1.Clinical observation on the treatment effect for pruritus in uremic patients by different dialysis modalities

Objective To study the effect of different dialysis modalities on pruritus in uremic patients .Methods Patients with maintenance hemodialysis who were suffered with severe cutaneous pruritus were randomly divided into hemodialysis group (HD group) ,hemodialysis combined with hemodiafiltration group (HD+ HDF group) ,hemodialysis and hemodiafiltration combined with hemoperfusion group (HD+ HDF+ HP group) .Plasma P3+ ,parathyroid hormone (PTH) and β2-microglobulin (β2-MG) were measured at pre-dialysis ,1 weeks ,4 weeks and 12 weeks after dialysis ,cutaneous pruritus was scored too .Results Compared with pre-dialysis ,the level of plasma P3+ ,PTH ,β2-MG and the scores of cutaneous pruritus were significantly lower at 4 weeks and 12 weeks after dialysis in HD+ HDF+ HP group (P<0 .05) .The were statistically significant difference in P3+ ,PTH ,β2-MG and cu-taneous pruritus scores among 3 groups after 12 weeks (P<0 .05) .Conclusion HD+ HDF+ HP is superior to HD+ HDF in effi-ciently clear P3+ ,PT H andβ2-M G ,and relief cutaneous pruritus and itching .
2.Application of Emotional Problems Factor of Chinese Personality Assessment Inventory II in Anxiety Disorders Patients

Objective:To evaluate the reliability and validity of Emotional Problems Factor of Chinese Personality Assessment Inventory II(CPAI2-E).Methods:Item analysis of the test was carried out;internal consistency reliability, retest reliability and structure validity, differential validity were examined.Results:①Through factor analysis, 20 subordinate factors were got.②The Cronbach ? coefficient of the total was 0.948 and each factor ranged from 0.755 to 0.896;re-test reliability of the total test was 0.718.Conclusion:CPAI2-E can be used as an effective and reliable tool for assessing anxiety disorders.

5.Evaluated on Expression of Endothelin-3 mRNA Detected by RT-PCR in Acute Pancreatitis Rats

Objective To investigate the relationship between gene expression of endothelin-3(ET-3) and inflammation of acute pancreatitis(AP) in rats.Methods Fifty-four rats were divided randomly into 4 groups:sham operation group,AP group,arterial injection group and vein injection group.AP was induced by reverse intra-bile duct infusion 4.5% sodium taurocholate,treated with low dose dopamine 〔5 ?g/(kg?min)〕 by injecting arterial or tail vein.Rats were sacrificed at 1,6 and 24 h after the induction of AP.The mRNA expression of ET-3 was evaluated by semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR) and pathological changes was observed in rats.Results Expression of ET-3 mRNA could be detected from 1 up to 24 h after the induction of pancreatitis.Expression of ET-3 mRNA of sham operation group was decreased significantly compared with other three groups.Expression of ET-3 mRNA showed a significant decrease by arterial injection dopamine than that by tail vein(P
6.Effect of SIRT1 regulating cholesterol synthesis in repairing retinal ganglion cells after optic nerve injury in rats

