Objective:To evaluate the therapeutic effect of combined procedure of surgical left ventricular repair (LVR) and skeletal myoblast transplantation for congestive heart failure after myocardial infarction. Methods: Forty-eight male adult Lewis rats, with a four-week old myocardial infarction created by left coronary artery ligation, were randomized into 4 groups: group Ⅰ (n=10) received phosphate-buffered saline (PBS) injection into the border zone of the infarction; group Ⅱ(n=11) received myoblast injection ; group Ⅲ(n=13) received left ventricular repair and simultaneous PBS injection; and group Ⅳ (n=14) received left ventricular repair and myoblast injection. Echocardiography was performed once a week until the fourth week after the treatment. Results: Left ventricle end diastolic diameter (EDD) and end systolic diameter (ESD) declined significantly one week after the LV repair in group Ⅲ and Ⅳ, then increased as time passed; on the fourth week, there were no statistical differences between group Ⅰ,Ⅱ and Ⅲ, but group Ⅳ was still much smaller than the other 3 groups(P

BACKGROUND:Recent studies have shown that platelet rich plasma has strong osteogenic ability and it has been widely used in the field of department of orthopedics and department of stomatology, but experimental study on avascular necrosis of the femoral head has been not reported.
OBJECTIVE:To observe the effect of platelet rich plasma gel combined with marrow core decompression in the repair of aseptic necrosis of rabbit femoral head.
METHODS:A total of 25 New Zealand white rabbits were obtained. Without dislocation of the hip joint, aseptic necrosis model was established by using liquid nitrogen cryotherapy on bilateral femoral head of rabbits. At 2 weeks after model establishment, rabbit models were randomly assigned. Model group with nine rabbits did not receive any treatment. Core decompression group with eight rabbits underwent core decompression treatment. Combination group with eight rabbits underwent core decompression combined with platelet rich plasma gel treatment. At 8 weeks after model induction, tissue sections of femoral head specimens were col ected and subjected to MRI and hematoxylin-eosin staining.
RESULTS AND CONCLUSION:(1) MRI examination:The model group showed low signal area of different forms in femoral head fat high signal, exhibited annular, ribbon and focal shapes. No evident changes in osteogenesis, no apparent minification of the defect cavity were detected at defect site in the core decompression group, showing long T1 and T2 signals. Defect cavity disappeared, and new bone of short T1 and T2 signals fil ed in the combination group. (2) Hematoxylin-eosin staining:Articular cartilage defect and repair existed in the model group, showing osteoarthritis-like changes. Empty bone lacuna of bone trabecula, hematopoietic celland adipocyte necrosis in bone marrow were detected in the core decompression group. Empty bone lacuna of bone trabecula, focal necrosis scattered in bone marrow were visible in the combination group. Results suggested that the outcomes of platelet rich plasma gel combined with core decompression were better than core decompression alone in treatment of rabbit models of femoral head necrosis.

Objective To observe the effect of laparoscopy combined with choledochoscopy in treatment of cholelithiasis clinical effect .Methods 80 cases diagnosed as cholelithiasis cases were divided into control group and observation group with 40 cases in each group .The control group used the traditional laparotomy incision gallbladder stone ;observation group were treated by laparos-copy combined with choledochoscopy .Results In the observation group ,the operation and the hospital stay were significantly shor-ter than the control group ,operation curative effect was obviously superior to the control group ,the incidence of postoperative com-plications was significantly lower than the control group .Conclusion The use of laparoscopy combined with choledochoscopy tech-nology in treatment of gallstones have good clinical effect .

Nucleic acids, as a next generation of biotechnology drugs, not only can fundamentally treat diseases, but also own significant platform characteristics in view of technology and production. Therefore, nucleic acid-based drugs have broad clinical applications in biomedical fields. However, nucleic acids are degradable and unstable, and have very low intracellular delivery efficiency in vitro and in vivo, which greatly limits their applications. In recent years, ionizable lipid-based lipid nanoparticles have shown promising application potentials and have been successfully applied to COVID-19 (Coronavirus Disease 2019) vaccines in clinic. Lipid nanoparticles demonstrate high in vivo delivery efficiency and good safety profile due to their unique structural and physicochemical properties, which provides many possibilities for their clinical applications for nucleic acid delivery in the future. This review focused on the characteristics of nucleic acid drugs and their delivery barriers, and discussed the approved nucleic acid drugs to illustrate the key aspects of the success of their delivery carrier system. In addition, problems to be solved in the field were highlighted.

Icarrin is the main monomer extracted and purified from Epimedium koreanum Nakai. Using dependentcell lines and LDH release assay,the synergistic effects of icarrin on inducing IL-2, 3, 6 and enhancing NK,LAK cell activity were studied. Results showed that icarrin can induce tonsil mononuclear cells to produce IL-2, IL-3,and IL-6 in collaboration with PHA in a dose-dependent manner. IL-2 and IL-6 peaked at 48h after reaction,and IL-3 peaked at 72h. Icarrin enhanced NK and LAK cell activity dose-dependentlylthe LAK activitypeaked on the 5th day. These results suggested that icarrin may be an effective BRM agent

Objective To express and purify the killer immunoglobulin-like receptor KIR3DL1 extracelluar domain.Methods pUC57-KIR3DL1 was used as template,KIR3DL1 extracelluar domain was amplified by polymerase chain reaction(PCR)and cloned into pGEM-T vector with A-T cloning technique.After DNA sequence analysis,the target fragment was inserted into the prokaryotic expression vector pET28a-DsbA to construct the recombinant vector pET28a-DsbA /KIR3DL1.The recombinant plasmid was transformed into E.coli BL21(DE3),and induced with IPTG.Bacterial pellets were resuspended in 8M urea and centrifuged to remove the insoluble material.The crude extract was purified by passing over a Ni-NTA-agarose column.After the inclusion body flowing through the Ni-NTA-agarose affinity chromatography was refolded successfully,it was purified by Superdex75 gel filtration and the purification effects of the fusion protein were identified by SDS-PAGE and western blot.Result Some fusion proteins were expressed in the supernatant,and the others were expressed in the form of inclusion bodies.The purity of fusion protein was over 95% after purification under denaturing condition.Conclusion The highly efficient expression of KIR3DL1 extracelluar domain laid the foundation for the further studies on exploration of the mechanism of immunization recognition between KIR3DL1 and its ligand.

Objective To evaluate the changes and clinic countermeasures of cough and expectoration in the patients with lung cancer after airway stent placements. Methods With the fibrobronchoscope, metal stents were placed in tracheas, main bronchia or right intermediary bronchia. It was compared that the degrees of cough and expectoration difficulties, the volumes and mucin content of sputa and lung functions before the placements, within 3 d and 1 to 2 months after operation. Results Within postoperative 3 days, the patients coughed more frequently, but VC, FEV_(1) and PEF were improved significantly. In postoperative 1 to 2 months, the degrees of cough were close to preoperative degrees, lung functions kept on improved conditions, but expectoration difficulties, volumes and mucin contents of sputa increased markedly, especially in the patients with tectorial membrane stents. Proper stents and reasonable treatment including use of antitussive, apophlegmatisant and antibiotics could decrease the degrees of cough within postoperative 3 days, and expectoration difficulties, volumes and mucin contents of sputa in postoperative 1 to 2 months. Conclusion To the patients with malignant central airway constriction, metal stent placement would improve ventilative functions but bring difficulties for cough and expectoration. The side complications would be relieved through corresponding treatments.
