1.Early diagnosis and treatment of diabetic macular edema

Diabetic retinopathy ( DR ) is one of the main blinding eye diseases for people over the age of 50, and diabetic macular edema ( DME) is the leading cause of vision loss is DR patients. The early diagnosis and early treatment is important. As OCT and FFA, mfERG, especially the retinal thickness, volume, retinal edema index quantitative indicators such as objective evaluation of macular edema, embodies the new progress of retinal imaging technology in recent years. OCT is a non -contact clinical application in recent years, noninvasive, high resolution of ophthalmic imaging examination, can do it on retinal ultrastructure observation and quantitative analysis, and the technology is relatively mature, become a routine inspection diagnosis of macular edema. Laser photocoagulation, intravitreous injection with Ranibizumab and vitrectomy is nowadays the important means for the treatment of intractable macular edema.
2.EIS-MS Performance and fingerprint of lipophilic constituents from Radix Salviae Miltiorrhiza

Objective To elucidate the ESI-MS performance of lipophilic tanshinones and to establish a fingerprint of lipophilic tanshinones of Radix Salviae Miltiorrhiza.Methods The lipophilic tanshinones were extracted by ultrasonic wave with 95% ethanol from Radix Salviae Miltiorrhiza and the extracts were analyzed directly in positive ion mode by electrospray ion trap mass spectrometry(ESI-MS).Results The lipophilic tanshinones were easily to form molecular ions ~+ and dimeric sodium adduct ions ~+ in positive ion mode,molecular ions of lipophilic tanshinones were fragmentated through lossing H_2O,CO and A-ring cleavages in ESI-MS~2.The ESI-MS fingerprints of lipophilic tanshinone extracts with 14 selected characteristic peaks analyzed with SPSS software were characteristic and stable.Conclusion Lipophilic tanshinones have similar ESI-MS performance;the ESI-MS fingerprint is a useful tool for a rapid and special identification of lipophilic tanshinones in selected traditional Chinese medicine.
4.Construction of in vitro models of human bone marrow-derived mesenchymal stem cells differentiated into nucleus pulposus-like cells in rabbits

BACKGROUND:There is notan effective method for inducing bone marrow-derived mesenchymal stem cells (BMSCs) differentiatied in to intervertebral disc cells.OBJECTIVE:To establish a model of human BMSCs (hBMSCs) differentiated in to nucleus pulposus-like cells in vitro.DESIGN,TIME AND SETTING:A single sample contrast.The experiment was performed at the Orthopaedics Institute and Central Laboratory,the Affiliated Hospital of Qingdao University Medical College,from August 2008 to March 2009.MATERIALS:Two New Zealand white rabbits with 8-week-old were used to culture nucleus pulposus cells.Human nucleus pulposus was obtained from a aged 16 years patient with T_(12)L_1,L_(1.2) congenital scoliosis.The informed consent was obtained from his patients,hBMSCs were purchased from Salye Biological Science and Technology Co.,Ltd.(Guangzhou,China).METHODS:hBMSCs and nucleus pulposus cells of rabbit were co-cultured in new-style culture model (in 6-well plates and inserts with track-etched membrane having 0.4 μm pores without arms) for 7 days,conventionally laminar culture,solitary hBMSCs and nucleus pulpesus cells culture were set up as controls at same time.MAIN OUTCOME MEASURES:The expressions of collagen Ⅱ and aggrecan mRNA were detected by RT-PCR and Western blot.RESULTS:hBMSCs in the conventionally laminar and single hBMSCs groups were negative to collagen Ⅱ and aggrecan.The collagen Ⅱ and aggrecan expression of hBMSCs were upregulated evidently in new-style culture group,which was similar to nucleus pulposus cells that from degenerate intervertebral disc of human,but these expression did not appear in conventionally laminar culture and solitary hBMSCs culture (P < 0.01).The difference among new-style culture group and other groups had no significance (P > 0.05).CONCLUSION:A new-style model of hBMSCs differentiation to nucleus pulposus-like cells is constructed in vitro,which can highly express collagen Ⅱ and aggrecan and provide a foundation for intervertebral disc degeneration cellular treatment.

