3.Study on the correlation between BODE and inflammatory factors in patients with chronic obstructive pulmonary disease

Objective To assess the significance and the relationship of BODE index score and inflammation factors in stable chronic obstructive pulmonary disease(COPD).Methods Sixty COPD patients in their stable condition were evaluated for BODE index score and the level of tumor necrosis factor-α(TNF-α),interleukin-8(IL-8) and C reactive protein (CRP) were determined.Results BODE index score in COPD patients was positively correlated with serum concentrations of TNF-α,IL-8 (r =0.455,P < 0.01 ; r =0.303,P <0.05),but not with CRP (r =0.111,P =0.398).IL-8 and TNF-α were both significantly negatively correlated to body mass index(BMI),force exhale volume of the first second (FEV1) and 6 minute walking distance (6MWD) (r=-0.417,P <0.01;r=-0.538,P<0.01;r =-0.419,P<0.01 for IL-8;and r=-0.262,P<0.05;r=-0.348,P<0.01;r=-0.334,P<0.01 for TNF-α).Conclusion The BODE index,as a simple multidimensional grading system for COPD,is closely related to system inflammation,which indicates that system inflammation may contribute to the systemic development of COPD.
4.Experimental research of hair follicle reconstruction with the aid of embryonic mice dermal cells.

OBJECTIVETo investigate the effect of embryonic dermal signal on the hair-inductive capacity of neonatal mice dermal cells which have been amplified in vitro.
METHODSEmbryonic mice dermal cells of embryonic day 14 were added to a chamber on the back of nude mice with neonatal mice dermal cells which had been amplified in vitro for 3 days and freshly isolated neonatal mice epidermal cells. The hair regeneration was compared between the groups with or without embryonic mice dermal cells. Meanwhile, chambers with following cells respectively were constructed as controls: embryonic mice dermal cells + neonatal mice epidermal cells; freshly isolated neonatal mice dermal cells + neonatal mice epidermal cells; amplified neonatal mice dermal cells only; embryonic mice dermal cells only; freshly isolated neonatal mice dermal cells only; neonatal mice epidermal cells only.
RESULTSThe number of regenerated hairs with the aid of embryonic mice dermal cells (207 +/- 15. 948) was significantly higher than that (67 +/- 8.963) in the group without embryonic mice dermal cells (n = 3, t = 7.653, P = 0.002).
CONCLUSIONEmbryonic dermal signal can enhance the hair-inductive capacity of neonatal mice dermal cells which have been amplified in vitro.

5.Research on automated detection technology of standard 12 lead synchronous ECG signal.

The parts of system automatically detecting the standard 12-synchronous-lead ECG (electrocardiograms) signal based on personal computer is introduced in this paper. The object-oriented programming method is adopted based on Windows System. We put forward methods to analyze QRS complex wave, P wave, T wave and ST fragment. In this paper the techniques such as ECG signal preprocessing, cardio wave parameters detecting and wave pattern cognizing are discussed too. Furthermore we use wavelet transform technique to analyze the wave pattern and get sound effect.

6.Roles of adenosine receptors in Alzheimer's disease.

As an important neurotransmitter, adenosine displays its functions by acting on the adenosine receptors. Recent studies have shown that the distribution, expression and balance among subtypes of adenosine receptors are closely related with cognitive activities, and changes of adenosine receptors play key roles in neurodegenerative disorders including Alzheimer's disease. It has been pointed out that prolonged activation of adenosine receptors by high level adenosine may lead to the disturbance of balance among adenosine receptor subtypes. This imbalance mainly performed as increased expression of A2a receptor and decreased expression of A1 receptor, and enhancement of the excitatory signals mediated by A2a receptor and weakened inhibitory signals mediated by A1 receptor. Changes of these two subtypes of adenosine receptors may lead to a lot of disorders of neurological activities which developed into dysfunction of cognition to the end. These findings imply that the potential of maintaining the balance among adenosine receptors on the treatment of AD would facilitate both the revealing of the mechanism and the cure of AD.

7.HRM identification of Chinese medicinal materials Mutong.

High resolution melting (HRM), an important technology for genotyping and mutation scanning, has broad prospects in the authentification of traditional Chinese medicine. This paper selected universal trnH-psbA primers and used HRM to establish a new methods for identification of Akebia herbs. PCR was conduct at the annealing temperature of 58 degrees C and 35 cycles. The range of the DNA template concentration, the primer concentration and the Mg2+ ion concentration were further analyzed. The results showed the Tm values of Caulis Akebiae was (81.84 ± 0.16), (85.28 ± 0.16) degrees C and Caulis Clematidis Armandii was (83.22 ± 0.19) degrees C and Caulis Aristolochiae manshuriensis was (81.67 ± 0.14) degrees C, (84.24 ± 0.10) degrees C with 5-125 mg - L-' DNA template, 0.4 μmol x L(-1) primer, 2.0 mmol x L(-1) Mg2+. This method can achieve the authentification of Akebia herbs and is simple, fast, high-throughput, visual.

