1.Ligand fishing: a strategy for rapidly screening bioactive compounds from organism extracts

Natural products are valuable resources for discovering new drugs. So far, screening bioactive compounds from organism extracts is still an important and challenging task. Traditional biometric guided method involves repeated fractionation steps and bioactivity tests, which are time-consuming, labor-consuming, and inefficient. Ligand fishing is a bioanalysis method for screening ligands from complex organism extracts based on intermolecular affinity interactions. It has the characteristics of strong specificity, high efficiency, and less requirement for sample pretreatment. In this review, we summarize the classification of ligand fishing strategy and its application in enzyme inhibitors screening. Finally, the development prospects of this technology are forecasted.
2.Comparision of the hypotension induced by diethylamine/nitric oxide and nitroprusside

Objective: To compare hemodynamic effects of infusion of diethylamine/nitric oxide (DEA/NO)and sodium nitroprusside(SNP)during controlled hypotension. Method: General anesthesia was induced in dogs. The twelve healthy adult dogs were randomly assigned into one of two groups. 0.00l% DEA/NO or 0.01% SNP was infused to induce mean arterial pressure(MAP) to decrease to 60% of baseline and be maintained for 30 min. During hypotension, MAP,central venous pressure (CVP), mean pulmonary artery pressure (MPAP)and cardiac output (CO) were measured and recorded. Arterial blood gas, blood lactate and urine output were also measured. Result: HR,CVP and CO were unchanged during hypotension period in both groups, MPAP decreased correspondently with MAP. There were no significantly changes in arterial blood gas,blood lactate and urine output in both groups during hypotension. Conclusion: Controlled hypotension achieved with infusion of DEA/NO has a rapid onset and short duration of action.The hemodynamics were similar to those of SNP.
3.Relationship between quinone oxidoreductase1 gene ns-cSNP and genetic susceptibility of esophageal cancer.

OBJECTIVETo explore the relationship between quinone oxidoreductase1 (NQO1) gene nonsynonymous cSNP and the genetic susceptibility of esophageal cancer.
METHODSPolymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and Allele-Specific PCR (AS-PCR) were employed to assess the polymorphism of NQO1 genes both in 106 patients with esophageal cancer and control subjects matched by age, gender and origin.
RESULTSIt was shown that no C/C genotype was found at 406 of NQO1. The allelic frequency of NQO1 609T was significantly higher in patients with esophageal cancer than in the control subjects (P < 0.005) and the individuals with 609T allelic genotype of NQO1 gene were at greater risk to develop esophageal cancer (OR = 4.76, 95% CI = 1.064 - 3.397). But Individuals with mutant allele of NQO1 465 genotype did not show the rising risk of esophageal cancer.
CONCLUSIONSThe NQO1 C609T polymorphisms should likely be associated with the genetic susceptibility of esophageal cancer.

4.Application of pathological diagnosis by using ultrasound-guided percutaneous biopsy for cervical lymph lesions

Objective To evaluate the clinical application value of pathological diagnosis by using ultrasound-guided percutaneous needle core biopsy (PNCB) for cervical lymph lesions.MethodsTwo hundred and ten patients with cervical lymph lesions underwent ultrasound-guided PNCB,and pathological diagnosis were made based on core biopsy material.The results of study were concluded by comparing the pathological diagnosis from core biopsy of the lymph nodes with those from excision biopsy. Results There were 210 patients underwent ultrasound- guided PNCB,98.6% (207/210) cases of core biopsies yielded adequate material at first time and 92.9% (195/210) cases of those had pathologic diagnosis.The accuracy of ultrasound-guided PNCB in differentiating benign from malignant lymphadenopathy was 99.4% ( 194/195 ).The diagnostic accuracy of metastatic tumor and lymphoma by ultrasound-guided PNCB was 96.4% (135/140) and 86.1% (31/36),respectively.Only 67.7% (21/31) patients with lymphoma could be classified by ultrasound-guided PNCB.ConclusionUltrasound-guided PNCB in patients with neck lymph lesions is a safe,convenient and quick procedure that has a high diagnosis accuracy.Ultrasound-guided PNCB can replace the open surgery for neck lymph node diagnostic method.Due to the complicated and diverse pathologic performance,lymphoma should be cut by open surgery to confirm the diagnosis and classification.
5.Influence of escharectomy during shock stage on the systemic and intestinal immune function in scalded rats.

