OBJECTIVESTo evaluate the diagnostic value of arterial spin labeling (ASL) technology in newborns with hypoxic ischemic encephalopathy (HIE).

METHODSeven full-term newborn infants without any history of asphyxia and other nervous system diseases were selected as the control and 33 full-term newborn infants were assigned into HIE group. The patients in HIE group were further divided into three subgroups (19 cases of mild, 6 cases of moderate and 8 cases of severe HIE) based on their clinical diagnosis. The control group and HIE group were examined with GE Signa EXCITE HD 3.0T superconducting MRI scanner with a head phase array coil. Both groups were scanned with conventional axial MRI (T1FLAIR, T2WI and T2FLAIR), 1HMRS (PRESS sequence) and ASL (FAIR). Original images of 1HMRS and ASL were processed by Functool software of ADW 4.3 workstation. ASL perfusion images were observed and the signal intensity values of the region of interest (bilateral gray, white matter and basal ganglia) of the two groups were quantitatively measured, and mean value were calculated and compared between groups. Statistical analysis was performed with SPSS 13.0 software, and statistically significant difference was set at P < 0.05.

RESULTThe perfusion images of two groups were obtained perfectly. The signal intensity values of bilateral gray, white matter and basal ganglia of control group were 125.34 ± 11.76, 73.42 ± 11.67 and 173.65 ± 15.49, respectively and there was a statistically significant difference between the different areas. The signal intensity values of bilateral gray, white matter and basal ganglia of HIE group were 153.47 ± 11.72, 71.35 ± 10.37 and 217.13 ± 12.51, respectively. There was a statistically significant difference (P < 0.05) in the average signal intensity value of gray matter and basal ganglia, but there were no statistically significant difference (P > 0.05) in white matter between the two groups.

CONCLUSIONASL Perfusion technique can assess HIE comprehensively and accurately. Furthermore, it can evaluate the brain damage of hypoxic ischemia. The results provide a strong basis for clinical treatment.

About one-quarter (26%) of the patients showed positive results for oligoclonal bands, and about half of them exhibited increases in the IgG index and 24-hour IgG synthesis rate. Magnetic resonance imaging showed white- and gray-matter lesions in 83% and 23% of the patients, respectively. Steroids were the main treatment, and full recovery occurred in 62% of the patients, with residual focal neurological deficits recorded in a few patients. After a mean follow-up period of 3.4 years, two patients exhibited recurrence and one patient exhibited a multiphasic course. One patient was diagnosed with multiple sclerosis (MS). CONCLUSIONS: With the exception of the seasonal distribution pattern and prior vaccine rate, the clinical profiles of ADEM in adult Chinese patients are similar to those in pediatric populations. No specific markers are available for distinguishing ADEM from MS at the initial presentation. Careful clinical evaluations, cerebrospinal fluid measurements, and neuroradiological examinations with long-term follow-up will aid the correct diagnosis of ADEM.

Angiogenic gene therapy and cell-based therapy for peripheral arterial disease(PAD) have been studied intensively currently. This study aimed to investigate whether combining mesenchymal stem cells(MSCs) transplantation with ex vivo human hepatocyte growth factor(HGF) gene transfer was more therapeutically efficient than the MSCs therapy alone in a rat model of hindlimb ischemia. One week after establishing hindlimb ischemia models, Sprague-Dawley(SD) rats were randomized to receive HGF gene-modified MSCs transplantation(HGF-MSC group), untreated MSCs transplantation (MSC group), or PBS injection(PBS group), respectively. Three weeks after injection, angiogenesis was significantly induced by both MSCs and HGF-MSCs transplantation, and capillary density was the highest in the HGF-MSC group. The number of transplanted cell-derived endothelial cells was greater in HGF-MSC group than in MSC group after one week treatment. The expression of angiogenic cytokines such as HGF and VEGF in local ischemic muscles was more abundant in HGF-MSC group than in the other two groups. In vitro, the conditioned media obtained from HGF-MSCs cultures exerted proproliferative and promigratory effects on endothelial cells. It is concluded that HGF gene-modified MSCs transplantation therapy may induce more potent angiogenesis than the MSCs therapy alone. Engraftment of MSCs combined with angiogenic gene delivery may be a promising therapeutic strategy for the treatment of severe PAD.

The co-occurrence of blue rubber bleb nevus syndrome (BRBNS) and ventricular septal defects is rare. Here we present a case of BRBNS in a 15-year-old boy who was born with multiple cavernous hemangiomas and a ventricular septal defect. Examinations revealed the presence of hemangioma lesions in the subcutaneous and mucosal tissues as well as in the cerebrum, nasopharynx, tongue, esophagus, gastric body, sigmoid colon and adrenal gland. Combined imaging modalities played an important role in the diagnosis of hemangioma lesions.

Objective:To investigate the surgical treatment of the tumors at cervicothoracic junction.Methods:A retrospective analyses was performed for 63 patients with tumors at the cervicothoracic junction receiving surgery from Mar 2008 to May 2020 in the Department of Thoracic Surgery, Zhongshan Hospital, Fudan University. Clinical data about manifestation, surgical approach, resection degree and pathological types were collected. There were 43 cases of asymptomatic patients and 20 cases of patients with ≥1 clinical manifestations. Twenty two patients receiving radical resection with video-assisted thoracoscopic surgery. Anterior approach was the most popular treatment in open surgery (24 cases, 38.1%), and 8 cases of anterolateral approach(6 cases of Hemiclamshell incisions, 2 cases of trap-door incisions), 1 case of posterior approach, 2 cases of posterolateral approach and 1 case of supraclavicular combined posterolateral approach.Results:Pathological examination suggested 61 cases of radical resection and 2 cases of microscopic residual. Neurilemmoma was the most common pathological type (27 cases, 42.9%), the second common pathological type was tumor originated from fibrous tissues (6 cases, 9.5%). The 3-year overall survival rate of those 63 patients was 88.9%, while the 5-year overall survival rate was 84.1%.Conclusion:Tumors involving the cervicothoracic junction are characterized as special location, complicated anatomy and various histopathological subtypes. Individualized approach and surgery improve safety and normalization of tumors at cervicothoracic junction treatment.

