1.Correlation analysis of Sirt1 gene variant and susceptibility to chronic obstructive pulmonary disease

Objective To explore the relationship between Sirt1 single nucleotide polymorphisms ( rs2236319 ) and chronic obstructive pulmonary disease ( COPD ) susceptibility. Methods The subjects included 149 patients with stable COPD in the First People′s Hospital of Jining outpatient or ward from March 2014 to March 2015, and 160 cases of the same period health check. Sirt1?rs 2236319 genotype distribution frequency was detected by real?time PCR,and the levels of inflammatory factors were compared,including IL?6 and CRP. Results The AA,AG,GG genotype frequency of COPD group were 59. 1%( 88/149) ,35. 6%( 53/149),5. 4%(8/149) respectively,of control group were 45. 0%(72/160),48. 8%(78/160),6. 3%(10/160) respectively,the allele A,G frequency of COPD group were 76. 8%(229/298),23. 2%(69/298) respectively,of control group were 69. 4%( 222/320 ) , 30. 6%( 98/320 ) respectively, there were statistically significant differences between the two groups(χ2=6. 23,6. 06,P<0. 05) . IL?6 and CRP levels were significantly increased in COPD group carrying AA genotype as compared to COPD group carrying AG+GG genotype((10. 30±0. 40) mmol/L,(10. 16±0. 36) mmol/L;(2. 56±0. 20) mg/L,(2. 46±0. 22) mg/L;F=4. 52,8. 04,P<0. 05);There were no significant differences in terms of IL?6 and CRP levels in control group carrying AA genotype as compared to control group carrying AG+GG genotype ( P>0. 05 ) . Conclusion The polymorphisms of Sirt1?rs2236319 may be an additional risk factor for COPD and associate with inflammatory factors. Furthermore the involvement of rs2236319 in the initiation and progression of COPD may relate to inflammatory processes.
2.Clinical study of pusher syndrome in stroke patients

Objective To investigate the incidence of the pusher syndrome in stroke patients, and the relationship between the syndrome and the neuropsychological symptoms and location of brain lesion, and to investigate the mechanism and the physiotherapy intervention of pusher syndrome. Methods Thirty nine patients(25 male, 14 female, 62.5?9.4 years old) with pusher syndrome were examined, 91 patients(57 male,34 female, 58.4?11.6 years old) without ipsilateral pushing served as control. The lesion areas, neuropsychological syndrome and the Barthel Index(BI) were recorded, and the physiotherapy were administered in the patients. Results The incidence of pusher syndrome was 30% in the patients studied, corresponding to 17% of the total number of stroke patients in the study period. 3.Prediction of peripheral blood lymphocytes radiosensitivity in normal tissue radiation damage in oncology patients with radiotherapy

Patients treated with radiotherapy (RT) might experience a large variation in normal tissues. Severe radiation damage in a minority of patients limits the doses that might be safe given to the majority. The possibility of predicting such radiation-induced damage would provide a better treatment schedule for the patients. Several predictive tests in peripheral blood lymphocytes such as initial DNA damage, radiation-induced apoptosis and genetic variation have been proposed to know the individual sensitivity of patients to the radiotherapy schedules. This study aimed to summarize the main studies regarding to this ifeld.
4.Standardization of Data Element in Medical Data Resource Share

5.Clinical features of ocular manifestations of 93 AIDS patients in Urumqi

AIM: To investigate the clinical features of acquired immune deficiency syndrome ( AIDS) patients associated ocular diseases in Urumqi and the relationship between ocular fundus manifestations and CD4+T cell count.
METHODS: The fundus of 93 AIDS patients were examined by indirect ophthalmoscopy. The clinical symptoms and CD4+T cell count of those patients with fundus changes were analyzed.
RESULTS: Thirteen patients were found having fundus changes which occurred in one eye of 4 patients and two eyes of 9 patients, respectively, and the total detection rate was 14. 0%. Seven patients had vision changes, and the main clinical features of retinal lesion were cotton wool spot and hemorrhage of retina. Four patients were diagnosed as retinitis with cytomegalovirus ( CMV ) infection and 9 patients were diagnosed as HIV related retinopathy diseases. Seven patients among 37 patients with CD4+T cell count ≤100cell/μL had fundus changes related AIDS, and the detection rate was 18. 9%; while 6 patients among 56 patients with CD4+T cell count >100cell/μL had fundus changes related AIDS, and the detection rate was 10. 7%. There was statistical difference between the two detection rates (P<0. 05).
CONCLUSION: No specificity was found of those patients with the clinical manifestation of HIV- related retinopathy, and those patients are easy to be missed diagnosis. A number of AIDS patients have fundus changes without any vision changes. Therefore, it is very useful for AIDS patients to be carried out the routine fundus examination for the early diagnosis and treatment.
6.Improving isoprene production by engineered heterologous mevalonate pathway in Escherichia coli.

Isoprene is an important precursor of synthetic rubber material. In our previous study, metabolic engineered Escherichia coli strain (BW-01) was constructed and used to produce isoprene. Based on the theory of protein budget, using synthetic biology strategies including the increased copy number of genes and rare codons, we regulated the expression of key enzyme to improve isoprene production in Escherichia coli strain. Under shake-flask conditions, isoprene productivity of the engineered strain (BW-07) increased by 73% compared with BW-01, reached 761.1 mg/L. It provides a reference for further studies.

