1.Staurospolin promotes the apoptosis of multidrug resistance tumor cell lines induced by adriamycin

Objective To investigate whether the protein kinase C inhibitor can promote the apopto- sis of multidrug resistance tumor cell lines which are induced by chemotherapy drugs.Methods Choose the KB/S(oral squamous cancer cell line)and KB/VCR(its multidrug resistant cell line)to compare the Adri- amycin-induced apoptosis with or without staurospolin(protein kinase C inhibitor).The apoptosis is stained with acridine orange,tested by flow cytometry,and approved by electron microscope.Results 36 hours after the treatment with 0.04 ?g/ml adriamycin,apoptotic cells of KB/S are 96.68%,and after 48 hours,the apop- totic cells of KB/VCR are 64.99%.When the concentration of adriamycin are augmented to 0.4?g/ml and 2.0?g/ml,the apoptotic cells of KB/VCR are 69.74% and 37.18% respectively.When treated with stau- rospolin together,the apoptotic cells of KB/VCR increased to 72.58%(?~2=4.5,P0.05)respectively.These results were testified by electron microscope and acridine orange-stain.Conclu- sion The resistance to apoptosis may be one of the mechanisms of multidrug resistance and the protein ki- nase C inhibitor may reverse this resistance by promoting the apoptosis of multidrug resistance tumor cells.
2.Construction and application of a multiplex PCR system for genotyping of human red blood cell antigens.

OBJECTIVEA reliable method for genotyping blood group antigens Dib, k, Jsb1910 and Jsb2019 was developed. Through screening for rare blood types, the National Rare Blood Bank of China may be enriched.
METHODSThe controls for allele detection of blood groups Dib, k, Jsb1910 and Jsb2019 were prepared via polymerase chain reaction (PCR)-mediated gene site-directed mutagenesis (SDM) technique. Sequence-specific primers were designed according to known single nucleotide polymorphism (SNP) sites of alleles of blood groups antigens Dib, k, Jsb1910 and Jsb2019, a multiplex PCR system was developed by optimizing PCR reaction system. And 4190 random healthy donors samples were screened for the blood group antigens.
RESULTSUsing SDM technique, controls for alleles in blood group Dib, k, Jsb1910 and Jsb2019 were successfully generated. And a multiplex PCR system for genotyping above blood groups was developed. After verification, the system has performed with good stability and reproducibility. Two Di (b-) samples have been discovered from 4190 samples, no k- and Js(b-) sample was found.
CONCLUSIONMultiplex PCR features rapid detection, high throughput and low cost, and can be used for screening for donors of rare blood types. Information of donors may be registered in a database, which in turn can help those with rare blood types or require long-term blood transfusion to obtain matched blood, thereby reduce the adverse reactions of blood transfusion.

Aquaporin 5(AQP5),as the main water transport protein in the body,can regulate lung diseases by regulating airway mucus secretion,pulmonary inflammation,and lung function.Programmed cell death(PCD)plays a crucial role in chronic obstructive pulmonary disease(COPD).AQP5 may affect the development of COPD by regulating PCDs.This article reviews the molecular regulatory mechanism of AQP5 on apoptosis,autophagy,iron death and pyroptosis in PCDs in recent years,and further discusses its effect on COPD in order to provide theoretical support for clinical prevention and treatment of COPD.
4.Effects of long-term microwave irradiation on NMDAR, BDNF and related molecular expression in their signal pathways in rat hippocampus

Objective To evaluate the effect of long-term microwave radiation on the expression of N-methyl-D-aspartate receptor(NMDAR),brain derived neurotrophic factor(BDNF) and related molecules in signal pathways in the hippocampus of rats.Methods Fifty male Wistar rats were exposed to microwave radiation at an average power density of 0,5,10,20 and 30 mW/cm2for 6 min/time,3 times/week,and for 6 weeks,which were sacrificed and the hippocampus was quickly removed at 14 d and 28 d after exposure.The changes in NMDAR (NR1,NR2A,NR2B),postsynaptic density protein(PSD)-95,cortactin,BDNF and tyrosine kinase receptor B (TrkB) in hippocampal neurons of each group were detected by Western blotting and image analysis techniques.Results Compared with the control group,the expressions of related proteins did not change significantly after microwave irradiation of 5 mW/cm2 at each time point.After 20 mW/cm2 microwave radiation,the expression of NR1 was increased at 14 and 28 d (P <0.05),the expression of NR2A was increased at 28 d (P < 0.05),but the expression of NR2B was decreased at 14 and 28 d (P < 0.05).At a average power density of 30 mW/cm2,the expressions of NR1,NR2A and PSD-95 and the expression of NR2B were decreased at 14 and 28 d(P <0.05),and cortactin,BDNF and TrkB were increased at 14 d after irradiation (P < 0.05).Conclusion The effect of different dosages of long-term microwave radiation on the proteins of NMDAR and its signal pathway related molecules is different.Microwave radiation may affect the NMDAR of postsynaptic information transmission through the BDNF-TrkB signaling pathways,which might play an important role in the impediment of learning and memory function caused by microwave radiation.
5.Near Infrared Spectral Analysis Based on Data Augmentation Strategy and Convolutional Neural Network

