1.Genetic Transformation of Phytoene Synthase Gene into Ginseng Callus Cells

Phytoene synthase is a rate-limiting enzyme in the pathway of carotenoid biosynthesis.To aim at establishing a transformation of Ginseng callus cells, elevating the nutritive value through encouraging the composition of corresponding carotenoid,taking Ginseng callus cells as acceptor,psy gene were transformed into cells via Agrobacterium-mediated. The factors affecting genetict ransformation were also investigated seperately from concentrations of A.tumefaciens, age of Ginseng callus cells, infection time and co-culture time. PCR,PCR-Southern and RT-PCR analysis from the transferred gene plants proved that psy gene has been transferred into Ginseng callus cells and can be expressed. The content of ?-carotene was increased by an average of 26 times.A base of improving and raising the contents of carotenoid in the Ginseng callus cell was established.
2.The relationship between self-esteem and childhood abuse, life events among violent male adolescents

Objective To study the relationship between self-esteem and childhood abuse, life events among male violent adolescent. Methods According to the modified overt aggression scale, juvenile delinquents were divided into violent group (n = 128 )and nonviolent group (n = 118 ). All the participants were investigated by Self-Esteem Scale (SES), Childhood Trauma Questionnaire-28 Item Short Form (CTQ-SF) and Adolescent SelfRating Life Events (ASLEC) and conducted correlation and regression analysis. Results ①Violent group had lower self-esteem scores ( 22.73 ± 3.30 ) than the nonviolent group ( 23.81 ± 3.30, P < 0.05 ). Violent group had higher scores in physical abuse, sexual abuse, total life events, interpersonal relationship, punishment, and other (8.30 ±4.07, 7.23 ±2.26, 54.48 ±18.60, 10.09 ±3.84, 14.43 ±5.87, 4.93 ±3.15, 9.93 ±3.64), compared to the nonviolent group (7.27 ± 3.27, 6.60 ± 2.09, 45.40 ± 18.45, 8.42 ± 4.13, 11.07 ± 5.75, 3.66 ±2.81, 7.84 ±3.66, P<0. 05). ②The self-esteem was significantly negative correlated with the childhood abuse and life events( r= -0. 143 ~ -0. 358, P< 0.01 ) among violent male adolescents. ③The physical abuse and punishment had significant prediction function of 15.6% on self-esteem. Conclusions Violent adolescents have low self-esteem, which is closely related to childhood abuse and adolescent life events.
3.Inhibitory effect of eicosapentaenoic acid on proliferation of human vascular endothelial cells induced by vascular endothelial growth factor

Background Eicosapentaenoic acid(EPA)function as the critical lipid mediators involved in several biological events in human body and play important role in suppressing the genesis of vascular endothelial growth factor (VEGF),migration and proliferation of vascular endothelial cells.Many ocular diseases were proved to be associated with neovascularization.Objecfive The purpose of this study was to investigate the inhibitory effect of EPA on the proliferation of human umbilical vein endothelial cells (HUVEC) indueed by VEGF. Methods HUVEC strain was cultured and passaged,and difierent concentrations of EPA were added to the medium with and without VEGF.The cultured cells were identified by antiofactor Ⅷ polyclonal antibody.The suppressing role of different concentrations of EPA on the proliferation of VEGF-induced or-uninduced HUVEC was assessed by MTT method.The influence of difierent concentrations of EPA on the cellular cycle of VEGF-induced HUVEC was assayed using flow eytometry.The expression of Flk-1,a receptor of VEGF,in the HUVEC Was detected by immunohistochemistry. Results Cultured HUVEC showed the ftlsiform in shape and presented with the cobblestone-like arrangement with the positive response for Ⅷ factor-related antigen.Various concentrations of EPA showed obviously inhibitory effect on VEGF-induced or-unindueed HUVEC at a dose-dependent manner (F=23.072.P=0.000).The inhibitory ability of EPA on VEGF-induced HUVEC was stronger than VEGF-uninduced HUVEC(F=41.417,P=0.000).In 24,48 and 72 hours,the action of EPA on the proliferation of HUVEC was gradually enhanced with the prolong of time(F=1.495,P=0.236).Cell cycle analysis indicated that EPA arrested VEGF-induced HUVEC in G0/G1 phase.The ratio of HUVEC in G0/G1 phase in EPA group was(75.83±1.56)%,and that in control groups was(68.62±1.44)%,showing a significant difference between them(t=-5.88,P=0.00),and no apoptosis of HUVEC was found in both groups.Flk-1 was strongly expressed in the cellular nucleus and cytoplasm in control group.However,the positive expressing intensity of Flk-1 in the HUVEC weakened,and the positive cell number was evidently less in EPA group. Conclusion EPA can inhibit the proliferation of VEGF induced HUVEC through arresting the synthesis of DNA of HUVEC and downregulate the expression of Flk-1 in HUVEC.These results suggest that EPA might exert an antiangiogenic effect.
4.Effects of insulin-like growth factor binding protein 4 on morphology and function of chick extraocular muscle