OBJECTIVETo investigate the influence of escharectomy during shock stage on systemic and intestinal immune function and its mechanism in scalded rats.
METHODSNinety-six Wistar rats were employed in the study of which 8 were used as normal control group. The donor skin from the trunk in twenty-four rats were preserved in liquid nitrogen. The other 64 rats were subjected to 30% full-thickness scalding, and they were randomly divided into A (n = 24, no treatment after scalding), B (n = 24) and C (n = 16) groups. Physiological saline was intraperitoneally injected (50 ml/kg) on the 24 post-scalding hours to the rats in the B and C groups. The rats in B group underwent escharectomy during shock stage, and the excision wounds were covered with the cryo-preserved alloskin. The rats in C group received the same treatment as in B group but at 72 post-scalding hours. The change in the proliferative ability of splenic lymphocytes, the plasma and intestinal tissue content of interleukin 2 (IL-2), the contents of sIgA in intestinal mucus, and the content of DAO in the intestinal tissue were observed on 2, 4 and 8 post burn days (PBD) in A and B groups and also on 4 and 8 PBD in C group, respectively.
RESULTSThe splenocytic proliferative ability, IL-2 level in the plasma and intestinal tissue, and the sIgA content in intestinal mucus in the rats in A, B and C groups were lower than that in control group at all time points (P < 0.05). The proliferative ability of splenic lymphocytes in B group on 4 and 8 PBD and in C group on 8 PBD respectively was similar to that in control group. Whereas the IL-2 content in plasma and in intestinal tissue was higher in B and C groups than that in A group (P < 0.01). The sIgA content in intestinal mucus in B group was twice of that in C group respectively [(3.51 +/- 2.14) mg/g vs (1.40 +/- 0.64) mg/g, (3.03 +/- 0.95) mg/g vs (1.52 +/- 1.26) mg/g (P < 0.05 or P < 0.01)] on 4 and 8 PBD. The DAO activity in the intestinal tissue in A group was lower than that in control and B group (P < 0.05) on 4 and 8 PBD.
CONCLUSIONEscharectomy during shock stage might be beneficial to the recovery of the systemic and intestinal immune functions in rats with scalding injury.

6.Discussion on Dunhuang Liao Fengxu Shouruo Formula for Treatment of Endometriosis-dysmenorrhea Based on Theory of Pain Caused by Cold-coagulation and Blood-stasis and Medical Cases

One of the mechanisms of endometriosis-dysmenorrhea is closely related to the changes of pelvic anatomy and hemodynamics. TCM holds that cold-coagulation and blood-stasis that alternate and resist the cell palace is one of the main pathogenesis of dysmenorrheas.Dunhuang Liao Fengxu Shouruo Formula(P.3930),a precise and appropriate formulating prescription, plays an important role in activating blood, dispelling cold, tonifying qi and nourishing blood, which can warm the maridians and disperse cold, nourish blood and dissolve stasis and it achieves significant results. And in this prescription, the method of decocting herbs in mutton soup embodies the local characteristic of foodtherapy combined with medication of ancient Dunhuang medical prescriptions.
7.Surgical resection of sternal tumors and reconstruction with titanium mesh.

OBJECTIVETo evaluate the use of titanium mesh reconstruction after sternal tumor resection.
METHODSFrom January 2007 to January 2011, 14 patients with sternal tumors were admitted into Peking Union Medical Hospital. The clinical characteristics, surgical resection, and technique of reconstruction were reviewed.
RESULTSOf the 14 patients, 3 had a metastatic sternal tumor, the primary sites of which were as follows: hepatic carcinoma in one case (metastasis 19 years after operation), breast carcinoma in another case (metastasis 5 years after operation), and renal carcinoma in the other case (found simultaneously). Two patients showed local involvement of the sternum: 1 had thymic carcinoma, and the other had myofibrosarcoma. The remaining 9 patients had primary tumors: 4 were osteochondroma, 3 chondrosarcoma, 1 eosinophilic granuloma, 1 non-Hodgekin's lymphoma. En bloc resection of the sternal tumor was performed in all the 14 patients. The defect was repaired with the titanium mesh adjusted to the shape of the defect and fixed with the stainless steel wire. Eleven patients were followed up for a period from 2 months to 4 years, during which no translocation or broken of the titanium mesh was observed.
CONCLUSIONSRadical en bloc excision remains the treatment of choice for sternal tumors. Sternum defect reconstruction using titanium mesh as a rigid replacement proves appropriate and effective.

