1. Study on apoptosis-inducing effect for mouse mammary cancer cells transfected with IL-23 gene in vitro and in vivo
Tumor 2008;28(10):842-846
Objective: To investigate the changes in apoptosis of mouse mammary cancer cells (MA-891) after transfection with IL-23 gene in vitro and in vivo and explore the anti-tumor mechanism of IL-23. Methods: IL-23 gene was transferred into two packing cell lines (ectotrophic ψ2 and amphotrophic PA317) in sequence by a retrovirus vector (LXSN), and the positive cellular clones that can produce retroviruses carrying IL-23 gene were screened by G418. The retroviruses were used to transduce the mIL-23 gene into mouse mammary cancer cells (MA-891). After being screened by G418, IL-23/MA-891 cells expressing IL-23 protein were obtained. The expression of IL-23 protein in IL-23/ MA-891 cells was detected by ELISA method. The proliferations of MA-891, LXSN/MA-891 and IL-23/MA-891 cells were detected by MTT assay in vitro. Mice were subcutaneously inoculated with IL-23/MA-891, LXSN/MA-891 and the parental MA-891 cells and the solid tumor formation was observed in vivo. The apoptosis of tumor cells from mice injected with different MA-891 cells was detected by TUNEL and flow cytometry analysis. The expressions of Fas and survivin were determined by RT-PCR and Western blotting in vivo. Results: IL-23 gene was successfully transfected into MA-891 cells. The stable IL-23/ MA-891 cells with over-expression of IL-23 was obtained. Transfection with IL-23 gene had significant effects on the proliferation and apoptosis of MA-891 cells in vitro (P > 0.05). But the tumor growth in mice was greatly inhibited and apoptosis ratio of tumor tissues was significantly increased in the IL-23 gene-transfected group compared with the control group (P <0.01). The expression of Fas on the surface of cells was up-regulated and the expression of survivin was significantly decreased (P <0.01). Conclusion: Transfection with IL-23 gene has no significant effects on the apoptosis of MA-891 cells in vitro. But IL-23 exerts its anti-tumor effect by up-regulating the expression of Fas and down-regulating the expression of survivin in vivo.

Chinese Journal of Endocrinology and Metabolism 2001;0(05):-
Objective To observe the expression of motilin precursor mRNA and location of motilin in human thyroid and to explore the clinical significance. Methods RT-PCR,Sourthern blot and immuno- fluorescence histochemical techniques were applied to detect the expressions of motilin precursor mRNA and motilin in human thyroid and to compare the difference of motilin peptide and motilin mRNA expression among tissues of normal thyroid, nodular goiter and medullary thyroid carcinoama.Results(1)The results of RT-PCR and Southern blot showed that motilin precursor mRNA was expressed in human thyroid.(2)A large number of cells showing collocation of motilin with calcitonin were observed in human thyroid, suggesting that motilin was expressed in C cells.(3)Both the motilin immunoreactive(MTL-IR)and the motilin mRNA were significantly increased in medullary thyroid carcinoma(P0.05).(4)Western blot results showed that the expression of motilin in thyroid medullary carcinoma was significantly higher than that in normal thyroid. However no difference was observed between thyroid nodular goiter and normal thyroid tissue. Conclusion MTL- IR positive cells and motilin mRNA expression are found in human thyroid.Motilin is located in C cells of thyroid. The expressions of motilin and motilin precursor mRNA are significantly increased in medullary thyroid cancer,no difference between thyroid nodular goiter and normal thyroid tissue.

Chinese Journal of Nephrology 2008;24(6):435-440
Objective To study whether the uremic toxins accumulated long-term in uremia patients may be involved in oxidation of protein by forming advanced oxidative protein products (AOPPs). Methods Malonylaldehyde (MDA), hippuric acid (HA) and p-cresol were used as the representatives of uremic toxins. Human albumin serum (HSA), plasma specimens from normal or uremia patients were incubated respectively with MDA (10 retool/L), HA (20 mmol/L) and p-cresol (10 retool/L) or PBS (20 retool/L, pH 7.4, as control groups) at 37℃ for 30 minutes or 24 hours, respectively. Those indices such as AOPPs, protein thiol groups (Pt-SH) and dityrosine were used as biomarkers of protein injury. High performance liquid chromatography (HPLC) was employed to identify the aggregation and cross-links of modified proteins. Results AOPPs levels in all groups containing poison compounds were significantly increased by 121.5%(P<0.05) compared to that in control groups. Uremic toxins also resulted in over 14.7% loss in Pt-SH (P< 0.05) and 119.2% increment in dityrosine, respectively (P<0.05). Meanwhile, the formation of HMW-AOPPs in a time-dependent manner was observed by HPLC and cross-linked protein levels were significantly increased by 148.45%~333.3% in comparison with control groups. Conclusion Uremic toxins can directly mediate the damage of proteins by inducing the formation of HMW- AOPPs in a time-dependent manner, which is also one of the mechanism of AOPPs production in vivo besides the activation of the myeloperoxidase-H2O2-Cl pathway.

Chinese Journal of Nephrology 2005;0(08):-
Objective To study the effect of oxidative modification of hydrochlorous acid (HOCl) on human serum albumin (HSA) and the relationship between the AOPPs and HOCl-treated HSA. Methods Purified HSA (60 mg/ml) was treated with HOCl (0, 1, 5, 10, 20, 30, 40, 50 and 60 mmol/L). Size-exclusion chromatography was applied to estimate molecular weights of oxidized products of HSA by HOCl and spectrum scan from 190 nm -400 nm was performed to observe the spectrum characteristics of all variants of HSA. Results Major products of HSA after exposure to HOC1 were dimer and hexmer of HSA. The first-order process could be employed to describe the oxidative dynamics of monomer and dimer of HSA oxidized by HOCl. To AOPPs formation mediated by oxidant was identified as pseudo first-order reaction. However, formation hexmer was much in accordance with second-order reaction. Hexmer was also a major contributor to AOPPs in all types of modified HSA. Spectral analysis showed that red shift of absorbance maximum of polymers of HSA occurred, suggesting that a possibility that polymers of HSA were cross linked by tyrosine residues in protein. Conclusions Protein aggregation is primary consequence of HSA after its exposure to HOCl. Hexmer of HSA is the major contributor to AOPPs.
