Objective To inquire clinical effects of hysteroscopic electroresection in the treatment of submucous hysteromyoma. Methods A retrospective analysis regarding curative effects of hysteroscopic electroresection in 72 cases of submucous hysteromyoma was carried out. Results The resected myoma was 1.5~7 cm in diameter, and the number of resected myoma was 1~3. The operation time was 10~90 min (mean, 30 min). The operation was accomplished on one session in all the 72 cases. No uterine perforation, bleeding and bowel injuries took place. Postoperative follow-up for 4~20 months in the 72 cases reported a satisfaction rate of 97 2% (70/72). Out of 6 patients who had pregnancy desire, 2 were pregnant with child and 1 of them had a delivery. Conclusions Hysteroscopic electroresection in the treatment of submucous hysteromyoma is safe and effective. It can markedly improve the menstruation and elevate the pregnancy rate.

Objective To investigate whether transforming growth factor-β1 (TGF-β1) could induce the expression of twist protein in the human kidney tubular epithelial cells (HKCs).Methods Human proximal HKCs were cultured in vitro and divided into three groups as follows:control group,TGF-β1group (10 ng/ml),and TGF-β1 + wortmannin group (10 ng/ml,10nmol/l,respectively).After cultured for 48 hours, cell immunofluorescene was used to observe the expression of E-cadherin and alpha-smooth muscle actin (α-SMA).Meanwhile,Western blotting was used to detect the protein expression of E-cadherin,α-SMA,and twist. Results Compared to control group, the protein expressions of E-cadherin were significantly decreased in T group (3.54 ± 0.17 vs 16.06 ± 0.50,P =0.001),whereas the protein expressions of α-SMA and twist were significantly increased in T group (α-SMA:14.78 ± 0.48 vs 3.75 ± 0.50,P=0.001 ;twist:14.24 ±0.14 vs 3.06 ±0.15,P =0.001).Compared to T group,a significant increase of the E-cadherin protein expression was detected in TGF-β1 + wortmannin group (15.88 ± 0.36,3.46 ±0.19,P =0.001),whereas,the protein expressions of α-SMA and twist were significantly decreased in TGF-β1 + wortmannin group (α-SMA:3.50 ±0.39 vs 15.0 ±0.24,P =0.001 ;twist:3.09 ±0.1 vs 14.04± 0.16,P =0.001).Cell imunofluorescence showed that the expressions of E-cadherin and α-SMA were consistent with the results of Western blotting. Conclusions TGF-β1 can induce the expression of twist protein in HKCs through Akt pathways.

Objective To study the effects of motor imagery therapy on functional performance of upper ex- tremity in stroke patients. Methods Sixty cases of stroke were divided into a treatment group(n=30) and a con- trol group(n=30).The control group was treated with routine rehabilitation training, while the treatment group was treated with motor imagery therapy in addition to the routine rehabilitation training. The Fugl-Meyer Assessment (FMA),upper extremity function test(UEFT)and simple test for evaluating hand function(STEF)were used to as- sess the upper extremity function before and after six months of treatment. Results After treatment, patients in both groups scored significantly better with the FMA scales,UEFT scales and STEF scales when compared to their scores obtained before the treatment(P

Basic & Clinical Medicine 2006;0(11):-
Pyruvate dehydrogenase kinases(PDKs 1-4) can regulate the activity of mitochondrial pyruvate dehydrogenase complex(PDC) to catalyse the oxidative decarboxylation of pyruvate, and then to link glycolysis to the tricarboxylic acid cycle and ATP production. In this review,we summarize up-to-date information of mechanisms regulating PDKs and the function of PDKs inhibitors in lowering blood glucose level, alleviate damage during heart ischemia and also triggering apoptosis in cancer cells.PDKs will be a possible pharmacological targets in diabetes, heart ischemia and cancer therapy.

Overlapping PCR technology was employed to splice IFN?and HSA genes in vitro. The spliced gene was inserted into Pichia pastoris secretory vector pPIC9K. The IFN?-HSA gene was designed for secretory expression under the control of promoter AOX1 and Mat a signal peptide in pPIC9K. The recombinant plasmid pPIC9K/IFN?-HSA was linearized by restriction enzyme SalI and transformed into Pichia pastoris KM71 by electroporation. The recombinant strains identified by G418 selection and confirmed by PCR analysis were induced by methanol to express fusion protein IFNp-HSA. SDS-PAGE and Western blot analysis of the fusion protein showed that the expressed fusion protein IFNp-HSA with an apparent 90kDa molecular weight had the antigenicity of HSA. The specific activity of culture supernatant was about 640IU/ml assayed by the standard amiviral activity test on WISH cells challenged with VSV virus.

AIM:To clarify if interferon-?(IFN-?), tumor necrosis factor-?(TNF-?)and interleukin-1?(IL-1?)can induce apoptosis of human airway smooth muscle cells (ASMCs) in vitro. METHODS: Human ASMCs were isolated and cultured in DMEM containing 10% fetal bovine serum. Passage 4-6 cell was used in the experiment. IFN-?,TNF-? and IL-1?, were used separately or together in the treatment of human ASMCs. The effects of IFN-?,TNF-? and IL-1? on the growth of the cells was detected by MTT method at the hour 0,24,48 and 72. Light microscopy and electron microscopy were used to examine the morphological change. DNA fragmentation was analyzed by agarose gel electrophoresis. SP immunohistological staing method was performed to detect the change of expressions of p53 , bcl-2 and bax gene. The apoptosis cell percentage were detected by in situ end labeling technique (TUNEL)of fragmental DNA. RESULTS: (1)IFN-? or IFN-? together with TNF-? and IL-1? decreased the number of viable cells in a time dependent manner. (2) Light and electron microscopic examination showed cell shrinkage, membrane blebbing, nuclear contraction, chromatin condensation and nuclear fragmentation in human ASMCs. (3) Agarose gel electrophoresis showed a characteristic"ladder"of DNA bands representing integer multiples of the internucleosomal fragments (about 180-200 bp) in cytokine cotreated human ASMCs. (4)The expression of p53 and bax gene in cytokine cotreated group was significantly higher than in control group, but the expression of bcl-2 gene was lower than in control group. (5)Stimultaneous treatment with IFN-?(4?10 5 U/L),TNF-?(4?10 5 U/L)and /or IL-1? (10?10 4 U/L) induced apoptosis of human ASMCs. Apoptotic index of human ASMCs in cytokine co-treated group was significantly higher than in control group ( P
