1.Nitoprusside, dopamine and dobutamine for treatment of refractory heart failure

Objective To observe the clinical effect of nitroprusside,dopamine and dobutamine for treatment of refractory heart failure. Methods 60 patients with refractory heart failure were randomly divided into control group and treatment group. Clinical symptoms, signs, echocardiography and other changes were observed. Results The total effective rate was 86.67% ,control group was 66.67% ,the total effective between the two groups was statistically significant(P < 0.05). After treatment, left ventricular ejection fraction (LVEF) improved significantly compared with the control group, the difference was statistically significant(P < 0.05). Conclusion The Sodium nitroprusside, dopamine and dobutamine for treatment of refractory heart failure was safe and effective method.
2.Relationship between maternal serum lipid levels and newborn weight in pregnant women with single positive diabetic screening test

Objective To investigate the relationship between maternal serum lipid levels and newborn weight in pregnant women with single positive diabetic screening test. Methods On the basis of 75 g oral glucose tolerance test (OGTT), 406 pregnant women during 24-28 gestational week whose 50 glucose challenge test (GCT)≥7.8 mmol/L were separated to gestational diabetes mellitus (GDM) group with 115cases, gestational impaired glucose tolerant (GIGT) group with 60 cases,and GCT (+) group with 231 cases.The serum levels of total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL-C),low density lipoprotein cholesterol(LDL-C), apolipoprotein A1 (ApoA1) and apolipoprotein B (ApoB) were measured at the same time. Results Serum TG concentration was significantly higher in GDM group than that in GIGT group and GCT(+) group(P<0.01).The level of HDL-C, ApoB in GDM group were lower than those in GIGT group and GCT(+) group (P<0.01). In the 231 women with positive diabetic screening but normal glucose tolerance who delivered at term, the incidence of macrosomia was significantly higher in those with TG levels higher than 3.5 mmol/L (25.0%, 11/44) than in mothers who TG levels were lower than 3.5mmol/L (11.2%, 21/187) (P=0.017). Multiple regression analysis (stepwise) showed that the independent risk factors of macrosomia was the level of TG and ApoB (F=7.677,P=0.006;F = 7.842,P = 0.001 ).Conclusion Among pregnant women with single positive diabetic screening test,maternal serum TG and ApoB are independently associated with neonatal birth weight.

Objective To examine the effects of acute hypobaric hypoxia on abilities of learning and memory,the water content in brain,the level of nitric oxide(NO) and endothelin-1 (ET-1) in hippocampus of mice.Methods The acute hypobaric hypoxia environment were made by putting the mice in a hypobaric chamber simulated at altitude of 6 000 meters for 8 hours.The capabilities of learning and memory of mice were detected by Morris water maze test.The content of water in hippocampus were examined by measuring the ratio of dry/wet weight,and the level of NO and ET-1 in brain was detected by colorimetric method.Results Morris water maze test showed that the mean escape latency of mice in the hypobaric hypoxia group were longer than that in the normobaric normoxia group((44.60±7.80) s vs (26.39±8.44)s,P<0.01),and the target quadrant residence time were decreased((19.78±2.74) s vs (22.98±6.14)s,P< 0.05).Compared with the normobaric normoxia group (NO:(2.37 ± 1.07) μmol/gProt,ET-1:(38.87 ± 6.17) ng/L),the water content in brain of mice in the hypobaric hypoxia group was increased (P< 0.05),meanwhile,both the level of NO ((4.48 ± 1.45) μmol/gProt) and ET-1 ((52.09±6.75)ng/L) in brain were increased too(P<0.01).Conclusion Hypobaric hypoxia can decrease the abilities of learning and memory of mice,and these changes might be related with the increased water content and the increased level of NO and ET-1 in hippocampus of mice.
6.Effects of cyclic tensile stress on metabolism and apoptosis of chondrocytes inrat

Objective To investigate the effect of cyclic tensile stress (CTS) on the metabolism inand apoptosis of rat chondrocytes.Methods Primary rat chondrocytes were cultured on a Bioflex plate for one day and then stretched cyclically for 24 hours at a frequency of 0.5 Hz using a Flexcell-5000T apparatus.The cells were divided into 5 groups according to their stretching ratio:0% (the control group),2%,6%,10% and 14%.After the stretching,Col Ⅱ,Aggrecan,MMP-13 and ADAMTS-5 mRNA were measured using qPCRs,and the NO and PGE2 levels were measured using ELISA kits.Moreover,TUNEL staining and Annexin V-FITC/PI were used to analyze the apoptosis of chondrocytes.Results Compared with the control group,the average levels of Col Ⅱ and Aggrecan mRNAdecreasedin 10% and 14% groups [(0.738±0.11) and (0.58±0.13),(0.75±0.11) and (0.55±0.09)].In those groups,the MMP-13 [(2.47±0.47) and (2.88±0.36)] and ADAMTS-5 mRNA level [(2.39±0.33) and (2.75±0.49)],the NO [(6.96±0.96) and (8.28±0.82)] and PGE2 level [(6.83±0.66) and (7.15±0.71)] had increased significantly.In the 6% group the average levels of Col Ⅱ(1.76±0.30) and Aggrecan mRNA (1.93±0.14)of 6% group were significantly higher than the control group,but the NO level of the former (3.07±0.20) was significantly lower than the control group (3.89 ± 0.33).The apoptosis rate of chondrocytesin 2% and 6% groups were (0.065±0.013) and (0.063 ± 0.147),without significant differences to that of the control group (0.045 ± 0.008).However,compared with the control group,apoptosis in the 10% and 14% groups [(0.135 ±0.026) and (0.184±0.020)] increased significantly.Conclusion The effect of cyclic tensile stress on chondrocyte metabolism and apoptosis was magnitude-dependent.Ten percent and 14% CTS can increase the catabolism and apoptosis of chondrocytes.Ten percent and 14% strain can increase the catabolism and apoptosis of chondrocytes.Cyclic 6% strain can increase the anabolism of chondrocytes,but 2% strain has no impact on metabolism or apoptosis.
7.Investigation on awareness status of malaria control knowledge of primary and secondary school students in Shandong Province