Background Researches have showed that the extraocular muscle injection of insulin-like growth factor(IGF) results in the thickness of extraocular muscle and enhancement of contractility,and lack of IGF probably is associated with pathogenesis of strabismus.If extraocular muscle injection of insulin-like growth factor binding protein 4(IGFBP-4),an inhibitor of IGF,can further ensure the effect of IGF is still under-investigation.Objective The aim of this study was to evaluate the effects of IGFBP-4 on the developing extraocular muscle.Methods IGFBP-4 of 2 μl(1 g/L)was injected into superior rectus muscles via supraorbital margin in the left eyes of 24 postnatal l-day-old chickens once every other day for five times,and the equivalent amount of normal saline solution was used in the same way in the fellow eyes as controls.The animals were sacrificed by overdose of anesthesia 2 days after the final injection and 12 animals were randomly selected for the maximal contractility test of superior rectus muscle.The other 12 pieces of superior rectus muscle of the chickens were obtained for the hematoxylin & eosin staining to evaluate the morphology of superior rectus muscle and calculate the myofiber diameter using imaging system.Results The maximal contractility was (1.602 ± 0.080) mN and(1.815±0.O10)mN in the IGFBP-4 injection group and normal saline solution group respectivily,showing a significant difference between the two groups(t =13.65,P<0.01).The maximal contraction force in cross-sectional area was (95.500 ± 7.420) mN/cm2 in the IGFBP-4 injection group and it was significantly lower than that of the normal saline solution group(101.510±5.220)mN/cm2(t =28.81,P<0.01).Histological examination showed that thin myofibers were obvious more in the IGFBP-4 injection group than those with saline injection.The mean diameter was (8.7 ± 0.5) μm in the IGFBP-4 injected group,and that of the normal saline group was(9.1 ±0.4) μm,there was no significant difference between them (t =0.75,P =0.46).However,significant differences were found in the number of different diameters of myofibers between the two groups(all at P<0.05).Conclusions IGF is one of the key nutrition factors in the development of extraocular musele.The extraocular muscle injection of IGFBP-4 may cause the dysfunction and morphologic abnormality of extraocular muscle.
5.Relationship between coping styles of patients with depression and childhood abuse

Objective To explore the relationship between coping styles and childhood abuse in adult patients with depression . Methods A case‐control study was conducted between 80 adult patients with depression and 80 health controls .In general ques‐tionnaire was designed to collect the demo‐graphic information ,Childhood Trauma Questionnaire‐28 Item Short Form (CTQ‐SF) was used to assess childhood trauma and Trait Coping Styles Questionnaire (TCSQ) was assigned to evaluate coping styles .T test , Mann‐Whitney U test ,Spearman Bivariate correlations and Multiple linear regression were employed to explore the characteristics of the major depressive patients′coping styles and the association between childhood abuse .Results (1) The patients with depres‐sion had both statistically significantly higher scores on negative coping style (t=9 .13 ,P<0 .01) and lower scores on positive cop‐ing style (t= -12 .01 ,P<0 .01)than the controls with childhood abuse .The patients with childhood abuse had both statistically significantly higher scores on negative coping style(Z= -6 .134 ,P<0 .01) and lower scores on positive coping style(Z= -5 .452 , P<0 .01) than the controls with childhood abuse .Inside the group of patients with depression ,the patients who had childhood a‐buse gained significantly higher scores in negative coping style than the others who had no early trauma (Z= -2 .159 ,P<0 .05) , however ,no conspicuous differences were found on the positive coping style scores .(2)The childhood abuse was both significantly positive correlated with the passive coping and negative related to the active coping (r=0 .562 ,P<0 .01;r= -0 .333 ,P<0 .05)a‐mong the patients of depression .(3)The total degree of abuse and physical abuse had significant prediction function of 34 .4% (P<0 .01)on the patients′ negative coping .Conclusion The coping styles of patients with depression were prone to be influenced by childhood abuse remarkably ,more severe abuse ,especially with the physical abuse ,more negative the coping styles were .

7.Effects of enhanced recovery after surgery on early cognitive function of patients after hepatectomy for liver cancer

