BACKGROUNDThe 3q26 chromosome region, where the human telomerase RNA gene (hTERC) is located, is a biomarker for cervical cancer and precancerous lesions. The aim of this study was to confirm the value of measuring hTERC gene gain in predicting the progression of cervical intraepithelial neoplasia grade I or II (CIN-I and -II, respectively) to CIN-III and cervical cancer.
METHODSLiquid-based cytological samples from 54 patients with CIN-I or CIN-II lesions were enrolled in this study. Follow-up was performed with colposcopy and biopsy within 24 months after the diagnosis of CIN-I or CIN-II. Copy numbers of the hTERC gene were measured by fluorescence in situ hybridization with a dual-color probe mix containing the hTERC gene probe (labeled red) and the control, the chromosome 3 centromere-specific probe (labeled green).
RESULTSAll patients whose lesions progressed from CIN-I or CIN-II to CIN-III displayed a gain of the hTERC gene, whereas patients where the hTERC gene was not amplified did not subsequently progress to CIN-III or cervical cancer. The signal ratio pattern per cell was recorded as N:N (green:red). The numbers of cells with the signal ratio pattern of 4:4 or N:≥5 in patients whose lesions progressed to CIN-III were significantly higher than those whose lesions did not progress. Significantly, none of the patients with a 4:4 signal ratio pattern regressed spontaneously.
CONCLUSIONSIn conclusion, measurement of hTERC gene gain in CIN-I or CIN-II patients using liquid-based cytological samples could be a useful biomarker to predict the progression of such cervical lesions. In addition, a 4:4 or N:≥5 signal ratio pattern may indicate the unlikeness of spontaneous regression of CIN-I or CIN-II lesions.

3.Cloning and sequence analysis of UreB of Helicobacter pylori isolated from children.

OBJECTIVETo clone UreB gene of Helicobacter pylori (H. pylori) isolated from children to pGEX-4T-1 expression plasmid, and do sequence analysis.
METHODSA pair of specific primer was designed according to H. pylori UreB gene in the GenBank. Using H. pylori strains isolated from children as a template, a UreB gene was obtained by PCR. After EcoR I and Not I digestion, the PCR production was linked with pGEX-4T-1 which was digested with the same enzymes. The recombinant plasmid was transformed into E.coli BL21 and identified by double enzyme digestion and sequence analysis. The sequence results were compared with the gene sequence in the GenBank.
RESULTSA UreB gene was successfully amplified from children's H. pylori strain GZCH1. It was 1710 bp in size. The objective band was identified by double enzyme digestion. DNA sequence showed that UreB was in the correct open reading frame. The sequence comparison analysis showed that DNA and amino acid sequence identities of UreB gene with other strains were 98%. The sequence of UreB of H. pylori strain GZCH1 was submitted to GenBank (accession number:FJ455126).
CONCLUSIONSUreB of H. pylori strain GZCH1 is successfully cloned to pGEX-4T-1, which provides a basis for research of oral H. pylori vaccine.

4. Facial changes of Mongolian nationality from youth to old age in China

Objective To explore the law of changes in the appearance of Chinese Mongolians with age. Methods Totally 24 human indexes of head and face from 4410 cases (1916 males and 2494 females) and 13 Mongolian ethnic groups were measured, and 12 craniofacial indexes were calculated. Correlation analysis was used to study the changes of craniofacial indexes and indexes with age. The method of analysis of variance was used to study whether the differences between the index values and index values of the three age groups were statistically significant. Results With the increase of age, the distance between the two mandibular angles became wider, the mouth widened, the width, height and length of the nose increased, the skin of the upper lip increased, the length and width of the ear increased, the width of the head, forehead, face, mastoid and tragus narrowed, the width of the eyes narrowed, the upper and lower lips thinned, the circumference of the head and the height of the head decreased, and the length of the head did not change significantly. With the increase of age, the Mongolian face becomes narrower and longer, the proportion of the height of the face in the whole head high school increases, the nose becomes wider, and the head appears to be relatively longer, lower and wider. The distance between the left and right frontotemporal points is relatively closer, the mouth cleft appears more slender, and the ratio of length to width of the ear does not change much. Conclusion With the increase of age, the facial features of Mongolian have changed.
5. Body composition of Mongolian in Western China