Near infrared spectroscopy(NIRS)technology combined with chemometrics algorithms has been widely used in quantitative and qualitative analysis of food and medicine.However,traditional chemometrics methods,especially linear classification methods,often yield unsatisfactory results when addressing multi-class classification problems.Convolutional neural network(CNN)is adept at extracting deep-level features from data and suitable for handling non-linear relationships.The modeling performance of CNN depends on the size and diversity of sample,while the collection and preprocessing of NIRS sample data is often time-consuming and labor-intensive.This study proposed a NIRS qualitative analysis method based on data augmentation strategies and CNN.The data augmentation strategy included two steps.Firstly,applying Bootstrap resampling and generative adversarial network(GAN)methods to augment three NIRS datasets(Medicine,coffee and grape).Secondly,combining the original samples(Y)with the Bootstrap augmented samples(B)and GAN augmented samples(G)to obtain three augmented datasets(Y-B,Y-G and Y-B-G).Based on this,a CNN model structure suitable for these datasets was designed,consisting of 2 one-dimensional convolutional layers,1 max-pooling layer,and 1 fully connected layer.The results showed that compared to the optimal models of partial least squares discriminant analysis(PLS-DA),support vector machine(SVM),and back propagation neural network(BP),the CNN model based on Y-B dataset achieved average accuracy improvements of 3.998%,9.364%,and 4.689%for medicine(Binary classification);the CNN model based on the Y-B-G dataset achieved average accuracy improvements of 6.001%,2.004%,and 7.523%for coffee(7-class classification);and the CNN model based on the Y-B dataset achieved average accuracy improvements of 33.408%,51.994%,and 34.378%for grapes(20-class classification).It was evident that the models established based on data augmentation strategies and CNN demonstrated better classification accuracy and generalization performance with different datasets and classification categories.
6.Comparative study on infrared radiation spectrum of yuan point and Xiahe point of the large intestine channel in the patient of ulcerative colitis.

OBJECTIVETo probe the diagnostic value of the infrared radiation spectrum of acupoint for ulcerative colitis (UC).
METHODSA high sensitivity PHE 201 infrared spectrum instrument was used to determine the infrared radiation spectrum of Hegu (LI 4) and Shangjuxu (ST 37) in 34 cases of UC.
RESULTSOf 59 waves detected, there were significant differences in infrared radiation intensity of 28 different waves between the healthy people and the patients with UC in right Hegu (LI 4) (P < 0.05 or P < 0.01) and 13 waves in left Hegu (LI 4) (P < 0.05); there were significant differences in 16 different waves in right Shangjuxu (ST 37) (P < 0.05 or P < 0.01) and in 17 waves in left Shangjuxu (ST 37) (P < 0.05 or P < 0.01); there was a significant difference in 18 waves between right and left Hegu (LI 4) of the patients (P < 0.05 or P < 0.01) and 7 waves between right and left Hegu (LI 4) of the healthy people (P < 0.05). There was a significant difference in 4 waves between right and left Shangjuxu (ST 37) of the patients and one wave between right and left Shangjuxu (ST 37) of the healthy people (P < 0.01).
CONCLUSIONBoth Hegu (LI 4) and Shangjuxu (ST 37) show changes of infrared radiation spectrum when the intestine gets lesion, and Hegu (LI 4) can better show the change.

7.Association of HLA-DPB1 alleles with chronic myelogenous leukemia in southern Chinese Hans.

To clarify the association between HLA-DPB1 alleles and chronic myelogenous leukemia (CML) in South Chinese, the allelic types of HLA-DPB1 were detected by sequence based typing (SBT) in 86 patients with CML and 82 healthy individuals from Southern China. The results showed that the frequencies of HLA-DPB1 * 1301 and DPB1 * 20011 were higher in patients with CML in comparison with those of healthy individuals. It is concluded that positive association may exist between certain HLA-DPB1 alleles and CML.

8.Therapeutic effect of PCI combined tirofiban hydrochloride on aged ASTEMI patients and its influence on cardiac function and myocardial microcirculation indexes

