1.Central pontine myelinolysis: a report of 5 cases with autopsy pathological analysis

Objective A series of 5 patients with central pontine myelinolysis (CPM) confirmed either by cranial MRI,CT image (4 cases) or by autopsy pathological analysis (1 case) was reported. The pathophysiological mechanism,the early diagnosis and the role in prevention were discussed. Methods The creteria obtained from clinical features, cranial MRI and CT imagine as well as autopsy pathological changes were thoroughly analyzed. Results ① All 5 case of CPM were occurring in patients who had already had somatic disease and serious systemic functional failure, especially in those who had had seriously water and ion disturbances, such as hyponatremia. ② In 4 of the 5 patients, psychiatric manifestation tended to show the earliest clinical features. ③ The pseudobulbaral paralysis appeared in 4 patients. ④ Cranial MRI and CT served as the most reliable techniques for diagnosing CPM cases when patients are still alive. ⑤ A variety of paralysis were found in all of the 5 CPM patients. ⑥Pathological analysis demonstrated that there is a symmetric unspecific myelinolysis in areas of basal pontine, caudal middle brain and the superior medulla (1 patient). Conclusion ① The occurrence of hyponatrimia and its too fast correction may be associated with the onset of CPM. ② CPM, a self-restrained disease can be cured from dangerous complications, such as pneumonia, respiratory failure and malnutrition when suitably treated.

Recently,the progress in employing transplanting stem cells to cure injured retina is very fast and has been continuously yielding exciting results.Various sources are used in the studies,including retina-derived cells such as M?ller cells and ciliary body cells,and non-retina-derived cells such as embryonic stem cells and brain-derived stem cells.This review briefly discusses the recent progress of these studies.
3.Effect of Tanshinone II A on Cytokines of Rats with Severe Acute Pancreatitis Lung Injury.

OBJECTIVETo explore the effect of Tanshinone II A on severe acute pancreatitis (SAP) lung injury (ALI) rats and its possible mechanism.
METHODSSD rats were injected with sodium taurocholate to induce SAP group, and then intervened with sodium tanshinone II A sulfonate ( STS group). Simultaneously a sham-operation group (SO group) was set up. There were 24 rats in each group. The survival state and wet-to-dry weight ratio of lung tissues were observed. Activities of myeloperoxidase (MPO) in lung were determined by MPO reagent kit. Pathologic changes of lung tissues were determined by Hofbuaer method. Expression levels of three cytokines, TNF-alpha, IL-1beta, and intercellular cell adhesion molecule-1 (ICAM-1) were detected by ELISA.
RESULTSThe survival state of rats in the SAP group was deteriorated. The wet-to-dry weight ratio, MPO activities, pathologic changes in lung tissues, and expression levels of TNF-alpha, IL-1beta, and ICAM-1 increased significantly more in the SAP group than in the SO group (P < 0.05). Compared with those in the SAP group, the survival state of rats in the STS group was improved; the wet-to-dry weight ratio, MPO activities, pathologic changes in lung tissues, and expression levels of TNF-alpha, IL-1beta, and ICAM-1 obviously decreased in the STS group (P < 0.05).
CONCLUSIONTanshinone II A had remarkable effect on SPA LI rats, which might be associated with changing cytokines levels and attenuating infiltration of lung inflammatory cells.

5.The application of large flat-panel multifunction digital X-ray machine in digestive system

Objective: To evaluate the application value of large flat-panel multifunction digital machine in digestive system. Methods: Exam the gastrointestinal tract and biliary with dynamic detector, and then observe its advantages and disadvantages. Results: All the 13 gastrointestinal cancer lesions and 15 inflammatory lesions were shown clearly, including mucosa. Biliary tomography found two small stones, which were undetected by the general X-ray photography. The excellent rate of the photo was improved to 98%. Conclusion: The advantages of the flat panel detector include the wide range of display, clear image of digital pulse fluoroscopy spot film, the improvement of the diagnostic accuracy and the reduction in radiation dose. The disadvantage is that it is more inconvenient than the remote cradle gastrointestinal machine in rotating the position of the patient.
6.Prolongation of murine cardiac allograft survival by dendritic cells treated with NF-?B decoy

