1.Seasonal decomposition and ARIMA methods in prediction of tuberculosis incidence in Urumqi,China

Objective To compare the accuracy of the seasonal time series decomposition method and autoregressive integrated moving average (ARIMA) in the prediction of incidence of tuberculosis(TB) in order to facilitate early-warning.Methods The seasonal decomposition model and ARIMA model were constructed by SPSS20.0 software based on time series of monthly TB incidence between January 2005 and December 2014 in Urumqi,China.The obtained models were used to forecast the monthly incidence in 2015 and compared with the actual incidence respectively.Results Between 2005 and 2014,the incidence of TB was higher during March,April and May in Urumqi.A linear fitting model and a cubic curve fitting model were constructed by the time series seasonal decomposition method.The mean absolute percentage error (MAPE) of each predicted monthly incidence in 2015 was 18.75% and 92.25%,respectively.The predicted values of the linear model were lower than actual values and the predicted values of the cubic curve model were higher than actual values.An ARIMA (2,1,1) (1,1,0)12 fitting model was established by ARIMA method.The MAPE of each predicted monthly incidence in 2015 was 9.46% and there were no significant differences between the predicted and actual values.Conclusion The ARIMA method is better than the seasonal decomposition method for predicting the monthly incidence of TB in Urumqi.
2.Treatment of Refractory Rheumatoid Arthritis by Huayu Tongbi Recipe Combined Methotrexate.

OBJECTIVETo evaluate the clinical efficacy and safety of Huayu Tongbi Recipe (HTR) combined methotrexate (MTX) in treating refractory rheumatoid arthritis (RRA).
METHODSTotally 167 RRA patients were assigned to the treatment group (73 cases) and the control group (94 cases) according to different therapeutic methods. Patients in the treatment group were treated with HTR combined MTX, while those in the control group were treated with leflunomide (LEF) combined MTX. Clinical signs and symptoms, RF, CRP, ESR, disease activity score 28 (DAS28), and safety indicators were compared between the two groups before treatment, at week 12 and 24 after treatment. The efficacy and safety indices were also evaluated.
RESULTSAt week 12 after treatment the total effective rate was 82.2% (60/73 cases) in the treatment group and 79.8% (75/94 cases) in the control group, showing no statistical difference between the two groups (chi2 = 0.15, P > 0.05). At week 24 after treatment the total effective rate was 78.1% (57/73 cases) in the treatment group and 755% (71/94 cases) in the control group, showing no statistical difference between the two groups (chi2 = 0.15, P > 0.05). There was statistical difference in the total effective rate between week 24 and week 12 in the control group (chi2 = 0.49, P < 0.05). Clinical signs and symptoms, RF, CRP, ESR, and DAS28 were significantly improved in the two groups after 12- and 24-week treatment (P < 0.01). There was no statistical difference in the improvement at week 12 after treatment between the two groups (P > 0.05). There was statistical difference in time of morning stiffness, tender joint numbers, swollen joint numbers, patient global assessment, RF, CRP, and DAS28 at week 24 after treatment between the two groups (P < 0.05). Besides, adverse reactions occurred less in the treatment group than in the control group (P < 0.01).
CONCLUSIONThe efficacy of HTR combined MTX was equivalent to that of LEF (10 mg per day) combined MTX, but with more stable therapeutic effects and less adverse reactions.

A patient developed persistent parkinsonism after intravenously injecting a high dose of methamphetamine.
Magnetic resonance imaging revealed bilateral hypoxic/ischemic basal ganglia damage, which could have
been caused by the vasoconstrictive effect of methamphetamine. This case adds some circumstantial
evidence to the association between methamphetamine and Parkinsonism.

4.Biological activity of the virulence factor cagA of Helicobacter pylori.

BACKGROUNDChina is one of the countries with the highest incidence of H. pylori and more than 9090 isolates possessed the cagA gene. This study was to evaluate the biological activity of the H. pylori virulence factor cagA isolated from Chinese patients.
METHODScagA DNA fragments were amplified from the genomic DNA and subsequently cloned into the mammalian expression vector for cell transfection and DNA sequencing. cagA protein, phosphorylated-tyrosine cagA and the complex of cagA precipitated with SHP-2 were identified respectively by western blot in the crude cell lysate from conditionally immortalized gastric epithelial cells at 48 hours after transfection with cagA DNA. In addition, the ability of induction of scattering phenotype was examined after transient expression of cagA in AGS cells.
RESULTSThe C-terminal half of cagA contained only one repeated sequence and three tandem five-amino-acid motifs glutamic acid-proline-isoleucine-tyrosine-alanine (EPIYA). Moreover, the amino acid sequence of D2 region in repeated sequence was aspartic acid-phenylanaline-aspartic acid (D-F-D) which was significantly distinguished from the three repeated sequences and aspartic acid-aspartic adid-leucine (D-D-L) in the western standard strain NCTC11637. Western blot revealed that cagA became phosphorylated in tyrosine site and bound with SHP-2 after transient expression of cagA DNA in gastric epithelial cells. Transient expression of cagA in AGS cells showed that cagA was able to induce the elongation phenotype although to a lesser extent than western strains.
CONCLUSIONScagA perturbs cell signaling pathways by binding with SHP-2. However, significant difference exists in amino acid sequence and biological function of cagA in Chinese compared with those of western countries.

