1.Experience on the prevention and treatment of endoscopic thyroidectomy complications

Objective To understand the characteristics of complications occurred after endoscopic thyroidectomy and its prevention and treatment.Methods Retrospective analysis on the clinical effects and complications of 105 cases of endoscopic thyroidectomy.ResultsThe average operative time was 85 minutes in the 105 cases.Five cases were transferred to open operation because of bleeding and 11 cases transferred for the intraoperative pathology of papillary carcinoma.After operation,subcutaneous ecchymosis occurred in 5 patients,subcutaneous emphysema in 10 cases,neck and anterior chest skin tautness and discomfort in 7 cases,parathyroid injury in 2 cases.No superior laryngeal nerve or recurrent laryngeal nerve injury was found in the 105 cases.ConclusionUnder strict control on operation indications,proficient and meticulous endoscope operative skills,familiarization with the anatomical structure of thyroid,and timely and proper treatment on postoperative complications can reduce and prevent the occurrence of complications.
2.Study on inhibitory effect of EGCG on Calcium oxalate nephrolithiasis in rats and its related mechanism.

In the study, the inhibitory effect of epigallocatechin gallate (EGCG) on Calcium oxalate nephrolithiasis and its possible mechanism were investigated. The rat Calcium oxalate nephrolithiasis model was induced through the combined oral administration of ethylene glycol and ammonium chloride, which was intervened with EGCG. Rat blood samples were collected to detect blood creatinine (Cr), blood urea nitrogen (BUN) and blood calcium. Rat urine samples were collected to observe and compare 24-hour urine volume, oxalic acid (Ox) and calcium in urine. Renal samples were collected to prepare tissue slices and observe the pathological changes in Calcium oxalate nephrolithiasis. The expression of osteopontin (OPN) in renal tissues was evaluated by Real-time PCR and Western blot. According to the results, compared with normal rats, rats in the nephrolithiasis model showed significant increases in Cr, BUN, urine Calcium, urine Ox and renal OPN expression (P < 0.05), but obvious decrease in 24-hour urine volume (P < 0.05); Compared with rats with nephrolithiasis, those processed with EGCG revealed remarkable declines in Cr, BUN, urine Calcium and urine Ox (P < 0.05), with significant rise in 24-hour urine volume (P < 0.05) in a concentration-dependent manner. Additionally, compared with the control group, nephrolithiasis rats showed significant pathological changes in Calcium oxalate calculus. After ECCG treatment, the renal pathological changes and OPN expression attenuated significantly in a concentration-dependent manner. The results showed that EGCG inhibits the formation of calcium oxalate nephrolithiasis in rats and shows a notable protective effect on renal functions.

3.Changes of Activated T Cell Disfunction in Children with Refractory Mycoplasma Pneumoniae Pneumonia

Objective To study disfunction of activated T cell from peripheral blood and explore its role on pathogenesis in patients with refractory mycoplasma pneumoniae pneumonia (MPP).Methods T cell subsets and expression of membrane molecule on activated T cell in peripheral blood were measured in 26 refractory MPP patients by using immunofluorescence and flowcytometry,and compared with those of 18 control healthy children.Results The percentage of CD4~+ cell from peripheral blood in refractory MPP children was significantly lower than that in control group(P0.05).Conclusions There are disfunctions of T cell in refractory MPP patients in acute period,and the abnormal T cell activated disfunction may play an important role on pathogenesis.
4.A clinical study of the prognostic value of modified Glasgow prognostic score in patients with gas- tric cancer

Objective To investigate the prognostic value of modified Glasgow prognostic score(mGPS)in patients with gastric cancer.Methods The clinical data of 600 patients with gastric cancerwho underwent surgical treatment were retrospectively reviewed.These patients were divided into themGPS0 group,mGPS1 group,and mGPS2 group according to the standard of mGPS.Postoperative survivals and risk factors that may affect patient's prognosis were analyzed among the three groups.Results There were 46 (7.7%)cases,33 (5.5%)cases and 521 (86.8%)cases in the mGPS0,mGPS1 andmGPS2 group,respectively.Except for gender(P =0.203),age,BMI,CEA,CA199,CRP,Alb,TNM stageand mGPS affected the over all survivals and there were significant differences among the groups(P <0.01).Gender,age,BMI,CEA,CA199 and TNM stage affected the mGPS and there were significantdifferences among the groups(P <0.01).Multivariate analysis identified that age(OR,1.319,95% CI1.068,1.629,P <0.01),TNMstage(OR,2.909,95% CI 2.616,3.234,P <0.01)and mGPS(OR,1.845,95% CI 1.184,2.875,P =0.007)were correlated with the postoperative death rate and they wererisk factors of poor postoperative prognosis(P <0.01).Conclusion mGPS can be used as an early,simple and effective prognostic predictor of postoperative survival for patients with gastric cancer.
5.Study on the Differential Expression of miRNA Screening and MiR-2 2 1 Regulates Apoptosis by Targeting PUMA Gene in Lung Cancer

Objective To investigate the identification of MiR-221 regulates apoptosis by targeting PUMA in lung cancer. Methods The express levels of miRNAs that collected from lung cancer tissue and lung benign lesion were tested by the particular kid they made,analyzed the correlation between individual miRNA.Results 9 6 possible regulated PUMA related miRNAs had been screened through the bioinformatics database,then customized it into the special kit to test the expression with real time fluorescent PCR.The results showed that 10 types miRNAs were up expression in lung cancer tissue,miR-NA221 were selected for deep analyze.Results with rank sum test showed statistical significance (P<0.05)in lung cancer and para cancer tissue.There were statistically significant differences in the expression between metastasis and nonmetastasis (P<0.05),but had nothing to do with the pathological types.Conclusion Real time fluorescent PCR test showed several types of PUMA related miRNAs up-expressed in tissue lung,which indicate miRNA have relationship with PUMA in gene express regulation and cancer generation and development.Pression situation and clinic pathological parameters.Expression of miRNA-221 in lung cancer was associated with tumor metastasis.
6.Correlation Analysis of Clinicopathological Features with Osteopontin and Aquaporin-1 Expression in Laryngeal Carcinoma