Objective To investigate the effects of enhanced recovery after surgery (ERAS) pathway on early cognitive function of post-hepatectomy patients after hepatectomy.Methods Ninety-six patients undergoing elective hepatectomy for liver cancer were randomly assigned into two groups:group ERAS,n =48 and group C,n =48.patients in group ERAS accepted perioperative and anaes-thetic optimization of enhanced recovery after surgery pathway,and patients in group C received peri-operative and anaesthetic management of normal surgery concept.The mini-mental state examination (MMSE)was used to test the cognitive function of the patients one day before operation (T0 ),one day after operation (T4 ),three days after operation (T5 )and seven days after operation (T6 ),and the blood levels of S100βprotein,neuron-specific enolase(NSE),IL-1β,IL-6,and TNF-α were de-tected at T0 ,30 minutes after operation (T1 ),6 hours after operation (T2 ),12 hours after operation (T3 )and T4 .Results Compared with group C,MMSE scores in group ERAS were increased at T4 ,T5 , the blood levels of S100βprotein,IL-1βand IL-6 in group ERAS were decreased at T1 ,T2 ,the blood levels of NSE in group ERAS were decreased at T2 ,T3 ,the blood levels of TNF-αin group ERAS were decreased at T1-T3 ;Compared with the baseline value at T0 ,MMSE scores were decreased at T4 ,T5 in group C and decreased at T4 in group ERAS,the blood levels of S100 βprotein in two groups were increased at T1-T3 , the blood levels of NSE in two groups were increased at T2-T4 ,the blood levels of IL-1β,IL-6,and TNF-αin two groups were increased at T1-T4 ;and the postoperative length of stay in group ERAS were decreased. Conclusion Enhanced recovery after surgery pathway is helpful in improving early postoperative cognitive function of patients after hepatectomy,and the mechanism may be related to decreased the blood levels of S100βprotein,NSE,IL-1β,IL-6 and TNF-α.
8.Protective Effect of Extract from Morindae Officinalis on the Oxidative Injury of Human Sperm Motility

ObjectiveTo observe the protective effect of extract from Morindae officinalis on the oxidative injury of human sperm motility.MethodsThe sperm specimens were cultivated in vitro, the reactive oxygen species (ROS) was made from hypoxanthine-xanzine xanzine (HX-XO), and sperm specimens were co-incubated with ROS and different dosages of extract and oxygen. The motor parameters were evaluated, the motility were analyzed by the test of capillary tube penetrating, and compared with the control group.ResultsThe sperm function were injured, the motility and penetrating ability decreased evidently under the action of ROS, and with a significant difference compared with the normal group ( P<0.001). The sperm motility and penetrating ability could be improved by all of the small, medium and massive extract compared with vitamin C, especially 0.25 and 0.5 g/ml Morindae officinalis had more evident action in the rate of progressive motility and vitality exponent.ConclusionThe ROS can injury the sperm motility and penetrating ability, the appropriate content of extract from Morindae officinalis can significantly intervene peroxidation in sperm by the ROS, and protect the sperm motility.
9.Genetic polymorphism of eleven canine STR loci.

OBJECTIVE:
To investigate the polymorphism of 11 canine STR loci.
METHODS:
A fluorescent multiplex system with 11 STR loci (PEZ1, PEZ2, PEZ3, PEZ5, PEZ6, PEZ8, PEZ12, FH2010, FH2054, FH2132 and FH2611) was constructed independently and performed to amplify 105 samples from dogs. The character of these loci was analyzed with the PCR data.
RESULTS:
The distributions of genotypes and allele frequencies of 11 STR loci were obtained. The total power of discrimination for the 11 loci in canine population was 0.9999999 and the cumulative probability of exclusion was 0.9330621. The observed heterozygosity and polymorphism information content (PIC) were 0.502 and 0.640, respectively.
CONCLUSION
Each of the eleven canine STR loci has a high genetic polymorphism and can be applied for the parentage testing and individual identification. The fluorescent multiplex system is a reliable method in forensic application.

10.The application of fluorescence in situ hybridization and automated image cytometry in the diagnosis of urothelial carcinoma of bladder.

OBJECTIVETo discuss the application of automated DNA image cytometry (ICM) and fluorescence in situ hybridization (FISH) in the diagnosis of urothelial carcinoma of bladder.
METHODSFrom August 2008 to March 2009, 60 volunteers with informed consent were divided into two groups, 40 patients proven as urothelial carcinoma of bladder by pathology and 20 healthy individuals as control. Urine was collected and tested by cytology, ICM and FISH.
RESULTSOverall sensitivity of FISH was significantly higher in detection of malignancy than that of ICM (82.5% vs 62.5%, P < 0.05) and that of urine cytology (82.5% vs 25.0%, P < 0.05), while ICM was more sensitive to diagnose urothelial carcinoma of bladder than urine cytology (62.5% vs 25.0%, P < 0.05). Specificities of urine cytology, ICM and FISH were 100% in diagnosis of urothelial carcinoma of bladder (P > 0.05). Sensitivities of urine cytology, ICM and FISH have no correlation with pathological stage (P > 0.05), but have significant correlation with grade (P < 0.05).
CONCLUSIONSICM and FISH have the same specificity as urine cytology in diagnosis of urothelial carcinoma of bladder, but they have significantly higher sensitivity than urine cytology. FISH has the highest sensitivity among three diagnostic methods. Therefore, FISH may become a newly non-invasive technique for the diagnosis and surveillance of urothelial carcinoma of bladder.