6.Detection of serum vitamin D in T2DM patients with retinopathy and the relevance to oxidative stress

Objective To detect the serum vitamin D levels in T2DM patients with retinopathy and to analyze the relevance to oxidative stress.Methods Totally 293 cases of T2DM were selected and they were divided into diabetic retinopathy (DR) group and non-diabetic retinopathy (NDR) group according to the results of fundus examination whether with or without DR.46 healthy volunteers were selected as control group (NC).The clinical information was collected,blood sugar such as fasting blood glucose (FBG),glycosylated hemoglobin (HbA1c),blood lipid index such as high-density lipoprotein cholesterol (HDL-C),total cholesterol (TC),triglyceride (TG),low density lipoprotein cholesterol (LDL-C),serum 25-hydroxyl vitamin D3 (25 (OH)D3),oxidative stress indicators such as malondialdehyde (MDA),superoxide dismutase (SOD) and reduced glutathione (GSH) were detected.The correlation between 25 (OH)D3 and clinical data,blood glucose,blood lipids,oxidative stress indicators was analyzed.Results 25(OH)D3 of the 3 groups had significantly difference based on statistical analysis (P< 0.05).In details,the 25(OH)D3 in DR and NDR group were significantly lower than that in NC group,and 25(OH) D3 in DR group was significantly lower than that in NDR group,and the differences had statistical significance (P<0.05).MDA,SOD,and GSH had significantly difference between the 3 groups (P<0.05).In details,MDA in DR group and NDR group were significantly higher than that in NC group.SOD and GSH were significantly lower than those in NC group,while MDA in DR group was significantly higher than that in NDR group.SOD and GSH were significantly lower than those in NDR group.The differences were statistically significant(P<0.05).Relevant analysis showed that 25 (OH)D3 was significantly positively related to HDL-C,SOD and GSH in DR patients,and was significantly negatively related to disease course,FBG,and MDA.Multiple stepwise regression analysis showed that totally HDL-C (X1),SOD (X2),and MDA (X3)were included into the model.The regression equation was Y=15.434+0.261X 1 +0.078X2-0.121X3.Conclusion T2DM patients with retinopathy have the oxidative stress injury and their 25(OH)D3 declines,which may be involved in pathogenesis of DR by reciprocal causation.
7.Effects of sense and antisense T-STAR gene on colon cancer cell line HCT-116

Objective To investigate the effects of the sense and antisense testes-signal transduction and activator of RNA (T-STAR) gene on the colon cancer cell line HCT-116. Methods The sense and antisense T-STAR gene was stably transfected into HCT-116 cells with lipofectamine. The expression level of T-STAR in those cells was detected by Western blotting and the growth velocity and proliferation of those cells by cytokinetics. Results The growth velocity and proliferation decreased after transfection of the sense T-STAR gene, but increased after transfection of the antisense T-STAR gene. Conclusion T-STAR gene can inhibit the growth velocity and proliferation of HCT-116 cells.
8.Transfection of T-STAR antisense gene can depress the telomerase activity in lung adenocarcinoma cell line A549

Objective To investigate the changes of telomerase activity after knocking down endogenous expression of T-STAR (testes-signal transduction and activator of RNA) gene in human lung adenocarcinoma cell line A549 by antisense strategy. Methods The mRNA and protein expression of T-STAR gene were determined by RT-PCR and Western blotting, and the telomerase activity was measured by PCR-ELISA, after transfection of T-STAR antisense gene into A549 cells with lipofectamine. Sense pcDNA-STAR and blank pcDNA3.1 transfection served as control. Results The expression of T-STAR gene was significantly inhibited at mRNA and protein level, and the telomerase activity was significantly decreased. Conclusion The down-regulation of telomerase activity may result from inhibition of T-STAR gene expression in lung adenocarcinoma A549 cells.
9.Relationship between Fatty Liver and Blood Lipids in Children with Simple Obesity

Objective To study the relationship between fatty liver and blood lipids in children with simple obesity and the precaution of this disease.Methods Forty-eight cases from 2 to 16 years old with simple obesity were divided into 2 groups.The first group included 19 cases with fatty liver(38.78%);the second group included 29 cases without fatty liver(61.22%).
