1.Isolation and culture of mouse embryonic fibroblasts and preparation of feeder layers

BACKGROUND:It is important to produce and save a large amount of high-activity feeder cel s for the culture of human embryonic stem cel s.
OBJECTIVE:To establish the optimal method for isolation and culture of Kunming mouse embryonic fibroblasts, and to evaluate the feasibility of preparing feeder layers for culture of human embryonic stem cel s.
METHODS:Embryonic fibroblasts were isolated and cultured by different concentrations of trypsin from Kunming mouse fetuses in vitro. The biological characteristics and growth rule of mouse embryonic fibroblasts were investigated, and then the feeder layers for human embryonic stem cel s culture were produced. The growth of human embryonic stem cel s on the prepared feeder layer was tested.
RESULTS AND CONCLUSION:The optimal fetal age for preparing Kunming mouse embryonic fibroblast feeder layer was 13.5 days. Kunming mouse embryonic fibroblasts at different concentrations grew wel with high purity and active proliferation by trypsin digestion method. There was no significant difference in the survival rate of cel s after cryopreservation for 2 weeks, 1 month, 3 months and 6 months. The cel s were proliferative from the second to fourth passage and declined sharply after the fifth passage. Human embryonic stem cel s which grew on Kunming mouse embryonic fibroblasts feeder layers were stil to remain the typical undifferentiated morphology and were strongly positive for alkaline phosphatase and periodic acid-Schiff after long-term subculture. The
mouse embryonic fibroblasts can be used as the stable and high-quality feeder cel s for human embryonic stem cel s.
2.Effect of culture supernatant of rat bone mesenchymal stem cells on biological characteristics of RSC96 cells

Objective:To study the effect of culture supernatant of rat bone marrow mesenchymal stem cells (BMSCs) on biological characteristics of RSC96 Schwann cells.Methods: BMSCs of Sprague Dawley rat were cultured and purified in vitro.The BMSCs culture supernatant medium (BMSC-CM) was collected from the third generation of BMSCs.Using MTT method to detect proliferation influence of BMSC-CM on RSC96 cells,and using plate cloning assay to analysis a single cell proliferation effect.The migration analysis of BMSC-CM on RSC96 cells were detected by scratch and TranswellTM chamber assay,Western blot tested Bax and Bcl-2 proteins changes in RSC96 cells.Results: The third generation of BMSCs was long spindle-shaped and vortex-like;the result of Osteogenesis staining was positive;lipid droplets appeared after adipogenesis induction.BMSC-CM inhibited the proliferation and migration of RSC96 cells.Meanwhile,Bcl-2 protein decreased and Bax protein increased in RSC96 cells after BMSC-CM treatment.Conclusion: BMSC-CM can promote apoptosis and inhibit the proliferation and migration of RSC96 cells.
3.Application of Ultracision Harmonic Scalpel in Laparoscopic Radical Gastrectomy

Objective To study the application of ultracision harmonic scalpel in laparoscopic radical gastrectomy.Methods Ten patients with gastric cancer were given laparoscopic-assisted radical gastrectomy by using ultracision harmonic scalpel.Results All operations were successfully performed with ultracision harmonic scalpel,and none of which converted into open surgery.The operation time was 300-492 min,mean(385?64) min.The blood loss was 100-500 ml,mean(401?70) ml.The number of harvested lymph nodes was 21-43,mean 31?6.The time for gastrointestinal function recovery was 3-6 d,mean(4.2?1.0) d.The time of patients' taking out-of-bed activity was 3-7 d,mean(4.5?1.3) d.The time of taking liquid food was 4-6 d,mean(5.0?0.9) d.No case had relapse or metastasis after 4-20 months(mean 12.6 months) of follow-up.Conclusions Laparoscopic radical gastrectomy by using ultracision harmonic scalpel is safe and feasible.Ultracision harmonic scalpel has the advantage of minimal invasion,less bleeding and shorter operation time,which is a very important equipment and useful for laparoscopic gastrointestinal surgery.

Objective To investigate the correlation between postoperative intraabdominal adhesions and tumor growth and metastasis and to evaluate the effect of PDLLA. Methods Sixty male Wistar rats were divided into tumor transplantation group ( A group) and non-tumor-transplantation group (B group). Phospholipid membrane vesicles from cells play key roles in cell communication, antigen presentation, and transmission of infectious agents.Extracellular vesicles(EVs) carry various proteins, mRNA, and microRNA(miRNA), like a truck to bring different molecular cargoes to remote cells.Furthermore, the encapsulated molecules by EVs can be prevented from being degraded by enzymes in body fluids, making EVs as a natural, novel drug delivery system.In this paper, we review the progress of EVs for biological application and drug delivery vehicles from the classification and composition, which provides a theoretical basis for seeking new drug delivery systems for the treatment of diseases.
6.Effects of intravenous anesthesia with sufentanil combined with propofol on hemodynamics and oxygen metabolism in patients with esophageal cancer under high altitude