8.Correlation of expression of Oct-4 and tumorigenesis and prognosis of colorectal cancer

Objective To study dynamic expression of Oct-4 during development of colorectal cancer (CRC) and investigate the association between Oct-4 expression and prognosis of patients with CRC.Methods CRC tissues, matched non-tumor tissues and colonic polyp tissues were collected and Oct-4 expressions were analyzed by quantitative reverse transcription-polymerase chain reaction (qRT-PCR), flow cytometry analysis (FCM) and immunohistochemistry (IHC).Medical records of patients with CRC were reviewed.Clinicopathological analysis was performed to assess the association between Oct-4 expression and certain chnicopathological parameters.Kaplane-Meier survival curve was conducted to evaluate association between Oct-4 expression and survival time of patients with CRC.Results The results of qRT-PCR showed that the relative expressions of Oct-4 mRNA in matched non-tumor, colonic polyp and CRC tissues were (0.23 ±0.16) × 10-5, (1.19 ± 0.76) × 10-5 and (3.79 ± 1.94) × 10-5 respectively, with a significant difference (F =8.633, P =0.001).IHC analysis displayed that the positive expression ratios of Oct-4 in matched non-tumor tissues, colonic polyp tissues and CRC tissues were 4.43%, 12.68% and 40.51% respectively, and a significant difference was discovered among them (x2 =66.311, P < 0.001).Oct-4 expre-ssion in CRC was significantly correlated with pathological stage (x2 =7.248, P =0.007), lymph node metastasis (x2 =4.888, P =0.027), distant metastasis (x2 =5.732, P=0.017) and TNM stage (x2 =4.174, P=0.041).Kaplan-Meier survival curve analysis demonstrated that Oct-4 positive cases had a shorter median survival time (37.0 months) compared with Oct-4 negative cases (76.0 months), with a significant difference (x2 =14.050, P =0.001).Conclusion The expression of Oct-4 is step-wise increased during development of CRC.The abnormal expression of Oct-4 may play certain roles in the development of CRC.Oct-4 may be an important biomarker for the clinical diagnosis, assessment of disease and prognosis in CRC.
9.Diversity Investigation of Actinomycetes Isolated from Radiation-polluted Soil

One hundred and fifty two actinomycetes were isolated from forty two radiation-polluted soil samples,using six different isolation media. Sixty cultures were chosen for 16S rRNA gene sequence and systematic analysis,which based on their morphology and ARDRA. Results of 16S rRNA gene sequences blasting showed that the strains were assigned to 12 recognized genera of actinomycetes,most of them fall within Streptomyces genus and a great deal of strains belonged to rare actinomycetes,which indicated a rich diversity of actinomycetes in the radiation-polluted soil.
10.Study on the aberrant methylation of SOCS-1 gene in diffuse large B-cell lymphoma

Objective To explore the significance of the suppressor of cytokine signaling-1 (SOCS-1)gene methylation in the genesis, development and prognosis of diffuse large B-cell lymphoma (DLBCL).Methods The methylation state of CpG island in SOCS-1 gene were detected by methylation-specific polymerase chain reaction (PCR) and the level of SOCS-1 gene was measured by the real-time PCR. The clinical data of 30 patients with DLBCL were collected, and according to the international prognosis index (IPI), they were divided into low risk group and high risk group. Results Aberrant methylation of SOCS-1 in 17 DLBCL patients (56.7 %) were positive, however, in control group aberrant methylation was negative(P <0.01).The methylation level of DLBCL patients with positive SOCS-1 methylation was higher than that of patients with negative (P <0.05). Combined with the clinical data, the positive rate of methylation in patients with high level of serum LDH or the numbers of extra-nodal lesions>l were significantly higher than that in patients with normal LDH level or the numbers of extra-nodal lesions ≤ 1, respectively. Hence, the positive rate of methylation in the high risk group of DLBCL was higher than that in the low risk group (P <0.05). Conclusion There were aberrant methylations of the SOCS-1 gene in the patients with DLBCL. The methylations of SOCS-1 can silence the gene expression, which indicates that SOCS-1 and its methylations play some role on genesis and development of DLBCL and can evaluate the prognosis of the patients with DLBCL.
