1.Performance verification of portable Celercare M1 analyzer in CRP determination

Objective To verify the analytical performance of the portable Celercare M1 analyzer in the C-reactive protein(CRP) quantitative determination.Methods According to the standard program of the American Clinical and Laboratory Standards Insti-tute (CLSI),The portable Celercare M1 analyzer was performed the verification on the CRP detection in the aspects of the linearity range,precision,accuracy,interference test and method comparison.Results The CRP detection showed good linearity in the range of 0.5-200 mg/L and the regression equation was Y =0.979X +0.456.The within-run coefficients of variation(CV)of samples with low and high CRP levels were 5.9% and 2.0%,respectively.The between-day CV of samples with low and high CRP levels were 6.3% and 2.2%,respectively.The accuracy of the assay was admirable,and bias of CRP results at low and high levels were 2.3% and 0.7%,respectively.No significance interference was observed when serum bilirubin ≤340 μmol/L,triglyceride ≤10 mmol/L and hemoglobin ≤8 g/L in this method.The regression equation of Celercare M1 analyzer(Y )and the IMMAGE 800 (X) for detecting CRP was Y =0.944X +0.206,R 2 =0.996(P <0.05).The relative bias of CRP results in three medical decision levels (3,10,100 mg/L)was 1 .0%,4.0% and 5.0%,repectively,which was lower than 1/2 of total error allowance.Conclusion The linearity,precision,accuracy and interference of the Celercare M1 analyzer in CRP determination are admirable and can be accepted in clinical practice.

Objective: To investigate the antitumor effect of lactobacillus peptidoglycan (PG) and explore its possible mechanisms. Methods: The inhibition for colon tumor growth, and the influence on the activities of CTL, NK, PM?, IL-1, IL-2, TNF-?, IFN-?and nitric oxide secretion were investigated in mice by cell culture, DNA agrose gel electrophoresis, respectively. Results: Growth in vivo of CT26 colon tumor was significantly inhibited by ip injection of PG. PG also induced significantly the activities of IL-1, TNF-?, IFN-?, CTL, NK, PM?and the production of nitric oxide, but had no effect on the activity of IL-2. Conclusion: The peptidoglycan derived from lactobacillus inhibited the colon tumor growth through activation of murine immunity. The antitumor effect of PG maybe partly if not all, induced, by the enhanced production of IFN-?, TNF-?and NO, through activation of NK cell and PM?. 4.Effect of dihydroarteminisin against Schistosoma japonicum in vivo

Objective To investigate the sensitivity of dihydroartemisinin against different developmental stages of Schistosoma japonicum, so as to understand the effect of dihydroarteminisin against on S. japonicum. Methods Mice were infected with 40 S. japonicum cercariae on the abdomen. Dihydroartemisinin was given intragastrically at different developmental stages of S. japonicum , and the mice were sacrificed 50 days post-infection, the adult worms were collected, and the worm reduction rates and female worm reduction rates were calculated. ① On the 2nd h, 3rd, 5th, 7th, 10th, 14th, 18th, 21st, 28th, 35th day post-infection, the mice were administered intragastrically with dihydroartemisinin at a single dose of 300 mg/kg, and the effect of dihydroarteminisin on different developmental stages of S. japonicum were observed. ② The mice were administered with different doses of dihydroartemisinin on the 7th or 35th day post-infection, and the dose-effect was explored. ③ The mice were administered on the 7th day post-infection and re-administered on the 35th day post-infection, respectively with different doses of dihydroartemisinin, and the effect was evaluated. Results The dihydroartemisinin at a single dose of 300 mg/kg had obvious effect on 7-day schistosomula and 35-day adult worms, with 64.81% and 60.47% of worm reduction rates and 73. 81% and 90.48% of female worm reduction rates, respectively. When the mice on the 7th day post-infection were administered with 200, 300, 400 and 600 mg/kg dihydroartemisinin, the worm reduction rates were 46.84% , 60.63% , 59.55% and 60. 21% , respectively, and the female reduction rates were 59.73% , 72.29% , 72.58% and 76.61 % , respectively. While in the mice on the 35th day post-infection, the corresponding rates were 47. 23% , 62.33% , 76.31% and 83.63% , respectively, and 59. 73% , 89. 36% , 89.65% and 93.96% , respectively. When the mice were treated twice with dihydroartemisinin on the 7th day and 35th day post-infection, the worm reduction rates were 58. 16% , 82.66% ,83.42% and 83.79% , respectively, and the female worm reduction rates were 68.69% , 90.43% , 93.74% and 94.63% , respectively. Conclusions Dihydroartemisinin has effect against S. japonicum, and the 7-day schistosomulum and 35-day adult worm are sensitive to the drug.
5.Comparative analysis of common biochemical assay results for venous blood and peripheral blood

