1.Evaluation of Communication Difficulty Perceived by the Children with Hearing Impairment Using APHAB

Objective To investigate the perception of communication difficulties of children with hearing impairment and their parents.Methods Using the abbreviated profile of hearing aid benefit (APHAB),perception of communication difficulty in daily situations of the children with hearing impairment and their parents was measured,then the scores were compaired.Results There was no significant difference in perception of communication difficulty between the children and their parents.On problems on ease of communication(EC) and aversiveness(AV), there was difference in the perception of communication difficulty between the children who did not wear hearing aids and their parents.Conclusion It's critical to teach the parents knowledge about hearing loss to reduce their communication difficuty.
2.Percutaneous laser disc decompression in the treatment of discogenic back pain

Objective To evaluate curative effects of percutaneous laser disc decompression(PLDD) in the treatment of discogenic low back pain.Methods A total of 36 patients with discogenic low back pain was included in the study.The PLDD was performed by using the DIOMED semiconductor laser treatment,with a power of 15 W,1 s-pulse duration,1 s-interval,and an energy of radiation of 800~1 200 J.Curative outcomes were evaluated based on the Visual Analog Scale(VAS) criteria. Results The operation time was 15~60 min(mean,30 min).The pain provocation was observed in 32 patients.Follow-up examinations were conducted in the 36 patients for 6~36 months(mean,11 months).The postoperative VAS scores were improved by ≥ 3 points in 18 patients and ≥ 2 points in 14 patients.Results revealed that 32 patients(88.9%) reported a good response and 4(11.1%) reported no response. Conclusions Percutaneous laser disc decompression is a safe,effective,and minimally invasive procedure that can be used for patients with discogenic low back pain.
3.Effect of high glucose on the expression of KLF6 in human lens epithelial cell

Background Epithelial-mesenchymaltransition (EMT)isamajorcontributortothe pathogenesis of posterior capsular opacification(PCO).Kruppel-like factor 6 (KLF6) is a zinc finger protein,which can be stimulated by high glucose in proximal tubule cells and involved in transforming growth factor beta (TGF-β)induced EMT of diabetic nephropathy.ObjectiveThis study was designed to investigate the effect of high glucose on the expression of KLF6 and its target genes( TGFB1,TGFBR1,COLIA1,HSP47) in human lens epithelial cells (LECs).MethodsHuman LECs(SRA01/04) were cultured and exposed to different concentration of glucose.The expressions of KLF6 mRNA and protein were analyzed by real time polymerase chain reaction( real time PCR) and western blot after treatment with high glucose.The expressions of KLF6 target genes were analyzed by real time PCR to evaluate the EMT of SRA01/04 cells.ResultsCompared with the control group(5.5 mmol/L),the relative mRNA levels of t-KLF6 and wt-KLF6 in SRA01/04 treated with high glucose(22.2,44.4,66.6 mmol/L) increased obviously (F =72.53,42.02,P<0.01 ).Then,the concentration of 22.2 mmol/L was used in the next experiments.The relative mRNA levels of t-KLF6 and wt-KLF6 increased to the peaks after treatment with high glucose for 12 h,and began to decrease after 24 h until lower levels after 48 h ( F =100.12,125.52,P < 0.01 ).Western blot showed that the expression of KLF6 protein was also upregulated by high glucose treatment.With the promotion of the expression of KLF6 gene,the relative mRNA levels of TGFB1,TGFBR1,COLlAl and HSP47 of treated cells also respectively increased after treatment for 12 h,and began to decrease after 24 h until nearly at the levels of the control groups after 48 h( F=6.73,162.35,64.39,12.05,P<0.05 ).ConclusionsIt was concluded that high glucose induced the expression of KLF6 in human LECs,and KLF6 transiently stimulated the expression of target genes TGFB1,TGFBRl,COLlAl and HSP47 which were mainly involved in the mechanism of EMT.
4.Clinical research on application value of flexible ureteroscope combined with extracorporeal shock wave lithotripsy in patients with kidney stones with a diameter of more than 2cm

Objective:To observe the reliability and clinical implement of flexible ureteroscope combined with extracorporeal shock wave lithotripsy to treat the kidney stones with a diameter of more than 2cm. Methods:The diagnosed 80 cases of kidney stones (all with a diameter of more than 2cm) patients in our hospital were chosen, randomly divided into two groups, URL group and PCNL group,each of them with 40 cases. Surgical curative effect, operation time, postoperative complications, hospitalization days and the change of NGAL levels and the change of urine levels Kim-1 preoperative and postoperative blood 3 days of two groups were observed and compared. Results: Comparing with the PCNL group, the surgical curative effect, operation time and postoperative complications of the URL group have no significant difference, but the hospitalization days between are significantly different(t=8.433, P<0.05);In terms of NGAL, Kim-1, the postoperative 1d of PCNL group is significantly higher than preoperative, NGAL postoperative 1d of URL group is significantly higher than preoperative (t=3.319, P<0.05). Conclusion:It was reliable that flexible ureteroscope combined with extracorporeal shock wave lithotripsy to treat the kidney stones with a diameter of more than 2cm, lower complication rate, worthy of clinical.
5.Quercetin suppresses HeLa cells by blocking PI3K/Akt pathway.

