Biodegradable PLGA [poly(lactide-co-glycolide acid)] have shown significant potential for sustained and targeted delivery of several pharmaceutical agents, including DNA. We reviewed the formulating approaches of PLGA nanoparticles/microspheres as DNA carriers and utilization for gene therapy and vaccine adjuvant.
4.Drainage tube placement via transnasal route for the treatment of mediastinal anastomotic fistula after surgery of esophageal cancer: clinical analysis of 6 cases

Objective To evaluate the clinical feasibility of performing the placement of drainage tube via transnasal route in treating mediastinal anastomotic fistula after surgery of esophageal cancer.Methods A total of 6 patients with mediastinal anastomotic fistula after surgery of esophageal cancer,who were admitted to authors' hospital during the period from August 2015 to January 2016,were included in this study.The diagnosis was confirmed by esophageal radiography and thoracic CT scan in all the 6 patients.Under X-ray monitoring,the drainage tube was inserted into the cavity of mediastinal fistula with the help of a guide wire guidance,and continuous negative pressure suction was adopted.The jejunum nutrition tube was inserted via the same nostril if feeding tube was not placed.Results Successful placement of fistula drainage tube and jejunum nutrition tube was achieved in all the 6 patients.In one patient the fistula drainage tube had to be re-placed as the drainage tube was obstructed five days after initial placement.Under X-ray monitoring the mean time used for the drainage tube placement was 33 min (range of 23-48 min).The procedure was well tolerated by all the 6 patients,and no procedure-related complications occurred.After continuous negative pressure suction that lasted for 6-40 days (mean of 23 days) the fistulae healed.Conclusion For the treatment of mediastinal anastomotic fistula after surgery of esophageal cancer,the placement of fistula drainage tube under X-ray monitoring is simple,safe and effective.(J Intervent Radiol,2017,26:250-252)
5.Effect of stellate ganglion block on brain-derived neurotrophic factor and serum interleukin-8 of dorsal root ganglion

Objective To investigate the effect of stellate ganglion block ~SGB) on formalin-induced nociceptive behavior of rabbits, the expression of brain-derived neurotrophic factor ~BDNF) of dorsal root ganglia ~DRG) and the concentration of serum interleukin-8 ~IL-8) in rabbits. Methods Twenty-four rabbits were used in this study. Catheters were inserted closely to right stellate ganglia of the animals, which were randomly divided into control group ~A), SGB group ~B) and pain model group ~C). Group B and C were administered with an injection of 0.5ml of 3% formalin into the right front paw subcutaneously, group A 0.5ml saline by the same way. 0.5ml of (0.25)% bupivacaine was administered via the catheter in group B at 10 minutes before injection, while group A and C were given 0.5ml of saline at the same time. Nociceptive behavior was observed for 60 minutes after injection, and assessed by weighted pain score. Serum IL-8 was measured radioimmunologically at 10 minutes ~T_0) before injection, 10 ~T_1), 60 ~T_2) and 120 minutes ~T_3) after injection. The expression of BDNF in the right DRG of cervical 7, 8 and thoracic 1 was investigated immunohistochemically at 120 minutes after injection. Results Formalin-induced nociceptive behavior in phase Ⅱ was relieved following SGB. The concentration of serum IL-8 in groups B and C at T_2 and T_3 was significantly increased than that at T_0 and T_1~P
6.The synergistic effect of amygdalin and HSYA on the IL-1beta induced endplate chondrocytes of rat intervertebral discs.