Objective To investigate the comparability of the results of common biochemical items detected by Celercare M1 an‐alyzer in the peripheral blood and the venous whole blood .Methods The samples of peripheral blood and venous whole blood were collected from subjects .The biochemical items including Mg2+ ,Cl- ,tCO2 ,K+ ,Na+ ,Ca2+ ,α‐HBDH ,LDH ,AST ,CK ,CK‐MB ,TP , ALB ,TBIL ,ALT ,GGT ,ALP ,UREA ,GLU ,UA ,CHOL ,and HDL‐C were determined by Celercare M1 analyzer ,and the results were compared .Results The tCO2 results of venous blood was significantly higher than that of peripheral blood (P<0 .05) .How‐ever ,the results of α‐HBDH ,LDH ,CK and CK‐MB of venous blood samples were significantly lower than those of peripheral blood samples ,and the difference was statistically significant (P<0 .05) .Conclusion The peripheral blood can replace venous blood for biochemical analysis on Celercare M1 analyzer ,except for the electrolyte test items and cardiac enzyme items such as α‐HBDH , LDH ,CK and CK‐MB .
6.Clinical significance of determination of serum neutrophil gelatinase associated lipocalin in the diagnosis of contrast induced nephropathy

Objective To discuss the clinical significance of serum neutrophil gelatinase associated lipocalin(NGAL)detection in the diagnosis of contrast induced nephropathy .Methods A total of 299 inpatients with contrast medium in department of inten‐sive care unit(ICU) ,department of cardiovascular disease ,department of urology from Guangdong Province Hospital of Traditional Chinese Medicine were enrolled ,and the fasting blood glucose and lipids were detected .Sera were collected before examination of contrast medium and 1 ,2 and 6 d after examination of contrast medium respectively .Serum creatinine(SCr)and cystatin C(CysC) were detected ,the concentration of NGAL was determined .CIN was defined as of increased by 44 μmol/L or 25% from baseline in SCr within 24-48 h .The NGAL detection variation tendency was analyzed in different time point .Results In 299 cases ,CIN oc‐curred in 28 patients ,the incidence rate was 9 .36% .The diabetes patients in the incidence of CIN was 16 .21% (12/74) ,no inci‐dence of diabetic patients was 7 .11% (16/225) .When compared with that of before contrast ,CIN in serum of patients with Cr and CysC coronary angiography in 2 d significantly increased after contrast medium ,returned to the level before angiography on 6 d .But the serum NGAL in angiography 1 d after it was significantly increased (P<0 .05) ,statistically significant differences compared with pre contrast ,and was still at a high level on sixth day .Conclusion Prediction of CIN level of serum NGAL in contrast after 1 d could be a good predictor for CIN ,and the prediction time is earlier than the serum levels of Cr and CysC .The level of serum NGAL can be used for early diagnosis of CIN .
7.Instructions of Neural Response Telemetry for Prediction of the Rehabilitation Effects and Mapping in Children Cochlear Implant

Objective To study the stability of NRT threshold and the correlation between NRT threshold and the value of behavior T ,C in order to instruct mapping in infant cochlear implant ,and to study whether the elic‐itation of NRT waveform can be applied to determine the effects of rehabilitation .Methods There were 159 cases with complete information in data in this study .150 out of the 159 cases coincided with the study requirements of the changes and correlation .We had collected the NRT thresholds of the electrodes of No .1 ,6 ,11 ,16 ,22 in switch-on ,one month after switched -on ,half a year and one year after switched -on .The analysis of variance method was adopted to compare the threshold changes of each electrode in different stages .We had collected the values of behavior T ,C in one year after switched -on .Pearson regression analysis was used to compare the correlation be‐tween NRT threshold in switch -on and this value of behavior T ,C .We applied MAIS assessment scale to make a questionnaire for all 159 patients during the follow -up at one year after switched -on .We had gathered the scores in three aspects ,the average score in questionnaire and the average hearing threshold of every child .All children were divided into four groups according to whether the elicitation of NRT waveform during - operation ,post-op‐eration .Results Regarding the NRT threshold ,there was no significant difference in the pairwise comparison of four time points(P>0 .05) .There was positive correlation between NRT threshold and behavior T ,C(P<0 .01) , but the correlation was wake .The group with no waveform on the during -operation and post -operation had sig‐nificant differences with the other groups in all scores and average hearing threshold (p<0 .01) .And there was no statistical significance among the other three groups (P> 0 .05) .Conclusion The NRT threshold remains steady from switched -on .A positive correlation exists between NRT threshold and the actual T ,C value .The NRT threshold can be used to estimate the T ,C value .But the accuracy of the measured value is less than the behavior method .We still need to use behavior method to measure as soon as possible .The NRT waveform appearance on the post-operation is more significant than that on the during -operation to prognosis of rehabilitation .
8.Homology of 18S small subunit ribosomal RNA gene among species and strains of Schistosoma and sensitivity of PCR assay to detect single cercaria

