2.Mechanism of radiosensitizing effects of endostatin on H-520 human lung squamous cancer cells

Objective To investigate the mechanism of radiosensitizing effects of endostatin on H-520 human lung squamous cancer cells.Methods H-520 cells was treated with endostatin and/or radiation.Colony-forming assays were used to indicate the radiosensitising effects.Cell cycle distribution and expression of phosphor-p38-MAPK were assayed by FCM,and cyclin D1,cdk2,cdk4 and survivin mRNA leveh were assayed by RT-PCR.Phosphor-Akt was evaluated by Western-blotting.Results Combination of endostatin and irradiation inhibited the proliferation of H-520 cells.According to the colony-forming assays,the D0,Dq,D10 and SF2 values of the combination groups were much lower than those of irradiation groups.The sensitization enhancement ratio(SER)was 1.51.G2/M arrest occurred after 4 Gy irradiation.The gene expression of cyclin D1,cdk2,ckd4 and survivin and phosphor-Akt protein were down-regulated after treatment.The expression of phosphor-p38-MAPK protein was also down-regulated after treatment with 200 μg/ml endostar.Conclusions Endostatin inhibits the growth of H-520 cells and radiosensitizes the cells by induction of G0/G1 arrest,cell apoptosis and down-regulation of gene expression of cyelin D1,cdk2,cdk4 and reduces the phosphorylation of Akt and p38-MAPK.
3.Information Security in TCM Budget Monitoring Platform

With constant development and application of new generation information technology such as big data, cloud computing and Internet of Things, traditional management style and thought patterns of TCM are being changed. It is particularly important to introduce information security into budget management of TCM projects. This article discussed security factors in TCM budget monitoring platform, organized key contents of information security construction, built information security model for monitoring platform, and analyzed security strategies for the construction of TCM budget monitoring platform, with a purpose to guarantee effective implementation of budget information management measures of TCM projects.
4.Spinal cord ependymoma presenting as acute paraplegia and subarachnoid hemorrhage: a case report and review of literature

Acute paraplegia is a rare presentation for a spinal cord ependymoma. Subarachnoid hemorrhage
(SAH) due to spinal ependymoma is also very rare. We report a 32-year-old woman who presented
with acute paraplegia and typical clinical signs of SAH with normal cerebral angiography, and further
diagnostic work-up revealed an spinal cord ependymoma as the source of the hemorrhage. There
is evidence that some spinal cord ependymomas have intratumoral hemorrhage, but most of these
bleedings occur without symptoms. We discuss the clinical and neuroradiological findings of this rare
case and review the literature related to this unusual presentation.431
5.Low-density lipoprotein cholesterol levels and lipid-lowering treatment status among young and middle-aged ultra-high-risk patients with acute coronary syndrome in China.

Objective: To assess low-density lipoprotein cholesterol (LDL-C) levels and use of lipid-lowering treatment among young and middle-aged ultra-high-risk patients with acute coronary syndrome (ACS) in China. Methods: The study was based on the"Improving Care for Cardiovascular Disease in China (CCC)-ACS"project, a collaborative registry by and Chinese Society of Cardiology (CSC) and the American Heart Association. Hospitalized-patients with ACS were consecutively enrolled from 159 tertiary and 82 secondary hospitals across China, related clinical information was collected. This study included young and middle-aged hospitalized patients (18-59 years) with ACS from November 2014 to December 2019 registered in CCC-ACS project. Ultra-high-risk was defined according to Chinese expert consensus on lipid management of ultra-high-risk atherosclerotic cardiovascular disease (ASCVD) patients of CSC. The mean LDL-C levels at admission, pre-hospital lipid-lowering therapy and proportion of patients with LDL-C target achieved were analyzed. Results: A total of 42 230 patients younger than 60 years with ACS were included in this study. The mean age was (50.4±6.9) years, and 86.8% (36 676/42 230) of the ACS patients were male. Among them, 86.9% (36 687/42 230) met the criteria of ultra-high-risk. The mean level of LDL-C at admission was (2.8±1.0)mmol/L, only 5.3 % (1 948/36 687) patients achieved the targeted goal of LDL-C<1.4 mmol/L. Among the ultra-high-risk ASCVD patients, 17.5% (6 430/36 687) received lipid-lowering drugs before hospitalization, 96.4% (6 198/6 430) of whom received statins monotherapy. Among patients receiving pre-hospital statins, only 9.9% (626/6 323) patients reached an LDL-C<1.4 mmol/L at admission. Conclusions: The majority of young and middle-aged hospitalized patients with ACS are ultra-high-risk patients for ASCVD in China. Pre-hospital lipid-lowering drugs use is lower in these ultra-high-risk ASCVD patients and most patients do not reach the new LDL-C target level at admission.