Objective To explore the body composition characteristics of four ethnic groups in Western Mongolia. Methods Bioelectrical impedance analysis was used on the Western Mongolia four ethnic groups 18 indices of body composition. which include Qinghai Khoshut tribe(166 males, 195 females), Xinjiang Chahar tribe (217 males, 226 females), Xinjiang Torgouts tribe(112 males, 127 females), Alxa Khoshut tribe (89 males, 144 females).Using principal component analysis method to explore the characteristics of the Mongolian body composition in the west of China. Results The body composition data of four ethnic groups were obtained. The gender difference of most indicators was statistically significant. The fat rate of female trunk and limbs was higher, while the muscle mass of male was larger. Principal component analysis showed that compared with the southern ethnic group, the northern ethnic group had larger bone mass, more developed muscles and higher level of fat development. On the principal component scatter plot, the distance between the mongols in Western China and other ethnic groups was relatively far, indicating that the body composition of the mongols in Western China had unique characteristics. Conclusion Among the Chinese ethnic groups, the Western Mongolians have large bone mass, well developed muscles and high body fat.
7.Influence of Dexmedetomidine Applied in Combined Acupuncture-medication Anesthesia on Vomiting After Thyroidectomy

Objective To observe the influence of dexmedetomidine applied in combined acupuncture-medication anesthesia on vomiting after thyroidectomy. Method Seventy patients who were going to receive thyroidectomy were randomized into group A and B, with 35 cases in each group. Combined acupuncture-medication anesthesia [electroacupuncture at Hegu (LI4), Neiguan (PC6) and Futu (ST32) plus intravenous infusions of sufentanil citrate injection (0.3 mg/kg) 20 minutes before operation]was adopted in both groups. Group A was additionally intervened by constantvelocity micropump infusion of dexmedetomidine hydrochloride injection (0.5 mg/kg) within 10 minutes before operation and remaining 0.4 mg/kg/min during operation, while group B was intervened by infusion of normal saline 10 minutes before operation. The observer's assessment of awareness/sedation (OAA/S) scores at different time points[lying in bed for 5 minutes before operation (T0), before the beginning of the operation (T1), 30 minutes after operation (T2), 60 minutes after operation (T3) and at the end of operation (T4)] in the two groups were observed. The additional times and total dose of sufentanil during operation, the number of cases using esmolol and urapidil, and the number of vomiting cases occurred within 2 hours after operation and 2~24 hours after operation in the two groups were recorded.Result The additional times and total dose of sufentanil during operation in group A were significantly different from those in group B (P<0.01). The utilization rate of esmolol and urapidil were respectively 31.4% and 14.3% in group A versus 77.1% and 65.7% in group B, and the between-group differences were statistically significant (P<0.01). The incidence of vomiting within 2 hours after operation and 2~24 hours after operation were respectively 20.0% and 17.1%in group A versus 54.3% and 42.9% in group B, and the between-group differences were statistically significant (P<0.01). The OAA/S scores at different time points (T1, T2, T3 and T4) in group A were significantly different from those in group B (P<0.01). Conclusion Dexmedetomidine applied in combined acupuncture-medication anesthesia can reduce the additional times and total dose of sufentanil during thyroidectomy, and it can reduce the incidence of postoperative vomiting as well.
8.Imaging anatomic characteristics of petrosal vein studied by Three-dimensional imaging technology of magnetic resonance

Objective To analyze the normal anatomy of petrosal vein and the space adjacent relationship between the petrosal vein and the homolateral trigeminal nerve by balanced turbo field echo (B-TFE) Cine-MR imaging and enhanced T1 high-resolution isotropic volume excitation (e-THRIVE)imaging.Methods Forty-one patients with facial spasm and epileptiform neuralgia were selected and taken a scan with ACHIEVA NOVA DUAL A-serial 1.5T MR machine using the BTFE and e-THRIVE series.The space adjacent relationship between the petrosal vein and the trigeminal nerve in cerebellopontine angle were observed.Results The 82 sides of petrosal veins and homolateral trigeminal nerves (41 cases) were displayed well.Petrosal veins were located in cavitas subarachnoidealis,partly in free state; the number of trunk ofpetrosal vein could be 1,2,3,respectively,responding to 70 (86%),10 (12%),2 (2%).The petrosal veins located in the dorsal-lateral of trigeminal nerve were found in 74 sides (37 cases,91%,by BTFE,e-THRIVE series),the petrosal veins located in the ventri-lateral of trigeminal nerve in 6 sides (3 cases,7%,by e-THRIVE series),and the petrosal veins located in the right upon trigeminal nerve in 2 sides (1 cases,2%,by BTFE series).Conclusion B-TFE MR imaging and e-THR/VE imaging,showing the petrosal veins and trigeminal nerves clearly and evaluating their relationship accurately,can provide information of topographic anatomy before microsurgical vascular decompression.
9.Inhibitory effects of emodin on drug-resistant HL-60/ADR cell proliferation and its induction of apoptosis.