The effect of amygdalin joint hydroxysafflor yellow A (HSYA) on the endplate chondrocytes derived from intervertebral discs of rats induced by IL-1beta and the possible mechanism were studied and explored. Chondrocytes were obtained from endplate of one-month SD rat intervertebral discs and cultured primary endplate chondrocytes. After identification, they were divided into normal group, induced group, amygdalin group, HSYA group and combined group. CCK-8 kit was adopted to detect the proliferation of the endplate chondrocytes. FCM was measured to detect the apoptosis. Real-time PCR method was adopted to observe the mRNA expression of Aggrecan, Col 2 alpha1, Col 10 alpha1, MMP-13 and the inflammatory cytokines IL-1beta. The protein expression of Col II, Col X was tested through immunofluorescence. Compared with the normal group, the proliferation of the endplate chondrocytes decreased while the apoptosis increased (P < 0.05). With down regulation of the mRNA expressions of Aggrecan, Col 2 alpha1 and up regulation of the mRNA expressions of Col 10 alpha1, MMP-13, IL-1beta (P < 0.05), the protein expression of Col II decreased while the protein expression of Col X increased. Compared with the induced group, amygdalin group, HSYA group, the combined group could inhibit the apoptosis and promote the proliferation (P < 0.05). They could increase the mRNA expressions of Aggrecan and Col 2 alpha1 while decrease the mRNA expressions of Col 10 alpha1, MMP-13 and IL-1beta (P < 0.05). They could also enhance the protein expression of Col II while reduce the protein expression of Col X. The effect of the combined group was significantly better than that of amygdalin and HSYA. Amygdalin joint HSYA could inhibit the degeneration of the endplate chondrocytes derived from intervertebral discs of rats induced by IL-1beta and better than the single use of amygdalin or HSYA.

7.Diagnosis of malignant obstructive jaundice by percutaneous transhepatic cholangial biopsy

Objective To explore the diagnostic value of percutaneous transhepatic cholangial biopsy for malignant obstructive jaundice.Methods Data of 42 cases suspected of malignant obstructive jaundice who underwent percutaneous transhepatic cholangial biopsy and percutaneous transhepatic cholangial drainage (PTCD) were retrospectively analyzed.And during the examination,the biopsy tissues were obtained for pathology.Results The pathologic results were obtained successfully in 41 of 42 cases and the success rate of operation was 97.62% (41/42).There were 31 cases with positive results.The positive rate was 73.81% (31/42).In 12 cases diagnosed as distal biliary tract obstruction by percutaneous transhepatic cholangiography (PTC),positive pathology results were found in 3 cases;the positive rate was 25.00% (3/12).In 30 cases diagnosed as perihilar biliary tract obstruction by PTC,positive pathology results were found in 28 cases;the positive rate was 93.33% (28/30).The accuracy of perihilar biliary tract obstruction was higher than that of distal biliary tract obstruction (x2=20.704,P<0.05).In all 42 cases,there were no severe complications during or after surgery.In the 31 cases with positive biopsy pathological results,6 cases underwent surgery after improved liver function.The postsurgical pathological results were agree with biopsy pathological results.Conclusion Percutaneous transhepatic cholangial biopsy is safe and efficient for malignant obstructive jaundice.The successful rate and positive rate are high,especially for perihilar biliary tract obstruction.
8.Chemical Constituents from Roots of Angelica apaensis with Cytotoxic Activity

Objective Obtaining chemical constituents of Angelica apaensis collected in Qiaojia County of Yunnan Province and detecting their anti-tumor activities in vitro. Methods The MeOH extract of was repeatedly subjected to column chromatography over silica gel, Sephadex LH-20 and/or RP-18 and semipreparative HPLC. Their anti-tumor activities in vitro were measured by MTT colorimetric assay. Results Eight compounds were isolated from the roots of this plant. Conclusion The major type of chemical constituents of in Yunnan is furocoumarin. These separated compounds showed weak anti-tumor activity in vitro.
9.PRELIMINARY STUDIES ON PHYSIOLOGICAL CHARACTERISTICS OF ALKALIPHILIC ACTINOMYCETES

pH, affects of different alkaline materials KOH, K 2CO 3, NaOH, Na 2CO 3 on the growth, and NaCl, KCl tolerance of 29 isolates from the saline and alkaline soils in Xinjiang and Qinghai Provinces of China and 1 type strain were studied. Results showed that only a few alkaliphilic actinomycetes were Na +-obligately dependent, and K +-sensitive. Some alkaliphilic actinomycetes were CO 3 2- -sensitive, and NaCl, KCl could inhibit their growth. 4 kinds of alkaline materials had no affect on growth of alkaliphilic Nocardiopsis, and these strains showed high tolerance to NaCl, KCl. So it was presumed that only K + and CO 3 2- obligately dependent alkaliphilic Actinomycetes maybe exist in alkaline environments.