5.Predictive value of radiological parameter for re-displacement of conservative treatment of distal radius fracture.

OBJECTIVETo analyze the radiological parameters of the conservatively in treating distal radius fracture and investigate whether the final re-displacement can be predicted after primary reduction.
METHODSFrom January 2013 to June 2014,212 patients with distal radial fracture conservatively were treated in our hospital, 107 patients of them were excluded because of their incomplete radiological parameters;the remaining 105 patients were available for radiological were assessed after injury, there were 56 male patients and 49 female patients in this study, the average age of the patients was 51 years old (ranged from 22 to 80 years). According to AO classification, there were 47 cases of type A2 and C1, and 58 cases of type A3, C2, C3. All patients were treated by closed reduction and below-elbow cast immobilization for 4 to 6 weeks. All patients were followed up for 3 to 6 months (means 4.5 months) by X-ray, all fractures were healed. Standard AP and lateral radiographic examination was conducted before reduction and after reduction and bony consolidation,the dorsal angulation and the radial angle were measured at each time point. The linear regression was used for the analysis to find out whether the final re-displacement can be predicted after primary reduction.
RESULTSAmong 105 patients,the significant correlations were found for the dorsal angulation between the reduction time and the end time (r = 0.82) and for the radial angulation between the reduction time and end time (r = 0.85).
CONCLUSIONThe dorsal angulation and the radial angulation after complete healing can be predicted from linear the regression functions. Due to the possibility of predicting the end result, whether the fracture should receive further conservative treatment or surgical treatment can be decided immediately.

6.Molecular identification of heparin from pigs by allele-specific PCR (AS-PCR) and amplification refractory mutation system (ARMS).

The aim of this study is to develop a convenient and effective method for the identification of heparin from pigs (include Sus scrofa domestica Brisson and Sus scrofa riukiuanus). Based on sequences of D-loop region of pigs and the other animals, two pairs of highly specific primers were designed for distinguishing heparin of pigs from other animals. The primers were employed to amplify D-loop region of DNA templates extracted from pig and seven other animal species that amounted to 49 samples. AS-PCR (allele-specific PCR) and ARMS (amplification refractory mutation system) were all suitable for fast identification of heparin from pig with anneal temperature at 54-56 degrees C in AS-PCR and with wider anneal temperature in ARMS,at 52-58 degrees C. An about 170 bp DNA fragments were amplified from separately pigs and whereas no DNA fragment was amplified from other samples under the same reaction condition.

7.The influence of controlling redox potential on ethanol production by Saccharomyces cerevisiae.

Redox electrode was used to control redox potential at four different levels (-50 mV, -100 mV, -150 mV, - 230mV) for the study of ethanol fermentation. The result showed that there was notably influence on the yield of ethanol, the formation of glycerol, the secretion of organic acid, biomass and the death of cell by controlling redox potential at different levels. For example: the biomass of ORP at -50 mV was 1.26, 1.86, 2.59 times higher than ORP at -100 mV, -150 mV, -230 mV respectively, the final glycerol concentration was 1.2, 1.1, 1.7 times higher but final ethanol concentration was 0.87, 0.49, 0.51 times lower compared to the latest three ORP control level. And take biomass, ethanol yield, glycerol concentration, and unconsumed glucose into consider, we get the conclusion that it is very favorable for ethanol fermentation by control ORP at 150 mV. So it give us a apocalypse that we can use redox electrode to control the ethanol fermentation exactly on bioreactor scale.

