3.Effects of ZPSJ Recipe on Insulin Sensitivity,Serum Resistin and Inflammatory Factors in Type 2 Diabetic Patients with Insulin Resistance

Objective To Observe the influence of Zaoshi Jiedu Zhupi Sanjing (ZPSJ) Recipe on insulin sensitivity, serum resistin and inflammatory factors related to the effects of Chinese herbal compound in type 2 diabetic patients with insulin resistance. Methods A single-blind randomized controlled clinical trial was conducted. 80 patients with 2 type diabetic patients with insulin resistance were recruited into a control group and a treatment group. The treatment group was treated with ZPSJ Recipe, and the control group was treated with metformin. Results In each group before and after treatment: blood glucose, serum insulin levels, HOMA-IR were significantly decreased (t=7.14, P<0.01 ) ; between the two groups, FPG, FINS showed no significant difference, while TG TCH, HOMA-LR showed significantly differences (t=2.62, P <0.05 ). Within the two groups before and after the treatment: serum IL-6, TNF-a, hs-CRP were decreased in treatment group, (t= 1.85, P<0.05 ) , while TNF-a, hs-CRP were decreased in the control group (t=2.56, P<0.05 ) , while IL-6 did not change significantly. The serum resistin levels were not statistically changed in both groups before and after treatment. Conclusion ZPSJ Recipe can regulate blood sugar, blood fat and improve insulin sensitivity in type 2 diabetic patients with insulin resistance.
4.Baculoviruses as Vectors in Mammalian Cells

The Baculoviridae are a large family of enveloped DNA viruses exclusively pathogenic to arthropods. Baculoviruses have been extensively used in insect cell-based recombinant protein expression system and as biological pesticides. They have been deomostrated to be safe to mammals, birds and fish. Recently, baculoviruses has been shown to transduce different mammalian cells in spite of the fact that they cannot replicate in mammalian cells (11, 73, 76). This has resulted in the development of baculoviruses as mammalian expression systems and even as vestors for gene therapy.
5.Ventilatory Treatment of Flail Chest with Pulmonary Contusion

Objective To evaluate the early therapeutic effect of flail chest with pulmonary contusion by using different mode of mechanical ventilation. Methods Twenty-nine patients of flail chest with pulmonary contusion were analysed retrospectively. All the patients were treated with the ventilator Bear1000. Two groups were established: invasion group was treated with SIMV+PEEP(8 cases) and CPAP+PSV (7 cases), noninvasion group was treated with NIPPV(14 cases). Results There was no death in all the patients. CPAP+PSV was more effective than SIMV+PEEP not only in decreasing breath rate and improving hypoxemia but also in decreasing peak inspiratory pressure(P
6.Baculovirus-mediated Expression of p35 Confers Resistance to Apoptosis in Human Embryo Kidney 293 cells

Baculovirus has many advantages as vectors for gene transfer. We demonstrated that recombinant baculovirus vectors expressing p35 (Ac-CMV-p35) and eGFP (Ac-CMV-GFP) could be transduced into human kidney 293 cells efficiently. The level of transgene expression was viral dose dependent and high-level expression of the target gene could be achieved under the heterogonous promoter. MTT assay suggested that both Ac-CMV-p35 and Ac-CMV-GFP did not have cytotoxic effect on human embryo kidney 293 cells. Cell growth curve showed the Ac-CMV-p35 and Ac- CMV-GFP transduced and non-transduced cells had similar proliferation rate, so baculovirus-mediated p35expression had no adverse effect on cell proliferation. In addition, baculovirus-mediated p35 gene expression protected human embryo kidney 293 cells against apoptosis induced by various apoptosis inducers such as Actinomycin D, UV or serum-free media. These results suggested that the baculovirus vector mediated p35 gene expression was functional and it could be widely used in molecular research and even gene therapy.
7.Correlation between contrast-enhanced ultrasound features and size of clear cell renal cell carcinoma

