Fluorescence resonance energy transfer(FRET)is increasingly used to study inter-and intramolecular interactions in living cells.Since being proportional to the concentration of the donor-acceptor complex.FRET value must be normalized to exclude the influence of the ratio and the concentration of donor and acceptor for comparison.Different from the intra.molecular FRET which is simplified by the fact that the COncentration of the donor is equal to that of the acceptor,the inter-molecular FRET is usually too complicated for most existing measurements to quantify exactly.We deduced the exact proportion of the donor-acceptor complex based on a unique characteristic of homodimer,a special kind of the intermolecular interaction,developed an exact quantification measurement of the FRET.We proved the novel method Can generate more reliable estimation of FRET value by comparison with other methods using a homodimer,estrogen receptor alpha(ERa),as a FRET pair.

Objective To explore the telemedicine education by using operation live telecast system.Methods Multi-channel video and audio signals were collected by video camera and transmitter phone and were connected with telemedicine education web after matrix treating and controlling.Results Telecommunication of live telecast was realized.Conclusion Operations can be telecast more widely,and the level of telemedicine education is raised.

Students generally felt difficult in studying during clinical practice and the orthopedics teaching quality was unsatisfactory due to the discrepancies existed among teaching of anatomy,imagie and orthopedics clinical practice concerning the time,content,requirement and method.Students' understandings of anatomy and image knowledge were improved through launching theoretical study by means of lectures and case discussion.Students' understandings of orthopedics diseases were promoted through preoperative diagnosis and intraoperative conformation using anatomy and image knowledge for the patients they were responsible for.In all,orthopedics clinical teaching quality was promoted through applying these measures.
5.Study of anatomy and variation of arteries of ankles in chinese adults by ultrasound

Objective To analyze the normal anatomy and variation of arteries around ankles in Chinese adults in and around Shaanxi province. Methods Aged 18-82 (48±15) years, 522 males and 557 females who came to Xijing Hospital for medical treatment or for a healthy check were randomly recruited to participate in this study. Their arteries around ankles were assessed by high-frequency color Doppler ultrasound. Then the anatomy and variation of the arteries were analyzed. Results There were totally 213 arterial variations in 175 subjects, including variations at single ankle in 137 subjects and variations at both sides in 38 subjects. The rate of arterial variation at ankle in persons was 16.2%(175/1 079) and was higher in females than that in males (21.5%, 120/557,χ2=24.028, P=0.000). The rate of arterial variation at ankle in limbs was 9.9%(213/2 158). There was no signiifcant difference for the rate of arterial variation between the left (5.1%, 109/2 158) and right (4.8%, 104/2 158) ankles (χ2=0.129, P=0.701). In variations of arteries at ankles, there were 131 cases with small posterior tibial arteries and peroneal arteries extending to the bottoms of feet, i.e., posterior tibial arteries substituted by peroneal arteries (typeⅠ, 6.1%, 131/2 158), 69 cases with small anterior tibial arteries and peroneal arteries extending to the dorsums of feet, i.e., anterior tibial arteries substituted by peroneal arteries (typeⅡ, 3.2%, 69/2 158), and 13 cases with both small anterior and posterior tibial arteries, and peroneal arteries extending to both the dorsums and bottoms of feet, i.e., both anterior and posterior tibial arteries substituted by peroneal arteries (typeⅢ, 0.6%, 13/2 158). There was no signiifcant difference in each type of variation rates between left and right sides. The rates of typeⅠvariation were 6.0%(65/1 079) and 6.1%(66/1 079) at the left and right ankles respectively (χ2=0.010, P=0.928);the rates of typeⅡvariation were 3.6%(39/1 079) and 2.8%(30/1 079) at the left and right ankles respectively (χ2=1.213, P=0.271);the rates of typeⅢvariation were 0.5%(5/1 079) and 0.7%(8/1 079) at the left and right ankles respectively (χ2=0.697, P=0.404). Conclusions Ultrasound can be used to depict anatomy and variation arteries around ankles. There is a high prevalence of anatomic variation of ankle arteries in population in and around Shaanxi province. Therefore it will be helpful for diagnosis vascular abnormalities with the knowledge of characteristics of normal anatomy and variation of ankle arteries.
6.Expression of urinary ptasminogen activator receptor in tissue and serum of patients with cervical cancer and clinical significance

