1. Preparation and characterization of DCA-LTSL

OBJECTIVE: To prepare decitabine long-circulating thermosensitive liposome(DCA-LTSL), screen and optimize its formulation, and evaluate its properties. METHODS: DCA-LTSL was prepared by modified reverse phase evaporation method, and the encapsulation efficiency was determined by minicolumn centrifugation-HPLC method. The influences of the concentration of PC, the weight ratio of PC to Chol, the weight ratio of DCA to PC and buffer salts were studied. Orthogonal design was adopted to obtain the optimal prescription using encapsulation efficiency as the main index. RESULTS: The best prescription was as follows; the concentration of PC was 5 g · L-1, the weight ratio of PC to Chol was 4 : 1, the weight ratio of DCA to PC was 40: 1 and the pH value of aqueous phase was 7.0.The encapsulation efficiency of DCA-LTSL was (44.50 ± 1.08)% . The Zeta potential was -8.34 mV. The mean diameter of DCA-LTSL was (140.25 ± 2.40)nm. The burst release effectiveness of DCA-LTSL was 42-43°C. CONCLUSION: The formulation of DCA-LTSL has been optimized with excellent in vitro drug release characteristics at the phase-thransition temperature.
2.An empirical study of skill points in pathology courses

Objective To investigate the characteristics and distribution of class teaching skill points in state high quality course pathology using Flanders interaction analysis system to analyze the teaching video of pathology course.Methods Teaching videos of state high quality course (pathology course) between 2006 and 2012 were analyzed by Flanders interaction analysis system.Frequencies of using teaching skill,presentation skill and interaction skill were statistically analyzed.Results The study found that frequencies of using teaching skill points in pathology course were decreased in the following order:presentation skill (53.94%),interaction skill (26.64%) and teaching skill (19.42%).Sensory focusing was most commonly used in presentation skill.Monitoring and expressing concern were most commonly used in interaction skill.Gestures and cases were most commonly used in teaching skill.Conclusions Teachers in pathology teaching should correctly handle the relationship among the presentation,interaction and teaching skills according to the teaching objectives and teaching content.
3.Study on the relationship of red blood cell distribution width and cardiovascular events in patients with A-cute ST-segment elevation myocardial infarction

Objective Recent studies suggest that the red blood cell distribution width ( RDW) may play a role in the diag-nosis and treatment of cardiovascular disease.The aim of this study was to investigate the correlation of red blood cell distribution width (RDW) with cardiovascular events in patients with Acute ST-segment elevation myocardial infarction (ASTEMI) in order to provide the basis for improving the diagnosis level and therapeutic effect. Methods Retrospective study was made on the clinical data of 189 patients with ASTEMI enrolled in Sichuan Provincial People′s Hospital from January 2013 to March 2014.The survival rates of patients with ASTEMI with different RDW were estimated by Kaplan-Meier and compared by Log-rank test and the prognosis factors were inves-tigated by Cox proportional hazard regression model. Results 97 patients′RDW levels were more than 13.7% and 92 patients′RDW levels were less than or equal to 13.7% in 189 ASTEMI patients. The hs-CRP, BNP, LVEF and HDL-C were significantly different be-tween two groups (P<0.05).The rates of recurrent myocardial in-farction, heart failure, sudden death cardiac in patients with highRDW were higher than that in patients with low RDW (22.7%vs 8.7%, 27.8%vs 10.9%, 15.5%vs 5.4%)(χ2 =6.915, 8.632, 5.019, P<0.05) .There were 60 patients with high RDW who experienced cardiovascular events and the 1-year cardiovascular event-free survival rate was 38.1%, and 20 patients with low RDW cardiovascular events was 79.3% in follow-up period (χ2 =30.959, P<0.001).Multivariate cox regression showed that age>65 (HR=2.43, 95%CI:1.09~5.44) and RDW>13.7%(HR=2.20, 95% CI:1.10~4.43) were risk prognostic factors. Conclusion There is a certain correlation between RDW and cardiovascular e-vents in patients with ASTEMI.The ASTEMI patients with high RDW hold higher risk of cardiovascular events and poorer prognosis.
4.Relationship between GABAA receptor and the analgesic、hypnotic and amnesic effects induced by GABA

Aim To investigate the effect of the GABAA receptor on the analgesia,hypnosis and amnesia induced by GABA in mice.Methods GABA was intracerebroventricularly(icv) injected and securinine,a GABAA receptor antagonist,was intravenously(iv) injected after the GABA intracerebroventricularly injection.Then four techniques including hot-plate test,acetic acid-induced writhing test,awaken test as well as step-through test & step-down test were employed to evaluate the effects on the pain index in hot-plate test(HPPI),the writhing times,the sleeping time(ST) and the latency & the number of errors.Results GABA increased the HPPI and ST,but decreased writhing times and the latency & increased the number of errors in conscious mice;the above effects are dose-dependent.Securinine could antagonize the effects stimulated by GABA.Conclusion GABA lead to analgesia,hypnosis and amnesia.GABAA receptor might be the important target for these effects.
5.Effects of Several Factors on the Chlamydospores Production of Trichoderma aureoviride T-33

The effects of several factors on the chlamydospores production of Trichoderma aureoviride T-33 during the fermentation were researched.Based on the results above, the orthogonal test was made to screen out the best prescription.The results showed that, the best single-factor conditions for the chlamydospores production of T.aureoviride T-33 were, liquid culture of oat powder, 30℃, pH4.0, 120r/min, 24 hours oscillate incubating as well as liquid culture volume of 80mL/bottle when the 250mL size triangle bottle was used.The result of orthogonal test showed that, the best prescription for temperature, pH and oscillating speed was 30℃, pH4.0 and 140r/min.3.37?10~ 7 spore/mL chlamydospores were obtained at this combined condition.
6.Effects of tree species on polysaccharides content of epiphytic Dendrobium officinale.