Objective To investigate the correlation of clinicopathological features with osteopontin and aquaporin-1 expres-sion in laryngeal carcinoma.Methods From August 2011 to December 2014 in Dongfeng General Hospital,80 laryngeal cancer specimens and adjacent normal tissue samples were selected,all samples were carried out osteopontin and aquaporin Immunohistochemical analysis expression and clinicopathological characteristics and correlation analysis.Results Osteopon-tin staining particles and aquaporin-1 were located in the stromal cells,yellow dye.In cancer tissue,osteopontin and aquapor-in-1 positive granules were mainly located in the cytoplasm of cancer cells,diffuse shading,coloring deeper place near nests. The positive expression rates of osteopontin and aquaporin-1 in the laryngeal carcinoma were 70.0% and 56.3% respective-ly,the positive rates in the adjacent tissues were 5.0% and 7.5% respectively that compared to the differences were statisti-cally significant (P <0.05).In 80 cases of laryngeal carcinoma.The expression of osteopontin and aquaporin-1 in different clinical stages,differentiation and lymph node metastasis tissues were statistically significant (P <0.05).Spearman correla-tion analysis showed that the osteopontin and aquaporin-1 in laryngeal carcinoma were positively correlated (P <0.05).Con-clusion Osteopontin and aquaporin-1 expression in laryngeal carcinoma were showed high status,and the clinical stage,de-gree of differentiation and lymph node metastasis were significantly correlated that can work together to promote tumor inva-sion and metastasis.
7.The correlation analysis of the prognosis of hand-foot-and-mouth disease and prealbumin

Objective To explore the curative effect of prealbumin (PA) in the hand-foot-and-mouth dis-ease and analysis its value in monitoring and prognosing this disease. Method Detect levels of serum PA of 390 ca-ses of children before, after and during the treatment and compare the changes during the treatment. Then observe the prognosis. Results There are significant difference of PV levels between cases of different prognosis (P < 0.05). Conclusion The PV levels of children of hand-foot-and-mouth disease will change significantly after effective treat- ment. There are clinical significance in observing and prognosing the hand-foot-and-mouth disease by detecting the se-rum PV levels.

9.Proliferation and migration of endogenous neural stem cells in rats after cerebral infarct

BACKGROUND: Conventional opinions believe tissues of central nervous system(CNS) cannot regenerate after injury when they have been developed and maturated. However, recent researches have validated that neural stem cells exist in human and adult animal nervous system, and most of them are in static status in vivo. The therapeutic effects of neural stem cells in cerebral infarct have been a key point in the researches.OBJECTIVE: To observe the reactive process of endogenous neural stem cells after cerebral infarct to explore the effects of endogenous neural stem cell in CNS traumatic rehabilitation, which provides theoretical gist for the self-restoration after cerebral infarct.DESIGN: A randomized controlled trial by using experimental animals as subjects.SETTING: Department of Neurosurgery of Peking Union Hospital.MATERIALS: The study was conducted in the Laboratory of Neurosurgery Department of Peking Union Hospital from March to October in 2003. Totally 82 healthy male Wistar rats with a body mass between 250 g and 300 g were selected.METHODS: Cerebral infarct model was established in rats by thread-ligation method. Rats were divided into post-infarct 1 day, 3 days, 7 days, 14 days and 28 days groups with 14 rats each. Control group was sham-operation group( n= 12). The expressions of bromodeoxyuridine (BrdU) and Nestin in the brain of the rats were dynamically detected by immunohistochemical method.MAIN OUTCOME MEASURES: The changes of BrdU and Nestin positive cell numbers after cerebral infarct in rats.RESULTS: Only very few BrdU and Nestin positive cells had survived in hippocampal dentate gyrus and subventricular zone(SVZ) area in control group. BrdU positive cell significantly increased compared with control group in hippocampus and SVZ area after 1 day of cerebral infarct( P < 0.05),which reached its peak on the 7th day( P < 0.05), started to reduce after 14 days but still in the level significantly higher than normal( P < 0.05), and closed to normal level after 28 days. And moreover, BrdU and Nestin positive cells were significantly more in infarct side than the opposite side ( P< 0.05), which migrated towards the opposite side through callus.CONCLUSION: Cerebral infarct can activate the proliferation in situ and migration of endogenous neural stem cell.
10.Proteomic analysis of hippocampus in the rat

Objective To analyze the protein expression in the rat hippocampus by the proteomic approach.Methods Proteins from hippocampal tissue homogenates of the rat were separated by two-dimensional gel electrophoresis(2-DE),and stained with colloidal Coomassie blue to produce a high-resolution map of the rat hippocampus proteome.Selected proteins from this map were digested with trypsin,and the resulting tryptic peptides were analyzed by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF-MS).The mass spectrometric data were used to identify the proteins through searches of the NCBI protein sequence database.Results 37 prominent proteins with various functional characteristics were identified.The identified brain protein classes covered metabolism enzymes,cytoskeleton proteins,heat shock proteins,antioxidant proteins,signalling proteins,proteasome-related proteins,neuron-specific proteins and glial-associated proteins.Furthermore,3 hypothetical proteins,unknown proteins so far only proposed from their nucleic acid structure,were identified.Conclusion This study provides the first unbiased characterization of proteins of the rat hippocampus and will be used for future studies of differential protein expression in rat models of neurological disorders.
