1.Study Progress in Inhalation Drug Delivery Systems

Objective:To review the research progress and practical achievements in inhalation drug delivery systems. Methods:Based on the recently published papers and international conference papers,the research on the application of inhalation drug delivery systems was summarized and analyzed. Results:The advantages of inhalation drug delivery systems were illustrated. The advantages and drawbacks of different inhalation devices were reviewed,the guidelines for the selection and application were introduced,and the development direction of inhalation therapy was reviewed as well. Conclusion:Inhalation drug delivery systems show good application prospect.
2.Analysis of Glucocorticoid Outpatient Prescriptions in a Hospital during 2013-2015

OBJECTIVE:To provide reference for rational use of glucocorticoid in the clinic. METHODS:The glucocorti-coid outpatient prescriptions collected from clinical departments of a hospital during Jan. 2013 to Jun. 2015 were analyzed statisti-cally in respects of glucocorticoid use,department distribution,DDDs,distribution of prescription diagnosis,etc. RESULTS:Of 15 000 prescriptions,there were 1 562 glucocorticoid prescriptions,with utilization rate of 10.4%;189 prescriptions were recog-nized as irrational ones,with irrational rate of 12.1%. The proportion of glucocorticoid in otorhinolaryngology department was the highest(27.66%),and irrational drug use was the highest in pediatrics department(16.12%);Dexamethasone injection was the most widely applied(44.88%);most of glucocorticoid prescriptions were used for acute bronchitis and bronchitis(267 pre-scriptions). CONCLUSIONS:The irrational glucocorticoid application has existed in the hospital,which deserved special atten-tion. Hence,prescription check and evaluation should be strengthened to promote the safety and efficacy of glucocorticoid in clini-cal application.
3.Autologous hamstring Rigidfix and Intrafix fixation for reconstruction of anterior cruciate ligament

BACKGROUND:Autologous hamstring Rigidfix and Intrafix has been frequently used to fix and reconstruct anterior cruciate ligament. However, it lacks of evaluation of middle and long period of clinical outcomes. OBJECTIVE:To retrospectively evaluate the medium term clinical outcomes of anterior cruciate ligament reconstruction with hamstring tendon autograft by Rigidfix and Intrafix. METHODS:The 39 cases of anterior cruciate ligament injury were subjected to anterior cruciate ligament reconstruction with hamstring tendon autograft by Rigidfix and Intrafix under arthroscope. They were fol owed up for 2 years or more. The clinical outcomes were evaluated using Lyshlom score scale, IKDC2000 and Tegner score scale. RESULTS AND CONCLUSION:In the fol ow-up, IKDC2000 score and Lyshlom score were significantly increased fol owing Rigidfix and Intrafix fixation than before treatment (P<0.01). Results indicate satisfactory clinical outcomes of anterior cruciate ligament reconstruction with hamstring tendon autograft by Rigidfix and Intrafix. However, long-term fol ow up is needed to verify the feasibility of extensive application.
4.Therapeutic Observation of Zhuang’s Medicated Thread Moxibustion plus Acupoint Thread Embedding for Allergic Rhinitis

Objective To observe the efficacy of Zhuang’s medicated thread moxibustion plus acupoint thread embedding in treating allergic rhinitis.Method By using random number table, 103 patients with allergic rhinitis were randomized into a treatment group (n=53) and a control group (n=50). The treatment group was intervened by Zhuang’s medicated thread moxibustion and acupoint thread embedding, while the control group was by acupoint thread embedding alone. The subjects were treated for 6 courses with a one-month follow-up. The therapeutic efficacies were compared between the two groups.Result The total effective rates were respectively 94.3% and 80.0% in the treatment group and control group.Conclusion Zhuang’s medicated thread moxibustion plus acupoint thread embedding has significant advantage in improving the symptoms of allergic rhinitis.

6.Effects of puerarin on MDA and MPO in myocardial in earlier period of scalded rats

Objective To observe the effect of puerarin on the level of malondialdehyde (MDA) and myeloperoxidase (MPO) in myocardial in scalded rat.Methods Eighty-eight Wistar rats were randomly divided into the recovery group (group R,n=40),the treatment group (group T,n=40) and the control group (group C,n=8) . The rats in the recovery and the treatment groups were subjected to 30% TBSAⅢdegree scald.Myocardial tissue samples from the group R and group T were harvested at 1,3,6,12,24 postburn hours for the determination of MDA and MPO. The morphological change in the myocardial tissue was observed with transmission electronic microscope.Results (1)In group R,MDA、MPO went up 1 hours after burns,and all attaining the top at 12 hours post burn (P<0.01).(2)In group T,the indexes above had the same trends as group R.But comparing with group R,MPO and MDA were much lower at 1,3,6,12 hours (P<0.05 or P<0.01).(3)Comparing with group R,the altrastructural changes were obviously alleviated at 24 hours in group T.Conclusion The production of MDA,MPO in severely burned rats can be inhibited by puerarin,which was beneficial in the management of myocardial injuries after severe burns.
7.Percutaneous endovascular angioplasty for the treatment of arteriosclerosis obliterans of the lower extremities showing no outflow tract visualization:a preliminary exploration

