The anti-tumor activity of fermentation and the ethanol extraction of ten Polyprous fungi against human lung cancer cell NCI-H460 were determined by MTr method,the effect of medium and time for cultivation on the anti-tumor activity were studied.The results showed that the fermentation liquid of Onnia tomentosa and Ceizene unicolon and the mycelium ethanol extract of Formitopsis pinicola had obvious an- ti-tumor activity,the tumor inhibiting ratio of mycelium ethanol extract of Formitopsis pinicola cultivated in Potato-woodehipping medium was 88.87% at the concentration for 500?g/mL,and the inhibiting ratio had little change when Formitopsis pinicola cultivated in different medi- um and for different days.

Objective To study the changes in somatosensory evoked potential caused by injection of adriamycin magnetic gelatin microspheres into subarachnoid space of rabbits. Methods Thirty rabbits were randomly divided into 5 groups: sham operation group (group S), gelatin microspheres 5mg (group C1), 15mg of gelatin microsphere (group C2) controlled groups, and adriamycin microspheres 5mg (group A1) and 15mg (group A2) groups. Background Sustained abnormal tear secretion in dry eye patients may lead to ocular surface and lacrimal glands in the state of long-term inflammatory cell infiltration.Lacrimal gland suffers immune attack by lymphoproliferative,and inflammation interferes normal gland secretion,in which chemokines and their receptors on lymphocytes play a key role.Objective This study was to investigate the inflammation mechanism of delayed allergy induced by Th1 cell in the development of dry eye.Methods Sixty eyes of 30 patients with dry eye and matched 30 healthy volunteers were enrolled in Affiliated First Hospital of Xinjiang Medical University from June to December in 2012.Schirmer Ⅰ test (S Ⅰ t),break up time (BUT) of tear and corneal fluorescence stain (FLS) were performed on the subjects,and conjunctiva epithelial cells were obtained using cytological method of conjunctiva imprinting.Positive cell rates of CC chemokine receptor 5 (CCR5) and CXC chemokine receptor 3 (CXCR3) were detected by flow cytology,and the relative expression levels of regulated upon activation of normal T cells exp ressed and secreted (RANTES),macrophage inflammatory protein (MIP)-lα and MIP-1β,monokine induced by interferon-γ (IFN-γ) (MIG),interferon-γ inducible protein 10 (IP10) and interferon-inducible T-cell α chemoattractant (I-TAC) mRNA were quantitatively assayed by real-time PCR.Differences of the positive rates of CCR5,CXCR3 and lignds were compared between dry eye group and normal control group.Relationship between the positive rates of CCR5,CXCR3 and BUT,S Ⅰ t,FLS scores was analyzed.Results The values of BUT,S Ⅰ t were (2.90±1.37) seconds and (4.00±2.49) mm/5 minutes in the dry eye group,which were significantly lower than (8.56±4.69) seconds and (11.31 ±5.23) mm/5 minutes in the normal control group (t =3.172,2.186,both at P<0.05).FLS scores,positive rates of CCR5 and CXCR3 were0.90±0.57,(3.38±0.66) % and (2.64±0.47)% in the dry eye group,showing significant elevations in comparison with 0.14±0.06,(2.12±0.21) % and (1.12±0.11) % in the normal control group (t=2.297,3.151,5.454,all at P<0.05).In the dry eye group,the masculine rate of CCR5 was negatively correlated with BUT and S Ⅰ t (r=-0.473,-0.385,both at P<0.05).The masculine rate of CXCR3 was negatively correlated with BUT and S Ⅰ t (r =-0.753,-0.684,both at P<0.05).No considerable correlations between the positive rate of CCR5 with the positive rate of CXCR3 or FLS scores (r =0.231,0.336,both at P<0.05).The relative expression levels of RANTES,MIP-1α,MIG and IP10 mRNA in the dry eye group were significantly higher than those in the normal control group (t =3.091,2.894,2.688,2.245,all at P<0.05),but there were no significant differences in the relative expression levels of MIP-1β and I-TAC mRNA between the two groups (t =0.512,1.979,both at P>0.05).The positive correlations were seen between the masculine rate of CCR5 with the relative expression of RANTES or MIP-1α mRNA (r=0.473,0.285,both at P<0.05),but there was no obvious correlation between the masculine rate of CCR5 and the MIP-1 β mRNA expression(r=0.214,P>0.05).In addition,the masculine rate of CXCR3 was positive correlated with the expressions of MIG and IP10 mRNA (r=0.553,0.314,both at P<0.05),whereas the masculine rate of CXCR3 was not related to the expression of I-TAC mRNA (r=0.364,P>0.05).Conclusions Dry eye is probably along with the long-term infiltration of inflammatory cells.The delayed allergy induced by Th1 cells and the nature killed cells is probably the primary cause to xerophthalmia.CCR5,CXCR3 and their ligands might be the regulative targets in the inflammation mechanism of dry eye.

