1.Relationship between the weight gain in the first and second trimesters and gestational glucose metabolism

Objective To study the difference of body weight gain in the first and second trimesters between pregnant women with abnormal gestational glucose metabolism and those with normal gestational glucose metabolism.Methods We retrospectively analyzed the data of pregnant women visited the obstetric clinic of Peking Union Medical College Hospital from January 2010 to May 2011,with or without abnormal glucose metabolism.The data of body weight gain in the first and second trimesters,body mass index (BMI) before pregnancy,BMI in the end of second trimester,age,obesity history,diabetes family history,insulin level in the oral glucose tolerance test (OGTT),and lipid profile of these women were collected.Results Altogether 216 pregnant women with abnormal glucose metabolism (case group) and 230 pregnant women with normal glucose metabolism (control group) were included in this study.In the case group,the increment of body weight in the first and second trimesters was 10.0 (8.1,12.5) kg,and the increment of BMI was (4.01 ± 1.43) kg/m2,BMI in the end of the second trimester was (25.84 ± 3.14) kg/m2;in comparison,the increments of body weight and BMI in the first and second trimesters were 9.5 (7.0,11.5) kg and (3.60 ± 1.43) kg/m2,respectively,BMI in the end of the second trimester was (25.1 ± 3.00) kg/m2 in the control group,all significantly lower than in the case group (all P < 0.05).The BMI before pregnancy was higher in the case group than in the control group [(21.84 ±2.99) kg/m2 vs.(21.50 ±2.82) kg/m2],but with no statistical significance (P>0.05).Compare with the control group,the case group had older age [(33.17 ±3.65) years vs.(31.68±3.36) years],higher fasting insulin level [11.2 (7.1,16.3) mU/Lvs.8.9 (6.5,13.7) mU/L],higher peak insulin level in OGTT [135.3 (99.7,208.0) mU/L vs.104.9 (75.6,144.4) mU/L],higher homeostasis model assessment for insulin resistance (HOMA-IR) [2.40 (1.49,3.47) vs.1.82 (1.28,2.71)],and triglyceride [2.62 (2.04,3.31) mmol/L vs.2.18 (1.81,2.81) mmol/L],all with significant differences (all P < 0.05).There were no significant differences in obesity history,family history of diabetes and history of menstrual disorder between the two groups (all P > 0.05).Conclusions The body weight gain is higher in the pregnant women with abnormal glucose metabolism than in those with normal glucose metabolism in the first and second trimesters.Large increment of body weight is likely to be a risk factor for abnormal gestational glucose metabolism.
2.The designing of will tendency scale for inpatients′ articipation in patients for patient safety and the research in its structure of the scale

Objective To design patient paticipatin in patient safety willness and behavior scale (PSWBS) and check its validity and explore structure model. Methods Based on the related literature at home and abroad , the theoretical model was developed on the basis of preliminary survey questionnaire and the formal scale. Preliminary survey by questionnaire was carried out on sample 1, through item analysis, exploratory factor analysis and confirmatory factor analysis , reliability analysis , correlation analysis , and binary content validity test scale evaluation. Results The scale was composed of four dimensions of 14 items , qualified for the theoraticl research of a scale in terms of its internal consistency reliability , content validity , structure validity associated with the school. Conclusion The scale is good at the reliability and validity , suitable for measuring the hospitalized patients′ participation in willingness to maintain their own medical treatment was safe , and mainly adopts propensity to maintain their own medical safety behavior.
3.Study on identification of four kinds of Gentianaceae Mongolian medicine Digeda with spectroscopy techniques.

To study the identification of Gentianaceae Mongolian medicine Digeda with spectroscopy techniques, near infrared spectroscopy and differential scanning calorimetry techniques were applied to study on the identification of 4 kinds of Gentianaceae Mongolian medicine Digeda, and characteristic spectrums obtained were systematically analyzed. In NIR study, the four species of Digeda exist some differences in 4 250-4 400 cm(-1) and 5 650-5 800 cm(-1) of one-dimensional spectra, and show significant differences in 4 100- 4 400 cm(-1), 4 401-4 900 cm(-1) and 5 400-5 800 cm(-1) of the second derivative spectra. DSC curves of them present distinct topological pattern, characteristic peak and peak temperature. Using near infrared spectroscopy and differential scanning calorimetry analysis can realize efficient and accurate identification of four kinds of Mongolian medicine Digeda, and provide scientific basis for the efficient and accurate identification of other Gentianaceae Mongolian medicine Digeda.

5.Characteristics of infrared radiation of meridians and acupoints.

Infrared radiation temperature and characteristics of infrared spectrum of meridians and acupoints are summarized in this article. The infrared radiant track along the running course of meridians might be a ubiquitous phenomenon of the vital activity. The high temperature bands below the meridian lines at the cuticular layer are often taken as the manifestation of the tract. It could be induced by various stimulations on acupoints with warm-needling as the best causative method. High temperature and low resistance are considered as 2 features of the acupoint zone. The adenosine-triphosphate energy metabolism of the acupoint zone is higher than the non-point zone, which indicates that essential physiological and pathological information are carried by infrared radiation temperature of acupoint and infrared spectrum of acupoint. However, it is realized that study on characteristics of infrared radiation only is far from enough to reveal the essence of meridians and acupoints. And much still remains to be done in strengthening basic studies of characteristics of infrared radiation, structure pattern of meridians and acupoints as well as volt-ampere characteristics.