Objective To investigate the clinical efficacy and value of percutaneous transluminal angioplasty (PTA) in treating arteriosclerosis obliterans (ASO) of lower extremity that shows no visualization of its vascular outflow tract. Methods A total of 19 patients with ASO of lower extremities showing no outflow tract visualization were included in this study. The patients included 11 males and 8 females with a median age of 66 years (48-79 years. The main clinical symptom was rest pain of the leg in all patients; 10 cases had different degrees of foot ulcer and gangrene. Preoperative multi-slice CT angiography and DSA examination were performed in all patents. After PTA, the patients were followed up for 12 months, the changes of the clinical symptoms and signs were recorded. The claudication distance, toe skin temperature, ankle-brachial index (ABI), toe-brachial index (TBI), and CT angiography or color Doppler examination were separately performed before and one, 3, 6 and 12 months after PTA. Results In the 19 patients (38 diseased limbs in total), ASO with no visualization of vascular outflow tract was detected in 54 arteries, 3%), posterior tibial artery (n=18, 33.3%), peroneal artery (n=9, 16.7%) and popliteal artery (n=2, 3.7%). The technical success rate of limb PTA was 89.5%, while the technical success rate of single artery PTA was 85.2%. After PTA, the skin temperature of all the diseased limbs that had been successfully treated was obviously improved, the pain was significantly relieved or disappeared. One, 3, 6 and 12 months after PTA, the claudication distance, toe skin temperature, ABI and TBI were strikingly improved when compared with those determined before PTA (P<0.05). Twelve months after PTA the vascular restenosis rate was 39.5% (15/38), the limb vascular patency rate was 55.3% (21/38), and the limb salvage rate was 81.6%(31/38). Conclusion For the treatment of lower extremities arterial obliterans with obstructed outflow tract, PTA is safe and effective in short-term period. Although its long-term restenosis rate is higher, this technique can effectively control the progress of the disease, relieve the clinical symptoms, and help improve the limb salvage rate of ischemic limbs.
8.The anti-aging study of human umbilical cord mesenchymal stem cells combined with lycopene treatment in the aging beagles

BACKGROUND:Previous tudies have shown that the anti-aging effects of stem cel s with lycopene are more significant, and can also significantly improve the aging body immune function.
OBJECTIVE:To investigate the anti-aging effects of human umbilical cord mesenchymal stem cel s combined with lycopene on the aging beagles.
METHODS:Sixteen aging beagles (6-7 years old) were randomly divided into two groups:aging control group and aging treatment group;young beagles (3-4 years old) were chosen as young control group. In the aging treatment group, human umbilical cord mesenchymal stem cel s combined with lycopene was given;while in the other two group, an equal amount of DMEM/F12 cel culture medium and sunflower oil was given. Each dog's general conditions were observed regularly during the whole progress. The changes of superoxide dismutase,
malondialdehyde, glutathione peroxidase in the serum were detected at regular time of the whole process, and the structure changes of each organ were observed at 24 weeks of treatment.
RESULTS AND CONCLUSION:(1) Before treatment, the levels of superoxide dismutase and glutathione peroxidase in the aging control and treatment groups were lower than those in the young control group (P<0.05), while the malondialdehyde content was higher than that in the young control group (P<0.05). However, there was no significant statistical difference between aging control and treatment groups (P>0.05). (2) For the aging treatment group at 24 weeks of treatment:the beagle fur became clearer and smoother, motility was strengthened, appetite became better;and the activity of superoxide dismutase in serum at 8 to 24 weeks of treatment increased significantly compared with before treatment (P<0.05), the activity of glutathione peroxidase significantly increased from 6 weeks (compared with treatment before, P<0.05), the malondialdehyde content decreased significantly from 4 weeks of treatment to the completion of the experiment (compared with before treatment, P<0.05). (3) After the experiment, the microscopic observation showed that compared with the aging control group, the tissues and organ structures of the aging treatment group were al clear, had no inflammatory infiltrates, no obvious necrosis and fibrosis lesions. These results were mainly consistent with the observations of young control group. The above results show that umbilical cord mesenchymal stem cel s combined with lycopene therapy on the natural aging beagles may enhance the activities of antioxidant enzymes, reduce malondialdehyde content, and their combination also can repair tissue structures and promote the functions, which has obvious anti-aging effects.
9.Analysis of haplotype and linkage disequilibrium of PPARγgene rs3856806, rs12490265, rs1797912, and rs1175543 in patients with metabolic syndrome in Kazakhs of Xinjiang