8.Influence of chronic prostatitis/chronic pelvic pain syndromes on male health.

OBJECTIVETo investigate the influence of chronic prostatitis/chronic pelvic pain syndromes (CP/CPPS) on male health.
METHODSWe conducted investigations among 200 cases of CP/CPPS, aged 20 -59 years and randomly selected from 650 out-patients, using an anonymous questionnaire including such items as the patients'age, height, body weight, disease duration and treatment, the results of EPS and semen determinations, and the scores on NIH-CPSI, IIEF-5 and Symptom Checklist (SCL-90).
RESULTSAccording to the 198 valid questionnaires collected, CPSI scores showed 56 (28.3%) cases to be mild, 98 (49.5%) moderate and 44 (22.2%) severe. Sperm density and motility (grade a + b) were reduced in 38 (19.2%) and 47 (23.7%) cases, respectively. IIEF-5 revealed ED in 41 (20.7%) cases, of which 58.5% were mild. Psychological abnormality was found in 94 (47.5%) cases, including anxiety, depression, sensitiveness and so on. NIH-CPSI scores were positively correlated with psychological symptoms and ED (r = 0.25, P < 0.05; r = 0.12, P < 0.05), but not significantly related with sperm density and motility (P > 0.05).
CONCLUSIONThe influence of CP/CPPS is obvious on male psychology but mild on penile erection and not significant on semen. Integrated physiological and psychological therapy is recommended for the treatment of CP/CPPS.

9.Expression and significance of RECK, metalloproteinase-9 and vascular endothelial growth factor-C in colorectal cancer.

OBJECTIVETo investigate the expression of reversion-inducing cysteine-rich protein with Kazal motifs (RECK), matrix metalloproteinase-9 (MMP-9), and vascular endothelial growth factor-C (VEGF-C) in colorectal cancer.
METHODSS-P immunohistochemistry was used to detect the expression of RECK, MMP-9 and VEGF-C in colorectal cancer specimen. Colorectal mucosal tissue at least 10 cm away from the tumor was used as control.
RESULTSThe positive expression of RECK in colorectal cancer was significantly lower than that in the controls(53.3% vs. 100%, P<0.05). Both MMP-9 and VEGF-C were over expressed compared with the controls(86.7% vs. 23.3% and 71.7% vs. 13.3% respectively, P<0.01). The expression of RECK was negatively correlated with that of MMP-9 and VEGF-C, and the expression of MMP-9 was positively correlated with that of VEGF-C. There were significant associations between the expression of these proteins and lymph node metastasis, distant metastasis, and TNM staging (all P<0.05).
CONCLUSIONSRECK expression is low in colorectal cancer, while MMP-9 and VEGF-C expressions are high. Combined testing of these 3 markers is important in the evaluation of tumor metastasis and invasion, and is helpful in the prediction of the prognosis.

10.Current application of placebo in Chinese clinical studies in past 30 years.

To analyze and discuss placebo-related information in clinical research literatures in the past 30 years, including placebo's dosage form, ingredients, preparation process and quality control. Effort were made to research the CNKI. full-text database to preliminary find 700 placebo-related clinical research literature, screen out 301 eligible articles by hand, read the literatures to extract placebo-related information and make statistics and discussions. According to the results, Chinese randomized placebo-controlled clinical studies were characterized by diverse dosage forms of placebo with lack of reports for components, as evidenced by the only 17 literatures describing placebo's preparation or specific composition among the 301 literatures. Placebo-controlled clinical trials covered a wide range of disease spectra, but with a specific tropism of diseases in terms of system classification. Although placebo plays a key role in blinded clinical studies, researchers made less records of placebo, perhaps because they paid less attention to placebo or more attention to the research process or restricted by other objective conditions. Moreover, placebo production, quality control and quality evaluation also need to be further standardized.
