AIM To establish an HPLC-ELSD method for the simultaneous content determination of gastrodin,parishin A,parishin B,ginsenosides Rg1,Re,Rf,Rb1 and Rd in Tiantai No.1 Tablets (Ginseng Radix et Rhizoma,Gastrodiae Rhizoma,Cistanches Herba,etc.).METHODS The analysis of methanol extract of this drug was performed on a 45 ℃C thermostatic Alltima C1scolumn (4.6 mm × 250 mm,5 μm),with the mobile phase comprising of acetonitrie-0.1% phosphoric acid flowing at 1.0 mL/min in a gradient elution manner.RESULTS Eight constituents showed good linear relationships within their own ranges (r > 0.999 0),whose average recoveries were 96.94%-97.93% with the RSDs of 1.06%-2.48%.CONCLUSION This simple,accurate and reliable method can be used for the quality control of Tiantai No.1 Tablets.
4.Twenty-One Cases of Type 1 Diabetes under 5 Years Old

Objective To explore the clinical characteristics and emergency treatment for type 1 diabetes(T1DM) and diabetes ketoacidosis(DKA) in children under 5 years old.Methods Twenty-one children under 5 years old with T1DM with 10 years were retrospecti-vely reviewed.The onset situation,clinical feature and treatment of DKA were analyzed.Results The cases of little children diabetes might not have typical symptoms.The positivity of islet antibody was lower.High morbidity of DKA was found in little children and DKA was often caused by infection.Conclusions Infection may be involved in the onset and progress of childhood T1DM.Emergency treatment for DKA may involve the injection of small dose insulin,correction of the disorder of water and electrolysis and regulation of acid-base.

6.Cloning and expression analysis of cinnamate 4-hydroxylase (C4H) reductase gene from Aquilaria sinensis.

The study aimed to clone the open reading frame of cinnamate 4-hydroxylase (C4H) from Aquilaria sinensis and analyze the bioinformatics and expression of the gene. One unique sequence containing C4H domain was discovered in our previous reported wound transcriptome dataset of A. sinensis. The open reading frame of C4H was cloned by RT-PCR strategy with the template of mixed RNA extracted from A. sinensis stem which treated by different wound time. The bioinformatic analysis of this gene and its corresponding protein was performed. C4H expression profiles in responds to MeJA (methyl jasmonate) application were analyzed by real-time PCR. The length of C4H open reading frame (ORF) was 1 515 bp, encoding 514 amino acids. The GenBank accession number is KF134783. Inducible-experiments showed that the genes were induced by mechanical wound as well as MeJA induction, and reached the highest expression level at 8 h and 20 h, respectively. The full-length cDNA of C4H and its expression patterns will provide a foundation for further research on its function in the molecular mechanisms of aromatic compounds and flavonoids biosynthesis.

7.Molecular mechanism for a case with ABO subtype Bx10/O01.

OBJECTIVETo explore the molecular mechanism of a case with ABO subtype Bx10/O01.
METHODSThe serological phenotype of the proband was determined with a conventional method, and the ABO genotype was determined by sequence-specific primer polymerase chain reaction (PCR-SSP). Exons 6 and 7 of the ABO gene were amplified with PCR and sequenced. The samples were collected from both parents and analyzed.
RESULTSThe proband's erythrocytes were detected with B antigens, along with the presence of anti-B in serum and absence of B substance in saliva. The genotype B/O of the proband was identified by PCR-SSP. Direct sequencing of the proband revealed 261delG/G, 297A/G in exon 6 and 526C/G, 657C/T, 703A/G, 796C/A, 803C/G, 829G/T, 930A/G heterozygote in exon 7, which was assigned as Bx10/O01 genotype. The Bx10 allele of the proband was inherited from his mother.
CONCLUSIONThe G to T mutation at position 829 of α -1,3-galactosyltransferase enzyme gene has resulted in the Bx10 phenotype.

8.Characteristics of auditory brain stem response in neonatal hyperbilirubinemia induced by different causes.

OBJECTIVETo explore the characteristics of auditory brainstem response (ABR) in neonatal hyperbilirubinemia induced by different causes.
METHODSA total of 88 neonates (176 ears) with hyperbilirubinemia were divided into several groups according to the causes and followed up after 42 d, and 15 normal neonatus (30 ears) were measured ABR and analyzed the results.
RESULTSThe thresholds of ABR in glucose-6-phosphate dehydrogenase were the highest in all the groups and had the longest incidence rate. The wave III, V latencies and III-V, I-V interwave intervals of the ABR were significantly difference and prolonged during test in comparison to the latencies in the control group (P < 0.05). The neonatal infections group had the longest wave and interwave intervals, followed by ABO incompatibility hemolytic diseases and the unknown cause groups, while the breastfeeding jaundice were the shortest in the groups of neonatal hyperbilirubinemia. The thresholds of ABR in the hyperbilirubinemia caused by several etiologies were significant abnormality when compared with the single etiology. However, they were similar in the wave latencies and interwave intervals of ABR. During the follow up, the ABR wave latencies and interwave intervals except for interwave latency I-III were significantly shorter.
CONCLUSIONSThe toxicity of hyperbilirubinemia to the auditory nervous system are related to the species and number of etiologies. The neonate hyperbilirubinemia caused by glucose-6-phosphate dehydrogenase, infections, ABO incompatibility hemolytic diseases and many etiologies are much more dangerous to the auditory system than the breastfeeding jaundice. The damages of hyperbilirubinemia to the auditory nervous system are reversible probably.

9.Study on tissue cultivation of adventitious roots of Pseudostellaria heterophylla.

OBJECTIVETo systematically optimize the cultivation conditions of adventitious roots of Pseudostellaria heterophylla.
METHODTissue cultivation technology and ultraviolet spectrophotometry were adopted to observe the effect of inoculum volume, sucrose concentration, inorganic salt concentration, number of cultivation days, gradual scale-up cultivation and bubble different angles of bioreactor on the growth of adventitious roots of P. heterophylla, and determine the content of constituents such as saponin, polysaccharide and amino acid.
RESULTThe propagation multiple of adventitious roots reached the maximum when the inoculum was 6 g in a 1 L culture shake flask. With the increase in sucrose concentration, the dry weight propagation multiples of adventitious roots followed an up and down trend. The inorganic salt concentration in a cultivation dish had a greater effect on the growth of adventitious roots, particularly 3/4 MS was the most favorable for the growth of adventitious roots. The growth curve of P. heterophylla was "S", with the biomass reaching the maximum at the 28th day.
CONCLUSIONThe inoculum volume, sucrose concentration, inorganic salt concentration, gradual scale-up cultivation and angles of bubble bioreactor had a significant effect on the growth of adventitious roots of P. heterophylla. The contents of saponin and amino acid in adventitious roots were higher than that in cultivated P. heterophylla, whereas the polysaccharide content were lower than that in cultivated P. heterophylla.