Objective To investigate the function of donor-derived dendritic cells (DCs) treated with NF-?B decoy in prolonging murine cardiac allograft survival time.Methods Donor bone marrow- derived DCs were treated with NF-?B decoy in vitro.Heterotopic abdominal heart transplantation was performed from BALB/c to C57BL/6 mice.Recipients were grouped according to different pretreat ments as follows:(1) Control group,infusion of PBS (0.2 ml) alone intravenously via the portal vein 7 days before heart transplantation;(2) CsA group,treated with sub-therapeutic CsA only for 7 days (10 mg?kg~(-1)?day~(-1)) through intraperitoneal injection after transplantation,and the same as control group before transplantation;(3) Control DCs group,infusion of only cultured 5th-day recipient DCs untreated with NF-?B ODN decoy;(4) Treated DCs group,infusion of recipient DCs pretreated with NF-~cB ODN decoy;(5) Combined treatment group,infusion of recipient DCs treated with NF-~cB ODN decoy before transplantation and intraperitoneal injection of sub-therapeutic CsA for 7 days (10 mg?kg~(-1)?day~(-1)) after transplantation;(6) Third party donor group,C3H/HeJ mice used as donor, and recipient (C57BL/6) was treated the same as combined treatment group.Every group had 8 mice and graft survival time was observed.Cytokines (IL-2,INF-?,IL-4 and IL-10) in recipient serums were analyzed by ELISA at 7th day after transplantation.Results The graft mean survival time (MST) in control group,CsA group,Control DCs group,treated DCs group,combined treatment group and third party donor group was 7 days,10.3 days,7.6 days,21.4 days,53.6 days and 9 days,respectively.There was significant difference in MST between treated DCs group and control group or control DCs group (P
7.The Progress of Tissue Engineering Heart Valve

The currently used heart valve prosthesis are still far from the ideal one in anticoagulation and longevity. The construction of tissue engineering heart valve leaflets are carried out recently. This article reviews the progress of tissue engineering heart valve leaflets in implanting materials, seeded cells, animal experiment and basic research. It also discusses the benefits and feasibility of tissue engineering heart valve leaflets.
8.Lymphocyte apoptosis induced by sulfur mustard in rat spleen

Objective To investigate the apoptotic effect induced by sulfur mustard on the lymphocytes of the rat spleen,and define the role of caspase-3 during the process.Methods Sulfur mustard was given by intraperitoneal injection in a dose of 3.5mg/kg.The animals were anesthetized and the spleens were harvested at different timepoints after intoxication.The histopathogy of spleen was studied with hematoxylin-eosin staining.Caspase-3 mRNA was detected with RT-PCR.Protein expression of caspase-3 was assayed with Western blotting,lymphocytes of rat spleen were isolated and cultured in vitro and they were challenged with sulfur mustard(100?mol/L).The effect of Ac-DEVD-CHO(a specific inhibitor of caspase-3)on sulfur mustard-induced apoptosis of the lymphocytes cultured in vitro was evaluated.Fluorescent probe labeled with Rhodamine 123 was used to study mitochondrial potential.Results The histology of rat spleen was affected after sulfur mustard intoxication,as evidenced by apoptosis of a part of lymphocytes.Protein and mRNA expressions of caspase-3 were increased significantly in the spleens of intoxicated rats as compared with that in control group.DNA ladder and 'sub-G1' peak of lymphocytes which were treated with sulfur mustard in vitro were partially improved by Ac-DEVD-CHO(the specific inhibitor of caspase-3).In addition,mitochondrial potential decreased in a time-dependent manner in the lymphocytes intoxicated by sulfur mustard in vitro compared with that in the control group.Conclusions The spleen is injured in the rat which is intoxicated by sulfur mustard.Lymphocyte apoptosis is one of the mechanisms splenic injury.Caspase-3 may be involved in the process of lymphocyte apoptosis induced by sulfur mustard.
9.Practicality and repeatability of B-mode ultrasonography on diagnosis and score of carotid atherosclerosis

