Objective The aim of this study was to assess the expression of protein products of Hugl-1 (human lethal-giant-lavae 1)in non-small cell lung cancer(NSCLC), to estimate their possible expression and to correlate the results of immunohistochemical analysis with various clinicopathologic parameters. Objective To study the application of native pelvis and ureter in kidney transplantation. Methods Between 1977 and 2001, 1 364 kidney transplantations were performed in our institution. 18 cases of ureteral complication after kidney transplantation were treated with native pelvis and ureter. The mean age of the patients was 42 years. In 10 patients with urine leakage, 3 cases underwent pyeloureterostomy using native ureter, 2 cases ureteroureterostomy using native ureter and 5 cases pyelopyelostomy using native pelvis. Five cases due to short ureter underwent ureteroureterostomy using native ureter. Three cases of ureteral stenosis were subjected to ureteroureterostomy using native ureter.Results Two grafts were rejected. Sixteen patients developed a normal function of kidney postoperatively (follow up: 3 months to 8 years). Conclusion Application of native pelvis and ureter might be a therapy of choice when secondary reconstruction by re-ureteroneocystostomy is not possible.

A prospective study of vitamin B6, B12, folate, and homocysteine levels was carried out in 7 kidney transplant(KT) recipients. The first sample for the basal level was drawn on the morning of the KT day before the start of cyclosporine injection. Thereafter, serial blood samples were taken every day until the serum creatinine level decreased below 1.5 mg/dl and then every 2 or 3days until discharge. The serum creatinine level decreased to below 1.5 mg/dl within 3days except for cases 4 and 6. The homocysteine levels decreased markedly in synchronisation with serum creatinine levels increased during the first 7days and then declined together with serum creatinine levels but went up again after a few days. The levels of B6, and vitamin B12 do not continue to decrease after the levels of serum creatinine have decreased to 1.5mg/dl, while homocysteine levels go up progressively. In conclusion, contrary to our expectation, the level of homocysteine rebounds a few days after KT following a transient decline. A deficiency of folic acid, vitamin B6 and vitamin B12 does not seem to cause hyperhomocysteinemia in KT recipients. It is necessary to pay attention to hyperhomocysteinemia in KT recipients, especially when the recipients have an atherosclerosis-related complication.

Background and purpose:Overexpression of human telomerase reverse transcriptase(hTERT)plays a critical role in the process of immortalization of tumor cells.This study aims to investigate siRNA's effects on endogenous hTERT mRNA level and growth of breast cancer cells by targeting hTERT with its specific siRNA.The ultimate goal was to further elaborate the possible mechanism of immortalization in breast cancer cells.Methods:Real-Time RT-PCR was used to determine the hTERT transcripts in various breast cancer cells.hTERT siRNA was transfected into MCF7 cells with lipofectamine 2000.Cell growth was illustrated by growth curve,and cells apoptosis percentage was measured with Flow Cytometry(FACS).The relative expression levels of immortalization-related genes were evaluated with Real-Time RT-PCR.Results:Overexpressions of hTERT were demonstrated in all tested breast cancer cell lines.The inhibitory effects of hTERT siRNA were shown on both hTERT mRNA levels and cell growth from day 4 post transfection.Apoptosis was induced by hTERT siRNA as well.Certain immortalization-related genes were reduced by more than 50%,such as RAC1,PCYT2,FDFT1 and ATP5G2.Conclusions:hTERT siRNA specifically inhibited hTERT at mRNA level as well as cell growth.The apoptosis and downregulation of immortalization-related genes due to hTERT siRNA was demonstrated.

Background and purpose:The effects of triiodothyronine(T3)on breast cancer remain unclear.The aim of this study was to investigate the expression and correlation of estrogen receptor-?(ER-?)and thyroid hormone receptor-?(TR-?)in breast cancer cell lines,and to determine the changes of relevant receptors expression and its possible mechanism due to T3 treatment.Methods:Real-Time PCR was used to analyze the expressions of ER-? and TR-? at mRNA level in seven breast cancer cell lines.The expression level of retinoid X receptor-?(RXR-?)and these two receptors after T3 treatment were determined as well.The gene profile was further determined in SK-BR-3 cells treated with T3.Results:The expressions of ER-? and TR-? were quite low in SK-BR-3,MDA-MB-231 and MDA-MB-435 cells,but high in MCF7,BT20,BT474 and T47D cells.ER-?,TR-? and RXR-? were upregulated due to T3 treatment in ER-?(-)cells,but not in ER-?(+)cells.In SK-BR-3 cells,T3 treatment led to up-regulation of E2F1,TGF-?1R,TGF-?2R and hTERT,but down-regulation of TGF-?1 and p21.Conclusions:The expressions of ER-? and TR-? are correlated in breast cancer cells;the effect of T3 treatment on hormone receptors is associated with endogenous ER-? level.

Objective To explore the therapeutic effects of Ni-Ti memory alloy embracing fixator on the multiple fracture of ribs and flail chest. Method From January 2006 to December 2009, the effects of operation and long-term follow-up after internal fixation operation with memory alloy embracing fixator in patients with multiple fracture of ribs and flail chest were observed. Results The hospital stay of patients treated with memory alloy embracing fixator was 10.31 ± 3.14 days and pain was significantly reheved after treatment ( P= 0.02) .The rate of post-operative complications was 17.65% such as atelectasis, pulmonary infection, etc. There were fewer longterm complications and less influence to daily activity of life. Conclusions It is a advanced and practical method for patients with the multiple fracture of ribs by using Ni-Ti memory alloy embracing fixator with slighter trauma,simple, safe, reliable, good histocompatibility and fewer post-operative complications.