Objective To analyze the correlation between the levels of urinary ptasminogen activator receptor (uPAR) and the clinicopathological characteristics of patients with cervical carcinoma and it's clinical significance. Methods SABC immunohistochemistry was employed to detect the expression of uPAR in 50 cases of cervical carcinoma tissue and 50 cases of normal cervical tissue. ELISA method was used to determine the serum uPAR levels for the patients with cervical carcinoma. Results There was no expressionfor uPAR in normal tissues, The positive expression rate of uPAR was 66 % (33/50). The uPAR level of cervix cancer tissues [(70.92±28.55) ng/100 mg protein]was significantly higher than those in normal tissues [(11.01±5.40) ng/100 mg protein], (P <0.001). and uPAR levels were closely related to clinical stages,lymphatic metastasis and differentiation degree (P <0.05), but not related to deep myometrial invasion and vascular embolization (P >0.05) (however, they were not related to patient's age, tumor growth type and the size of tumor. Significant difference of uPAR level was observed between the patients with cervical carcinoma [(2.38±0.29) ng/ml]and in healthy controls [(0.50±0.16) ng/ml](P <0.001). Single factor analysis indicated that, before the treatment, the serum uPAR levels were closely related to clinical stages, lymphatic metastasis, vascular embolization, and deep myometrial invasion (P <0.05-P <0.01). However, they were not related to differentiation degree (P >0.05). Multifactor regression analysis showed that the pretreatment serum uPAR levels of patients were related to clinical stages (P =0.000), cavum pelvis lymphatic metastasis (P =0.000) and deep myometrial invasion. Patients with cervical carcinoma showed a dramatic drop in serum uPAR levels after treatment, which were significantly different compared to their pretreatment uPAR levels (P <0.001). Linear correlation analysis showed that there was a positive correlation between serum and tissue uPAR levels, (r =0.801, P <0.001). Conclusion There were high expression of uPAR in serum and tissues with cervical carcinoma. Pretreatment serum uPAR levels were closely related to patients' clinical stages,cavum pelvis lymphatic metastasis and deep myometrial invasion, serum uPAR levels may be an important tmnor marker for the diagnosis, detection, prognosis of cervical carcinoma.
7.Expression of HIF-2αin non-small cell lung cancer tissue and its relationships with MVD,Ki67,and GST-π

Objective:To investigate the expression of hypoxia-inducible factor-2α (HIF-2α)in non-small cell lung cancer (NSCLC)tissue,and to analyze its relationships with angiogenesis,cell proliferation and chemotherapy resistance. Methods: Total 112 cases of NSCLC and 20 cases of normal lung tissues were selected, immunohistochemical method was used to detect the expressions of HIF-2α,CD31,Ki67 and GST-π in 112 cases of cancer tissue and 20 cases of normal lung tissue,and the correlations of HIF-2α expression with microvessel density (MVD),Ki67, and GST-π were analyzed.Results:The positive expression rate of HIF-2α in NSCLC tissue was significantly higher than that in normal lung tissue (P < 0.05 ), the expression rate of HIF-2α in 112 cases of NSCLC was 47.3% (53/112).The MVD in HIF-2α protein high expression NSCLC group (31.1 ± 14.7)was higher than that in HIF-2αprotein low expression NSCLC group (24.3±15.8)(P <0.05).The cases of high expression of Ki67 in HIF-2αhigh expression group occupied 54.7% (29/53),and it was higher than that in HIF-2αlow expression group (16/59,27.1%);there was significant difference (r = 0.281,P = 0.003).The high expression of HIF-2α had no obvious correlation with the expression of GST-π (r = 0.122,P = 0.202). Conclusion:HIF-2αmay play an important role in the carcinogenesis and development of NSCLC by promoting the angiogenesis and enhancing the cell proliferation of NSCLC,but it may have no correlation with chemotherapy resistance.

Objective To use ELISA detecting self‐developed hepatitis B five internal quality control products .Methods Hepa‐titis B five positive sera by the detection of ELISA were diluted through optimal ratio ,and were homemade indoor quality control materials .Results Self‐control materials and commodities were simultaneously detected by ELISA ,and the test results were com‐pared ,the two were no significant difference(P>0 .05);Self‐control materials continuously detected by ELISA ,its batch variation were less than 15% ,and stability was in line with the requirements .Conclusion Self‐developed hepatitis B five indoor quality con‐trol materials are made simply ,have good stability ,are satisfied control effect ,and have promotional value .

10.Primary clinical study of a novel multi-layer flat-plate bioartificiai liver for patients with liver failure

Objective To investigate the safety and therapeutic effects ot a novel multi-layer flat-plate bioartificial liver (BAL) for patients with liver failure.Methods Thirty-eight patients with liver failure from Dec.2010 to Dec.2011 were treated with a novel BAL based on multi-layer flatplate bioreactor and the co-cultured cells of the porcine hepatocytes and mesenchymal stem cells.A total of 48 treatments was performed,4 h each time.The clinical signs and symptoms,liver function,ammonia,coagulation function and complete blood count were evaluated before,during and after the treatment.DNA in the collected PBMCs was extracted for PCR with PERV specific primers and the porcine specific primer Sus scrofa cytochrome B.The RT activity was detected as well.Levels of xenoantibodies (IgG,IgM) were determined by using ELISA kit. Titers of complement were quantified by CH50 kit.Results All treatment procedures were completed successfully without any adverse reaction. All samples presented negative PERV DNA and RT activity. The levels of antibodies were similar before and after treatment.Treatment was associated with a temporary decline in levels of complement,and then the levels were recovered quickly.The clinical symptoms such as acratia,anorexia and abdominal distension were improved.The stage of hepatic encephalopathy in 16 patients was decreased. The liver function and ammonia was reduced disproportionately. Seven patients in all were bridged to liver transplantation,2 patients died and 2 patients gave up the treatment,and the others were turned better.After the outcome judgment according to the standard developed by the Artificial Liver Group,and Chinese Association of Infectious and Parasitic Diseases,there were 9 patients with clinical healing,25 patients with improvement and 4 patients with no effect,and the cure-improvement rate was 89.5%.Conclusion The novel multi-layer flat-plate BAL could be used as a safe and effective therapy for patients with liver failure.
