2.Lipoprotein lipase gene mutations and the risk of cardiovascular diseases in children with obesity.

OBJECTIVETo inquire into the relationship between lipoprotein lipase (LPL) gene D9N, N291S and S447X polymorphisms and the development of cardiovascular diseases in children with obesity.
METHODSThe polymerase chain reaction (PCR) and restriction fragment length polymorphism (RLFP) techniques were used to detect three common mutations of LPL gene exon D9N, N291S and S447X in 157 obese children and 175 normal controls. Plasma lipid and lipoprotein levels between children with different genotypes were compared.
RESULTSThe D9N and N291S gene mutations were not detected in either the obese or the control groups. There were no significant differences in the frequency of S447X gene mutation between the two groups. There were no significant differences in the levels of plasma lipid and lipoprotein between children with S447 and X447 genotypes.
CONCLUSIONSD9N and N291S gene mutations may not be risk factors associated with cardiovascular diseases in children with obesity. S447X gene mutation might not play an important role in the development of cardiovascular diseases in childhood.

3.Clinical study on gefinitib in the treatment of non -small -cell lung cancer harboring mutations of epidermal growth factor receptor

Objective To discussed clinical efficacy and safety of gefinitib compared with chemotherapy in the treatment of non-small-cell lung cancer ( NSCLC) harboring mutations of epidermal growth factor receptor( EGFR) .Methods A total of 43 NSCLC patients harboring mutations of EGFR were included in this study and divided into treatment group ( n=22 ) and control group ( n=21 ) .Patients in the treatment group received gefinitib 250 mg? d-1 orally, and those in control group were administered with docetaxe 60 mg? m-2 plus cisplatin 80 mg? m-2 intravenously until progression or unbearable adverse reactions.The data of objective response rate, progression free survival and adverse reactions were compared between the two groups.Results The objective response rate and disease control rate was 59.1%, 90.9%in treatment group and 33.3%, 76.2% in control group, respectively.The objective response rate was significantly higher in treatment group compared to that in control group ( P<0.05 ) .The median progression free survival time was 11.95 months and 6.90 months in treatment group and control group, respectively, which showed that the survial time in the treatment group was much longer (HR=0.49, 95%CI:0.24-1.01, P<0.05).The main grade 3-4 adverse reactions were rash and diarrhea in treatment group and leucocytopenia and anemia in control group.The incidence rate of leucocytopenia was significantly higher in control group than that in treatment group (P<0.05).Conclusion Gefinitib treatment for NSCLC harbouring mutations of EGFR has longer progression free survival and better efficacy.
4.Sero-epidemiologic study on the relationship between Chlamydia pneumonia infection and hypertension in Chinese Mongolian population.

OBJECTIVETo explore the relationship between Chlamydia pneumonia (CP) infection and hypertension as well as the interaction of traditional risk factors and CP infections on hypertension in Chinese Mongolian population.
METHODS1430 inhabitants living in Tongliao city, Inner Mongolia were selected as research subjects, including 488 hypertensives and 942 normotensives. Enzyme-linked immunoassay technique was used to test CP IgG antibody in the blood serum and SPSS 13.0 Microsoft was used to analyze the data.
RESULTSThe prevalence rates of hypertension were significantly different between CP IgG positive and negative groups, However, the trend was attenuated after adjusting age, sex, drinking, smoking, body mass index (BMI) and dyslipidemia. The CP infection rate was significant different in diastolic blood pressure groups, which was significantly higher in 80-84 mm Hg than that in the lowest DBP group. The interaction of chronic CP infection and other traditional risk factors were associated with hypertension in Chinese Mongolian ethnic groups. The ranking with significant sequence from high to low were BMI, dyslipidemia, alcohol intake, sex and age.
CONCLUSIONCP infection was not related to hypertension in Chinese Mongolian ethnic groups but the interaction with other traditional risk factors would increase the risk of developing hypertension.

5.188Re-labeled herceptin inhibits proliferation of breast cancer cell line SKBR-3 in vitro.

OBJECTIVETo investigate the inhibitory effects of (188)Re-labeled herceptin on the proliferation in vitro of breast carcinoma cell line (SKBR-3) overexpressing HER-2/neu proto-oncogene.
METHODSHerceptin was radiolabeled with (188)Re through a direct labeling method. SKBR-3 cells were cultured with (188)Re-Herceptin at different radioactivity doses (3.7x10(4), 18.5x10(4), 37x10(4), 55.5x10(4) and 74x10(4) Bq/ml) or with (188)Re-nmIgG and (188)ReO(4)(-) for comparison. The cell proliferation inhibition was determined with MTT colorimetric assay.
RESULTS(188)Re-Herceptin could markedly inhibit the growth of SKBR-3 cells in a radioactivity dose-dependent fashion, while the effect of (188)Re-nmIgG and (188)ReO(4)(-) showed rather poor inhibitory effect in vitro. The 50% inhibition doses (IC(50)) of (188)Re-Herceptin, (188)Re-nmIgG and (188)ReO(4)(-) were 76.1x10(4) Bq/L, 139.2x10(4) Bq/L and 175x10(4) Bq/L, respectively.
CONCLUSION(188)Re-Herceptin can effectively inhibit the growth of in vitro cultured breast cancer cells overexpressing HER-2/neu, and shows much potential for clinical use in beast cancer radioimmunotherapy.

6.Establishing an experimental guinea pig model of dermatophytosis Using Trichophyton rubrum.

