phobia with the oxytocin levels and the relationship of drug effects with oxytocin levels.Methods Twenty seven 16-26 years old social phobia patients and thirty one normal controls were tested by an enzyme-linked immunosorbent assay to evaluate the oxytocin levels,and the Liebowitz Social Anxiety Scale was used to evaluate the symptoms severity of social phobia in patient group.Paroxetine was used to treat the patients with the drugs of 20mg per day,the Clinical Global Impression Scale were used to evaluate the drug effects after 4 weeks treatment.Results Mann-Whitney test showed there were significantly differences in oxytocin level between patients and controls ( (202.93 ± 145.06) pg/ml vs ( 152.29 ± 101.89 ) pg/ml,Z =- 1.307,P=0.030).Multiple liner regression analyses showed that the level of oxytocin was an impact factor of the severity of social phobia symptom (adjusted R2 =0.158,F=5.888,P=0.023 ).Logistic regression analyses showed that the level of oxytocin was an impact factor of the effective of drug treatment( OR=3.132,P =0.029 ).Conclusion This study indicate that the value of plasma oxytocin maybe significantly differences between social phobia and normal controls and the plasma oxytocin level maybe a factor which influence the symptoms severity and the effective of drug treatment in social phobia patients.

Acute myeloid leukemia (AML),the most common disease in acute leukemia,is a highly heterogeneous invasive hematological disease.The t(8;21)(q22;q22) translocation is the most common chromosomal translocation in AML,generating AML1-ETO fusion gene and encoding AML1-ETO fusion protein.This article summarizes the two-hit hypothesis in AML occurrence,the pathogenesis of t(8;21)AML,all features involved in t(8;21)AML,and the function of the components in AML1-ETO fusion protein,providing important basic information for the treatment and prognosis of t(8;21)AML.Meanwhile,this article also summarizes the progress of next generation sequencing technique in leukemia,providing a new technique for the accurate therapy of (8;21)AML.