This study was conducted to set up a common mechanism for varying phases of focal segmental glomerulosclerosis(FSGS) by comparing the morphological differences between human FSGS and changes in 5/6 renal ablation animal model, which has been accepted as experimental prototype for hyperfiltration theory as pathogenesis of FSGS. Both the human and the experimental rats showed very similar changes such as segmental glomerulosclerosis, vacuole formations or inclusion of small granules of podocytes, appearance of foamy cells in the capillary lumina, eosinophilic deposits along the mesangial area, and focal atrophy of tubules with associated interstitial fibrosis. The halo, frequently seen in human FSGS, is due to detachment of visceral epithelium from basement membrane, however, did not appear in the experimental rat specimen. On the other hand, the foamy cells and hyalinization were more frequently noted in the rat series and even involved the arterioles. The mesangial proliferation never appeared in the rat series occasionally found in human FSGS. In conclusion, the pathogenesis of FSGS cannot depend solely on the hyperfiltration theory of hemodynamic derangement, but has complex impairment of visceral epithelium and cells forming the constituents of basement membrane.

8.Detection and significance of phosphatidylinositol 3-kinase in adipose tissue of polycystic ovary syndrome patients with insulin resistance

Objective To investigate the expression of phosphatidylinositol 3-kinase(PI-3K)in adipose tissue of polycystic ovary syndrome patients(PCOS),and explore molecular mechanisms of insulin resistance(IR)in PCOS.Methods Samples from patients with PCOS with IR(n=19),PCOS without IR (n=10)and controls(n=15)were collected.Serum fasting insulin(FIN)and fasting plasma glucose (FPG)were measure& Insulin resistance index was calculated using homeostasis model assessment (HOMA)to analyze the relationship between these markers and IR.Western blot technique was used to detect the PI-3K p85 subunit.Gene expression of PI-3K p85 subunit was detected by reverse transcription polymerase chain reaction(RT-PCR)method.Kinase activity was detected by immunoprecipitation,thin- layer chromatography and gamma scintillation counting.Results(1)The levels of FIN[(25.2?3.8) mU/L]and HOMA-IR(1.6?0.3)in PCOS with IR were significantly higher than those in PCOS without IR[(13.4 +3.8)mU/L,0.9?0.3]and controls[(9.5 +2.6)mU/L,0.5?0.3;all P<0.05).(2) There was no significant difference in the protein(0.65?0.10)and gene expression(0.92?0.12)of PI-3 K p85 subunit in PCOS with IR compared with PCOS without IR(0.72?0.10,1.01?0.10)and control groups(0.73?0.14,1.00?0.12;P>0.05).(3)PI-3K activity in PCOS with IR(81%)and PCOS without IR(89%)was significantly decreased(P<0.01,P<0.05)and negatively correlated with HOMA- IR(r=-0.69,P<0.01;r=-0.62,P<0.05).Conclusions No significant difference in the protein and gene expression of PI-3K p85 subunit in PCOS with IR is found.The decreased PI-3K activity may lead to IR of PCOS.

10.Fluorescence enhancement of flavoxate hydrochloride in alkali solution and its application in pharmaceutical analysis.

Fluorescence enhancement reaction of flavoxate hydrochloride (FX) in strong alkali solution was studied, the mechanism of the reaction was investigated, and a novel fluorimetric method for analysis of FX in drug sample was established. FX has no intrinsic fluorescence, but it can slowly produce fluorescence in strong alkali solution. Heating can promote the fluorescence enhancement reaction. In 3D fluorescence spectra of the decomposition product of FX, two fluorescence peaks, located respectively at excitation wavelengths λex/ emission wavelength λem =223/410 nm, and 302/410 nm, were observed. Using quinine sulfate as a reference, fluorescence quantum yield of the decomposition product was measured to be 0.50. The structural characteriza- tion and spectral analysis of the decomposition product reveal that ester bond hydrolysis reaction of FX is firstly occurred during heating process, forming 3-methylflavone-8-carboxylic acid (MFA), then a cleavage reaction of the γ-pyrone ring of MFA occurred, producing α, β-unsaturated ketone. This product includes adjacent hydroxyl benzoic acid group in its molecule, which can form intramolecular hydrogen bond under alkaline condition, so that increase the conjugate degree and enhance the rigidity of the molecule, and thereby cause fluorescence enhancement. Based on this fluorescence enhancement reaction, a fluorimetric method was proposed for the determination of FX. A linear calibration curve covered the concentration range 0.020 3-0.487 µg · mL. The regression equation was I(F) = 23.9 + 5357.3 c, with correlation coefficient r = 0.999 7 (n = 8), detection limit D = 1.1 ng · mL(-1). The method was applied to the analysis of FX tablets, with a spiked recovery rate of 100.2%. The reliability of the method was verified by a UV-spectrophotometric method.
