AIM: A HPLC fingerprint has been established of six active principles in Cistanche tubulosa in nine different regions in the Xinjian Uygur Automomous Region in China. METHODS: Chromatographic condition included a symmetry C_(18)(4.6 mm?250 mm,5 ?m) column and the gradient elution was adopted with ratio of acetonitrile-0.095% H_3PO_4 from 8()∶92 to 12()∶88 in 0-18 min and 12()∶18 to 19()∶81 in 18-40 min,19()∶81 maintain for 50 min.The detection wavelength was at 330 nm and the flow rate was 1.0 mL/min and detection time was 90 min. RESULTS: Eleven common characteristic peaks, including salidroside, bartsioside, echinacoside, cistanbuloside A,tabuloside A and acteoside were taken as fingerprint peaks the precision and the accuracy were in accordance with the chromatographic requirement. CONCLUSION: The method is stable, reliable, precision and provides a scientific basis for the quality standard for Cistanche tubulosa.
3.Clinical application research of laparoscopic ISR of anal cancer surgery in low location
China Modern Doctor 2014;(27):107-110
Objective To study the various aspects of the outstanding advantages that intersphinctefic resection, anal sphincter resection (ISR) was applied to the clinical ultra-low position rectal cancer anal surgery (SSO) treatment. Methods From 70 patients with rectal cancer in our hospital were divided into two groups at random in accordance with the operation mode and the average. Patients in one group were treated by laparoscopic ISR ultra-low a SSO (called a mirror abdominal group), and patients in another group were treated by opening the IRS ultra-low a SSO (control group), compared the clinical outcomes. Results From the point of the analysis of the clinical operation, all patients with abdominal lens group line TME (total mesorectum excision), no open treatment conditions, the average op-eration time and intraoperative blood loss were significantly and hospital stayed lower than control group in patients (P< 0.05), the main postoperative complications for colon membrane prolapse and crissum eczema (not severity complica-tion), and two groups of patients with postoperative bowel and anal pressure determination results had no significant dif-ference (P> 0.05). Conclusion Laparoscopic IRS ultra-low rectal cancer SSO wound was small, safe and feasible, for the short-term treatment of patients with rectal cancer curative effect was good and being worthy of application.

Adult pluripotent stem cells, such as mesenchymal stem cells derived from bone marrow and adipose tissue are capable of multilineage differentiation. Although autologous stem cell transplantation is an effective alternative to organ transplantation, the loss of cell viability and differentiation confinement of implanted cells has largely impaired the therapeutic efficacy. To produce biomaterial-free liver construct to integrate into living tissue, we isolated adipose mesenchymal stem cells and subjected them to a delicate culture configuration to mediate the hepatocyte differentiation. The differentiated hepatocyte-like cells were then inoculated onto poly (N-isopropylacrylamide) (PNIPAAm) grafted cell culture dish. By lowering the culture temperature to 20 degrees C, cells detached from the dish surface into a complete cell sheet. Hematoxylin and eosin staining and immunohistochemistry results showed that cell sheet was composed of 2-3 layers of cells and extracellular matrix was maintained intact. As compared with traditional cell harvest using trypsin digestion, cell sheet fabrication causes no damage to cell membrane and extracellular matrix. Hence, cell sheets would form a better interaction with tissues in situ, and a higher cell viability and therapeutic efficiency would be expected.

BACKGROUND AND OBJECTIVEAtelectasis is a common complication after thoracotomy, and it may threaten patients' life if it was not treated correctly and properly. The aim of this article is to explore and discuss the prevention and treatment for atelectasis during the perioperative period, and also to explore new methods for reducing the perioperative mortality due to atelectasis after thoracotomy.
METHODSWe retrospectively reviewed the medical records of 374 lung cancer patients who underwent thoracotomy in our department between Jan 2007 and Nov 2009.
RESULTSAtelectasis occurred in 14 patients among all the 374 lung cancer patients who underwent thoracotomy. All the atelectasis returned to reexpansion after treatment.
CONCLUSIONThe incidence of atelectasis in these series is relatively low compared with the reports in literatures. Good perioperative preparation and perioperative treatment can remarkably decrease the incidence and mortality of atelectasis after thoracotomy in the treatment of lung cancer.

OBJECTIVE To investigate the effects of andrographolide (Andro) on angiogenesis in rats with diabetic foot and to explore its mechanism of action based on the Hippo-Yes-associated protein (YAP) signaling pathway. METHODS The rat model of type 2 diabetes was established by using low-dose streptozotocin combined with high-fat and high-glucose diet. On the basis of successful modeling, the rat model of diabetes foot was established by scalding. Model rats were randomly divided into 5 groups with 12 rats in each group: model group, Andro low-dose, medium-dose, and high-dose groups (1, 10, and 20 mg/kg), as well as inhibitor group (20 mg/kg Andro+100 mg/kg of verteporfin, an specific inhibitor of Hippo-YAP signaling pathway); other 12 healthy rats were included in the Control group. Rats in each group were intragastrically and intraperitoneally injected with solvents or corresponding drugs, once a day, for 2 consecutive weeks. The wound healing, fasting blood glucose (FBG) and fasting insulin (FINS) were detected in rats after medication. HE staining was performed to observe the tissue damage and capillary number of rat trauma; the number of endothelial progenitor cells (EPCs) in peripheral blood of rats was counted by using flow cytometry; the contents of serum total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C) in rats were determined by fully automatic biochemical analyzer; the expressions of hypoxia- inducible factor 1α (HIF-1α), vascular endothelial growth factor (VEGF) and Hippo-YAP signaling pathway-related proteins in the traumatic tissues of rats in each group were detected by Western blot. RESULTS Compared with Control group, the wound healing rate, capillary number, the proportion of EPCs, HDL-C content, as well as the protein expression levels of HIF-1α and VEGF and the phosphorylation levels of mammalian Ste20-like kinase 1, large tumor suppressor gene 1 and YAP proteins were significantly reduced in the model group, while the FBG, FINS levels and TC, TG and LDL-C contents were significantly increased (P<0.05). Compared with model group, the above indexes were significantly reversed in Andro low-dose, medium-dose and high-dose group, in a dose-dependent manner (P< 0.05); verteporfin attenuated the above reversal effect of Andro (P<0.05). CONCLUSIONS Andro has the effects of lowering blood glucose and blood lipids, promoting blood vessel formation and wound healing in rats with diabetic foot, and its mechanism of action may be related to the activation of Hippo-YAP signaling pathway.
