1.Abnormalities of sex chromosome in patients with mental disorders

Subject To explore the features of the abnormalities of the sex chromatin and the sex chromosome of the patients with mental disorders. Methods The sex chromatin in the cells of buccal mucous membrane of the 2 784 patients with mental disorders were examined by using a blinding and random way. The chromosome karyotypes in peripheral blood were further examined in those whose measure showed abnormal. The comparison group consisted of 1 069 normal persons.Results Compaired with the control group,the average frequency of Y chromatin in the male patients [(28.0±6.6)%] and the average frequency of X chromatin in the female patients [(22.3±8.0)%] rose significantly ; the average frequency of X chromatin in the female patients of the schizophrenia subgroup and the affective disorder subgroup were higher than that of neurosis and psychogenic mental disorder subgroup. X chromatins of eight male patients were positive and the karyotypes of the abnormal X chromosome were 47,XXY in 3 cases and 47,XXY/46,XY in 5 cases, the rate of 47,XXY was 7.7 , it rose significantly to the control group. X chromatin of two female patients were negative and the karyotypes of the abnormal X chromosome was 45,X/46,XX, the rate of 45,X was 1.2 ; one female patient had double X chromatin and the karyotypes of the abnormal X chromosome was 47,XXX/46,XX,there is no abnormal sex chromatin and sex chromosome in the control group.Conclusions The higher rate of the sex chromatin might be closely associated with the tendency of heredity of mental disorders , and the rate of the lacking X chromosome in females and the increasing X chromosome in males rise in the patients with mental disorders.
2.Study on the Protective Effect Mechanism of Total Flavones from Prunus cerasifera Fruits for Alcoholic Liver Disease in Rats

OBJECTIVE:To study the protective effects of total flavone from Prunus cerasifera fruits(PCE)on alcoholic liver disease(ALD)in rats,and provide experimental basis for developing new medicines for anti-ALD. METHODS:40 rats were ran-domly divided into normal group(distilled water),model group(distilled water),silibinin group [positive control,30 mg/(kg·d)] and PCE high-dose,low-dose groups [80,40 mg/(kg·d)],8 in each group. All rats were intragastrically administrated(10 mL/kg) every morning,once,for 6 weeks;meanwhile,except for normal group,rats in other groups received 50% alcohol(10 mL/kg) once intragastrically every afternoon to induce ALD model. After administration,heart,liver,spleen and other organ indexes,sub-cutaneous fat,brown fat,abdominal fat indexes were determined,as well as serum biochemical indexes [glutamate transaminase (ALT),aspartate aminotransferase(AST),total bilirubin(TBIL),direct bilirubin(DBIL),indirect bilirubin(IBIL),gamma-glu-tamyl transpeptidase(GGT),and total cholesterol(TC),triglyceride(TG),high-density lipoprotein(HDL),low-density lipopro-tein (LDL)] level and liver biochemical indexes [superoxide dismutase (SOD),malondialdehyde (MDA),glutathione peroxidase (GSH-Px),TC,TG] and TC,TG levels in feces;pathological changes of liver and kidney tissues were observed. RESULTS:Compared with normal group,heart,liver,spleen indexes,subcutaneous fat,abdominal fat indexes in model group were in-creased(P<0.05 or P<0.01),brain index and brown fat index were decreased(P<0.05);HDL level and HDL/TC ratio in serum were decreased,other serum indexes were increased(P<0.05 or P<0.01);SOD,GSH-Px levels in liver tissue were decreased, other above-mentioned liver biochemical indexes were increased(P<0.01);TC,TG levels in feces were increased(P<0.01);liv-er and kidney showed obvious lesions. Compared with model group,the above-mentioned indexes in silibinin group and PCE high-dose group were significantly improved (P<0.05 or P<0.01);ALT,AST in serum and MDA level in liver tissue in PCE low-dose group were significantly decreased(P<0.01),and SOD,GSH-Px levels in liver tissue were significantly increased(P<0.05);lesion degree of liver and kidney and lipid accumulation in liver were reduced in administration groups. CONCLUSIONS:PCE may play a role in anti-ALD by anti-oxidation,promoting liver cell regeneration and regulating lipid metabolism.
3.Effects of glycyrrhizin on activities of peritoneal macrophages of mice