7.Dual-sided reading versus single-sided reading:comparison of image quality and radiation dose between the two computed radiography systems

Objective To assess and compare the difference in image quality and exposure dose between single-sided reading image plate(IP)and dual-sided reading IP.Methods A contrast-detail phantom CDRAD 2.0 was exposed by single-sided and dual-sided reading IP with different mAs sets.The entrance surface doses were recorded for all images.Images were then presented to two radiologists on a high resolution monitor of diagnosis workstation.The image quality figure(IQF)was measured for each image. Statistical analysis was performed using Spearman's correlation test and Wilcoxon signed-rank test to compare the difference in image quality and exposure dose between single-sided IP and dual-sided reading IP. Results With different tube current dosage of 5.6,12.0,20.0,25.0,and 40.0 mAs,IQF values of single-sided reading IP were 47.95,37.68,34.31,28.61,and 24.65,respectively,while those of dual- sided reading IP were 38.83,29.81,29.65,25.16,and 21.43,respectively.The IQF difference between them showed statistical significance(P
8.The Influence of Several Chief Parameters in Electrospinning on the Morphology and the Fiber Diameter of PLGA Fiber Scaffolds

To prepare PLGA fiber scaffolds by electrospinning process and investigate the influence of preparation parameters on the structure of the scaffolds. With the compound of THF and DMF as the solvent, the PLGA fiber scaffolds with different surface morphology were fabricated via altering PLGA solution concentration, flowing rate and applied electric field strength. The morphology and diameter of the fibers were observed using a scanning electron microscope (SEM). The biocompatibility of cell-scaffold complex was also evaluated by seeding human dermal fibroblasts onto the PLGA fiber scaffolds, including cell adhesion and proliferation. The results show that the diameter of fibers and the bound of distributing increase with the increase in the concentration of PLGA solution. As the flowing rate increases, the diameter of fibers increases. However, with the increase in the applied electric field strength, no significant difference in the diameter of the fiber can be observed. Furthermore, both the increase in the concentration of PLGA solution and applied electric field strength in the volume range of current investigation can lead to the reduction in the beads formation within the scaffold. The results of in vitro cell culture on the PLGA scaffolds also confirm that the PLGA fiber can support the adhesion and proliferation of huaman dermal fibroblasts.
9.Effects of Cdc20 mutation on growth of mouse embryonic fibroblast

Objective Investigation of biological characteristics of Cdc 20AAA/+APCmin/+ mouse embryonic fibroblast(MEFs) indicate the effect of Cdc20AAA/+on growth of mouse embryonic fibroblast and the possible mechanism . Methods MEFs of Cdc20AAA/+APCmin/+, Cdc20AAA/+, APCmin/+ and WT genotype were harvested from embryos for analysis.The growth characteristics of Cdc20AAA/+APCmin/+, Cdc20AAA/+,APCmin/+and WT mouse embryonic fibroblast were analyzed through growth curve analysis and foci formation assay .Separation of sister chromatid and the presence of aneuploid were detected by karyotype analysis .Results Cell proliferation assays showed that Cdc 20AAA/+APCmin/+cells grew at an accelerated rate compared with APC min/+MEFs(P<0.01).Foci formation assay showed that the clone forming ability was significantly increased .Cdc20AAA/+APCmin/+MEFs showed a significant increase in the frequency of aneuploid compared with WT MEFs , which had a karyotype of 38 and contained prematurely separated sister chromatids .Conclusion Cdc20 carrying a null allele (Cdc20AAA/+) may accelerate the growth and proliferation of APC min/+MEFs and present the growth characteristics of the tumor cells .The possible mechanism may be associated with chromosome instability .
10.Antitumor efficacy of irinotecan-loaded galactosyl modified lipid bilayer-coated mesoporous silica nanoparticles against hepatocellular carcinoma cells.

The purpose of this study is to prepare galactosyl modified lipid bilayer-coated mesoporous silica nanoparticles (GPEM) to enhance the antitumor efficacy against hepatocellular carcinoma cells. The irinotecan (CPT-11) loaded mesoporous silica nanoparticles (MSNs) was coated with the Gal-P123 modified functional lipid bilayer by thin-film dispersion method. Nanoparticles were characterized with particle size, zeta potential, morphology and drug release in vitro. Afterwards, the cell uptake, intracellular concentration of CPT-11, cell apoptosis rate and cytotoxicity were evaluated on human hepatocellular carcinoma cell line Huh-7. The results showed that MSNs were coated with intact lipid bilayers and the nanoparticles had clear core-shell structure. GPEM is stable with the mean particle size of (78.01 +/- 2.04) nm. The low leakage rate in normal physiological conditions in vitro is contributed to the protection of stable lipid bilayer, and the fast drug release in acid environment due to the destruction of the lipid bilayer. On the cell level, the vector could improve the intracellular CPT-11 concentration by 4 times because of the functional lipid bilayer. The high CPT-11 concentration led to the increasement of apoptosis rate by 48.6%, and the reduction of half maximal inhibitory concentration (IC50) values of CPT-11 by 2 times, indicating stronger cell cytotoxicity.
