1.Expressions of MCM5 and Ki67 in cervical intraepithelial neoplasia and cervical cancer and their clinical significance

Objective To study the expressions of MCM5 and Ki67 in cervical intraepithelial neoplasia (CIN) and cervical cancer and their clinical significance. Methods Immunohistochemistry (S-P method) was used to detect the expressions of MCM5 and Ki67 in 50 CIN and 49 cervical cancer. Results The positive expression rate of MCM5 and Ki67 in normal cervical epithelium, CIN Ⅰ-Ⅱ, CIN Ⅲ and cervical cancer were 26.7%, 72.0%, 80.0%, 97.9% and 10.0%, 32.4%, 84.0%, 97.9% respectively. The authors investigated and discussed therapeutic mechanism of small knife needle in treating closed myofascitis on the basis of pathomechanism of modern medicine and acupuncture theory of TCM among numbers of clinical cases and experimental data. Therapeutic mechanism lies in 6 aspects: (1) Relieve the energy crisis of tenderness point on muscular fasciae; (2) Affect nervous system and reduce induction of harmful stimulating signal; (3) Inhibit aseptic inflammatory reaction on muscular fasciae; (4) Regulate dynamic equilibrium of soft tissue by cutting scar and releasing the conglutination; (5) Increase patients' regional threshold of feeling; (6) Reduce tension and pressure of soft tissue of tenderness point so as to relieve extrusion of nervus cutaneous.

4.Simultaneous Determination of Chlorogenic Acid and Cefazolin in Rat Plasma With HPLC

OBJECTIVE:To establish a HPLC method for simultaneous determination of chlorogenic acid and cefazolin in rat plasma.METHODS: Chromatographic determination was performed on spherisorb C18 column with methyl alcohol- 0. 1mol/L potassium ethydrogen phosphate(30: 70v/v, pH 3.0)as the mobile phase and a flow rate of 1.2ml/min.Detection wavelengthes were UV 275nm for chlorogenic acid and 326nm for cefazolin.The concentrations of chlorogenic acid and cefazolin in rat plasma were detected.RESULTS: The calibration curves were linear with the correlation coefficients of 0.997 and 0.998, repectively.The RSDs for the between- day and within- day were both lower than 4% for chlorogenic acid and 6% for cefazolin, respective.The mean recoveries were 93.30%, 93.35% and 96.40% for chlorogenic acid, and 93.41%, 96.60% and 92.36% for cefazolin at large, middle and low dosages, respectively.The concentrations of cefazolin in rat plasma were decreased in coadministration with chlorogenic acid.CONCLUSION: The study provides a simple method for simultaneous determination of the plasma levels of chlorogenic acid and cefazolin.
5.miR-335 inhibits migration and invasion of human osteosarcoma cell line MG-63 by targeting ROCK1

Background and purpose:MicroRNAs are endogenous small RNAs and involved in target gene regulation in post-transcription level. As a tumor suppressor, microRNA-335 (miR-335) was participated in the occurrence and progression in many types of human tumors. This study aimed at investigating whether miR-335 can regulate cell migration and invasion by negative targeting Rho associated coiled-coil forming protein kinase (ROCK1) in human osteosarcoma cell line MG-63.Methods:The speciifc binding ability of miR-335 to ROCK1 3’-untranslated region (UTR) was theoretically predicted and detected by the luciferase reporter gene assay. Western blot and real-time PCR were used to evaluate the effect of miR-335 on the expression of ROCK1 at protein and mRNA levels. Cell migration and invasion assays were performed by transwell chambers in MG-63 cells intervened by over-expression of miR-335 and knockdown of ROCK1.Results:Luciferase reporter gene assay showed that miR-335 could target ROCK1 3’-UTR. Lower miR-335 but higher ROCK1 was expressed in MG-63 cells. The results of Western blot showed that ROCK1 protein expression was decreased by transfection of miR-335 mimic or siROCK1. Transwell chambers results showed decreased cells invading through the substrate membrane after over-expression of miR-335 or knockdown ofROCK1 gene.Conclusion:miR-335 inhibits the migration and invasion ability of MG-63 human osteosarcoma cell line, which may through targeting ROCK1 3’-UTR and subsequent down-regulating ROCK1 expression.
6.The expression and correlation of miR-335 and ROCK1 in osteosarcoma

