4.HCV-RNA testing of 28098 blood donors and follow up of some positive cases

Objective To evaluate the significance of HCV-RNA testing of blood donors by using reverse transcriptase polymerase chain reaction (RT-PCR) analysis, and investigate HCV “window period” in blood donors.Methods Test blood donors for HCV-RNA by RT-PCR, and some positive cases were followed up. Results Among 28098 routine test negative blood donors, 77 (0.27%) were HCV-RNA positive. Eleven of the 77 HCV-RNA positive donors were followed up and 9 of them were later found to have increased ALT and/or positive anti-HCV. The interval between positive HCV-RNA test and abnormal serological tests was at least 4 weeks.Conclusion RT-PCR can identify HCV infected donors at an early stage. This test should become a routine test for blood donors to further ensure blood safety.
5.Regulatory T cells in peripheral blood of chronic renal insufficient

Objective:To investigate the significance of regulatory T cells in peripheral blood of chronic renal insufficientce.Methods:The peripheral blood samples were collected from 40 patients with chronic renal insufficient.The ratios ot CD4+T cell in lymphocyte and CD4+CD127-Treg and CD4+CD25+CD127-Treg in CD4+T were detected by flow cytometry.Results:The number of CD4+T in lymphocyte of chronic renal insufficient was higher than in healthy control group and there wasn’t significantly difference of the CD4+CD25+CD127-Treg ratios in CD4+T between chronic renal insufficience and healthy central.The ratio of CD4+T cells in lymphocytes of chronic renal insufficience was lower than in healthy control group except compensatory stage.There was no correlation between CD4+T cell ratios in lymphocytes,CD4+CD127-Treg or CD4+CD25+CD127-Treg ratios in CD4+T cells and the values of BUN,Cr among the hypertension patients.Conclusion:The number of CD4+T cells increases,and CD4+CD127-Treg decreases in the patients with chronic renal insufficience and their immune functions are shown in disoroler .
6.Diagnostic Value of Pathogenic Detection on Acute Respiratory Infection in Children by Multi-IgM Investigation

Objective To investigate the pathogens of acute respiratory infection of children.Methods A total of 159 children with acute respiratory infection who were hospitalized in our department from August 2005 to January 2006 were involved in this study.The serum IgM antibody of 18 pathogens were detected by indirect immunofluorescence test.The 18 pathogens included respiratory syncytial virus(RSV),adenovirus(ADV),influenza A(H1N1,H3N2)and B viruses,parainfluenza viruses(PIV) type 1,2,3 and 4,coxsackie virus B1(CBV1),coxsackie virus A7(CAV7),echovirus(ECHO7),haemophilus influenzae(HI),klebsiella pneumoniae(KP),bordetella pertussis(BP),bordetella parapertussis(BPP) and legionella pneumophila serotype 1 and 12.Results The evidence of specific IgM was obtained in 103 of 159 patients(64.78%).Influenza A was found in 66 cases(64.08%),influenza B in 49 cases(47.57%),enterovirus in 26 cases(25.24%),RSV in 18 cases(17.48%),PIV in 11 cases(10.68%),and co-infection in 66 cases(64.08%),1/ 3 of them were co-infected with influenza A and B.Conclusions Viruses are the most common agents of acute respiratory infection.Influenza virus is predominant among them.

OBJECTIVETo investigate the therapeutic effect of primary reconstruction of skin avulsion injury with bilateral anterolateral thigh flaps combined with thorax umbilicus flap or latissimus dorsi flap.
METHODSFrom June 2005 to Aug. 2011, 4 cases with skin avulsion injury on both feet were treated. The bilateral anterolateral thigh flaps, including with anterolateral thigh cutaneous nerves, were transferred to cover the feet plantar. The thorax umbilicus flap or latissimus dorsi flap were used to cover the feet dorsum.
RESULTSAll the skin avulsion injury were reconstructed primarily. All the flaps survived completely with good cosmetic and functional results. The patients were followed up for 6 months to 2 years with good sensory recovery (two point discrimination: 14-18 mm).
CONCLUSIONThe skin avulsion injury on both feet can be primarily reconstructed by bilateral anterolateral thigh flaps combined with thorax umbilicus flap or latissimus dorsi flap.

9.Genome-wide analysis of DNA 5-hydroxymethylcytosine in uremia patients using hMeDIP-chip

Objective To investigate the role of the 5-hydroxymethylcytosine (5-hmC) DNA modification in the onset of systemic lupus erythemosus (SLE),we compared tihe levels 5-hmC between SLE patients and normal controls.Methods With informed consent,whole blood was obtained from patients,and genomic DNA was extracted.Using hMeDIP-seq analysis and validation by quantitative real-time quantitative polymerase chain reaction (RT-PCR),we identified the differentially hydroxymethylated regions that were associated with SLE.Results There were 1 701 genes with significantly different 5-hmC levels at the promoter region in the SLE patients compared with the normal controls.The CpG islands of 3 826 genes showed significant difference at 5-hmC levels in SLE patients compared with the normal controls.Out of the differentially hydroxymethylated genes,three were selected for validation,including TREX1,CDKN1A,and CDKN1B.The hydroxymethylation levels of these three genes were confirmed by quantitative RT-PCR.Conclusion Our studies indicate that there are significant alterations of 5-hmC in SLE patients;these differentially hydroxymethylated genes may contribute to the pathogenesis of SLE.Such novel findings show the significance of 5-hmC as a potential biomarker or promising target for epigenetic-based SLE therapies.
10.Clinical application of lateral superior genicular composite tissue flap.

OBJECTIVETo investigate the clinical effect of lateral superior genicular composite tissue flap for tissue defect.
METHODSThe axis line of flap is the lateral thigh vertical midline. The cutaneous branch is inserted 4 cm near the femoral lateral epicondylus. The anterior border is the elongation line along patellar lateral border. The posterior margin is the hinder margin of femoral biceps. The lower border is the horizontal line along the upper line of patella. The composite flaps were used in 18 cases with soft tissue defects in extremities, 11 cases with clacaneus tenden defects and 16 cases with bony nonunion. Results From Mar. 2002 to Sept. 2013, 45 cases were treated with the composite tissue flaps. The flaps size ranged from 6 cm x 3 cm to 17cm x 9 cm. All the flaps survived completely. Blood supply crisis happened in 2 cases, which was released by reanastomosis. The patients were followed up for 1 - 2. 5 years with satisfactory aesthetic and functional results. All the bone defect and nonunion were healed. Good healing was also achieved in donor sites. 8 months after operation, knee joint function is evaluated as good by hospital special surgery knee score (HSS).
CONCLUSIONLateral superior genicular compostie tissue flap can be used to reconstruct soft tissue defect, bone defect and tenden calcaneus defect in one stage.