Objective To research the homology of 18S small subunit ribosomal RNA(18S-rRNA) gene about Chinese Mainland and Philippine strains of Schistosoma japonicum,and Schistosoma mansoni,and the possibility to establish the PCR assay based on the gene for detecting the cercaria in a low density level. Methods The genomic DNAs of Chinese Mainland and Philippine strains of S.japonicum,and S.mansoni were extracted. The PCR assay was used to detect the identical target DNA elements in the above genome team and the homology of their genes was compared. The single cercaria was respectively treated with the method of heating in boiling water,the method of treating with ammonia and the method of treating with NaOH,HCl and ethanol,and the single treated cercaria and the single cercaria without treating were used as the templates to amplify the target DNA by using the PCR assay,and the detection rates of the PCR assay to detect the single cercaria treated with the different methods were calculated and compared. Results With the genomic DNAs of Chinese Mainland and Philippine strains of S.japonicum and S.mansoni as the templates,the target DNA element of which sequence length was 469 bp was all amplified by PCR. The target DNA was all amplified by PCR to the single cercaria treated with ammonia and the method of treating with NaOH,HCl and ethanol. However,only 50 percent of specimens of the single cercaria without treating and the single cercaria treated with the method of heating in boiling water were amplified to the target DNA by PCR. Conclusions The 18S-rRNA gene has the general homology among the species and strains of Schistosoma. The sensitivity of the PCR assay to detect the low density cercaria treated with ammonia or the method of treating with NaOH,HCl and ethanol is higher than that of the single cercaria without treating or treated with the method of heating in boiling water.
9.Effect of Sorafenib in Increased VEGF in Stimulating Effect of Walker-256 Transplanted Liver Cancer Model in SD Rats

[Objective] To investigate association between the time point of sorafenib administered and suppress effect on tumor growth secondary to the increased expression of vascular endothelial growth factor (VEGF).[Methods] Fifty SD rats were performed intrahepatic implantation using tumor tissues from subcutaneous tumors in nude mice which were administered Walker 256 tumor cells.Ten days after the procedure,MR scans were used to choose forty SD rats with successful hepatic tumor transplantation among fifty experimental animals.Then they were randomly divided into four groups:(A,control group) mere injection of vascular endothelial growth factor (VEGF);(B) administration of sorafenib 72 hours prior to VEGF injection;(C) administration of sorafenib together with VEGF injection;(D) administration of sorafenib 72 hours later to VEGF injection.The tumor growth and median survival time of rodents were observed and compared.After each experimental animal died,immunohistochemical (IHC) methods were applied to detect the expression of VEGF in tumors.[Results] Ten days after the administration of sorafenib,MR showed significant growth of hepatic tumors,the tumor size in experiment group were significiant smaller,than control group (5.4 cm) with statistical significance.Median survival time of four groups were (19.6 ± 1.8) d,(31.2 ± 7.0) d,(27.4 ± 4.9) d,and (26.5 ± 4.6) d,respectively,which indicated that animals in sorafenib groups lived longer than those in control group (P < 0.05).Differences can be obseverd in sorafenib groups with statistical significance existing (P < 0.05).Harvest hepatic tumor tissues from dead animals and HE staining as well as IHC examination were performed.The expression of VEGF in four groups were 88.3 ± 13.6,42.8 ± 8.0,71.9 ± 15.7,and 73.6 ± 13.7.There were statistical significance between control group and sorafenib groups.And further in sorafenib groups,the expressions of VEGF also varied greatly.[Conclusion] Sorafenib can extend the survival time,reduce tumor angiogenesis.And we can conclude that administration of sorafenib before the transient increased expression of VEGF offers survival benefits than that after the evaluation of VEGF levels.
10.The Factors Affecting the Rehabilitation Efficacy after the Operation of Adults Cochlear Implantation

Objective We will discuss the related factors affecting the rehabilitation efficacy after the opera-tion of adults cochlear implantation .Methods Nijmegen Cochlear Implant Questionnaire'was adopted .There were questionnaire surveys on 29 adult patients with cochlear implantation .The individual scores and total scores of all subjects were collected together with the personal information ,including age ,bilateral effects and other factors . They were grouped in accordance with different grouping standards of different affecting factors .Results Compared to the communication manner of long -term hearing loss ,the rehabilitation efficacy of the group using lip -reading preoperatively was greater than the group using sign language or writing ;the rehabilitation efficacy of the group with the aggregate time of using CIs more than two years was superior to that similar group of less than two years , and the average P within the group of these two factors was <0 .05 .There was no statistical significance among group comparisons for other factors as the P was >0 .05 .Conclusion Ages ,the bilateral deafness ,and auditory dep-rivation have no obvious effects on the rehabilitation efficacy after the operation .The patients with postlingual deafness can improve their hearing with proper rehabilitation .Lip-reading preoperatively has positive effects on the rehabilitation effica-cy after the operation ;the length of using CIs has become the major factor affecting the rehabilitation efficacy .
