1.Expressions of CD147 and EGFR protein and their relationship with metastasis and prognosis of gastric cancer

Objective To investigate the expressions of extracellular matrix metalloproteinase inducer molecule CD147 and epidermal growth factor receptor (EGFR) protein in gastric cancer and their relationship with the pathological behavior and prognosis.Methods Immunohistochemical staining was used to detect the expressions of CD147 and EGFR protein in 63 cases of gastric cancer and 40 cases of paracancerous normal tissues.The study was also combined with analysis of the pathological behavior and clinical follow-up survey of gastric cancer.Results The positive expression rates of CD147 and EGFR protein in gastric cancer were 77.8% (49/63) and 42.9% (27/63),respectively,which were higher than those in paracancerous normal tissues 25.0% and 0 (x2 =27.851,P =0.000;x2 =23.233,P =0.000).In gastric cancer,the expressions of CD147 and EGFR were closely related to the depth invasion (x2 =5.992,P =0.014;x2 =13.148,P =0.000),TNM stages (.x2 =8.481,P=0.004;x2=14.589,P=0.000),lymph node metastasis (x2 =5.270,P=0.022;x2=20.261,P =0.000) and prognosis of patients (x2 =5.606,P =0.018;x2 =4.982,P =0.026).The expression of CD147 was also significantly correlated with Ki-67 expression (x2 =10.481,P =0.001).CD147 expression was positively correlated with EGFR expression(r =0.309,P =0.014).Conclusion The expressions of CD147 and EGFR are closely related to carcinogenesis,metastasis and survival time of gastric cancer.Detecting the expressions of CD147 and EGFR may be prognostic indicators of gastric cancer.

Objective To isolate and purify the antibacterial peptides from larvae secretion of housefly (Musca domestica) and study their partial characteristics. Methods Protein isolation and purification were performed by routine process, namely, ultrafiltration, solid phase extraction (SPE) and reversed-phase high-performance liquid chroma-tography (RP-HPLC). The minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) of the antibacterial peptides were examined. The antibacterial effect of peptides was studied in nutritive medium with different pH value(pH 5.0-10.0), divalent cations (Mg2+: (0.5?10-3~10.0?10-3)mol/L, Na+、K+: (10?10-3~100?10-3)mol/L), and serum content(12.5%~75%). Results Molecular weight of the peptides was about Mr 3 000-30 000 after ultrafiltration. The fractions eluted with 20%, 30%, 70%, and 80% of acetonitrile (ACN) all showed antibacterial activity by solid phase extraction. The fractions eluted with 70% ACN showed strongest and stablest antibacterial activity which was further purified by RP-HPLC. Two sub-fractions appeared at around RT 15.5 min and 18.5 min were obtained with antibacterial activity. The MIC to those standard Escherichia coli, Pseudomonas aerugirwsa, Staphylococcus aureas, and Bacillus subtilis was 32.7380, 16.3688, 65.4750 and 32.7380?g/ml respectively. To improve 3-ketosteroid-△1-dehydrogenase(KSDH) activity and the transformation level for androst-4-ene-3,17-dione, 3-ketosteroid-△1-dehydrogenase gene(ksdD) from Arthrobacter simplex was cloned into plasmid pWB980 and expressed in B. subtilis WB600 under the control of promoter P43. The molecular weight of expressed enzyme was about 55kDa by SDS-PAGE analysis. The activitities assayed by spectrophotometrical method of intracellular and extracellular soluble enzyme were 110?0.5mU and 15?0.6mU per milligram of protein respectively. The transformation rate of androst-4-ene-3,17-dione by the B. subtilis recombinant cells was 45.3%. Compared with Arthrobacter simplex, the enzyme activity of KSDH expressed in B. subtilis was improved about 30 fold, and the transformation level of androst-4-ene-3,17-dione by the B. subtilis recombinant cells was improved about 10 fold. The recombinant B. subtilis cells used in biotransformation of steroids provided a new way for steroid medicines production.
9.Optimization of Cultural Conditions of Probiotic Bacteria Lactobacillus casei Zhang for High Cell Density Cultivation

To get high cell density,pH-stat fermentations of L.casei Zhang were carried out in the medium optimized previously under different cultural conditions.The influences of neutralizing agents,the concen-tration of buffer salts and glucose in the medium,pH,aeration and fed batch fermentation on the growth of L.casei Zhang were investigated.Based on the values of maximum specific growth rate,biomass and viable cells count in different cultural conditions,the optimal growth condition was regarded as follows:Glucose level of the medium was 80 g/L ~100 g/L;The pH was kept constant at 5.9 with aqua ammonia and anae-robic condition was kept by sparged with nitrogen periodically;The temperature was set at 37?C and cul-tured for 10 h ~12 h.The biomass and viable cells' number of L.casei Zhang under this culture conditions were 7 g/L and 3.5?1010 CFU/mL respectively,and were 7 times as high as that before optimized,which can meet the requirements of probiotics.

At the present time, training student to have innovative and applied ability has become the object of higher education in China. In this paper, it was proposed that teacher was obligated to create certain atmospheres in classroom to achieve this goal, including friendliness, harmoniousness, encouragement, happiness, discussion, exploration, etc. At the same time, student-centered study should be encouraged. Through these measures, the spirit of innovation will be inspired, the applied ability will be trained, the capability of self-study will be enhanced.