Objective To investigate the effect of haplotype and linkage disequilibrium of PPARγgene rs3856806, rs12490265, rs1797912, and rs1175543 in patients with metabolic syndrome ( MS) in Kazakhs of Xinjiang.Methods MALDI-TOF-MS was used to detect PPARγgene rs3856806, rs12490265, rs1797912, and rs1175543 genotypes in 489 subjects ( including 245 MS and 244 controls ) .Results ( 1 ) The frequencies of rs3856806T, rs12490265A, rs1797912C and rs1175543G alleles for MS group in Kazakhs were all significantly lower than those for controls [ rs3856806T allele:12.53% vs 17.01%; rs12490265A allele: 31.84% vs 38.52%;rs1797912C allele:35.31%vs 43.24%;rs1175543G allele:40.61%vs 47.54%(all P<0.05)].(2)Significant linkage disequilibrium were observed between PPARγgene rs1797912 and rs1175543, rs12490265, and rs1175543 polymorphisms.(3)AGCC and GAAT were significantly different between MS and control group in Kazakhs(both P<0.05).(4) Carrying rs3856806T, rs12490265A, rs1797912C, rs1175543G was 0.267 times that of carrying rs3856806C, rs12490265G, rs1797912A, rs1175543A.Conclusions The PPARγgene rs3856806, rs12490265, rs1797912 and rs1175543 polymorphisms were associated with metabolic syndrome in Kazakhs.There were very strong linkage disequilibrium between PPARγgene rs1797912 and rs1175543, rs12490265 and rs1175543 polymorphisms, The AGCC haplotype and GAAT haplotype may serve as protective factors of metabolic syndrome.Carrying rs3856806T, rs12490265A, rs1797912C, and rs1175543G may confer lower risk of MS in Kazakhs.
10.Human umbilical cord mesenchymal stem cells combined with immunotherapy for the treatment of type 1 diabetic mice

BACKGROUND: Type l diabetes mellitus is a T cell-mediated autoimmune disease resulting in pancreatic islet cell damage. In this study, immunotherapy was used to deal with type l diabetes mellitus and stem cell transplantation was used to repair damaged islet p cells, attempting to explore a new treatment for type l diabetes mellitus. OBJECTIVE: To observe the efficacy of human umbilical cord mesenchymal stem cells combined with immunotherapy for the treatment of type l diabetic mice. METHODS: Fifty BALB/c Foxp3-DTR-EGFP positive mice were selected, six of which were randomly selected as normal control group and the remaining of which were intraperitoneally injected with streptozotocin and diphtheria toxin to prepare an animal model of type l diabetes mellitus. After successful modeling, randomization was performed in model mice and there were four groups: model group (normal saline), immunotherapy group (subcutaneous injection of dexamethasone (10 μg) and insulin (10 μg) mixture), cell transplantation group (injection of human umbilical cord mesenchymal stem cells (1 X106 cells per mouse) through the tail vein, and combined treatment group (the combination of immunotherapy and cell transplantation as described above). At 4 weeks after treatment, changes in blood glucose, C-peptide, body mass, pancreatic histopathology and insulin-positive area were observed in each group. RESULTS AND CONCLUSION: (1) Compared with the normal control group, the blood glucose level of the model group increased (P < 0.01) the C peptide level and body mass decreased (P < 0.01), and the islet was severely atrophied, with decreased number of islet cells and reduced insulin-positive area. (2) Compared with the model group, the blood glucose level of the immunotherapy group decreased (P > 0.05), the C-peptide level and body mass did not change significantly (P > 0.05), the islet cells increased in number, and the insulin-positive area increased. (3) Compared with the model group, the blood glucose level of the cell transplantation group and the combined treatment group decreased (P > 0.05), the C peptide level and body mass increased (P < 0.05), the islet cells increased in number, and the insulin-positive area increased. These findings reveal that either human umbilical cord mesenchymal stem cells or immunotherapy can improve the islet function of type l diabetic mice, and the combination treatment has better outcomes.