Objective:To investigate the effect of Glycyrrhizin on the cytokines derived from peritoneal macrophages in mice.Methods:Glycyrrhizin was intraperitoneally administered 24 hr before the peritoneal macrophages (PMs) were harvested.The harvested PMs were then stimulated in vitro with lipopolysaccharide (LPS).The levels of tumor necrosis factor (TNF)-?,interleukin-12 (IL-12) p70,interleukin-10 (IL-10) and macrophage colony-stimulating factor (M-CSF) from culture supernatants were measured by ELISA.Results:Glycyrrhizin suppressed LPS-induced TNF-? production and increased LPS-induced IL-12 p70 production by PMs significantly. The production of IL-10 and M-CSF by PMs were not effected by Glycyrrhizin pretreatment.Conclusion:These findings demonstrate the ability of Glycyrrhizin to suppress LPS-induced TNF-? poduction and to enhance IL-12 production by peritoneal macrophages.
4.Expression of CXC chemokine receptor 4 in clear cell renal cell carcinoma

Objective To investigate the expression of CXC chemokine receptor 4 (CXCR4) in clear cell renal cell carcinoma (ccRCC) and its relationship with the clinical pathological parameters of ccRCC.Methods The expression of CXCR4 was detected by immuno-histochemistry method in 63 cases of ccRCCs,20 cases of para-carcinoma tissues and 20 cases of normal renal tissues.The correlation between expression level of CXCR4 and clinical pathological parameters of ccRCC patients were analyzed,and the clinical significance of its expression in ccRCC was evaluated.Results The positive expression rate of CXCR4(49.2%) in ccRCCs was significantly higher than that in para-carcinoma tissues (15%) and normal renal tissue (10%),and the difference was statistically significant (P < 0.05).The expression level of CXCR4 and the clinical stage and pathological grade of ccRCC were correlated (P < 0.05),and was associated with lymph node transfer (P < 0.05).The CXCR4 negative group overall survival rate [55.2% (16/29)] and the average survival time(46 months) was significantly better than the positive group [38.5% (10/26),32 months;P < 0.05].Conclusions The expression level of CXCR4 in ccRCC is correlated with the clinicopathological parameters and prognosis.CXCR4 is expected to be an important marker for diagnosis and prognosis evaluation of renal cell carcinoma.
5.Mutational analysis of NPHS1 gene in children with sporadic steroid-resistant nephrotic syndrome in Southern Chinese Han ethnic group

Objective To elucidate the mutations of NPHS1 gene in children with sporadic steroid-resistant nephrotic syndrome (SRNS) in Southern Chinese Han ethnic group.Methods Peripheral blood samples were collected for genetic analysis from 40 patients with sporadic SRNS and 50 healthy volunteers as control.Genomic DNA was isolated from peripheral blood leucocytes.Twenty-nine exons and exon-intron boundaries of the NPHS1 gene were amplified by polymerase chain reaction.Mutational analysis was performed by DNA sequencing directly.Results Seven variants,928G>A(D310N),2677A>G (T893A),2869G>C (V957L),IVS8+30C>T,IVS21+14G>A,IVS25-23C>T and *142T>C,of NPHS1 gene were found in 6 of 40 children with sporadic SRNS,whereas they were not found in 50 healthy controls.2677A >G,IVS8 +30C >T,IVS21 +14G>A,IVS25-23C >T and *142T>C were novel.Moreover,thirteen already reported NPHS1 polymorphisms,294C>T,349G>A,IVS3+15C>T,IVS3+61A>G,803G>A,IVS8+68A>G,1339G >A,1802G >C,2223C >T,2289C >T,IVS24 +36C >T,3315G>A and IVS27 +45C >T,were detected in some patients and controls. Conclusions NPHS1 mutations in 6 of 40 children with sporadic SRNS in Southern Chinese Han ethnic group (15%) are detected.NPHS1 mutations are existed in Southern Chinese children,so it is necessary to perform the mutation analysis of NPHS1 gene in those children patients.