To reveals the effects of tree species on polysaccharides content of epiphytic Dendrobium officinale. The polysaccharides content of D. officinale attached to living tress in wild or stumps in bionic-facility was determined by phenol-sulfuric acid method. There were extremely significant differences of polysaccharides content of D. officinale attached to different tree species, but the differences had no relationship with the form and nutrition of barks. The polysaccharides content of D. officinale mainly affected by the light intensity of environment, so reasonable illumination favored the accumulation of polysaccharides. Various polysaccharides content of D. officinal from different attached trees is due to the difference of light regulation, but not the form and nutrition of barks.

7.Optimization of b-value in breast diffusion-weighted 1.5 T MR imaging ZHANG Jing

ObjectiveTo optimize the b-value of breast diffusion-weighted MRI (DW-MRI) at 1.5T by applying a range of b values and comparing the apparent diffusion coefficient (ADC) and signal-to-noise ratio (SNR) on a phantom,disease-free breast tissues,and benign and malignant lesions.Methods A phantom and 32 women with pathologically confirmed malignant ( 18 ) and benign ( 14 ) lesions were examined using EPI-DWI with different b values on a 1.5 T MR scanner.The b-value of EPI-DWI was 0,50,100,200,400,600,800,1000,1200,1400,1600,1800,2000,2200,2400,and 2600 s/mm2,respectively.The SNR and ADC values of the phantom,disease-free breast tissues,and benign and malignant lesions were measured.The correlation between the b-value and ADC or SNR of each image was analyzed.ResultsThe SNR of DWIdecreased as the b-valueincreased,showing aninversecorrelation (r =-0.802,P <0.01 ).The ADC values of benign and malignant lesion decreased as the b-value increased (r =-0.923 and -0.855,P <0.01 ).The maximum difference in ADC between malignant and benign lesions was observed when the b-value is between 800 and 1000 s/mm2 and diminished when the b-value was greater than 1400 s/mm2.ConclusionFor good image quality and valid differentiation between malignant and benign lesions,the optimized b-value of DWI at 1.5 T is between 800 s/mm2 and 1000 s/mm2.
8.Quantitation of mitochondrial DNA A1555G mutation by real time amplification refractory mutation system quantitative PCR

Objective To develop a real time amplification refractory system(RT-ARMS-qPCR) quantitative PCR method with SYBR Green I to assess the mtDNA A1555G mutation. Methods A specific fragment flanking mtDNA 1555 site was amplified with PCR and ligated into a pGEM Easy T vector. Serial dilutions of the plasmid DNA were quantified the actual copy numbers were assessed using RF-ARMS-qPCR with SYBR Green I. RF-ARMS-qPCR was established with mismatched base pairs at 3' in the primer todetect the copy number of mtDNA containing wild or mutant mtDNA. The specificity of amplified products was checked by melting curve analysis. Results The intra- and interassay variation was 1.34% and 1.96%, respectively when the assay was used to detect 1 copy/ul recombinant template of plasmid. Thequantitative standard curve showed that the assay had good linear correlation from 102 copies/ul to108 copies/ul. This assay could be served for the quantification of other samples. There was significantcorrelation between frequency of mutant mtDNA and phenotype (r=0.771, P = 0.003) in hearing lossgroup. Conclusions The established assay can be used to detect quantitatively mtDNA A1555G mutation byRF-ARMS-qPCR. This assay is specific, stable and accurate. There is significant correlation betweenquantification of mtDNA AI555G and the severity of hearing loss.
9.Improved methods for monitoring sleep state and respiratory rhythm in freely moving rats.

OBJECTIVETo improve the method for monitoring sleep state and respiratory rhythm of SD rats, providing a solution for rats' chewing on the wires, signal loss and instability problems in the animal model of sleep apnea syndrome (SAS).
METHODSWe improved monitoring electrodes of both electrocorticogram (ECoG) and electromyogram (EMG), signal circuit and animal operation.
RESULTSOperation time was shortened and wound exposure time was reduced, which made it easier for postoperative recovery. The ECoG and EMG signals were more stable with sharp image, and signal circuit lines had better conductivity and material durability, achieving continuous monitoring for a long time and high success rate. We could precisely distinguish the sleep wake state and the sleep apnea events in rats according to these signals.
CONCLUSIONThe improved method is more reliable and practical to test the small animal model of SAS, and is more easily to operate and the signals are more stable.