OBJECTIVETo construct an animal model infected by Trichophyton rubrum.
METHODSThree different strains of Trichophyton rubrum were separated from clinical specimen for the infection of guinea pigs. Corticosteroids were given before and after the construction of animal model to facilitate the infection. Direct microscopy, culture, and histopathologic methods were adopted to verify the construction.
RESULTSTen days after the inoculation of Trichophyton rubrum, with the intervention of corticosteroid, the guinea pigs were examined. Prominent scales and inflammation could be seen on the inoculation site of the Trichophyton rubrum infected guinea pig. Scales and hairs of Trichophyton rubrum infected guinea pig dealt with 10% potassium hydroxide, hypha out of the hair and microconidia or hypha in the hair shaft could be seen. Seven days after the inoculation of scales and hair on SDA plate, cultures of Trichophyton rubrum showed that the colonial morphology were identical to the original dermatophytes. PAS staining of infected guinea pig skin tissue showed that hypha and microconidia could be seen in the infundibula and hair root.
CONCLUSIONWith the intervention of corticosteroid, a stable guinea pig model infected by Trichophyton rubrum were successfully constructed.

7.Effect of altitude chronic hypoxia on liver enzymes and its correlation with ACE/ACE2 in yak and migrated cattle.

OBJECTIVETo investigate the difference of liver enzyme levels and its correlation with serum ACE/ACE2 among yak and cattle on Qinghai-Tibetan plateau, and to further explore the biochemical mechanism of their liver of altitude adaptation.
METHODSThe serum samples of yak were collected at 3,000 m, 3,500 m, 4,000 m and 4,300 m respectively, meanwhile the serum samples of migrated cattle on plateau (2,500 m) and lowland cattle (1,300 m) were also collected. The levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), cholinesterase (CHE), gamma glutamyl transferase (GGT), alkaline phosphatase (ALP), serum lipase (LPS), angiotensin converting enzyme(ACE), angiotensin converting enzyme-2 (ACE2) in serum were measured by using fully automatic blood biochemcal analyzer. We analysed the differences of the above enzymes and its correlation with ACE/ACE2. We used one way analysis of variance (ANOVA).
RESULTSThe levels of ALT in 4,000 m group and 4,300 m group of yak increased significantly compared with other groups, there were no statistically significant differences in AST, CHE, GGT, ACE/ACE2 levels of yaks at different altitudes. As compared to lowland cattle, the serum levels of AST and CHE were increased, the level of LPS and ACE was decreased significantly, respectively, and especially, the ratio of ACE/ACE2 of migranted cattle reduced nearly two times. The levels of LPS were significantly correlated to the ratio of ACE/ACE2 in yak (r = 0.357, P < 0.01), and a high correlation between ALP and ACE/ACE2 in lowland cattle( r = 0.418, P < 0.05), But the biggest contribution rate of the ratio of ACE/ACE2 was only 17.5% for the changes of the levels of liver enzyme.
CONCLUSIONThe results indicated that with the altitude increased did not significantly influence the changes of liver enzymes' activities in mountainous yaks but not in cattle. However, all above these changes weren't actually correlated to the ratio of ACE/ACE2.

9.Expert survey for Chinese medicine syndrome characteristics of different clinical types of coronary artery disease based on the Delphi method.

OBJECTIVETo carry out expert survey for traditional Chinese medicine (TCM) syndrome characteristics of different clinical types of coronary artery disease (CAD).
METHODSBy using Delphi method, we carried out two rounds of nationwide expert surveys for modern TCM characteristics of syndrome elements and syndrome types of CAD.
RESULTSBased on expert consensus, qi deficiency, blood stasis, phlegm turbidity, qi deficiency blood stasis, and intermingled phlegm and blood stasis are common TCM syndromes for different clinical types of CAD. Of them, qi stagnation, blood stasis, phlegm turbidity, heat accumulation, cold coagulation, yang deficiency, deficiency of both qi and yang were more often seen in patients with unstable angina than in those with stable angina. Qi deficiency, yin deficiency, and deficiency of both qi and yin were less seen. We could see more excess syndrome and less deficiency syndrome (such as qi deficiency, yin deficiency, etc.) in acute ST-segment elevation myocardial infarction (STEMI) than acute non-ST-segment elevation myocardial infarction (NSTEMI). Qi deficiency, blood stasis, water retention, yang deficiency, phlegm turbidity, yin deficiency, Xin-qi deficiency, and qi deficiency blood stasis induced water retention are the most common TCM syndrome types of CAD heart failure (HF). Blood deficiency, yin deficiency, heat accumulation, deficiency of both Xin and Pi, deficiency of both qi and blood, deficiency of both qi and yin, yin deficiency and fire hyperactivity were more often seen in CAD arrhythmias.
CONCLUSIONSTCM syndrome distributions of different clinical types of CAD have common laws and individual characteristics. Results based on the expert consensus supplied evidence and support for clinical diagnosis and treatment of CAD.

10.Application of FTIR spectroscopy to the analysis of eleven kinds of Dendrobium.

OBJECTIVETo establish an FTIR method for the analysis of Dendrobium.
METHODUsing fourier transform infrared spectrometer to record the characteristic spectra of eleven samples of Dendrobium, and to compare the spectra by PCA (principal component analysis).
RESULTThe FTIR spectra of the upper part of the stem displayed significant differences between fresh and dried samples of Dendrobium. On the other hand, differences were observed in the spectra of the middle and lower parts of stems of D. guangxieuse when compared to other species.
CONCLUSIONThe method of applying PCA to FTIR analysis is a rapid and dependable method for comparing samples of Dendrobium.
