2.Association of galectin-3 expression with biological behaviors of squamous carcinoma of esophagus

Objective To investigate the galectin-3 expression in squamous carcinioma of esophagus and analyze its relationship with clinicopathological characteristics.Methods S-P immunohistochemistry was used to detect the expression of galectin-3 in 80 paraffin-embedded specimens of esophageal carcinoma and 22 specimens of normal tissues adjacent to the benigh lesions of esophagus.RT-PCR was performed to detect galectin-3 mRNA expression in 36 pairs of fresh squamous carcinoma tissues and corresponding adjacent normal mucosa.Results Out of 80 specimens of squamous carcinoma of esophagus,62 specimens had positive expression of galectin-3,with the positive rate of 77.5%(62/80)that was significantly higher than 27.27% in normal esophageal tissues(6/22,P
3.Neuroprotective effects of magnesium sulfate on spinal cord injury in rats

BACKGROUND: Available evidence suggests that following spinal cord injury, obvious reduction of Mg2+ level occurs in both the serum and the injured spinal cord, which has significant effects on cell membrane permeability, vascular regulation as well as secondary spinal cord injury.OBJECTIVE: To investigate the protective effects of magnesium sulfate (Mg2SO4) administered via intraperitoneal injection on the injured spinal cord of SD rats.DESIGN: A randomized controlled experiment.SETTING: Department of Orthopedics, Renmin Hospital of Wuhan UniversityMATERIALS: Forty-eight adult male SD rats were allocated randomly into experiment group and control group with 24 rats in each.INTERVENTIONS: The experiment was conducted in the Laboratory of Department of Orthopedics, Renmin Hospital of Wuhan University from April to August 2002. One hour after establishment of spinal cord injury models, the rats in the controlled group were injected intraperitoneally with normal saline, while those in the experiment group were given intraperitoneal injection of 300 mg/kg Mg2SO4. At each time point of 4, 8 and 24 hours after the treatment, 6 rats were selected from each group for measuring free Ca2+ concentration in the cells at the site of injury with spectrofluorometer . The activities of superoxide dismutase (SOD) in the spinal cord were detected by means of xanthine oxidation and thiobarbituric acid was used to determine the concentration of malondialdehyde (MDA). Lowered SOD activity and decreased MDA level were considered to suggest the protective effect of Mg2SO4 against spinal cord injury. Inclined plane test was used to assess the spinal cord function at 8, 24 hours and 1 week after the injury, in which the rat was made to stand on an inclined plate covered by a piece of rubber and the plate was inclined gradually until the rat was no longer able to stay in the original position for 5 s, and the inclination of the plate was recorded. The test was performed 3 times for each rat and the plate inclinations were averaged. An increased inclination indicated improvement of the spinal cord function.MAIN OUTCOME MEASURES: ① Intracellular Ca2+ concentration at the injury site. ② Changes in SOD activity and MDA concentration in the spinal cord. ③ Results of spinal cord function evaluation of the rats.RESULTS: Intracellular Ca2+ concentration at 8 and 24 hours after the injury was significantly lower in the experimental group than in the control group [(376.5±36.2)×10-9vs (425.9±32.7)×10-9 mol/L and (316.3±13.9)×10-9vs (350.2±29.4)×10-9 mol/L, respectively, P < 0.05]. Compared with the control group at each time point, MDA concentration was significantly decreased, while SOD activity of SOD was increased in the experiment group (P < 0.01). The improvement of spinal cord injury was not obvious in the experiment group and was significantly higher than that in the control group until I week after the injury [(53.3±4.3)° vs (44.3±5.7)°, P < 0.05].CONCLUSION: Intraperitoneal Mg2SO4 injection may significantly lower the concentration of intracellular Ca2+ at the injury site and alters the product of lipid peroxidation, suggesting its neuroprotective effect against spinal injury so as to lighten secondary spinal injury in rats.
4.Development of the electrohydraulic lithotripter

This paper presents the components and the principles of an internal electrohydraulic lithotripter.Cooperating with the cystoscope,the lithotripter introduces the high-pressure discharge pulse into the bladder through the microelectrode.Then the little,easily-moved stone is targeted,and the re-discharge of the lithotripter 5mm away from the stone results in the shock wave to perform lithotritization.
5.Effects of Hawthorn Leaf Procyanidins on Over Expressions of ICAM-1 and E-selectin in Human Umbilical Vein Endothelial Cells Induced by TNF-α

