1.Establishment of proteomic techniques and two-dimensional electrophoregram of human hepatocellular carcinoma cell line QGY-7701

Objective:To establish proteomic technique system of human hepatocellular carcinoma cell line QCY-7701. Method:Two different lysis buffer were taken to extract cell proteins. After two-dimensional gel electrophuresis (2-DE) and PDQuest analysis, 10 good-matched protein spots were chosen to be identified by MALDI-TOF-MS. Results:A steady 2-DE electrophregram of hepatocellular carcinoma cell line QGY-7701 was established. Connexin43,the base of electrical and intercellular chemical signal communication in the myocardial cell,can promote the growth and development of the heart.The decreasing of the number of connexin 43 and its' distribution changes may lead to the changing of the conduction velocity and direction,meanwhile,increase the occurrence of ventricular arrhythmias.Mesenchymal stem cells transplantation and drug therapy can improve the levels and distribution of connexin43 in ill myocardium,and can reduce the occurrence of ventricular arrhythmias.
5.Effect of Radix astragali on the expression of myocardial Cx43 in rats with dilated cardiomyopathy

Objective To observe the effect of Radix astragali (RA) on myocardial connexin-43 in rats with dilated cardiomyopathy (DCM). Methods The dilated cardiomyopathy model in rat was established through intraperitoneal injection with adriamycin. The rats in the model group were randomly divided into RA group and the model control group according to different methods of administration. Rats in RA group were gavaged with Astragalus particles and double-distilled water, and rats in model control group and normal control group were gavaged with an equal amount of double-distilled water daily for four weeks. At the end of 12 weeks, the left ventricular ejection fraction (LVEF) was measured with echocardiogram. The Cx43 mRNA level was tested by reverse transcriptase polymerase chain reaction (RT-PCR). Immunohistochemical method was used to observe myocardial Cx43 expression and distribution. Results Compared with the model control group, the Cx43 mRNA and protein expressions and LVEF were increased signiifcantly in RA group (P<0.05). The disorders in distribution of myocardial Cx43 improved in RA group in contrast to the model control group. Conclusions Radix astragali can improve myocardial Cx43 expression and distribution in DCM rats, and can further improve the cardiac function.
6.A simple microfluidic chip technology for assaying electrotaxis of cancer cells

Objective To develop a simple microfluidic chip technology for analyzing the electrotaxis of cancer cells . Methods The basic structure of the proposed microfluidic electrotaxis chip included a straight microchannel and liquid storage pools located on both sides of the microchannel .Two platinum electrodes were inserted into the liquid pools to create a controllable direct current ( DC ) field in the microchannel .The distribution and strength of the DC field in the microchannel was analyzed by the finite element analysis software COMSOL multiphysics and experiment tests .Finally, the electrotactic behavior of the rhabdomyosarcoma RD cells in the DC field of different strength was characterized using the accumulated distance, average velocity, x forward migration index ( xFMI) and y forward migration index ( yFMI) as quantitative parameters.Results The results of element analysis and experiments showed that the structure of the designed microfluidic electrotaxis chip was able to guarantee a uniform and strength-controllable DC field in the microchannel .The experiment of cell electrotaxis showed that the RD cells migrated toward the anode of the DC field .Meanwhile , the values of xFMI and accumulated distance for RD cells increased with the enlargement of the DC field , with the strength ranging from 188 to 1320 V/m.Conclusion The microfluidic chip technology developed in this paper for assaying the electrotaxis of cancer cells is simple and easily implementable , and it can be used for studies of the electrotactic behavior and underlying mechanisms of various cancer cells and normal cells in the future .

9.The relationship between serum vitamin D level and obesity

Serum 25-(OH)D3concentration was determined in subjects with different types of obesity. The serum 25-(OH)D3levels in overweight or obese and central obese subjects were lower than that in non-obese subjects.(P<0.01). In a multiple linear regression analysis serum 25-(OH)D3was independently and negatively correlated with body mass index and waist circumference.
10.HYPERLIPIDEMIA IN COPPER DEFICIENCY RATS

In order to investigate the interelationship between copper deficiency and lipidemia, an experiment was done in weanling rats fed normol and copper deficient diet for one month. At the end of experimental period, the growth rate, the contents of copper, zinc, cholesterol and triglyceride in plasma and the activity of monoamine oxidase (MAO) in plasma were measured. The differences between the two diets were compared by student's test and the relation between plasma copper and plasma cholesterol or MAO was ascertained by regression analysis.The results showed that a lower body weight gain and a highly significant hypercholesterolemia were found in copper deficiency rats and the plasma cholesterol concentration correlated reversely with the plasma copper (r = -0.724, p