8.Study on the influence of ROI size for SNR measurement during the test of MR quality control

Objective:To explore the influence of region of interest(ROI)size on signal-to-noise ratio(SNR)measurement during the test of magnetic resonance(MR)quality control(QC),so as to provide references for selecting ROI in conducting SNR measurement during MR QC test.Methods:According to the national health industry standard"Specification of image quality test and evaluation for medical magnetic resonance imaging(MRI)equipment"(WS/T 263-2006),this study utilized Magphan SMR170 performance test phantom(abbreviation:SMR170 phantom)to perform QC test on MR equipment after conducted three times tests.The slices of SNR measurement were selected on the obtained QC images,and 41 circular ROIs,which were incremental from 200mm2 to 4200mm2 as 100mm2,were sequentially chosen in the central region of the images on the SNR slices.The SNR was calculated according to formula,and then,the SNR curve that changed with the increasing of ROI size was formed.Finally,the influence of ROI size on SNR was obtained.Results:In conducting the QC test for MRI equipment by SMR170 phantom,the influence of the selected ROI size of central region of image on SNR was significant.The SNR fluctuation could not be really reflected when ROI>1000mm2 because the basically stable SNR leaded to the SNR change could not be timely found.When ROI<1000mm2,the signal means slowly increased with the increasing of ROI size,and the increase amplitude of noise was larger,and the SNR faster decreased and was smaller and smaller with ROI increase,and it tended towards stability at finally.Conclusion:When SMR170 phantom is used to conduct QC test for MR equipment and SNR is measured,it is recommended to select≤1000 mm2 of ROI size of the central region of the images,and to choose the same ROI for each measurement,which can really reflect the SNR change and ensure the result of each measurement has comparability.
9.Effect of berberine on microglia activation in rats with Alzheimer's disease

Objective To observe the effect ofberberine chloride on microglia activation and expression of peroxisome prolifcrator-activated receptor gamma(PPAR γ)in a rat model of Alzheimer's disease(AD).Methods Eighteen adult male Sprague-Dawley rats were randomized into normal control,β-amyloid 4o(Aβ40)and Aβ40+berberine chloride groups(n=6).Rat models of AD were established by injection of Aβ40(5 μg)into the bilateral hippocampus,and in Aβ40+berberine chloride group,berberine chloride(50 mg/kg)was given intragastrically once daily for 14 days.Immunohistochemistry,real-time polymerase chain reaction and Western blotting were used to detect the expressions of CD11b and peroxisome proliferator-activated receptor γ(PPAR γ)in the rats.Results The numbers of CD11b positive cells were 74.0±13.4,121.5±19.9 in the Aβ40 and Aβ40+berberine chloride groups,respectively,and the relative copy numbers of CD11b mRNA 4.08±2.43,5.52±1.83,which were significantly increased compared those with normal control group.The numbers of PPARγ positive cells were 42.5+5.6,31.7±8.7,the relative copy numbers of PPARγ mRNA 16.3±13.5,10.8±7.5,and the relative expression of PPARγ protein 0.18±0.08,0.09±0.05,which were significantly decreased compared those with normal control group(93.2±11.3,40.6±17.1,0.31±0.11).Berberine further increased CD11b expression and decreased PPARγ expression in the hippocampus.Conclusion Berberine can increase microglia activation in AD rats by inhibiting the expression of PPARγ.
10.Imaging anatomic characteristics of petrosal vein studied by Three-dimensional imaging technology of magnetic resonance

Objective To analyze the normal anatomy of petrosal vein and the space adjacent relationship between the petrosal vein and the homolateral trigeminal nerve by balanced turbo field echo (B-TFE) Cine-MR imaging and enhanced T1 high-resolution isotropic volume excitation (e-THRIVE)imaging.Methods Forty-one patients with facial spasm and epileptiform neuralgia were selected and taken a scan with ACHIEVA NOVA DUAL A-serial 1.5T MR machine using the BTFE and e-THRIVE series.The space adjacent relationship between the petrosal vein and the trigeminal nerve in cerebellopontine angle were observed.Results The 82 sides of petrosal veins and homolateral trigeminal nerves (41 cases) were displayed well.Petrosal veins were located in cavitas subarachnoidealis,partly in free state; the number of trunk ofpetrosal vein could be 1,2,3,respectively,responding to 70 (86%),10 (12%),2 (2%).The petrosal veins located in the dorsal-lateral of trigeminal nerve were found in 74 sides (37 cases,91%,by BTFE,e-THRIVE series),the petrosal veins located in the ventri-lateral of trigeminal nerve in 6 sides (3 cases,7%,by e-THRIVE series),and the petrosal veins located in the right upon trigeminal nerve in 2 sides (1 cases,2%,by BTFE series).Conclusion B-TFE MR imaging and e-THR/VE imaging,showing the petrosal veins and trigeminal nerves clearly and evaluating their relationship accurately,can provide information of topographic anatomy before microsurgical vascular decompression.
