1.Technology of supercritical CO_2 extraction process of Dragon's Blood

Object To study the technology of supercritical CO_2 extraction of effective components from Dragon's Blood which has the inhibiting function on ?-glucosidase. Methods The influence of the extraction pressure, temperature, time and fluid velocity on surplus extraction rate and the improvement of inhibition was mainly discussed. Results The optimal experimental parameters were 15 MPa extraction pressure, 45 ℃ extraction temperature, 30 kg/h fluid velocity and 2 h extraction time. Conclusion The results show that SFE-CO_2 can enormously increase the surplus extraction rate and the inhibitory activities on ?-glucosidase.
2.Analysis of Correlation between the Number of Macrophages in Deep Burn Wound and the Concentrations of sTNFR and sVCAM-I in Wound Fluid

Objective To detect the concentrations of (soluble TNF receptor type,sTNFR-I, sTNFR-II) and (soluble vascular cell adhesion molecule-1,sVCAM-I) in deep burn wound fluids and the number of macrophages in different stage of human deep burn wound, and to analyse the correlation between them. Methods The wound fluid was collected with sponge during the dressing change, and then the concentrations of sTNFR-I, sTNFR-II and sVCAM-I were detected by ELISA. The tissues of burn wound were collected at operation, and the tissue slices were dyed with HE.The macrophages were marked by immunohistochemistry with antibody of CD68, and then the number of macrophages was counted under the microscope. Results The concentrations of sTNFR-I and sTNFR-II in burn wound fluid and the number of macrophages in burn wound were significantly increased two days postburn, which maintained at high levels until the wound healed. The concentration of sVCAM-I was decreased immediately after the burn until the wound healed. The correlation coefficient between the concentrations of sTNFR-I, sTNFR-II, sVCAM-I and the number of macrophages were 0.95, 0.97 and -0.37, respectively. Conclusion The number of macrophages has a strong correlation with sTNFR-I and sTNFR-II in human deep burn wound, and has a weak correlation with sVCAM-I,which suggests that the macrophages may play a cooperative role with sTNFR in burn wound healing.

4.Sterol extracts from Begonia Sinensis Rhizome against respiratory inflammation.

The acute and chronic respiratory tract inflammation models were made to investigate the effect and mechanism of sterol extracts from Begonia Sinensis Rhizome (BSR). The first model of acute lung injury was made with Kunming mice by inhaling cigarette smoke, then the mice were treated with different concentrations of BSR sterol extracts. Lung tissue morphology was detected by HE staining, TNF-alpha/MPO were detected by Elisa, and cPLA2 protein were, detected by Western blotting respectively. Results showed that in model group, lung sheet became real, alveolar space shrank or disappeared, alveolar septum was thickened, plenty of inflammatory cells were infiltrated, capillary blood vessels were congestive and the expression of TNF-α, MPO, cPLA2 increased; after administration, a small amount of inflammatory cells were infiltrated, alveolar septum became obvious, capillary congestion status was significantly relieved and the expression of TNF-α, MPO, cPLA2 decreased (P < 0.05). The second model of chronic respiratory tract inflammation in BALB/c mice with bronchial asthma was induced by OVA, then the mice were treated with different concentrations of BSR sterol extracts. Lung tissue morphology was detected by HE staining, indexes such as IL-4, IL-5, IL-13 were detected by Elisa, and the cPLA2 protein expression was detected by Western blotting respectively. Results showed that in model group, a lot of inflammatory cells around lung vessels and bronchi exuded, bronchial goblet cells proliferated and the expression of IL-4, IL-5, IL-13, cPLA2 increased; after administration, inflammatory and goblet cell hyperplasia reduced, the expression of IL-4, IL-5, IL-13, cPLA2 also decreased (P < 0.05). The above results showed BSR sterol extracts could resist against respiratory inflammation by inhibiting cPLA2 in a dose-dependent manner.

5.Effect of saikosaponin on GABA of hippocampus of chronic kindling induced by pentylenetetrazol in rats

Objective To study the effect of active ingredients(saikosaponin) of bupleurum chinensis D C.on GABA of hippocampus of chronic kindling induced by pentylenetetrazol(PTZ) in rats.Methods Forty-eight healthy Sprague-Dawley rats were randomly divided into six groups with eight in each group: normal control(Group A), normal saline(NS) group(Group B),sodium valproate(VPA) group(Group C),group of large-dose saikosaponin(Group D),group of medium-dose saikosaponin(Group E),and group of small-dose saikosaponin(Group F).Except Group A to be controlled,the other groups were given different treatments when chronically kindled by PTZ.The process of the test lasted for 4 weeks.After 4 weeks' administration the brain tissues were sampled,sliced and stained by enzyme immunohistochemistry(EIH).The results were analyzed according to the positive cell population and gray value.Results The positive cell population of CA1,CA2 and DG in group B was significantly different from that in the other groups(P
6.PREPARATION AND RELATED STUDY OF DECALCIFIED BONE MATRIX PARTICLES WITH ADRIAMYCIN IMPREGNATED BONE CEMENT DRUG DELIVERY SYSTEM IN VIVO

To prepare decalcified bone matrix particles(DBMP)with adriamycin(ADM) impregnated bone cement(BC) drug delivery system(DDS), its drug delivery characteristics were studied and analysed in vivo. DBMP were made according to the methods of Urist et al. ADM was putting into DBMP by vacuum absorption and freeze drying techniques. Then the DBMP with ADM were impregnated into BC in 1∶1 to form DDS. The DDS was implanted into the greater trochanter of femur in rabbits and the release test was done in vivo. The DDS could gradually release ADM for over 12 weeks in local bone tissues, maintained stable release for 4 weeks. The maximum ADM in local bone tissues was 29 times higher than that of the same intrarenous dose, while the maximum ADM in plasma was only 1/10 of the same intravenous dose. The DDS has a good sustained release function, and can meet the needs of local chemotherapy for limb salvage treament of bone tumor.

A cardiac stimulator for rapid atrial pacing animal model was designed in this paper. According to the needs of clinical research, output pulse parameters of the cardiac stimulator can be designed. The cardiac stimulator will be controlled through magnet. Efficiency of the cardiac stimulator was validated by animal experiments.
