OBJECTIVETo elucidate the necessary and research of accelerating basic research of Chinese standard pieces as standard materials.

METHODAccording to over 10 years accumulated experience and be keenly aware of the author, the evaluation method of standardized processing technology and Chinese pieces quality, aimed at consummated the standard material of the quality evaluation of Chinese herbal pieces at the current situation, and inaccordance with the need of improving quality standard system of Chinese herbal pieces, illustrate the necessity of accelerating basic research of Chinese standard pieces as standard materials; from the technical specification for collecting and processing of raw materials, and the technical specification, homogenized sample, packaging, storage and etc., for processing of candidate standard pieces, determine the methods and steps of technical specifications for standard pieces as the standard substance, determine the methods and steps of technical specifications for standard pieces as the standard substance.

RESULT AND CONCLUSIONTo speed up the basic research of standard of Chinese medicine pieces as of standard material is very necessary. The research objective is to specificate the processing technical for a number of standard pieces, to identify technical specifications and to ascertain the guiding principle and technical specification of decoction pieces as standard substance. This research will provide basic scientific data relevant national departments to apply for the accreditation of the standard substance.

Objective To investigate formalin-induced nociceptive behavior and c-fos expression of interbrain following stellate ganglionic block (SGB) in rabbits. Methods Catheters were inserted closely to right stellate ganglia in rabbits by operation.One week later,twenty-four rabbits were randomly divided into three groups, each group containing 8 animals. groups A,B and C received 0.5ml of normal saline,3% formalin and 3% formalin stimulation by intraplantar injection , respectively. 10 minutes before stimulation,0.5 ml of 0.25% bupivacaine was administered via the catheter in group B, while in groups A and C 0.5 ml of normal saline was applied. Nociceptive behavior was observed for 1 hour using weighted pain score. 2 hours after stimulation, whole interbrain was removed to immunohistochemically detect the expression of c-fos. Results Formalin-induced nociceptive behavior in phase 2 was relieved following SGB, c-fos expression level of hypothalamus in group B was significantly lower than that in group C(P0.05). Conclusion SGB could relieve formalin-induced nociceptive behavior, and downregulate formalin-induced c-fos expression of hypothalamus, which may be related to the mechanisms of SGB for the treatment of inflammatory pain.

To explore the effective components represented by fingerprint contributed to allelopathic effect of different Salvia miltiorrhiza aqueous concentration on seeds and seedlings of radish, grey relational analysis was used to establish the chromatography-efficacy relation. The results show that 15 peaks devote high allelopathic contribution to radish seeds and seedlings. The study will provide a new concept for allelochemicals screening and study.

Objective To explore the impact of the variable number of tandem repeats of monoamine oxidase A gene (MAOA-uVNTR) on the intensity of brain activation during the recognition of facial expression in patients with depression and healthy controls.Methods 28 cases of depression,as well as 33 healthy controls who were matched in gender, age and years of education were divided into different genotypes with the methods of polymerase chain reaction (PCR) amplification and 1.5% agarose gel electrophoresis separation.61 cases were scanned to compare the intensity of brain activation in the recognition of happy, sad and neutral facial expression.Results In healthy controls,cases with high-activity genotype showed increased activation in left cuneus,left inferior frontal gyrus, right medial frontal gyrus and left inferior parietal lobule in comparision with carriers of low-activity genotype.In the depressed, compared with patients with low-activity genotype, cases with high-activity genotype decreased activation in bilateral putamen, left postcentral gyrus, left fusiform gyrus, right superior temporal gyrus and right thalamus.Conclusion Healthy controls with high-activity genotype shows the trend of priority for the identification of negative emotion,this genotype may be one of the risk factors for normal people suffering from depression.Patients with high-activity genotype is associated with the inhibitory of positive emotional state, this may attribute partly to the emotional symptoms in such kind of patients more serious.

Objective To observe the expression of SGK in decidua in patients with unexplained recurrent spontaneous abortion(URSA) and to investigate its role in the course of apoptosis.Methods The expression of SGK was detected by immunohistochemistry in decidua of URSA(abortion group,n =50) and normal first trimester pregnant women(control group,n =30).The apoptotic index was detected by TUNEL.Results Compared with the control group,the positive expression of SGK decreased significantly in the abortion group,and the difference was statistically significant(x2 =6.78,P ＜ 0.05).The average apoptotic index of the abortion group was (8.19 ± 3.58) ％,which was significantly higher than (2.87 ±1.07)％ of the control group(t =7.94,P＜0.05).Conclusion Intensive apoptosis,the decreasing positive expression rate of SGK in decidua cells may play critical roles in URSA.

