1.Detection of Treponema pallidum by immunohistochemistry in secondary syphilitic lesions

ObjectiveTo evaluate the diagnostic value of immunohistochemistry(IHC) for the identification and localization of Treponema pallidum(TP) in secondary syphilitic lesions.MethodsSkin tissue specimens were obtained from the lesions of 25 patients with secondary syphilis and 15 patients with dermatoses unrelated to TP infection,followed by fixation and embeding.IHC using a polyclonal antibody against TP and Warthin-Starry (W-S) silver stain were carried out respectively to detect TP in these specimens.Results TP was detected in 80.00% (20/25) of the specimens by IHC,44.00% by W-S silver stain (Fisher's Exact Test,P =0.046).Of the 20 IHC-based TP-positive specimens,all harbored TP in the epidermis,11 also in the dermis.The density of TP was associated with the types of skin lesions,and sequentially decreased from condyloma latum to papules,maculopapules and maculae(x2 =15.694,P =0.011 ).Spirochetes were not seen in any of the control lesional specimens.ConclusionsIHC is superior to traditional W-S silver stain for detecting spirochetes in secondary syphilitic lesions,and is of great value to the diagnosis of secondary syphilis.The accurate localization of TP by IHC may facilitate the study on the formation of syphilitic lesions.
2.THE 3D VISUALIZATION OF THE PULMONARY VENOUS SYSTEM OF CHINESE PEOPLE

Objective To build a digitized 3D model of the pulmonary venous system of Chinese people for the purpose of providing anatomic information of the pulmonary venous system for anatomic teaching,imaging diagnosis and surgical operation of pulmonary diseases. Methods The primal images from the first Chinese visible human data set of chest were converted into image sequence only cantaining the sections of pulmonary veins after manual registration and image segmentation.Then these images were imported into 3DMed software to build a 3D model of the pulmonary venous system through threshold registration algorithm. Results The three dimensional structures of the pulmonary venous system were reconstructed entirely.All reconstructed structures could be displayed individually or jointly,and could be rotated continuously on any plane.Conclusion The branches and the spacial characters of the pulmonary venous system can be clearly shown on the reconstructed 3D model.It will help anatomy teaching and provide morphological data for image diagnosis and lung operations.
3.Determination of the Content of Ceftazidime in Renal Dialysate Effluent in Burn Patients

OBJECTIVE:To determine the content of ceftazidime in renal dialysate effluent in burn patients.METHODS:HPLC was used.The sample was separated on Bondapak-C18 analysis column with a mobile phase of mixture of methanol-ammonium acetate(17∶83,V/V).The flow rate was 1.0ml/min.The UV detector wavelength was set at 254nm.RESULTS:A linearity was obtained from 0.5~100?g/ml with a good correlation(r=0.9 999).The RSDs of inter-day and intra-day precision were not more than 3.The average recovery was 100.30%.CONCLUSION:The method is simple,rapid and accurate.
4.Mycoplasma contamination decreases 28S and 18S RNC-rRNA via p38 MAPK pathways in human cell lines

AIM:To evaluate the effect of mycoplasma on the rRNA composition of mature ribosomes in human cell lines.METHODS:We isolated ribosome nascent-chain complex ( RNC) from multiple mycoplasma-positive-negative human cell lines.RNC-RNA was acquired from each cell line through RNA extraction, followed by agarose gel separation, fluorescent visualization and gray-scale value measurements on the rRNA bands.Western blot analysis was performed to in-vestigate the MAPK pathway alterations.RESULTS:In various human cell lines derived from different tissues, we found that as compared with mycoplasma-negative cells, mycoplasma-positive cells showed aberrant RNC-rRNA band patterns, featured by the decreases in 28S and 18S eukaryotic rRNAs and the increases in 16S and other unknown prokaryotic rRNAs.When cultured without ciprofloxacin, mycoplasma-negative HBE cells acquired mycoplasma contamination as ob-served with such characteristic abnormal rRNA bands.The ciprofloxacin treatment was able to recover the RNC-rRNA bands of the mycoplasma-contaminated cells to the normal pattern.The results of Western blot analysis on total and phos-phorylated proteins indicated that p38 pathway was significantly deactivated in mycoplasma-infected A549 cells, while ERK1/2 pathway was not significantly altered.CONCLUSION:Mycoplasma contamination severely alters the RNC-rRNA composition via p38 MAPK pathway, thus seriously impacting on the host cell translational behaviors.
5.Lentivirus-mediated inhibition of tumor necrosis factor-alpha effect in expression of Caspase-3 and Bcl-2 in spinal cord contusion rats

