2.Effect of GPS On Stimulating Murine Lymphocyte Proliferation

Glycyrrhiza uralensis Fish Polysacchoride(GPS)was observed to have mitogenic activity on murine spleen cell proliferation in vitro,The results showed that this mitogenic activity of GPS may be directed to B Iymphocytes,i.e.GPS is a new mitogen of murine B lymphocytes.Furthermore,it was found that B lymphocytes stimulated by GPS was M? depedent and T cell-indepedent.On the contrary,antibodies and IL-2 production was inhibited by GPS in vivo.

Objective To establish a rat model of pulmonary hypertension induced by left-to-right shunt and explore the influence of high pulmonary blood flow on pulmonary vascular collagen remodeling.Methods Abdominal aorta and inferior vena cava shunting was produced in rats. Pulmonary artery meanpressure (PAMP) of each rat was measured by using a right cardiac catheterization.Pulmonary artery collagen Ⅰ and Ⅲ were detected using immunohistochemisty.Results After 11 weeks of shunting the Qp/Qs was 3.3∶1.0,indicating a large shunt. Diabetes mellitus has became a growing public health problem,its acute and chronic complications threaten the patient's dramatically.HbA1c is recognized as the gold standard for blood glucose evaluation in patients with diabetes It has reported that the incideut rafe of diabetes related will decline remarkably when HbA1c decline by 1%,so the detection accuracy of the HbA1c is very important in the diabetes patient's management.Uncertainty of measurement can reflect the detation accuracy,which will benifite the value of HbA1c.
7. Preparation and evaluation of lung targeting microspheres for protionamide in vitro and in vivo

OBJECTIVE: To testify the feasibility of targeted delivery of protionamide to lung through the iv administration of the biodegradable poly(lactic-co-glycollic acid) (PLGA) microspheres. METHODS: Based on the single-factor study, the preparation of protionamide-PLGA microspheres was optimized. The surface morphology of the microspheres was observed by scanning electron microscope (SEM). The mean diameter and the size distribution of microsphere, the drug loading, the incorporation efficiency, drug release in vitro, stability and tissue distribution after intravenous administration were also evaluated, respectively. RESULTS: Protionamide-PLGA microspheres were regular and spherical in shape. The average particle size was (9.86 ± 1.38) μm and over 81.53% of the microspheres was in the range of 7-15 μn. The drug loading and encapsulated ratio was (32.70 ± 0.37)% and (8.48 ± 0.24)%, respectively. The in vitro drug release could be well fitted by the Higuchi equation (Q=4.4303t1/2 ± 7.7241, r=0.9913). Compared with the aqueous formulation, the drug level in lung of BALB/C mice in microsphere group was much higher, and also kept for longer time. CONCLUSION: The protionamide-PLGA microspheres prepared in this study show significant sustained release and lung targeting effect.
8.Measurements of teeth, dental arch and base bone in Mongolia teenages

Objective: To obtain the data of dental measurements in healthy Mongolia teenages.Methods: Normal dentognathic models were obtained by cephalometric roentgenography in 56 teenages (30 boys and 26 girls) aged 10~17 years. The width of the tooth crown, the size of the dental arch and dental base bone were measured with a vernier. The measurments were statistically analysed.Results: The tooth crown in boys was wider than that in girls( P 0.05). There was no significant difference in Bolton index and Point index between boys and girls.Conclusion: The dental measurments may be refrences for dental clinic and research.
9. Synthesis and anxiolytic effects of 2-aryl imidazo1,2-apyridine-3- acetamide derivatives

Objective: To search for new compounds with better water-solubility and higher antianxiety activities. Methods: A series of 2-arylimidazo[1,2-a] pyridine-3-acetamide derivitives were designed and synthesized. The anxiolytic activities were evaluated by BZDR competitive binding assay in vitro and the elevated-plus maze test in mice, the structure-activity relationship (SAR) has been studied. Results and Conclusion: Twenty-eight new compounds have been synthesized. Their structures were confirmed by 1H NMR and MS. According to the results of BZDR affinity test, compounds I1, I 8, I10, I13, I19 showed as good affinity as the positive control (Ro5-4864). The corresponding inhibition was 87%, 89%, 85%, 89% and 76% respectively at the concentration of 100 nmol/L, while that of Ro5-4864 was 82%. I8 and I10, which display better water-solubility and better BZDR affinity in vitro, show significant antianxiety effects in vivo.
10.Effect of high glucose on the expression of KLF6 in human lens epithelial cell

Background Epithelial-mesenchymaltransition (EMT)isamajorcontributortothe pathogenesis of posterior capsular opacification(PCO).Kruppel-like factor 6 (KLF6) is a zinc finger protein,which can be stimulated by high glucose in proximal tubule cells and involved in transforming growth factor beta (TGF-β)induced EMT of diabetic nephropathy.ObjectiveThis study was designed to investigate the effect of high glucose on the expression of KLF6 and its target genes( TGFB1,TGFBR1,COLIA1,HSP47) in human lens epithelial cells (LECs).MethodsHuman LECs(SRA01/04) were cultured and exposed to different concentration of glucose.The expressions of KLF6 mRNA and protein were analyzed by real time polymerase chain reaction( real time PCR) and western blot after treatment with high glucose.The expressions of KLF6 target genes were analyzed by real time PCR to evaluate the EMT of SRA01/04 cells.ResultsCompared with the control group(5.5 mmol/L),the relative mRNA levels of t-KLF6 and wt-KLF6 in SRA01/04 treated with high glucose(22.2,44.4,66.6 mmol/L) increased obviously (F =72.53,42.02,P<0.01 ).Then,the concentration of 22.2 mmol/L was used in the next experiments.The relative mRNA levels of t-KLF6 and wt-KLF6 increased to the peaks after treatment with high glucose for 12 h,and began to decrease after 24 h until lower levels after 48 h ( F =100.12,125.52,P < 0.01 ).Western blot showed that the expression of KLF6 protein was also upregulated by high glucose treatment.With the promotion of the expression of KLF6 gene,the relative mRNA levels of TGFB1,TGFBR1,COLlAl and HSP47 of treated cells also respectively increased after treatment for 12 h,and began to decrease after 24 h until nearly at the levels of the control groups after 48 h( F=6.73,162.35,64.39,12.05,P<0.05 ).ConclusionsIt was concluded that high glucose induced the expression of KLF6 in human LECs,and KLF6 transiently stimulated the expression of target genes TGFB1,TGFBRl,COLlAl and HSP47 which were mainly involved in the mechanism of EMT.