To explore the effect of quercetin on the proliferation and apoptosis of HeLa cells, HeLa cells were incubated with quercetin at different concentrations. Cell viability was evaluated by MTT assay, cell apoptosis was detected by Annexin-V/PI double labeled cytometry and DNA ladder assay. Cell cycle was flow cytometrically determined and the morphological changes of the cells were observed under a fluorescence microscope after Hoechst 33258 staining and the apoptosis-related proteins in the HeLa cells were assessed by Western blotting. The results showed that quercetin significantly inhibited the growth of HeLa cells and induced obvious apoptosis in vitro in a time- and dose-dependent manner. Moreover, quercetin induced apoptosis of HeLa cells in cell cycle-dependent manner because quercetin could induce arrest of HeLa cells at G0/G1 phase. Quercetin treatment down-regulated the expression of the PI3K and p-Akt. In addition, quercetin could down-regulate expression of bcl-2, up-regulate Bax, but exerted no effect on the overall expression of Akt. We are led to conclude that quercetin induces apoptosis via PI3k/Akt pathways, and quercetin has potential to be used as an anti-tumor agent against human cervix cancer.
6.Relationships of electrophysiological characteristic between speech evoked auditory brainstem response and auditory mismatch negativity.

OBJECTIVE:
To investigate the relationships of electrophysiological characteristics between speech evoked auditory brainstem response (s-ABR) and auditory mismatch negativity (MMN), so as to provide more clues for the mechanism of speech cognitive behavior.
METHOD:
Thirty-three ears in 33 normal hearing adults were included in this study. Their s-ABR were recorded with speech syllables /da/ at 80 dB HL intensity. Meanwhile, two MMNs were recorded with 1 kHz frequency deviant extent and 40 dB intensity deviant extent in them. The electrophysiological characteristics of s-ABRs and MMNs, as well as the relationships of MMN latencies between s-ABR parameters including latencies in time domain, fundamental frequency(F0) and first formants(F1) in frequency domain were analyzed statistically.
RESULT:
MMN latency of frequency deviance showed a negative correlation tendency with s-ABR transient components, and it showed a positive trend with sustained components of s-ABR. While MMN latency of intensity deviance showed a positive correlation with s-ABR latency of peak V, A and D respectively, and it negatively showed a correlation with s-ABR latency of other peak s and amplitude of F0 and FI respectively. Only the s-ABR latency of peak F and MMN latency of frequency deviance, and the F0 amplitude of s-ABR and MMN latency of intensity deviance were moderate correlation statistically.
CONCLUSION
It was probably the neurons of frequency deviant MMN unmatched the characteristics of frequency with the neurons of s-ABR transient component, but well matched the characteristics of frequency with the neurons of s-ABR sustained component. Similarly, the neurons of intensity deviant MMN probably matched the characteristics of intensity with neurons of different components of s-ABR or not. These results may formed as a valuable clue for further investigation of speech perception and temporal processing abilities.

10.Detection of high-risk human papillomavirus types-16 and-18 in laryngeal squamous cell carcinoma

Objective To investigate the relationship between human papilloma virus(HPV)infection and the development of laryngeal squamous cell carcinoma(LSCC).Methods To elucidate the role of HPV in the development of LSCC,we employed polymerase chain reaction(PCR)based on four pairs of primers an4 in situ hybridization(ISH)to screen the HPV infection in 84 ISCC tissues.Results Using HPV L1 general primer amplification,HPV DNA was detected in 23(27.4%)of the 84 LSCC samples.However,when specific primers for HPV-16 or-18 were used to amplify E6 and E7 in all samples,29 cases(34.5%)were positive for HPV-16,while 6 cases(7.1%)were positive for HPV 18.Coinfeetion of HPV-16 and-18 were found in 4cases (4.8%).Overall,HPV type 16 and 18 infections were present in 36.9% of the LSCC samples.In addition,the positive rate of HPV 16 E6 mRNA was 30.9%(26/84)in LSCC by ISH with digoxin-labeled sense probes of HPV 16 E6.Conclusion High-risk HPV-16may be an etiologic factor in the development of laryngeal squamous cell carcinoma, while the complicated molecular mechanism of HPV16 inducedtumorgenesis needs a further study.
