2.Effects of Azithromycin on Hyperoxia Induced Lung Injury in Neonatal Rats

Objective To investigate the effects of azithromycin on hyperoxia induced lung injury in neonatal rats.Methods Ninety 2-day-old neonatal rats were randomly divided into 3 groups (30 cases in each group):air control group,hyperoxia model group,azithromycin treated group.The rats in air control group were exposed to indoor air,and the rats in hyperoxia model group and azithromycin group were exposed to 900 mL?L-1 O2 for 14 days.From 2-days-after birth,rats in treated group received intraperitoneal injection of azithromycin (200 mg?kg-1)per day,rats in air control group and hyperoxia model group received intraperitoneal injection of normal saline.Ten rats in each group were executed and their lung tissues were taken out and were observed under a optical microscope on the 3rd,7th,14th days from the start.The expression of interferon gamma(IFN-?),connective tissue growth factor(CTGF) and matrix metalloproteinase-9 (MMP-9) in the lung tissues were detected by immunohistochemistry.Results There was no expression of IFN-? positive cells in air control group,but the expressional intensity level was gradually enhanced as the time of inhaling hyperoxia prolonged in hyperoxia model group,it reached the peak on the 7th day,and then obviously declined on the 14th day,but still higher than that of air control group.The expressional intensity level of IFN-? in azithromycin treated group was lower than that of hyperoxia model group in each time segment,there were significant differences among them(Pa

4.Clinical observation of head and body acupuncture and moxibustion on stroke

ObjectiveTo observe effect of head and body acupuncture and moxibustion on stroke.Methods183 stroke patients were randomly divided into 3 groups, head acupuncture and moxibustion, body acupuncture and moxibustion, and head body acupuncture and moxibustion. After two months treatment, effects of 3 groups were evaluated.ResultsThere were no differences between head group and body group, body group and head body group, but there was significantly difference between head body group and head group (P<0.05).ConclusionThe head body acupuncture and moxibustion can gain the best clinical effect on stroke patients compared with simply head or body acupuncture and moxibustion.
5.An analysis of values of massage to facilitate the rehabilitation of the women in puerperium

Objective To discuss the valuable influence of the total-body massage on the facilitation of the rehabilitation for the women in puerperium.Methods Both Chinese traditional medical total-body massage and the biological information theories and its methodology were employed in dealing with 50 cases of puerperants without contraindications,who had labored naturally 12 hours before and who had received cesarean sections 48 hours before. The results were compared with the control group in which conventional preventive health care for puerperants was adopted.Results Notable differences exist in the aspect of lochia,defecation,lactation and emotions between the two groups.Conclusion Significant benefits can be obtained from the massage to facilitate the rehabilitation of the women in puerperium while promoting their quality of lives and broadening the coverage of services of the preventive health care for the puerperants.
6.The preventive effectiveness in reducing tooth decay and decalcification of different concentration of fluoride toothpaste for orthodontic patients.

OBJECTIVETo investigate the preventive effectiveness in reducing tooth decay and decalcification of different concentration of fluoride toothpaste for orthodontic patients.
METHODS86 patients were divided into the first test group and the second test group. The patients of the first test group brushed tooth with 1.1% sodium fluoride and acidulated phosphate gel. The patients of the second test group brushed tooth with 0.243% sodium fluoride in a silica base. The extent of facial tooth decay and decalcification of the twelve upper and lower teeth from right cuspid to left cuspid was scored blindly and independently by four observers after 12 months of product use. The scores were rated either one (having tooth cavity/decalcification) or zero (no tooth cavity/decalcification). Four observer's readings were averaged per tooth, and then per patient for the two treatment groups.
RESULTSAfter 12 months of product use, the mean caries score of the first test group was 0.326, and the mean caries score of the second test group was 0.490. There was significant difference between them.
CONCLUSIONA gel system containing 1.1% sodium fluoride and acidulated phosphate provides a clinically better efficacy in reducing tooth decay and decalcification than does a toothpaste containing 0.243% sodium fluoride in a silica base under and adjacent to orthodontic brackets used in orthodontic therapy.

7.Generation and Expression of Recombinant Eukaryotic Expression Plasmids of PAX3 Gene and Its Significance

Objective To study exogenous expression and subcellular localization of wild type (WT ) and mu-tant PAX3 proteins in vitro by generating their expression plasmids for further study of pathogenesis of Waarden-burg syndrome (WS) .Methods The plasmids pECE-PAX3 and pcDNA3 .0-HA were ligased after they were cut by double enzyme digestion using molecular cloning technique to generate recombinant eukaryotic expression plasmid pcDNA3 .0-PAX3-HA ,which was as a template to generate expression plasmids pcDNA 3 .0 -H80D -HA and pcDNA3 .0-H186fs-HA of novel mutations H80D and H186fs of PAX3 gene .All constructs were verified by di-rect nucleotide sequencing .NIH3T3 cells were transfected transiently with the expression plasmids of PAX3 ,H80D and H186fs respectively .The exogenous expression of WT PAX3 protein and mutant H80D ,H186fs proteins were analysed using Western blot assay ,while their subcellular distribution were observed using immunofluorescence as-say .Results The DNA sequences of expression plasmids of PAX3 and its mutant H80D ,H186fs were correct . Both WT and mutant PAX3 proteins were detected at the expected size .WT PAX3 and H80D proteins were only lo-calized in the nucleus ,whereas H186fs protein showed aberrant localization in both cytoplasm and nucleus .Conclu-sion We successfully generated the recombinant eukaryotic expression plasmids of PAX 3 gene and its mutants and drew preliminary conlusion of gene mutation having effect on subcellular distribution of WT PAX 3 proteins in vitro , which lays experimental basis for further study of the moceluar mechanism of WS caused by PAX3 gene mutations in China .
8.Influence of aqueous humor on growth of bovine corneal endothelial cell in vitro