6.Radiosensitization of microRNA-17-92 on human mantle cell lymphoma cells in vitro

Objective To investigate the effects of microRNA-17-92 on radiosensitivity of human mantle cell lymphoma cells. Methods Tetracycline-regulated pRevTet-On expression system was established to generate cell line Z138c-miR-17-92 with over-expressed miR-17-92 and cell line Z138c-TMP2. Cell proliferation was measured by 3 H-TdR incorporation and viable cell counting stained with typan blue. Cell cycle distribution was analysed by flow cytometry(FCM). Results More viable and proliferous cells were counted in group miR-17-92,when exposure dose was greater than 2 Gy and incubation time was longer than 48 h under the same condition (t = -3. 12 and -3.28,P <0. 05). The percentage of G2/M cells in group TMP2 was increased while no obvious cell cycle arrests were found in group miR-17-92 at 2 and 4 Gy (t = 2. 885, P < 0.05 ). When cells were incubated for 96 h, higher percentage of propidium iodide (PI) positively stained cells were found in group TMP2 (24. 02% vs. 36. 16% )compared with group miR-17-92 (6.49% vs. 11.39% ) at 2 and 4 Gy, respectively( t = - 17.59, - 4. 972, P < 0.05 ).Conclusions Overexpression of microRNA-17-92 decreased the radiosensitivity of human mantle cell lymphoma cells by inhibition of cell cycle changes and cell apoptosis.
7.Current Situation and Thinking of Statistical Management of TCM

Objective To understand the current situation of statistical management of TCM;To analyze existing problems; To provide references for policy formulation and measure implementation for statistical management of TCM. Methods Literature research, field research, comparative analysis, questionnaire surveys, expert consultation, and Fisher's exact test were used in the study. Results It was found that China’s statistical management of TCM had the problems of imperfect system, unestablished investigation system, incomplete statistical index system, and nonstandard statistical management. No statistical difference was found in the three regions of east, middle and the west. Conclusion Management and investigation system should be established in statistical management of TCM; pilot projects should be conducted before they are spread;information sharing and data mining should be strengthened.
8.Radiosensitization of cetuximab on human tongue cancer cell line Tca8113

Objective To investigate the mechanism of radiosensitization by cetuximab (C225) on human tongue cancer Tca8113 cell line in vitro.Methods Tca8113 cell line with and without C225 treatment received 6 MV X-ray irradiation of different doses (0, 2,4,6, 8 and 10 Gy). Cell proliferation,cell-cycle distribution and clonogenic survival were analyzed through cell counting,MTT,colony formation assay,and flow cytometry,respectively.Results After irradiation of different doses,the growth inhibition rates in C225 group were higher than control (t =- 15.6 - -3.0,P<0.05),the radiobiological parameters (D0,Dq,N,and SF2 ) in C225 group were lower than control so that SER of C225 group was 1.353,and the proportions of G0/G1 cells in C225 group were higher than control ( t =-7.64,-7.89,-4.78,P <0.05 ) at 4,6,8 Gy.When the irradiation doses increased,the early phase apoptosis in both groups increased at first and then decreased with the maximum difference at 4 Gy [(7.96±0.36)% in C225 group and (4.13 ±0.29)% in control group,t =-12.75,P<0.01 ].Conclusions C225 has radiosensitization effect on Tca8113 cell line,possible through Go/G1 arrest and induction of apoptosia.
9.Changes of key enzymes involved in testosterone synthesis in testes of early diabetic mice

Objective Type 1 diabetes mice model was established to investigate the changes of key enzymes involved in testosterone synthesis in testes of early diabetic mice.Methods Tatolly 20 male C57 mice were randomly divided into two groups:control and diabetic groups,and the diabetes mice were ip administered with a single dose of 150 mg/kg Streptozotocin.Four weeks after confirmation of diabetic model,the serum and testis were collected for further study.The qRT-PCR method was used to measure the expression of LHR and steroidogenesis synthetase StAR,P450scc,3β-HSD6,P450c17a1,and 17β-HSD3 mRNA.ELISA assay was performed to measure the levels of testosterone and luteinizing hormone (LH) in serum,and the enzymatic activities of 3β-HSD1,1P450c17 and 17β-HSD3 in testis tissue.Results Compared to control group,the levels of testosterone and LH of diabetic group declined significantly (P < 0.05) after four weeks.The mRNA levels of LHR,StAR,CYP11a1,Hsd3b6,CYP17a1 and Hsd17b3,and enzymatic activities of 3β-HSD6,P450c17 and 17β-HSD3 were also decreased significantly compared with control group (P < 0.05,0.01 and 0.001).Conclusion Expression of key enzymes of testosterone synthesis in testis of early diabetic mice decreases significantly.
10.The synthesis of purine derivatives and its inhibitory activity on CD38 NADase.

CD38 is a multifunctional enzyme expressed in a variety of mammalian tissues, its catalytic activity was involved in a wide range of physiological processes. Based on the reported inhibitor of human CD38 NADase, 33 purine derivatives were designed and synthesized. The biological activity assay showed that compounds 20 and 38 exhibited almost the same extent of inhibitory activities on human CD38 NADase as the lead compound H2. The results also revealed that small substituents at C-6 of purine ring gave no obvious effect on inhibitory activity, but phenylpropionyl moiety at N-2 could affect the binding mode of the compound with CD38. This study provides a reliable basis for future rational design of inhibitors for CD38.