Objective To examine the relationship between the A118G polymorphism of mu-opioid receptor(OPRM1) gene and analgesic effects of oxycodone hydrochloride in patients with severe cancer pain. Methods Fifty-nine patients with severe cancer-induced pain were divided into 3 groups by genotype(AA group,AG group and GG group,23,28,8 patients, respectively). They were orally treated with oxycodone hydrochloride sustained-release tablets,and the treatment dosage and adverse reactions(including nausea,vomiting,dizziness,constipation,etc.)were compared between groups. Results The variation of allele frequencies(118G)was 37.3%.The dosage of oxycodone hydrochloride sustained-release tablets used in AA, AG and GG groups was(27.0±14.3),(36.4±22.5)and(55.0±35.1)mg,respectively,and the differences were statistically significant between groups( P = 0. 01). On the part of adverse reactions,the incidence of nausea and vomiting,dizziness, constipation was 28.8%,22.0%,and 52.5%,in AA,AG and GG groups,respectively,and there was no significant difference between groups(P>0.05). Conclusion The analgesic effect of oxycodone hydrochloride is affected by opioid receptor gene polymorphism.Patients with G allelic variation(AG or GG genotype)require larger doses of oxycodone hydrochloride than those with AA genotype.However,adverse reactions are not associated with polymorphism.

Objective To explore the early diagnostic significance of anti-cyclic citrullinated peptide(CCP) antibody and hidden rheumatoid factor immunoglobulin M(HRF-IgM) detected by ELISA in patients with juvenile rheumatoid arthritis(JRA). Methods The synthesized CCP was used as the antigen to detect anti-CCP antibody. Anti-CCP antibody and HRF-IgM were detected dynamically in 27 patients with early diagnosed JRA and their specificity and sensitivity were determined for early diagnosed JRA by calculating the positive predicting value (PPV) and negative predicting value (NPV). Results The total positive rate of anti-CCP antibody and HRF-IgM were 58.5 % and 65.0 % respectively in patients with JRA. The sensitivity of HRF-IgM was more predominant than that of anti-CCP antibody. There was a positive correlation between the positive rate of antibodies and the subtype of JRA. The specificity of anti-CCP antibody for predicting early JRA was superior to that of HRF-IgM. When using these two tests in combination, the PPV predicting rate of early JRA was 93.7 % . Conclusions Both anti-CCP antibody and HRF-IgM are elevated in patients with JRA, which show positive correlations with the subtype of the disease. The specificity of anti-CCP antibody testing is considerably higher for diagnosing early JRA, and when it was used together with HRF-IgM testing, the PPV for JRA can be raised further.

Sepsis is an infection induced systemic inflammatory response syndrome and is a major cause of morbidity as well as mortality in intensive care units. A growing body of evidence suggests that the activation of a proinflammatory cascade is responsible for the development of immune dysfunction, susceptibility to severe sepsis and septic shock. The present theories of sepsis as a dysregulated inflammatory response and immune function, as manifested by excessive release of inflammatory mediators such as high mobility group box 1 protein (HMGB1), are supported by increasing studies employing animal models and clinical observations of sepsis. HMGB1, originally described as a DNA-binding protein and released passively by necrotic cells and actively by macrophages/monocytes, has been discovered to be one of essential cytokines that mediates the response to infection, injury and inflammation. A growing number of studies still focus on the inflammation-regulatory function and its contribution to infectious and inflammatory disorders, recent data suggest that HMGB1 formation can also markedly influence the host cell-mediated immunity, including T lymphocytes and macrophages. Here we review emerging evidence that support extracellular HMGB1 as a late mediator of septic complications, and discuss the therapeutic potential of several HMGB1-targeting agents in experimental sepsis. In addition, with the development of traditional Chinese medicine in recent years, it has been proven that traditional Chinese herbal materials and their extracts have remarkable effective in treating severe sepsis. In this review, we therefore provide some new concepts of HMGB1-targeted Chinese herbal therapies in sepsis.
