4.Optimization of Lipase Production Conditions by Geotrichum candidum Y162 Using Single Factor-response Surface Methodology

The fermentation conditions of lipase production by Geotrichum candidum Y162 were optimized. Initially, the most suitable carbon olive oil, nitrogen source soybean flour and NH4Cl, salt BaCl2 and MgCl2 were selected according to single factorial experiments respectively. Based on the result, screening methodology Plackett-Burman design was used to evaluate the effects of twelve factors related to lipase production and three statistically significant factors olive oil, BaCl2 and NH4Cl were selected. The path of steepest ascent was used to approach the optimal region of lipase production subsequently. Then, the optimal combined concentration for maximum enzyme activity were further optimized by response surface methodology and determined as follows: olive oil 2.35%, BaCl2 0.36%,and NH4Cl 4.69%.The optimization of culture conditions of G.candidum Y162 led to a 2.25-fold increase in lipase production relative to initial result 14.16 U/ml, which indicate that single factor in combination with response surface methodology is an effective method for optimization of lipase production conditions by G.candidum Y162.
5.The clinical value of serum tumor specific growth factor and vascular endothelial growth factor in early diagnosis of thyroid cancer

Objective To discuss the clinical value of serum tumor specific growth factor (TSGF) and vascular endothelial growth factor (VEGF) in early diagnosis of thyroid cancer.Methods Eighty thyroid cancer patients (thyroid cancer group),120 nodular goiter patients (nodular goiter group),and 80 healthy adult (blank control group) were enrolled in this study.The level of TSGF and VEGF in two groups were detected by ELISA and compared.Results The level of TSGF and VEGF in thyroid cancer group and nodular goiter group were significantly higher than those in blank control group (P < 0.05).Conclusion Serum TSGF and VEGF have some clinical value in early diagnosis of thyroid cancer.
6.Staurospolin promotes the apoptosis of multidrug resistance tumor cell lines induced by adriamycin

Objective To investigate whether the protein kinase C inhibitor can promote the apopto- sis of multidrug resistance tumor cell lines which are induced by chemotherapy drugs.Methods Choose the KB/S(oral squamous cancer cell line)and KB/VCR(its multidrug resistant cell line)to compare the Adri- amycin-induced apoptosis with or without staurospolin(protein kinase C inhibitor).The apoptosis is stained with acridine orange,tested by flow cytometry,and approved by electron microscope.Results 36 hours after the treatment with 0.04 ?g/ml adriamycin,apoptotic cells of KB/S are 96.68%,and after 48 hours,the apop- totic cells of KB/VCR are 64.99%.When the concentration of adriamycin are augmented to 0.4?g/ml and 2.0?g/ml,the apoptotic cells of KB/VCR are 69.74% and 37.18% respectively.When treated with stau- rospolin together,the apoptotic cells of KB/VCR increased to 72.58%(?~2=4.5,P0.05)respectively.These results were testified by electron microscope and acridine orange-stain.Conclu- sion The resistance to apoptosis may be one of the mechanisms of multidrug resistance and the protein ki- nase C inhibitor may reverse this resistance by promoting the apoptosis of multidrug resistance tumor cells.
7.Clinical analysis of incipient neonatal hyperbilirubinemia at Nanjing areas

Objective To investigate the epidemiological characteristics of incipient neonatal hyperbilirubinemia. Methods The clinical data of admitted neonates with hyperbilirubinemia were retrospectively analyzed from June 2012 to May 2013. Results Two hundred and eight-four neonates with hyperbilirubinemia were enrolled and the ratio of male:female was 1.51:1. For the causes of hyperbilirubinemia, the incidences of ABO hemolytic and sepsis were higher in term infants than those in preterm infants, and the incidences of pneumonia, necrotizing enterocolitis and intracranial hemorrhage were higher in preterm infants than those in term infants (P<0.05). Compared with the preterm infants, the term infants had jaundice appearance and peak at earlier time, shorter duration of jaundice, faster decline rate of jaundice, higher levels of albumin and indirect bilirubin at the peak of jaundice (P<0.01). In the term infants, the time of jaundice appearance and peak were earlier in hemolytic group than those in non-hemolytic group (P<0.05). In preterm infants, the peak of transcutaneous bilirubin was higher in hemolytic group than that in non-hemolytic group (P<0.05). Six cases with bilirubin encephalopathy had abnormalities cranial MRI imaging, and the MRI was not entirely consistent with the peak level of bilirubin. Conclusions There are clinical differences between hemolytic and non-hemolytic hyperbilirubinemia in both term and preterm infants.
8.Comparison of the different formulas in intraocular lens power calculations on high myopia after LASlK with cataract patients

AlM: To compare the different formulas in intraocular lens(lOL) power calculations on high myopia after LASlK with cataract patients who received the phacoemulsification and intraocular lens implantation ( Phaco+lOL) .
METHODS:This was a retrospective study. Data from 102 eyes of 61 cataract patients with high myopia, who received Phaco+lOL after LASlK, were collected. Patients were divided into three groups by formulas in lOL power calculations before surgery: Holladay group ( 11 cases, 21 eyes), Haigis-L group (30 cases, 47 eyes) and SRK-T group (20 cases, 34 eyes). The ratios of equivalent lOL power after LASlK were predicted and the accuracy of their predictions were compared 3mo after surgery. lt was an index of reliability of the formulas outcomes.
RESULTS:The ratios of equivalent lOL power after LASlK using the Holladay, Haigis-L and SRK-T formulas were 0. 86 ± 0. 41D, 0. 43 ± 0. 30D and 1. 27 ± 0. 58D, respectively . There was no significant difference between Holladay group and SRK-T group ( t=-0. 271, P=0. 625>0. 05 ). However, significant difference was found between Haigis-L group and Holladay group ( t=2. 249, P=0.047<0. 05), Haigis-L group and SRK-T group (t=6. 012, P=0. 031<0. 05).
CONCLUSlON:The Haigis-L formula is more accurate than the SRK-T and Holladay formulas in predicting lOL power for cataract patients with high myopia after LASlK.
9.Knockdown of ClC-2 Gene Expression Inhibits the Growth of BT-325 Human Glioma Cells

ObjectiveTo observe the growth of BT-325 human glioma cells after interfering volume-regulated chloride channel ClC-2 gene.MethodsTwo expression recombinant vectors of ClC-2 gene were designed and constructed. The primary plasmid, pSUPER.puro-shRNA, and the two recombinant plasmids, pSUPER.puro-shRNA-ClC-21 and pSUPER.puro-shRNA-ClC-22, were transfected into BT-325 cells by LipofectamineTM2000 (Groups: control, PP1 and PP2, respectively). The mRNA expression of ClC-2 gene was detected with reverse transcription polymerasse chain reaction (RT-PCR), the cellular survival was determined with MTT assay, and the cell cycle was measured with flow cytometry (FCM). ResultsClC-2 mRNA expression and the growth of the cells in PP1 and PP2 groups were significantly lower than that of control group. The cell cycle progression was blocked in G1 phase (PP1 and PP2 vs control,P<0.01). ConclusionThe growth of BT-325 human glioma cells is prevented by knockdown of ClC-2 gene expression, which may be one of the novel targets to inhibit growth of human malignant glioma cells.
