2.Histochemical studies on Echinorhynchus gadi (Acanthocephala).

The distribution of glycogen, polysaccharide, mucopolysaccharide, lipid and nucleic acid has been studied in Echinorhynchus gadi(Acanthocephala). The results were summarized as follows: Glycogen and polysaccharide was demonstrated by Bauer PAS reaction technique and was found in fertilization membrane in ovum, central nuclear mass in acanthor and lemnisci, hypodermis in cystacanth. Mucopolysaccharide was demonstrated by Mowry alcian blue staining technique and was found in outer membrane, fibrillar coat, fertilization membrane and inner membrane in acanthocephalan ova. Lipid was demonstrated by Smith Nile blue stain and Lison Sudan black B staining technique and was found roughly parallel to that of polysaccharide. Nucleic acid was demonstrated by Rosenbeck Feulgen reaction, Taft methylgreen-pyronin stain and Diengdoh acridine orange staining technique and found in central nuclear mass in acanthor, also, was found in lemnisci, proboscis and hypodermis in cystacanth.

3.A Case of CHAP Syndrome.

CHAP syndorme consists of choreoathetosis, orofacial dyskinesia, affective change, hypotonia, and pseudobulbar signs. CHAP syndrome was firstly des- cribed with onset 1 to 7 days after induced profound hypothermia and complete circulatory arrest for con- genital heart lesion repair. Since then, only a few cases have been reported until now. Case We report a patient who demonstrated all of the elements of the CHAP syndrome in severe cachexic state. Brain SPECT(99mTC-ECD) shows decreased perfusion in left fronto-parieto-temporal lobe. Comments The pathogenesis of CHAP syndrome is obscure. However our case led us to suggest that more essential component pre- disposing to the CHAP syndrome is hypoxia rather than hypothermia. Furthermore, it seems to be related with circulatory insufficiency in extrapyramidal nervous systems.

5.Development of a novel antigen capture-ELISA using IgY against porcine interleukin-6 and its application.

Interleukin-6 (IL-6) is introduced as a marker of disease. At present, a variety of method may be used to quantify expression of this protein. Antigen capture-ELISA is a sensitive and accurate quantification method previously used with ovine, rat, and human IL-6 proteins. However, it has never been reported to quantify porcine IL-6 protein using capture ELISA. In this study, we generated and characterized a set of IgY and mono-specific polyclonal antibodies to recombinant porcine IL-6 (rpIL-6), and combining these with a sensitive and specific capture-ELISA for a diagnostic purpose. cDNA encoding the mature protein coding region of porcine IL-6 was cloned and expressed with pQE-30UA expression vector. rpIL-6 was then expressed and purified by using Ni-NTA resin. Protein mass of 24 kDa was found with SDS-PAGE and the identity of the protein was confirmed by Western-blot. Production of polyclonal antibodies against rpIL-6 was performed using the purified rpIL-6 in mice and hens. An antigen capture-ELISA was developed with the antibodies after their extraction. To compare the IL-6 level in the different sanitary state of farms, pig sera were randomly collected and concentration of IL-6 in the sera was measured with the antigen capture-ELISA. The capture-ELISA with the optimal concentration of antibodies, in this study, was able to detect about 10 ng/ml of rpIL-6. IL-6 levels determined with the capture-ELISA in pig sera showed positive correlation with the sanitary states of the farms. These results suggested that the developed antigen capture-ELISA could be a good tool for the screening of microbial infection in pig farms.

9.Measurement of Posterior Laxity of Normal Knee in Korean Adults by means of KT-2000 Knee Ligament Arthrometer

We measured normal range and mean value of the posterior laxity of normal knee which might be reference for objective and accurate diagnosis, decision of operative indication and position of knee during fixation of ligament, and role of quadriceps building in posterior cruciate ligament injury patients. These results were obtained by measurement and analysis of posterior laxity of normal knee by means of KT-2000 Knee Ligament Arthrometer. We measured 184 Korean adults (100 males, 84 females), and their average age, height, weight, and thigh circumference were 29.8 years, 165.8cm, 59.8kg, 41.2cm, respectively. At 25 degrees of knee flexion, mean posterior displacement of normal knee was 21.2mm with 15lb posterior tibial loading, 2.41mm with 20lb posterior tibial loading. And when quadriceps were tense, laxity was 0.79mm and 0.95mm at the same degree of knee flexion. At 75 degrees of knee flexion, the results were 0.57mm, 0.58mm, 0.26mm, and 0.28mm respectively. Right to left difference of posterior displacement with 20lb posterior tibial loading was 0.07mm at 25 degrees of knee flexion and 0.06mm at 75 degrees of knee flexion. We thought that above 75 degrees of knee flexion is suitable for knee position at the time of graft fixation because 75 degrees of knee flexion markedly decreased(75%) posterior displacement compare to 25 degrees of knee flexion. When quadriceps were tense, remarkable decrease of posterior laxity of normal knee was checked(58%), so we noticed quadriceps building is important to management of posterior cruciate ligament injury patients.

10.The Effect of The Coadministration of Vitamin E in Cyclosporine A(CsA) Induced Acute Nephrotoxicity in Rats.

Cyclosporine A(CsA) induced nephrotoxicity is a common clinical problem in the patients who underwent organ transplantation and subsequent immunosuppressive therapy. The precise pathophysiology of CsA induced nephrotoxicity still remains uncertain though many hypothesized mechanisms were reported. Therefore, no effective strategy is currently available to counter the nephrotoxic effects of CsA while cyclosporine G or cyclosporine microemulsion was tried to reduce the toxic effects. Recently, CsA was reported to induce lipid peroxidation in renal cortical mitochondria, which may cause mitochondrial damage. To examine the hypothesis of lipid peroxidation in the pathogenesis of acute CsA induced nephrotoxicity, antioxidant vitamin E was co-administerated in the CsA induced acute nephrotoxicity. Sprague Dawley male rats weighing from 250 to 400 gm were heminephrectomized 7 days prior to the experiments. The rats were divided into 3 groups and each group consisted of 7 rats. In group I(control group), the rats were given sterilized olive oil intraperitoneally, in group II(CsA group), CsA 50 mg/kg of body weight/day intraperitoneally, and in group III(CsA with vitamin E), CsA 50 mg/kg of body weight/day intraperitoneally with vitamin E 10 mg/kg of body weight/day intramuscularly for 7 days respectively. From the 6th day, all experimental animals were placed in a metabolic cage collecting urine for the measurement of 24-hours urine thromboxane B2, the metabolite of thromboxane A2. On the 7th day, at the sacrifice of the experimental animals, blood samplings for the measurement of blood CsA level and serum creatinine and left nephrectomy for morphological study were performed. Comparison of mean serum creatinine levels between 2 study groups revealed 171.7+/-164.2 micro Mol/L in group II and 53.4+/-13.9 micro Mol/L in group III(p<0.05). Mean 24-hour urine thromboxane B2 levels were 39.0+/-6.9 ng/24 hours in group I, 74.8+/-22.6 ng/24 hours in group II, and 34.5+/-8.0 ng/24 hours in group III. Urine thromboxane B2 was significantly(p=.0026) lower in group III. The differences of morphological changes in group II and III can be summarized as a diminution of PAS(+) granules under the light microscope and a diminution of the numbers of secondary lysosomes in the proximal tubular epithelial cell under the electron microscope. In conclusion, this animal study provides an evidence that CsA-induced acute nephrotoxicity is related with lipid peroxidation and the antioxidant, vitamin E was considered to ameliorate CsA-induced acute nephrotoxicity in rats.