Objective To investigate the expression and correlation of microRNA‐335(miR‐335) and ROCK1 in osteosarco‐ma tissue and osteosarcoma cell lines MG‐63 and U2‐OS .Methods Eighteen paired tissue specimens of osteosarcoma and matched normal tissue were chosen clinically .Two osteosarcoma cell lines MG‐63 and U2‐OS were chosen as experimental group while the human osteoblast cell line hFOB 1 .19 as control group ,Quantitative real‐time PCR were used to evaluate the different expression of miR‐335 and ROCK1 mRNA in osteosarcoma tissue and cell lines;miR‐335 mimic was transfected into MG‐63 and U2‐OS ,while Quantitative real‐time PCR and Western blot were used to evaluate the effect of over‐expressed miR‐335 on ROCK1 mRNA and protein level .Results The result of real‐time PCR indicated a special lower expression of miR‐335 but higher ROCK1 ,and the cor‐relation between miR‐335 and ROCK1 was negative .Western blot showed that miR‐335 could decrease ROCK1 protein expression . miR‐335 mRNA showed a hyper‐expression while lower ROCK1 mRNA and protein after transfection of miR‐335 mimic .Conclusion The expression of miR‐335 and ROCK1 shows a negative correlation in both tissue and cell level;over‐expression of miR‐335 can decrease ROCK1 expression .
9.The Progress of Tissue Engineering Heart Valve

The currently used heart valve prosthesis are still far from the ideal one in anticoagulation and longevity. The construction of tissue engineering heart valve leaflets are carried out recently. This article reviews the progress of tissue engineering heart valve leaflets in implanting materials, seeded cells, animal experiment and basic research. It also discusses the benefits and feasibility of tissue engineering heart valve leaflets.
10.Behavior problems and family factors in children of off-farm workers of Shanghai

Objective To survey prevalence of behavior problems in children of off-farm workers and effects of family factors on children' s behavior problems.Methods The study was done in the whole 7 schools for off-farm workers' children in Shanghai Baoshan district,including all 3 ~ 6 grades students of primary school and 1 ~3 grades students of junior high school.Totally 4350 students finished survey (95.4%).Achenbach' s child behavior checklist ( CBCL,parent' s version) and general family questionnaire were used.Results ( 1 ) Prevalence of behavior problems in children was 11.2% ; 12.8% 12 ~ 16 years children had behavior problems( 14.7%boys and 10.4% girls) ; 11.4% of one-child children had behavior problems compared with that of 11.2% child with siblings children ( x2 =0.038,P > 0.05 ).(2) Total score ( 20.64 ± 17.67 ) and factor scores of thought problems,depressed,hyperactivity,delinquent rule-breaking behavior were higher than those of Chinese norm (P <0.05 or P < 0.01 ) among 6 ~ 11 years boy group.Total score ( 17.23 ± 14.73 ) and scores of thought problems,depressed,social problems,hyperactivity,delinquent rule-breaking behavior were higher than those of Chinese norm (P < 0.01 ) among 6 ~ 11 years girl group.Among 12 ~ 16 years boy group,total score (20.44 ± 19.32) and all factor scores of CBCL were higher than those of Chinese norm(P<0.01 ).Total score ( 17.02 ± 16.72) and scores of immature,delinquent rule-breaking behavior,brutality were higher than those of Chinese norm(P < 0.05 or P <0.01 ) among 6 ~ 11 years girl group.(3)Multi-regression analysis showed that poor parent-child relationship was the most important predict factor for higher score of CBCL among all family risk factors (P < 0.05 ).Conclusion Children of the off-farm workers and their families need special mental health care program.