Objective:To explore therapeutic effect of percutaneous coronary intervention(PCI)combined tirofiban hydrochloride on aged patients with acute ST elevation myocardial infarction(ASTEMI)and its influence on cardiac function and myocardial microcirculation.Methods:A total of 90 aged ASTEMI patients undergoing PCI in our hospital from Jul 2013 to Mar 2016 were selected.According to hospitalization order,they were randomly and e-qually divided into pure PCI group and combined treatment group(received tirofiban based on pure PCI group), both groups were treated for one month.ST segment regression degree,corrected TIMI frame count(CTFC)and TIMI flow grade after PCI,contrast agent acoustic peak intensity in myocardial microcirculation(PI)and serum CK-MB peak concentration before and after PCI,LVEF,LVEDd and LVEDV before and six months after PCI, and incidence rate of adverse events were measured and compared between two groups.Results:Compared with pure PCI group after PCI,there were significant rise in ST segment regression degree[(43.8 ± 7.1)% vs.(66.2 ± 8.2)%],TIMI flow grade[(2.1 ± 0.5)grade vs.(2.9 ± 0.6)grade]and PI[(7.1 ± 1.1)vs.(8.6 ± 1.2)],and sig-nificant reductions in CTFC[(27.3 ± 8.0)frame vs.(18.9 ± 6.6)frame],and serum CK-MB peak level[(296.5 ± 58.1)U/L vs.(199.3 ± 32.4)U/L]in combined treatment group,P= 0.001 all.Compared with pure PCI group on six months after PCI,there was significant rise in LVEF[(54.2 ± 8.3)% vs.(61.1 ± 8.0)%],and signifi-cant reductions in LVEDd[(48.1 ± 7.7)mm vs.(41.3 ± 8.1)mm]and LVEDV[(85.4 ± 10.6)mm3vs.(80.2 ± 10.4)mm3]in combined treatment group,P<0.05 or <0.01.Total incidence rate of adverse events of com-bined treatment group was significantly lower than that of pure PCI group(8.89% vs.26.67%),P=0.001.Con-clusion:PCI combined tirofiban hydrochloride can significantly improve myocardial microcirculation and cardiac function with low incidence rate of cardiovascular adverse events.The mechanism may be related to improving effect of tirofiban hydrochloride on myocardial microcirculation.
9.Stature estimation of teenagers by limb long bones with computerized radiography.

OBJECTIVE:
Relative parameters of upper limb bones, tibia and fibula were measured with computed radiography and used to establish the mathematical models for stature estimation of teenagers (from 14 to 18 years old) of Han population in Sichuan Province.
METHODS:
The upper limb bones, tibia and fibula of 194 subjects were taken computerized radiography on normal position and were measured the lengths between relative landmarks. The body height of each subject was recorded. Linear regression equations for stature estimation between body height and the lengths of upper limb bones, tibia and fibula were established.
RESULTS:
Forty-two single linear regression equations and 4 multiple regression equations were obtained. The coefficients of correlation(r) were 0.689-0.917 and the standard errors of estimate(SE) were between 3.075 and 5.485 cm. All of the equations were statistically tested and diagnosed with good applicability.
CONCLUSION
These equations could be used to estimate the body height of Sichuan Han population aged from 14 to 18. The lengths of the upper limb bones, tibia and fibula measured on the CR films could be useful to stature estimation of the adolescence and the forensic personal identification.

10.Clinical trial of recombinant human interferon α1b injection combined with vidarabine injection in the treatment of hand foot and mouth disease

Objective To observe the clinical efficacy and safety of re-combinant human interferon α1b injection combined with vidarabine injec-tion in the treatment of hand foot and mouth disease (HFMD).Methods A total of 94 children with HFMD were randomly divided into control and treatment groups with 47 cases per group.Control group received re-combinant human interferon α1b injection 6-20 μg· d-1, qd, intramus-cular injection.Treatment group was given vidarabine injection 5 -10 mg· kg-1· d-1, qd, intravenous drip, on the basis of control group . Two groups were treated for 5 days.The clinical efficacy , levels of serum immunoglobulin M (IgM), IgG, IgA, creatine kinase(CK), creatine ki-nase isoenzyme ( CK-MB), lactate dehydrogenase ( LDH) and lactate dehydrogenase isoenzyme (LDH-1), and adverse drug reactions were compared between two groups .Results After treatment, the total effective rates of treatment and control groups were 97.87%(46 cases/47 cases)and 72.34%(34 cases/47 cases) with significant difference (P <0.05).After treatment, the main indexes in treatment and control groups were compared : IgM were ( 1.44 ±0.19 ) and ( 1.27 ±0.16 ) g· L-1, IgG were ( 9.08 ±1.28 ) and (8.47 ±1.17 ) g· L-1, IgA were ( 1.09 ±0.14 ) and ( 0.91 ±0.10 ) g· L-1, CK were ( 72.27 ±10.06 ) and (97.25 ±12.63 ) U· L-1, CK -MB were ( 19.94 ±2.66 ) and ( 32.14 ±4.17 ) U· L-1, LDH were (207.48 ±28.13) and (313.26 ±42.27)U· L-1, LDH-1 were (53.39 ±7.17) and (78.27 ±10.81)U· L-1, the differences were statistically significant ( P<0.05 or P<0.01 ) .The adverse drug reactions of treatment group were leukopenia, skin pruritus and rash during the infusion of adenosine monophosphate , while those in the control group were leucopenia.The total incidences of adverse drug reactions in treatment and control groups were 4.25%and 2.13%without significant difference ( P >0.05 ) . Conclusion Recombinant human interferon α1b injection combined with vidarabine injection has a definitive clinical efficacy in the treatment of HFMD , without increasing the incidence of adverse drug reactions .