Objective To observe the effects of hawthorn leaf procyanidins (HLP) on over expressions of ICAM-1 and E-selectin in human umbilical vein endothelial cells (HUVEC) induced by TNF-α,and clarify the mechanism of HLP's anti-inflammation effect. Methods HUVEC were cultured in vitro. MTT assay was used to detect cell viabilities. The expressions of ICAM-1 and E-selectin in HUVEC were detected by flowcytometry. Results Up to 200 mg/L, HLP showed no significant decrease in cell viabilities; the expression levels of ICAM-1 and E-selectin in the model group significantly increased, compared with that in the normal group; 10, 20, 30, 40, 50 mg/L HLP inhibited the expression elevations of ICAM-1 and E-selectin in concentration-dependent manner; and there were statistical significances in 40, 50 mg/L HLP groups, compared with the model group. Conclusion HLP can inhibit the over expressions of ICAM-1 and E-selectin of vascular endothelial cells induced by TNF-α, which possibly underlies HLP's anti-inflammation effect.
6.Correlations between quality of life and sagittal parameters in ankylosing spondylitis after kyphosis correction

Objective To explore the impact of the ratio between pelvic tilt (PT) and sacral slope (SS) on the life quality of patients with ankylosing spndylitis (AS) after kyphosis correction. Methods From November 2008 to May 2011, 33 AS pa?tients were reviewed, including 31 males and 2 females, aged from 19 to 58 years old (average, 36 years old). The thoracolumbar kyphosis angle was 35.23° ± 13.98° (range, 15.12°-74.37° ) and the lumbar lordosis angle was 8.68° ± 18.27° (range,-23.70°-62.15°). The Scoliosis Research Society (SRS)?22 questionnaire was used to evaluate the quality of life and the Oswestry disability index (ODI) was used to evaluate the condition of pain. The pelvic incidence (PI), PT, SS, sagittal vertical axis (SVA) and osteoto?my angle were obtained from standing lateral full?spine radiographs. The correlations were analyzed from the subjective grading and the sagittal parameters in AS patients. Results The osteotomy site was in L1 (5 cases, 21.00°-54.59° , average 32.59° ± 13.44° ), L2 (19 cases, 28.63°-66.24° , average 37.89° ± 9.26° ), L3 (9 cases, 31.78°-60.90° , average 47.05° ± 9.20° ), respectively. The range of osteotomy angle was 39.59°±10.82° (range, 21.00°-66.24°). The subjective grading and spino?pelvic parameters were improved significantly after operation except PI, only postoperative PT/SS (0.93±0.65) and ODI standing (0.60±0.75)(r=0.681, P<0.05), osteotomy angle (39.59°±10.82°) and satisfaction of management (3.33±0.49)(r=0.478, P<0.05)had correlation with the subjective grading. Conclusion Compared with the change of PT, SS and SVA, the change of PT/SS is more closely related to the quality of life after operation in AS patients with kyphosis, which should be pay attention to by surgeon when designing operative schemes.
7.Preparation and characterization of mono PEGylated recombinant human interferon omega.

The amino group PEGylation of rhIFNomega with monomethoxy polyethylene glycol succinimidyl succinate (mPEG-SS, 20 000) was investigated, and the modified mixture was separated and purified by ion exchange chromatography and gel filtration chromatography. Under the optimized purification conditions, the average content ofmono PEG-rhIFNomega in the collect liquid reached 182 microg x mL(-1). The average purified yield of mono PEG-rhIFNomega exceed to 22%, and the purity of mono PEG-rhIFNomega was greater than 98% by SDS-PAGE and RP-HPLC. Relative molecular mass of mono PEG-rhIFNomega was 43 790 detected by MALDI-TOF MS. The apparent molecular mass measured by SDS-PAGE was about 60 810. The purified PEG-rhIFNomega has the characteristics of typical PEGylated protein. Activity reservation rate of mono PEG-rhIFNomega was 15.0%, while the antigenicity decreased by at least 64 folds. In addition, the acid stability, thermal stability and stability in serum and trypsin solution of mono PEG-rhIFNomega were markedly better than those of the rhIFNomega. The pharmacological properties of mono PEG-rhIFNomega were significantly improved. The prepared PEG-rhIFNomega might be developed to a novel safe and long-acting interferon.
8.Effect of asymptomatic hyperuricemia on the postoperative functional recovery in patients with knee joint re-placement

