Objective To investigate the genes differentially expressed in prostate cancer and to determine some prostate cancer associated genes for diagnosis and treatment prostate cancer. Methods cDNA microarray chips containing 4 366 human genes in full length were used to investigate the differential gene expression patterns of samples from human prostatie cancers and normal prostate tissues. Results A total of 287 genes differentially expressed with prostatie cancers and normal prostate were screened out, comprising 165 unknown genes and 122 known genes.Among the known genes we further identified 56 genes which were markedly differentially expressed (20 up-regulated and 36 down-regulated genes). Conclusions cDNA microarray chips for analysis of genes expression patterns is a powerful method for identification of novel prostatie cancer associated genes.

Objective To evaluate the safety, biocompatibility and efficacy of transcatheter closure of atrial septal defect(ASD)with no stainless-steel-screw occluder in canine model. Methods The device was constructed from superelastic Nitinol wires tightly woven into two flat disks and sewed with polyester fibers inside, with a pliable loop on the right-atrial-disk of the device, connecting to the delivery cable.ASD was created by transcatheter puncture and balloon dilatation and then closed by occluder under fluoroscopy in the catheterization laboratory. The location and the influence of the implanted device on function of tricuspid valve and mitral valve were evaluated by echocardiography. At 1,2,3 and 6 months after the operation, the animals were killed and autopsy was conducted. Results Eight dogs with puncture-produced ASD underwent ASD closing procedure successfully. The occluder showed no influence on the function of MV and AV demonstrated by echocardiogram. The two disks of the implanted device were covered with a smooth intact neogenesis layer in all dogs.Endocardial cells fully covered the surface of the two disk without inflammating reaction 3 months later. There was no evidence of corrosion on the surface of the nitinol wire removed from the dog after 6 months.Light microscopic examination of the liver, kidney, lung and spleen showed no evidence of embolization and inflammation. Conclusion Transcatheter ASD occlusion with new-type occluder is safe, feasible, effective and good biocompatibility with a good prospective clinical application.

Objective To investigate the association of tumor necrosis factor (TNF)-α, heat shock protein (HSP)70-2 gene polymorphisms and susceptibility of acute pancreatitis(AP). Methods Using case-control method,The gene polymor?phism of TNF-α and HSP70-2 was detected by PCR-RLFP in 72 patients with AP and 71 healthy controls. Results There were no significant differences in proportion of TNF-αgenotype and alleles between AP and control groups (P>0.05). There were no significant differences in TNF-αgenotype and alleles between severe acute pancreatitis (SAP) and light acute pancreatitis (MAP) of AP group (P>0.05). There were no significant differences in white blood cell count, C-reactive pro?tein (CRP), amylase, three acyl glycerin and glucose between TNF-a and HSP70-2 gene of AA type and GA+GG type pa?tients (P>0.05). The HSP70-2 genotype GA+GG proportion was significantly higher in AP group than that of control group (69.4%vs 49.3%). The ratio of patients with G allele was significantly higher in AP group than that of control group(46.5%vs 31.7%). The ratio of patients with GA+GG type AP was significantly higher in SAP patients than that of MAP patients of AP group(81.0% vs 53.3%). There was no significant difference in G allele between SAP and MAP patients (P>0.05). Conclusion TNF-α polymorphisms is not associated with acute pancreatitis. There is an association between HSP70-2 polymorphisms and acute pancreatitis. Carrying the G allele increases the possibility of a severe acute pancreatitis ,which is one of the genetic susceptibility factors of severe acute pancreatitis.