Objective To investigate the effect of intravenous anesthesia with sufentanil combined with propofol on hemodynamics and oxygen metabolism in patients with esophageal cancer under high altitude.Methods 60 patients with esophageal cancer radical surgery was selected from March 2015 to March 2017 in our hospital and were divided into two groups according to the random number table,30 patients in each group.Patients in the control group were given with fentanyl and propofol intravenous anesthesia,and patients in the observation group were given sufentanil combined with propofol anesthesia.The occurrence of restlessness during the recovery of anesthesia in the two groups were observed,the spontaneous breathing recovery time,extubation time and postoperative Mini-mental State Examination (MMSE) score were recorded,and the indexes of hemodynamics and oxygen metabolism were measured at the time of before induction of anesthesia (T0),tracheal intubation (T1),skin incision (T2),thoracotomy (T3),free esophagus (T4),esophageal catheter removal time (T5).Results The spontaneous respiration recovery time and extubation time of the observation group were significantly shorter than those of the control group (P < 0.05).The degree of restlessness in the observation group was weaker than that in the control group (P < 0.05).Mean arterial pressure (MAP) in the control group at T2-T4 were significantly higher than those in T0 (P < 0.05),but MAP and heart rate (HR) in the observation group at T1-T5 were significantly lower than those of T0 (P < 0.05).Compared with T0 group,oxygen saturation (SpO2),oxygen saturation of mixed venose blood (SvO2) and oxygen delivery (DO2) at T1-T5 were significantly decreased (P < 0.05),oxygen consumption (VO2) at T2-T4 was significantly increased (P < 0.05),especially in the control group (P < 0.05).The MMSE scores of the two groups were significantly lower than those before the operation (P < 0.05),and the scores in observation group was significantly higher than the control group (P < 0.05).Conclusions The intravenous anesthesia with sufentanil combined with propofol is more helpful in maintaining hemodynamic stability and oxygen metabolism balance than intravenous anesthesia with fentanyl combined with propofol and has little effect on postoperative anesthesia recovery and cognitive function.
7.Progress of guanylate binding protein 5 in tumors

Malignant tumor ranks as a leading cause of human death. Guanylate binding protein 5 (GBP5) belongs to the family of interferon-γ-inducible large GTPase superfamily and participates in various pathological and physiological processes. A growing number of studies have indicated that GBP5 is involved in the malignant progression of tumors. This article mainly reviews the biological characteristics of GBP5 and its expression, regulation and function in tumors, in the hope of providing a theoretical basis for the diagnosis and treatment of tumors.

To understand the possibility of schistosomiasis transmission on the East Route Project of the South-to-North Water Diversion Project, a survey of endemic status of schistosomiasis in Weishan Lake area was conducted. A cluster sampling of 2086 boatmen were screened with DDIA Kit, and the positive ones were examined with Kato-Katz technique. Meanwhile, a questionnaire survey about schistosomiasis control was carried out to collect the data about the boatmen's general information and knowledge , attitude and practice (KAP). The results showed there were no schistosomiasis patients. However, there exist potential risk factors for schistosomiasis transmission because of the bad hygiene habits and the poor knowledge of schistosomiasis prevention in boatmen.
9.Cockroach as a Vector of Blastocystis sp. is Risk for Golden Monkeys in Zoo

Blastocystis sp. is a kind of protozoa living in the intestinal tract of human and animals, which will cause intestinal diseases such as diarrhea, abdominal distension and vomiting. This paper was aimed to understand the infection of Blastocystis sp. In golden monkeys and the transmission path in North China. Thirty-seven feces samples from golden monkeys and 116 cockroach samples from Shijiazhuang Zoo were collected from July to October 2019 for PCR analysis of Blastocystis sp. Genetic diversity analysis was further conducted on the samples with positive PCR results. The results showed that the infection rate was 48.7% (18/37) in golden monkeys and 82.8% (96/116) in cockroaches, respectively. The genetic evolution analysis based on small subunit ribosomal RNA demonstrated that three subtypes (ST) of Blastocystis sp. including ST1, ST2, and ST3 existed in the intestinal tract of golden monkeys, while only ST2 was detected in the intestinal tract of cockroaches. This paper may provide supports for the quarantine and control of Blastocystis sp. for the zoo in Northern China.
10.Constructing an early-warning model for mortality risk in heat stroke patients based on Fisher discriminant analysis

Objective:To establish an early warning model to assess the mortality risk of patients with heat stroke disease.Methods:The case data of patients diagnosed with heat stroke disease admitted to the comprehensive ICU of Shanshan County from January 2016 to December 2020 were selected. According to the short-term outcome (28 days) of patients, they were divided into death group (20 cases) and survival group (53 cases) . The relevant indicators with statistically significant differences between groups within 24 hours after admission were selected. By drawing the subject work curve (ROC) and calculating the area under the curve, the relevant indicators with the area under the curve greater than 0.7 were selected, Fisher discriminant analysis was used to establish an assessment model for the death risk of heat stroke disease. The data of heat stroke patients from January 1, 2021 to December 2022 in the comprehensive ICU of Shanshan County were collected for external verification. Results There were significant differences in age, cystatin C, procalcitonin, platelet count, CKMB, CK, CREA, PT, TT, APTT, heart rate, respiratory rate and GLS score among the groups. Cystatin C, CKMB, CREA, PT, TT, heart rate AUC area at admission was greater than 0.7. Fisher analysis method is used to build a functional model.Results:The diagnostic sensitivity, specificity and AUC area of the functional model were 95%, 83% and 0.937 respectively. The external validation results showed that the accuracy of predicting survival group was 85.71%, the accuracy of predicting death group was 88.89%.Conclusion:The early warning model of heat stroke death constructed by ROC curve analysis and Fisher discriminant analysis can provide objective reference for early intervention of heat stroke.