The study was aimed to investigate the effects of emodin on the proliferation and apoptosis of adriamycin-resistant HL-60/ADR cells, and to explore the underlying mechanism. The cell viability and colony formation were detected by MTT assay and colony formation assay respectively. Apoptotic cells were tested by means of cell cycle analysis, mitochondrial transmembrane potential levels, caspase-3 activity detection, Annexin V FITC/PI staining and TUNEL labeling. RT-PCR was used to analyze the bcl-2 and c-myc mRNA expressions. The protein expressions of Bcl-2, c-Myc and caspase-3 precursor were determined by Western blot. The results showed that HL-60/ADR cell growth was significantly inhibited by emodin in dose and time dependent manners. Cell colony formation obviously decreased with IC50 5.79 micromol/L. G0/G1 phase cell population increased while G2/M phase cells decreased in 40 and 80 micromol/L groups compared with control group (p < 0.01), and no significant difference of cell cycle was observed in 20 micromol/L group (p > 0.05). The typical hypo-diploid peak (apoptotic peak) appeared in each dose group. The levels of mitochondrial transmembrane potential of HL-60/ADR cells decreased and caspase-3 activity increased when incubated with emodin for 12 and 24 hours respectively. Apoptosis occurred in a dose-dependent manner, and its earlier and later stages were identified by Annexin-V FITC/PI staining and TUNEL labeling methods respectively. The expressions of bcl-2, c-myc mRNA and Bcl-2, c-Myc, caspase-3 precursor protein were all down-regulated in a time-dependent manner after treatment with emodin at different times. It is concluded that emodin efficiently inhibits growth and induces apoptosis on HL-60/ADR cells, which may be related with the down-regulation of mitochondrial transmembrane potential and expressions of bcl-2 and c-myc, as well as up-regulation of caspase-3 activity.

10.Fluorescence in situ hybridization in uncultured amniocytes for detection of aneuploidy in 4210 prenatal cases.

BACKGROUNDAlmost all reported fluorescence in situ hybridization (FISH) kits for prenatal diagnosis use probes from foreign (non-Chinese) countries. The aim of this study was to analyze the reliability of domestic (Chinese) FISH probe sets to detect aneuploidies of chromosomes 13, 18, 21, X, and Y related to prenatal diagnosis in 4210 cases.
METHODSCytogenetic karyotyping was carried out as a standard prenatal diagnostic test, and amniotic fluid cell interphase FISH analysis was performed using two sets of probes (centromeric probes for chromosomes 18, X, and Y, and locus-specific probes for chromosomes 13 and 21) provided by GP Medical Technologies, Beijing, China. Then we compared the two results and found the performance characteristics for informative FISH results of aneuploidies by the domestic kit probes.
RESULTSIn 4210 cases, 4126 cases generated karyotype results and 133 abnormal karyotypes (including 97 aneuploidies) were found. The FISH results of 98 cases (among them, 31 cases gave normal cytogenetic results) were uninformative. The rate of abnormal cases was 3.2% (133/4126). For the abnormal karyotypes, the rate of aneuploidy was 72.9% (97/133). Among the 97 aneuploidies, there were 58 cases of trisomy 21 (58/97, 59.8%), four cases of trisomy 13, 23 cases of trisomy 18, and 12 cases of sex chromosomal aneuploidies. The total concordance of the two methods was 97.9% (95/97; two cases were mosaics that had a low percentage of abnormal cells), and the concordance of trisomy 21, 13, and 18 by the two methods was 100%.
CONCLUSIONSThe two sets of the domestic FISH kit probes are reliable for prenatal diagnosis. The results demonstrate that FISH is a rapid and accurate clinical method for prenatal identification of chromosome aneuploidies.
