2.Study on the mechanism of miRNA-26a inhibiting ox-LDL-mediated apoptosis of HAECs
Chongqing Medicine 2016;45(36):5052-5055
Objective To investigate the role of miR‐26a in ox‐LDL‐mediated apoptosis of HAECs in endothelial cells and its mechanism .Methods Various concentrations of ox‐LDL were added in HAECs culture .Cell cytotoxicity and apoptosis were moni‐tored by MTT and TUNEL assay ,and expression level of miR‐26a examined by qRT‐PCR .Overexpression of miR‐26a mimic in HAECs ,MTT and TUNEL staining were used to detect the activity and apoptosis of ox‐LDL .The 3′UTR of luciferase reporter vector pMIR‐PTEN was constructed and the predicted target gene of miR‐26a was identified by luciferase activity assay .QRT‐PCR and Western blot were used to detect the mRNA and protein expression of PTEN .Results ox‐LDL could mediate the toxic death and apoptosis of HAECs cells ,and decrease the expression level of miR‐26a in HAECs cells .Overexpression of miR‐26a mimic could inhibit the cytotoxicity and apoptosis of ox‐LDL cells after HAECs .Transfection of miR‐26a mimics significantly inhibited lu‐ciferase activity (P<0 .05) .The expression of mRNA and protein in HAECs cells was significantly down regulated by transfection of miR‐26a analog (P<0 .05) .Conclusion MiR‐26a can inhibit the cytotoxicity and apoptosis of ox‐LDL cells after HAECs inhibi‐tion ,and the possible mechanism of action is to down regulate the expression of PTEN .The study suggests that miR‐26a may be a potential target for the treatment of atherosclerosis related to apoptosis .

Objective To control stem blight disease of Schizonepeta tenuifolia caused by Phytophthora nicotianae. Methods The antagonist effect of 13 Trichoderma strains(including T.viride and T.hamianum)was evaluated upon mycelia growth of P.nicotianae.Trichoderma strains with high antagonistic activities against the pathogen were used to control stem blight of S.tenuifolia in the field. Results Of 13 Trichoderma strains tested,T.viride strain M3 showed maximum mycelia growth inhibition(83.2%)to the pathogen, followed by T.viride strain Tv04-2(78.2%)and then T.harziamum strain ThB(65.0%),in vitro.Fungal cell wall degrading enzymes, protease, and β-1,3-glueanase were analyzed qualitatively and quantitatively in further study.T.viride strains M3,Tv04-2,and T.harzianum strain ThB efficiently against P.nicotianae were used to control stem blight of S.tenuifolia in the field, and T.viride strain M3 showed the best biocontrol potential. Conclusion Trichoderma spp.can be used as alternatives of pesticides to control stem blight, one of the serious soilhome diseases of S.tenuifolia caused by P.nicotianae.However,though T.viride strains Tv04-2 aad T.harzianum strain ThB are also highly against P.nicotianae in vitro, the controlling efficacy of them on stem blight disease is not as excellent as T.viride strains M3 in the field.

Objective By using optical coherence tomography (OCT) to observe the residual subretina fluid changes in fovea that may explain visual recovery after anatomically successful repair of rhegmatogenous retinal detachments (RD). Design Prospective observational case series. Participants 34 patients(35 eyes) with successful retinal reattachment after macula involving RD repair. Methods 34 patients were followed up at 3 days, 15 days, 2 months and 6 months after sclera buckling. The best corrected visual acuity(BCVA), eye fundus status were recorded. The OCT images through the center of the fovea were obtained. Main Outcome Measures The OCT images and BCVA after RD repair. Results The BCVA and OCT images were obtained 3 days to 6 months postoperatively. There were 3 eyes, 5 eyes, 9 eyes and 23 eyes respectively with residual subfovea fluid absorbed totally at 3 days, 15 days, 2 months and 6 months. In the residual subforvea fluid group, the mean visual acuity increased at 0.124?0.013. In the subfovea fluid absorbed group, the mean visual acuity increased at only 0.019?0.006. Conclusions After anatomically successful repair of RD performend with sclera buckling, the minimal residual subforvea fluid will exist for a long time and will be absorbed slowly. This may explain slow increase of postoperative visual acuity after anatomically successful repair.

Objective To determine the effect of tumor necrosis factor ?(TNF ?) on p38 mitogen activated protein kinase (MAPK) activation in human pulmonary microvascular endothelial cells (HPMEC) in vitro Methods HPMEC was isolated and cultured in vitro Expression and activation of p38 MAPK were detected by western blot and immune complex kinase assay Results TNF ? induced phosphorylation and activation of p38 MAPK in HPMEC in a time/dose dependent manner which was suppressed by SB203580,a specific inhibitor of p38 MAPK Conclusion TNF ? plays a important role in activation of p38 MAPK and signal transduction in HPMEC

Journal of Acupuncture and Tuina Science 2007;5(6):321-323
Different from the traditional teaching method, PBL (Problem-Based Learning) teaching mode is emphasized to foster the students' skills in spontaneous study and problem solving and the analyzing and comprehensive thinking ability. The authors believe that in the application of the PBL teaching method, the teachers' functions should be stressed on how to select the initial moment, provide the topics, excite the interesting points, search information, make comments and evaluate the students' ability, i.e. to offer fish and teach how to catch fish as well.

Objective To evaluate the therapeutic efficacy of intratympanic dexamethasone injections combined with prednisone in patients with idiopathic sudden sensorineural hearing loss. Methods A total of 71 patients diagnosed with sudden hearing loss were treated with intratympanic dexamethasone injections plus prednisone (B group) or prednisone alone (A group). Hearing was evaluated by pure tone audiogram performed before initial treatment and at 4 weeks following the final treatment. Results The total recovery rate after the treatment was 81.8% in the B group and 55.3% in the A group. The diflference between two groups was statistically significant (P < 0.05). Conclusion The present study suggests that sudden sensorineural hearing loss patients treated with intratympanic dexamethasone combined with prednisone have a higher likelihood of hearing recovery than those treated with prednisone alone.

Objectives To detect the loss of heterozygosity(LOH)frequencies of microsatellite loci D9S171and D9S1604in p16gene of psoriatic keratinocytes, and to study the correlation between mi-crosatellite LOH of p16gene and the development of psoriasis. Methods By the use of polymerase chain reaction(PCR)-denaturing polyacrylamide gel electrophoresis-silver staining,LOH was detected with23sam-ples of keratinocytes from psoriatic lesions and non-lesion skin. Results LOH was identified at loci D9S171and D9S1604in5and10out of23keratinocyte samples from LOH-exhibited psoriatic lesions, and in2and3of keratinocyte samples from psoriatic non-lesion skin, respectively. The frequency of LOH at D9S1604was significantly higher in psoriatic lesion samples than that in psoriatic non-lesion skin(P