AIM: To investigate the repair mechanism associated with cholesterol synthesis regulated by silent information regulator 1 (SIRT1) in rat model of optic nerve damage.METHODS: Preparation of optic nerve damage in 70 rats was randomly divided into normal group (10 rats), resveratrol treatment group ( experimental group 30 rats) and PBS buffer control group ( 30 rats ) . The experimental group and control group was further divided into 3 subgroups ( each group 10 rats ) , respectively. After 7, 14, 21d injected resveratrol or PBS, optic nerve injury were observed, then the rats were sacrificed. Retina was segregated; the surviving retinal ganglion cell ( RGCs ) was counted. Dissection of optic nerve, cholesterol content of them were tested; RT-PCR was used to detect mRNA expression of SIRT1, SREBP2 and HMGCR; Western blot assay was used to test the protein expression levels of SIRT1, cholesterol regulatory element binding protein 2 (SREBP2) and HMGCR.
RESULTS:The numbers of RGCs and cholesterol levels of rat model with optic nerve injury decreased significantly (P<0. 01). The mRNA and protein expression levels of SIRT1, SREBP2 and HMGCR were all decreased in a time-dependent manner (P<0. 05). Three components of the three time points, with time injuries were aggravated, and the extent of damage was significantly reduced in the treatment group compared with the control group. But in resveratrol treatment group, the cholesterol levels and mRNA or protein expression of SIRT1, SREBP2, HMGCR in optic nerve were significantly restored in a time-dependent ( P<0.05 ) . The number of surviving RGCs restored significantly in resveratrol treatment group (P<0. 01) in a time-dependent manner.
CONCLUSION:Up-regulating the expression of SIRT1, SREBP2 and down- regulating HMGCR by resveratrol could repair the injury of optic nerve through promoting the synthesis of cholesterol in neurons and retinal ganglion cells in the repair process. SIRT1 may be as a promising new target for treatment on optic nerve damage.
7.Cognition about Rehabilitation among Parents with Retarded Children Aged 0~6 in Beijing

Objective To investigate the cognition about rehabilitation among parents having retarded children aged 0~6 in Beijing. Methods237 retarded children diagnosed in Disabilities Sampling among 0~6 Years Children in Beijing in 2004 were followed up 3 years later, and their parents were investigated with questionnaire about their rehabilitation knowledge. Results25.99% of these children had received rehabilitation services in the 3 years at least once, and was higher in city than in country. 38.39% and 16.52% of their parents knew their children need early rehabilitation and family training. ConclusionIt is necessary to communicate the knowledge of rehabilitation to the parents of retarded children.
9.Effects of RNAi-mediated Cofilin-1 Gene Silencing on proliferation and invasiveness in hepatocellular carci-noma Huh-7 cells

[Abstract ] Objective Cofilin-1 is involved in the pathogenesis of various tumours .However, the expression and effect of Cofilin-1 in hepatocellular carcinoma is not clear .The aim of this study is to observe the Cofilin-1 gene expression in human hepatocel-lular carcinoma (HCC) tissues, and to explore the effect of Cofilin-1 gene expression on invasion and metastasis of HCC HuH-7 cells. Methods Real-time quantitative PCR was used to assess the Cofilin-1 gene expression in human HCC tissues and normal tumor-ad-jacent tissues.The specific small interfering RNA ( siRNA) of Cofilin-1 sequence was synthetized in vitro , and was transfected into HCC HuH-7 cells using liposome transfection.The experiment was divided into Cofilin-1-siRNA group, Ctrl-siRNA group and un-transfected group.Western blot assay was used to detect the protein expression of Cofilin-1.Migration and invasion experiments in vitro were used to investigate the invasive ability of transfected cells. Results Compared with the adjacent liver tissue , Cofilin-1 gene ex-pression in human liver cancer tissue was significantly increased (0.698 ±0.156 vs 3.523 ±0.412, P<0.05).The expression of Cofilin-1 protein in Cofilin-1-siRNA group was 0.558 ±0.033, which was lower than that in Ctrl-siRNA group ( 0.933 ±0.015 )
and in untransfected group (0.961 ±0.020) (P<0.05).The results of migration and invasion experiments in vitro showed that the amount of migration and invasion cells in Cofilin-1-siRNA group were significantly lower than Ctrl-siRNA group or untransfected group (58.50 ±1.78 vs 79.00 ±1.33, 74.50 ±1.35,P<0.05; 36.50 ±0.83 vs 60.20 ±1.60, 51.50 ±1.14, P<0.05). Conclusion Cofilin-1 is highly expressed in HCC, and the invasion and metastasis of HCC HuH-7 cells is suppressed by inhibiting the Cofilin-1 gene expression.