Acute myeloid leukemia (AML),the most common disease in acute leukemia,is a highly heterogeneous invasive hematological disease.The t(8;21)(q22;q22) translocation is the most common chromosomal translocation in AML,generating AML1-ETO fusion gene and encoding AML1-ETO fusion protein.This article summarizes the two-hit hypothesis in AML occurrence,the pathogenesis of t(8;21)AML,all features involved in t(8;21)AML,and the function of the components in AML1-ETO fusion protein,providing important basic information for the treatment and prognosis of t(8;21)AML.Meanwhile,this article also summarizes the progress of next generation sequencing technique in leukemia,providing a new technique for the accurate therapy of (8;21)AML.

9.The Factors Affecting the Rehabilitation Efficacy after the Operation of Adults Cochlear Implantation

Objective We will discuss the related factors affecting the rehabilitation efficacy after the opera-tion of adults cochlear implantation .Methods Nijmegen Cochlear Implant Questionnaire'was adopted .There were questionnaire surveys on 29 adult patients with cochlear implantation .The individual scores and total scores of all subjects were collected together with the personal information ,including age ,bilateral effects and other factors . They were grouped in accordance with different grouping standards of different affecting factors .Results Compared to the communication manner of long -term hearing loss ,the rehabilitation efficacy of the group using lip -reading preoperatively was greater than the group using sign language or writing ;the rehabilitation efficacy of the group with the aggregate time of using CIs more than two years was superior to that similar group of less than two years , and the average P within the group of these two factors was <0 .05 .There was no statistical significance among group comparisons for other factors as the P was >0 .05 .Conclusion Ages ,the bilateral deafness ,and auditory dep-rivation have no obvious effects on the rehabilitation efficacy after the operation .The patients with postlingual deafness can improve their hearing with proper rehabilitation .Lip-reading preoperatively has positive effects on the rehabilitation effica-cy after the operation ;the length of using CIs has become the major factor affecting the rehabilitation efficacy .
10.Study on application effect of cyclooxygenase-2 inhibitor in perioperative pain management in patients with laparoscopic cholecystectomy

Objective To investigate the application effect of selective cyclooxygenase-2 (COX-2) inhibitor in perioperative preemptive on-demand analgesia of the patients with laparoscopic cholecystectomy under enhanced recovery after surgery(ERAS).Methods The clinical data in 206 cases of gallstones undergoing selective COX-2 inhibitor for preemptive analgesia (new type analgesic group) from June to December 2015 and 198 cases of gallstones undergoing tramadol postoperative analgesia (traditional analgesia group) in the biliary surgery department of West China Hospital of Sichuan University were retrospectively analyzed.The intraoperative anesthesia schemes in the two groups were consistent.The same pain resolution scheme was adopted after operation.Then the VAS pain score,pain relief drug use rate,adverse reactions,analgesic satisfaction and hospitalization time were compared between the two groups.Results The VAS scores at postoperative 2,6,12,24 h in the new type analgesia group were lower than those in the traditional analgesia group,the difference was statistically significant (P<0.05);the analgesic drug use rate in the new type analgesia group was lower than that in the traditional analgesia group (14.56% vs.44.95%,P<0.05),and the use rate of tramadol hydrochloride and pethidine hydrochloride was lower than that in the traditional analgesia group (P<0.05).The incidence rate of adverse reactions in the new type analgesia group was lower (2.43% vs.36.36%,P<0.05).The incidence rate of nausea and vomiting in the new type analgesia group was lower than that in the traditional analgesia group (P<0.05),and the incidence rate of other complications had no statistically signifiwas higher than that in the traditional analgesia group (P<0.05);the average hospital stay and postoperative hospital stay had no statistical difference between the two groups (P>0.05).Conclusion COX-2 inhibitors can effectively reduce perioperative pain degree in the patients with laparoscopic cholecystectomy,reduces the use frequency of analgesic drugs,shortens the hospital stay time and increases the patient satisfaction.cant difference (P>0.05).The perioperative patient analgesia satisfaction the in the new type analgesia group