Objective To study the cytotoxic effect of cytokine-induced killer cells (CIKs) cocultured with U251 tumor cell antigen-loaded mature dendritic cells (DCs) against U251 cell line. Methods The DCs and CIKs were derived from the cord blood mononuclear cells (CBMCs) of the same donor. The DCs were challenged with U251 tumor cell antigen, and cocultured with the CIKs to induce the cell complex Ag-DC-CIK. The mature DCs were identified by morphological and phenotypic analyses. MTT assay was performed to detect the cytotoxic effects ofCBMCs, CIKs, antigen-loaded CIKs (Ag-CIK) or the cell complex Ag-DC-CIK in U25 ! cells. Results The mature DCs derived from the CBMCs highly expressed the costimulatory molecules CD86 (82.66%) and CD40 (69.40%), and moderately expressed CD83 (57.49%) and CD80 (51.14%). The cytotoxic activity of the cell complex Ag-DC-CIK against U251 cells (58.8%) was significantly higher than those of CBMCs (29.71%), CIKs (39.89%), and Ag-CIK (49.92%). Statistical analysis indicated significant difference in the cytotoxic activity between any two of the groups (P<0.05). Conclusion The DCs loaded with the tumor cell antigen can enhance the cytotoxic effect of the CIKs against the target tumor cells, which sheds light on a new approach of immunotherapy for intracranial tumors.

Objective To analyze the clinical and imaging features of a family(3 patients)with familial hemangioblastoma,and their diagnosis and prognosis.Methods The detailed data about clinical and imaging features of all patients diagnosed as familial hemangioblastoma,admitted to our hospital from October 2004 to May 2010,were analyzed,and the lesions of other regions,besides the tumor lesion,were observed.Results No lesions of other regions were noted in these 3 patients.Cranial MRI showed that 2 had cystic and solid tumor and 1 had solid tumor;,total removal was performed on these patients under microscope; regular follow-up was given and no recurrence was noted.Conclusion Familiar hemangioblastoma is serious hereditary disease; and MRI is the most important detective method; microsurgical operation is the most important therapy.Early diagnosis and treatment should be given to the patients with familiar hemangioblastoma due to its high recunence rate,having difficulty in operation and its trend to combining with other lesions of the other parts.

【Objective】 To investigate the feasibility of using the indirect labeling method to label small molecular peptide cNGQGEQc with ¹⁸⁸Re and to observe the inhibitory effect of ¹⁸⁸Re-cNGQGEQc on lung adenocarcinoma cell line A549 in vitro. 【Methods】 ¹⁸⁸Re-cNGQGEQc and ¹⁸⁸Re-cNAQAEQc(negative polypeptide radiolabeled with 188Re) were prepared by indirect labeling method with ethylene dicysteine(EC) as the bifunctional chelating agent. The labeling rate, specific activity, radiochemical purity(RCP) and octanol-water partition coefficient were determined, and the stability in normal human serum was evaluated. CCK-8 assay was used to detect the inhibitory effects of ¹⁸⁸Re-cNGQGEQc, ¹⁸⁸RecNAQAEQc and free ¹⁸⁸ReO4^- with different radioactive doses on A549 lung cancer cell proliferation. Then the relative inhibitory rates and 50% inhibiting concentration(IC50) of the three peptides were calculated and compared. 【Results】 The labeling rate, specific activity and log P value of ¹⁸⁸Re-cNGQGEQc were(90.24±1.58) %, (4.07±0.14) TBq·mmol/L^-1 and(-2.90±0.03), respectively. The RCP of ¹⁸⁸Re-cNGQGEQc purified by using a Sep-Pak C18 column and after being placed in normal serum for 24 h at 37 ℃ were(98.42±0.32) % and(89.08±0.94) %, respectively. CCK-8 assay results revealed that ¹⁸⁸Re-cNGQGEQc significantly inhibited the proliferation of A549 lung cancer cells in a dose-dependent and positively-correlated manner. IC50 value of ¹⁸⁸Re-cNGQGEQc was 44.88×10⁷ Bq/L, significantly lower than 93.45× 10⁷ Bq/L for ¹⁸⁸Re-cNAQAEQc and 99.60×10⁷ Bq/L for ¹⁸⁸ReO4^- (P<0.01) . 【Conclusion】 It is feasible to use EC as the bifunctional chelating agent to label small molecular peptide with ¹⁸⁸Re and this labeling method has high labeling rate and good in vitro stability. The in vitro experiment confirms that ¹⁸⁸Re-cNGQGEQc can effectively inhibit the proliferation of lung adenocarcinoma cell line A549. These results provide the experimental basis for further in vivo application of small molecular peptides for the targeted therapy of lung cancer.

In the present study, we used a proteomics approach based on a two-dimensional electrophoresis (2-DE) reference map to investigate protein expression in the ovarian tissues of pubertal Swiss-Webster mice subjected to carbon ion radiation (CIR). Among the identified proteins, ubiquitin carboxy-terminal hydrolase L1 (UCH-L1) is associated with the cell cycle[1] and that it influences proliferation in ovarian tissues. We analyzed the expression of UCH-L1 and the proliferation marker proliferation cell nuclear antigen (PCNA) following CIR using immunoblotting and immunofluorescence. The proteomics and biochemical results provide insight into the underlying mechanisms of CIR toxicity in ovarian tissues.