Objective To evaluate the effect of asymptomatic hyperuricemia on the postoperative functional recovery in osteoarthritis (OA)patients with knee joint replacement.Methods Among the 294 patients who recieved knee joint replacement in our hospital from Feb-ruary 2013 to September 2015,a total of 187 patients were included in this study with 79 cases in the hyperuricemia group and the other 108 cases in the control group.The WOMAC index of the two groups were analyzed before the surgery,2 weeks,3 months,and 6 months after the knee joint replacement surgery.Multivariable linear regression was performed to test the effect of impact factors on the variation of WOMAC index.Results The WOMAC index of the two groups significantly increased 2 weeks after surgery,and the difference was statistically signifi-cant(P <0.05).And the WOMAC index of the control group was higher than that in the hyperuricemia group 2 weeks and 3 months after sur-gery with statistically significant difference(P <0.05).Conclusion The uric acid level before surgery was a influence factor of WOMAC index, and high uric acid level before knee joint replacement has negatively impact for the postoperative functional recovery in the osteoarthritis patients.
9.A novel endoscopic classification system to determine the invasion depth of early esophageal cancer

Objective To develop a novel endoscopic classification system to determine the invasion depth of early esophageal cancer.Methods The esophageal lesion was endoscopicaUy stained with Lugol's iodine first,then methylene blue.According to the growth pattern,height and cup depth under endoscope,the lesions were classified into 5 types,including surface diffusion growth,intra-lumen growth,intra-wall growth,bi-direction growth and mix growth types.The lesions were then removed by endoscopic mucosa reection or surgery,the precise invasion depth of the lesion was determined pathologically and the results were compared with the endoscopy classification.Results The data of 44 cases of esophageal mucosal cancer and 34 cases of esophageal sub-mueosal cancer were included.With the criteria of mucosal cancer as surface diffusion growth,intra-lumen growth <5mm,bi-direetion growth <2mm and intra-wall growth <0.5 mm,the diagnostic specificity was 89.1%(41/46) and sensitivity was 93.2%(41/44).With the criteria of submucosal cancer as intra-lumen growth≥5 mm,hi-direction growth≥2mm,intra-wall growth≥0.5 mm and mix growth type,the diagnostic specificity was 90.6%(29/32) and sensitivity was 85.3%(29/34).The overall diagnostic accuracy in differentiating esophageal mucosal cancer from esophageal submueosal cancer by endoscopic classification was 89.7%(70/78).Conclusion This endoscopic classification system is effective in differentiating esophageal mucosal cancer from submucosal ones.
10.Follow-up study on pedicle fixation for treatment of thoracolumbar burst fractures in patients with osteoporosis

Objective To explore the effect of pedicle fixation for treatment of thoracolumbar burst fractures in patients with osteoporosis,and to provide more evidence for the treatment.Methods Retrospectively analyzed the clinical data of 121 patients with osteoporotic vertebral burst fracture from June 2012 to October 2015.And these patients were divided into two groups according to different operation methods, namely the control group (n=56) who were given short segment fixation and the observation group (n=65) who were given single segment fixation.The visual analogue scale(VAS),Oswestry disability index(ODI),vertebral height,kyphotic angle and bone mineral density of the two groups were analyzed before surgery and 3 days,1 month,3 months and 12 months after surgery.Results The VAS score,ODI score,vertebral height,and Cobb angle of the injured vertebra were significantly improved in both of the two groups,and the difference was statistically significant (P<0.05).The VAS score of the observation group was better than that of the control group 3 days after surgery with statistically significant difference (P<0.05).But there was no significant difference 3 months,6 months and 12 months after surgery(P>0.05).The ODI score of the observation group was better than that of the control group 3 days and 3 months after surgery with statistically significant difference (P<0.05),and there was no significant difference between the two groups till the end of follow-up.Pedicle fixation at the injured vertebra significantly improved the vertebral height and Cobb angle with statistically significant difference (P<0.05).And the anti-osteoporosis treatment significantly increased the bone mineral density (P<0.05).Conclusion Pedicle fixation at the injured vertebra is useful in pain relief as well as function and anatomical structure restoring.And anti-osteoporosis treatment is necessary for the bone mineral density increase.