Objetcive To analyze the contrast-enhanced ultrasound (CEUS)features in clear cell renal cell carcinoma (CCRCC) of different size and to discuss the diagnostic value of CEUS in CCRCCs. Methods The contrast-enhanced and conventional US features of 80 CCRCCs confirmed pathologically were retrospectively analyzed. Samples were divided into three groups by diameters: small CCRCCs (≤30 mm), medium CCRCCs(>30 mm) and large CCRCCs(>50 mm). The tumoral vascularity, lesion homogeneity and presence of an anechoic rim were observed on conventional US. The enhancement of echogenicity, homogeneity and pseudocapsule were evaluated on CEUS. Results Among the 80 renal masses, 32 were small CCRCCs, 28 were medium CCRCCs while the large CCRCCs were 20. On conventional US, 18.8%(6/32) of small CCRCCs, 71.4%(20/28) of medium CCRCCs and 95%(19/20) of large CCRCCs demonstrated as heterogeneous and the differences were highly statistically significant, whereas there were no differences in the tumoral vascularity and the presence of pseudocapsule sign among the three groups. On CEUS, 28.1%(9/32) of small CCRCCs, 85.7%(24/28) of medium CCRCCs and 100%(20/20) of large CCRCCs showed a homogeneous enhancement. The incidence of pseudocapsule sign in medium tumors was higher than small and large groups(71.4%,50% and 25%,respectively). There were statistically significant differences among the three groups in the enhanced homogeneity and the presence of pseudocapsule sign. However, all the three groups revealed mainly isoechoic and hyperechoic and there were no differences among them. Compared with conventional US, CEUS depicted significantly the increased tumoral vascularity (51.3% vs. 87.5%) and pseudocapsule sign(22.5% vs. 51.3%). Eight lesions demonstrated heterogeneous contrast enhancement on CEUS while homogeneous on conventional US, but there were no differences in lesion homogeneity between conventional US and CEUS findings. Conclusions Different sizes of CCRCC showed distinct CEUS features in the enhanced homogeneity and the presence of pseudocapsule sign. CEUS is more effective on improving the sonographic characteristics of tumoral visualization and may provide important information of US findings for the diagnosis of renal cell carcinoma.
8.Significance of fiberoptic bronchoscopic brush cytology in lung carcinoma diagnosis

Objective To evaluate the significance of fiberoptic bronchoscopic brush cytology in the diagnosis and histological classification of lung carcinoma.Methods Data of 309 patients with lung carcinoma were retrospectively analyzed.Both bronchoscopic cytology and histology diagnosis were available.The positive rate of bronchoscopic cytology and tissue biopsy were calculated respectively.The classification accuracy of cytological diagnosis for lung carcinoma was evaluated.In tissue biopsy standard,evaluated the significance of bronchoscopic cytology in diagnosis and histological diagnosis.Results The positive rate of bronchoscopic cytology and tissue biopsy were 86.1% (266/309) and 83.8% (259/309),respectively.Bronchoscopic cytology combined with bronchial biopsy could obviously improve the positive rate to 94.2% (291/309) in lung carcinoma diagnosis.Taking the tissue biopsy histological type as a standard,the cytotyping accuracy for brush method was 85.1%(74/87) in squamous carcinoma,82.4%(108/131) in adenocarcinoma and 100%(11/11) in small cell carcinoma for higher.However,the accuracy in diagnosing poorly differentiated carcinomas was only 12.2% (5/ 41).Conclusion Fiberoptic bronchoscopic brush cytology plays an stable and important role in diagnosing lung carcinomas and histological type determination.However,it has limited use in diagnosing poorly differentiated carcinomas.
9.Comparison of the effects of ursodeoxycholic acid, dexamethasone, and S-adenomethionine on intrahepatic cholestasis induced by ethinylestradiol in pregnant rats

Objective To investigate the effects of ursodeoxycholic acid (UDCA), dexamethasone (DEX), and S adenomethionine (SAMe) on the pregnant rats with intrahepatic cholestasis induced by ethinylestradiol (EE). Methods A total of 50 15 day pregnant Wistar rats were randomly divided into five groups: control group, EE group, UDCA group, DEX group, and SAMe group. In EE group, rats were subcutaneously injected with EE (2.5 mg?kg -1 ?d -1 ) for 5 d. In the control group, rats received subcutaneous injection of appropriate volume of propylene glycol (PG) for 5 d. In UDCA, SAMe, and DEX groups, rats were administered with the same dose of EE plus UDCA (100 mg?kg -1 ?d -1 ), SAMe (100 mg?kg -1 ?d -1 ), and DEX (1 mg?kg -1 ?d -1 ) for 5 d, respectively. The serum levels of aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatases (ALP), glycocholic acid (GC), and total bilirubin were measured after cesarean. Morphologic changes of the liver of pregnant rats and the growth and development of the fetal rats were observed. Stent-assisted angioplasty of intracranial and extracranial arteries is a complex pathophysiologic process.Therefore,understanding of related factors is significant for guiding clinical treatment and reducing restenosis rate.Intravascular stent as a foreign body leads to platelet activation and aggregation,cytokine secretion,resulting in thrombosis and leukocyte aggregation at vessel injury site to produce cytokine and inflammatory reaction.In addition,the smooth muscle cells migrate to injury site and proliferate,leading to intimal hyperplasia,vessel wall reconstruction and intra-stent restenosis.Moreover,the restenosis following stenting is related to stent type and medicine supportive treatment.Drug-eluted stent with anti-inflammation and vascular smooth muscle cell proliferation can reduce body inflammation intensity and duration,and postoperative medicine supportive treatment can reduce incidence of intravascular restenosis.