7.All-tram retinoic acid markedly inhibits over-excitement of renin-angiotensin system in 5/6 renal ablation rats

ObjectiveTo investigate the effect of all-trans retinoic acid (atRA) on the renin-angiotensin system (RAS) in 5/6 renal ablation model. MethodsAtRA was administered to 5/6 renal ablation rats by three dosages: 5 mg·kg-1·d-1 (n=8), 10 mg·kg-1d-1 (n=8) and 20 mg·kg-1 d-1 (n=8) and vehicle (vehicle group, n=8) for 10 weeks. Healthy rats consisted of shamoperation group (n =8). The level of renin and angiotensin Ⅱ in renal tissues were measured by radioimmunoassary. The level of angiotensin type 1 receptor (AT1R) in remnant renal cortex was measured by Western blot. The mRNA expression levels of two subunits of activative protein 1(AP-1),c-jun and c-fos was quantitated by real-time PCR. ResultsAfter 10 weeks of atRA treatment by gavarge, artery blood pressure decreased (P<0.05). AtRA reduced the levels of renin (P<0.05) and angiotensin Ⅱ (P<0.05) in kidney and down-regulated the expression of AT1R protein in renal cortex. Larger dose of atRA (20 mg·kg-1·d-1) performed higher activity in inhibiting renin and AT1R. Compared with vehicle group, atRA could significantly inhibit the expression of renal c-jun and c-fos mRNA (P<0.05). Conclusion atRA can decrease the over-expression of main components of RAS.
8.Chitosan/PVA nerve conduits repair sciatic nerve defect in rats

ObjectiveTo investigate the effects of chitosan/PVA nerve conduits which used for repairing sciatics nerve defect in rats.MethodsTwenty-seven rats were divided into 3 groups randomly,with 9 rats in each group. Firstly, the 15mm defects in the left sciatic nerves were made in the rats and were respectively repaired with chitosan/PVA conduits graft (group A), the silicon conduits graft (group B),and autografts (group C). At 12 weeks after the operations, the left sciatic nerves were taken out, and the comparative evaluation was made on the repairing effects by wet weight of gastrocnemius and soleus muscles, histological examination,computerized imaging analysis and True Blue retrograde tracing. ResultsThe wet weight of gastrocnemius and soleus muscles showed no significant difference between the chitosan/PVA graft and autograft groups (P > 0.05). The wet weight of gastrocnemius and soleus muscles in significant difference between the chitosan/PVA graft and the silicon group at 12 weeks after the operation(P < 0.05). The nerve fiber density showed no statistically significant differences between the chitosan/PVA and autograft groups(P> 0.05).The regenerative nerve fiber in group B had normal morphological and structural characters under transmission electron microscope.True Blue-labeled neuron cell bodies were found within both anterior horn of gray matter in the spinal cord and dorsal root ganglions (DRGs) ipsilateral to the operated side of the tested rats on illumination with ultra-violet light 1 week after the injection of True Blue.Conclusion Chitosan/PVA nerve conduit can effectively promote the nerve regeneration and myelinization of rat sciatic nerve, which is expected to substitute for autograft to repair nerve defects succesfully.
9.Regulation of P38 and MKK6 kinase on heat-induced Raw264.7 cell apoptosis

Objective To elucidate the role of P38 signaling pathway on heat-induced apoptosis in monocytic cell line Raw264.7.Methods Raw cells were transfected with constitutively active mutant MKK6b(E)and dominant negative mutant P38(AF),or the empty cloning vector pcDNA3 and apoptosis was detected by flow cytometric analysis.Results The ectopic expression of P38 mutant was confirmed by immunostaining with the antibody against the Flag-epitope tag.Expression of MKK6b(E)led to a marked increase in P38 kinase activity in transfected cells and induced a 4-fold increase in the number of apoptotic cells as compared to that in cultures of control transfected cells.Meanwhile the expression of MKK6b(E)increased the apoptotic rate of Raw cells induced by heat.In contrast,the dominant-negative mutant P38(AF)inhibited Raw cells apoptosis induced by heat.Conclusion The activation of the MKK6-P38 MAP kinase signaling pathway is required for heat-induced apoptosis in Raw264.7 cells.
10.Pharmacokinetic studies of scutellarein and glucuronide scutellarin in rats

Aims To establish a UPLC-MS/MS meth-od for the determination of plasma concentration of scutellarein and its metabolite and to study their phar-macokinetics in rat plasma. Methods The analysis was achieved by BEH C18 column with a mobile phase composed of 0 . 1 % formic acid in acetonitrile and 0 . 1% aqueous formic acid using step gradient elution. A TQD tandem mass spectrometry equipped with electros-pray ionization source was used as detector and opera-ted by multiple reaction monitoring( MRM) positive ion mode. After intravenous injection of scutellarein, the concentrations of scutellarein and its major metabolite glucuronide scutellarin in rat plasma were determined at different time points. The pharmacokinetic parame-ters were calculated by DAS 2. 0 software. Results Good linearity was achieved for scutellarein, the ex-traction recovery was between 80 . 5 % to 90 . 5 %, the precisions and accuracy were good. The result showed the pharmacokinetic profiles of scutellarein and glucu-ronide scutellarin both fit to the two-compartment mod-el. Conclusion The above mentioned method is spe-cific, rapid, sensitive and suitable for the pharmacoki-netic studies of scutellarein and its metabolite.