Objective To study the practicality and repeatability of B-mode ultrasonography on diagnosis and score of carotid atherosclerosis. Methods Ninety-nine B-mode ultrasonography pictures of carotid atherosclerosis were obtained from field investigation. According to the established standard(normal scored as 0, other scored as 1 - 7 by the severity), the 99 pictures were diagnosed and scored once together by three trained inexperienced members of the research team. Ten days later, these pictures were diagnosed and scored respectively, by the three members.The diagnostic results were analyzed with statistical methods to test the feasibility and repeatability of this set of diagnostic and scoring system. Results The joint diagnostic results by the three group members were regard as standard with scores 0 in 27 cases, 1 in 27 cases, 2 in 10 cases, 3 in 4 cases, 4 in 13 cases, 5 in 8 cases, 6 in 6 cases, and 7 in 4 cases. Independent diagnostic results by member A was 0 in 29 cases, 1 in 24 cases, 2 in 11 cases, 3 in 2 cases, 4 in 12 cases, 5 in 11 cases, 6 in 6 cases, and 7 in 4 cases. Independent diagnostic results by member B was 0 in 29 cases, 1 in 22 cases, 2 in 14 cases, 3 in 4 cases, 4 in 13 cases, 5 in 5 cases, 6 in 8 cases, and 7 in 4 cases. Independent diagnostic results by member C was 0 in 28 cases, 1 in 25 cases, 2 in 8 cases, 3 in 5 cases, 4 in 14 cases, 5 in 10 cases, 6 in 4 cases, and 7 in 5 cases. Comparison of the 4 diagnostic scores, the difference was not statistically significant(F = 0.019, P > 0.05). Joint diagnostic results of the three members were compared with the standard and correlation coefficient were 0.977,0.987,0.932, respectively(all P < 0.01 ). The correlation coefficient of diagnostic results of each member were 0.969,0.935,0.928, respectively (all P <0.01 ). Diagnostic results of each member were consistent with the standard and the compliance rate were 88.9% (88/99) ,90.9%(90/99) and 86.9%(86/99), respectively. Conclusions B-mode ultrasonography is a non-injure method for diagnosis of carotid atherosclerosis in epidemiology investigation. The method is easy to grasp and has a good repeatability.
10.Pharmacokinetic studies of scutellarein and glucuronide scutellarin in rats

Aims To establish a UPLC-MS/MS meth-od for the determination of plasma concentration of scutellarein and its metabolite and to study their phar-macokinetics in rat plasma. Methods The analysis was achieved by BEH C18 column with a mobile phase composed of 0 . 1 % formic acid in acetonitrile and 0 . 1% aqueous formic acid using step gradient elution. A TQD tandem mass spectrometry equipped with electros-pray ionization source was used as detector and opera-ted by multiple reaction monitoring( MRM) positive ion mode. After intravenous injection of scutellarein, the concentrations of scutellarein and its major metabolite glucuronide scutellarin in rat plasma were determined at different time points. The pharmacokinetic parame-ters were calculated by DAS 2. 0 software. Results Good linearity was achieved for scutellarein, the ex-traction recovery was between 80 . 5 % to 90 . 5 %, the precisions and accuracy were good. The result showed the pharmacokinetic profiles of scutellarein and glucu-ronide scutellarin both fit to the two-compartment mod-el. Conclusion The above mentioned method is spe-cific, rapid, sensitive and suitable for the pharmacoki-netic studies of scutellarein and its metabolite.