Objective To understand the awareness status of malaria control knowledge of primary and secondary school stu-dents in malaria endemic areas of Shandong Province,so as to provide the baseline information for formulating the malaria control strategy. Methods A cross-sectional survey was conducted in 5 TypeⅡmalaria-endemic counties in Shandong Province. A struc-tured questionnaire survey was administrated to students in primary and secondary schools. Results A total of 1 884 students were completed with the questionnaires and all were effective. The total awareness rate of malaria control knowledge was 65.9%, and 54.7% of the students knew the transmission route of malaria and only 31.5% of the students were aware of the preventive methods of malaria. There were significant differences of the awareness scores of malaria control knowledge among different malar-ia endemic areas but there were no significant differences between the awareness rates of male and female students and between two different education levels. Conclusions The awareness of malaria control knowledge of primary and secondary school stu-dents in Shandong Province is poor;therefore,the health education of malaria control knowledge for primary and secondary school students should be strengthened.
8.Epidemiological analysis of malaria prevalence in Shandong Province in 2014

Objective To understand the malaria epidemic situation and characteristics in Shandong Province in 2014,so as to provide evidences for taking targeted strategies and measures for malaria elimination. Methods The data of malaria epi?demic and malaria cases of Shandong Province in 2014 were collected from Information Management System for Infectious Dis?eases Report and Information Management System for Parasitic Diseases Control and Prevention. The epidemiological character?istics of malaria situation and the diagnosis and treatment of malaria cases were analyzed by Microsoft Excel 2007. Results A total of 150 malaria cases were reported in Shandong Province in 2014,which increased by 14.50% compared with that in 2013 (131 cases). All the cases were imported from other countries except one imported from Yunnan Province in China,and most of the imported cases were from Africa(142 cases,95.30%). All the cases were confirmed in Shandong Provincial Reference Labo?ratory and there were 121 cases(80.67%)infected with Plasmodium falciparum,16 cases(10.67%)infected with P. vivax,9 cases(6.00%)infected with P. ovale and 4 cases(2.67%)infected with P. malariae. The cases were mainly distributed in Tai’ an(40 cases,26.67%),Yantai(17 cases,11.33%),Weihai(13 cases,8.67%),Jining(11 cases,7.33%)and Heze(11 cases,7.33%)cities. Conclusions There have been no local malaria cases reported in Shandong Province in the continuous 3 years. However,the reported number of imported malaria cases shows an increasing trend in the province in recent years,and the species of infected Plasmodium are diverse. Imported malaria from other countries is the key of malaria control in Shandong Province at present.

Combining the structural features of picotamide and linotroban, a series of N,N'-bis-(halogenophenyl)-4-methoxybenzene-1, 3-disulfonamides were designed and synthesized on the basic principles of drug design. The structures of target compounds were confirmed by IR, 1H NMR and HR-MS, and the in vitro antiplatelet aggregation activity was evaluated by Born turbidimetric method with adenosine diphosphate (ADP) as the platelet aggregation inducers. The assay results showed that twelve compounds (4b, 4f, 4l, 5b, 5d-5g, 5j, 5k, 5m and 5n) were found to have superior anti-platelet aggregation activities than the positive drug picotamide. The preliminary structure-activity relationship (SAR) has been explored.
10.Effect of 9602 prescription on brain energy metabolism in cerebral ischemia reperfusion mice

AIM: To explore the influence of herbs (9602 prescription) on brain energy metabolism in cerebral ischemia reperfusion mice.METHODS: Ischemia reperfusion in cerebral injury model was duplicated in mice. Using nuclear magnetic resonance (NMR) and high performance liquid chromatography (HPLC), the brain metabolism were measured. The influence of 9602 prescription on cerebral energy metabolism in ischemia reperfusion mice model was dynamicly observed. RESULTS: Phase Ⅰ: The spectrum of NMR showed that after 10 min of ischemia, the PCr peak dropped significantly, while the Pi peak rose significantly in both the control and the “9602” group. There was no remarkable difference between the two groups. After reperfusion the PCr peak in the control group continued dropping slowly and remained at a low level (55.50?14.94) after 10 min of reperfusion, while after reperfusion the fallen PCr peak in the “9602” group started rising till 76.72?13.37 (P0.05).