Background The construction of tissue-engineered corneal endothelium needs the functional seeding cells,so how to culture a large amount of functional corneal endothelial cells (CECs) is an urgent problem to be solved.Objective The aim of this study was to evaluate the role of aqueous humor on bovine CECs in vitro.Methods Aqueous humor of 1.2 ml was collected from the anterior chamber of bovine and sterilized,and the liquid supernatant was obtained.The bovine CECs were isolated from bovine cornea and then cultured in low glucose Dulbecco Modified Eagle Medium with 10% fetal bovine serum (FBS) in vitro.Aqueous humor was added into the medium with the final concentration of 2.5%,5.0%,l0.0%,15.0% and 20.0%,respectively,and no aqueous humor was added in the control group.Cell counting kit-8 (CCK-8) assay was used to detect the absorbency value of CECs for the evaluation of cell proliferation.Progression of the cell cycle was analyzed by flow cytometry (FCM).After confluence of the cells was reached,1 ml plastic spear tip was used to scratch the cell single layer,and the cells were incubated consequently in medium with 10% FBS and with or without aqueous humor for 24 hours.Healing area of the cell single layer was measured.The cells were incubated at a density of 6 × 105 cells/ml and cultured using medium with or without 10.0% aqueous human for 5 days,and the number of the cells was analyzed by DAPI fluorescence technique.Results Under the phase-contrast microscopy,the confluent CECs showed a slabstone-like and hexagonal appearance.CCK-8 assay revealed that the absorbance values of CECs was significantly different among the various culture groups (F=4.051,P =0.007),and the absorbance value in different concentrations of aqueous human culture groups was significantly higher than that in the control group (P < 0.01).FCM showed that the percentage of the cells in S-G2 phases was (34.80-±3.13)% in the 10.0% aqueous humors group and (23.06±1.13)% in the control group,showing a significant difference (t =-5.729,P=0.005).The scratch test showed that the healing area of the cell signal layer was (0.116±0.019) mm2 in the 10.0% aqueous humors group and (0.358 ±0.049) mm2 in the control group,showing a significant difference (t =13.842,P =0.000).The density of cells in the 10.0% aqueous humor group was (1439± 1 10)/field,which was more than (1162±45)/field in the control group (t =-11.020,P=0.000).Conclusions Aqueous humor at the concentration of 10.0% promote the growth and proliferation of bovine CECs.The result suggests that 10.0% aqueous humor can be used as a promoting agent during the culture of CECs.
9.Clinical analysis on the pupil diameter changes of cataract patients during phacoemulsification

To study the changes of pupil diameter and its effects during phacoemulsification. METHODS: Block design, prospective study. 60 cataract patients (60 eyes) were selected, which divided into two groups according to the age, young age group (group A, 20 eyes of 20 patients) and older age group (group B, 40 eyes of 40 patients). Two groups were all given compound tropicamide eye drops for fully mydriasis before operation, computer software to process images during operation, and according to the cornea amplification ratio measure pupil diameter during the operation phase: before the corneal tunnel incision ( t1 ) , after injection of sodium hyalurantae ( t2 ) , before artificial lens implantation (t3), at the end of the surgery ( t4 ). Statistical analysis of data using SPSS18. 0 software.RESULTS: The pupil diameter measurements of t1-t4 were followed respectively:group A were 8. 36 ± 0. 65, 8. 97±0. 50, 8. 67±0. 63, 8. 72±0. 96mm; group B were 7. 73± 0. 58, 8. 23 ± 0. 59, 7. 89 ± 0. 16, 7. 70 ± 0. 63mm. Overall comparisons between the two groups, the pupil diameter measurements had differences during the operation phase ( F = 26. 696, P< 0. 05 ). Comparisons between the two groups at different time points: the pupil diameter of t1-t4 in group A was larger than that in group B, with statistically differences (P<0. 05). The pupil diameter of each groups had a tendency to change in different operation phrases, with no statistically differences (F=2. 617,P>0. 05). Comparisons in a group at different time points: the pupil diameter of t2 was larger than that in t1, with statistically differences ( P>0. 05), the pupil diameter of t3 and t4 were decreased in group B, with statistically differences (P<0. 05).CONCLUSlON: Compound tropicamide eye drops are given for mydriasis before operation, young patients with cataract and senile patients with cataract can achieve the effect of surgery for mydriasis, the pupil stability of phacoemulsification in senile patients with cataract are lower than that in young patients with cataract.
10.Expression and Clinical Significance of Cyclin Kinase Inhibitor P21~(WAF1) and P27~(KIP1) in Children with Acute Leukemia

Objective To investigate the expression of cyclin kinase inhibitor P21~(WAF1) and P27~(KIP1)in children with acute leukemia and its clinical significance.Methods A total of 32 hospitalized children with acute leukemia(AL) were included in this study.Their bone marrow samples were collected before chemotherapy and individual patient was detected after complete remission(CR).The method of immunocytochemistry was used to estimate the expression of P21~(WAF1) and P27~(KIP1).Both positive percentage and intensity of the cells were counted.Results Findings showed that the positive ratios of P21~(WAF1) and P27~(KIP1) in total samples,ALL samples and AML samples were lower than the control group(P