Objective To explore the change of apoptosis factor Caspase-3 and Bcl-2 in the injured segment of rat with spinal cord injury after inhibiting lentivirus expression of inflammation factor TNF-α. To study the relationship between Caspase-3, Bcl-2, Bax and TNF-α in spinal cord injury. Mthods Spinal cord contusion model was prepared by Allen method. The relation between tumor necrosis factor alpha and Bcl-2, was predicted by the method of GeneMANIA bioinformatics. The RNA which was packaged by lentivirus constructed the RNA interference model of tumour necrosis factor alpha. After interference of tumor necrosis factor alpha, we used the method of QRT-PCR to assays the mRNA expression of Caspase-3 and Bcl-2 in spinal cord and detect of the localization of Caspase-3 and Bcl-2 by immunohistochemistry. Statistical analysis with SPSS17.0. Results SD rats had paraplegia and urinate retentaion because of spinal cord injury. The result of QRT-PCR showed that in the seventh day after SCC, the expression of Caspase-3 reduced significantly (P < 0.05) and Bcl-2 did not change significantly (P > 0.05). Immunohistochemistry experiment results showed that Caspase-3 Bcl-2 and Bax immunoreactive cells were observed in the neurons and glial cells of both white matter and gray matter in the spinal cord. The results were the same with QRT-PCR.. Conclusion TNF-α in rats after SCC can effectively regulate the ratio of Bcl-2 and Bax , and then regulate the expression of Caspase-3 , which may affect the function of apoptosis and function recovery after spinal cord injury.
6.Study on water-soluble iron, heavy metals and harmful elements of Magnetitum.

OBJECTIVETo measure the contents of the water-soluble iron, five heavy metals and harmful elements in Magnetiturn and provide a basis for the quality control and safety evaluation of Magnetitum.
METHODIron (Fe), lead (Pb), cadmium (Cd) and copper (Cu) were determined by atomic absorption spectrometry (AAS); arsenic (As) and mercury (Hg) were determined by atomic fluorescence spectrometry (AFS).
RESULTThe mean content of element iron is 764.30 mg x kg(-1). The contents of five water-soluble heavy metals and harmful elements in Magnetitum were within the safety range. The recovery of the standard addition was in the range of 93.7% - 110.6%, and the RSD was less than 5.0%.
CONCLUSIONAnalyzing the water-soluble iron, heavy metals and harmful elements in Magnetitum is effective to the quality control and the safety evaluation of magnetitum.

7.Crystal structure and functional implication of the RUN domain of human NESCA.

NESCA, a newly discovered signaling adapter protein in the NGF-pathway, contains a RUN domain at its N-terminus. Here we report the crystal structure of the NESCA RUN domain determined at 2.0-Å resolution. The overall fold of the NESCA RUN domain comprises nine helices, resembling the RUN domain of RPIPx and the RUN1 domain of Rab6IP1. However, compared to the other RUN domains, the RUN domain of NESCA has significantly different surface electrostatic distributions at the putative GTPase-interacting interface. We demonstrate that the RUN domain of NESCA can bind H-Ras, a downstream signaling molecule of TrkA, with high affinity. Moreover, NESCA RUN can directly interact with TrkA. These results provide new insights into how NESCA participates in the NGF-TrkA signaling pathway.

Objective To observe the clinical efficacy and safety of 5% imiquimod cream in topical treatment of anogenital warts. Methods A randomized, double-blind, parallel placebo-controlled clinical study was conducted. Patients with anogenital warts were instructed to apply the test drug topically and then clean the drug with water 6 ~ 8 hours later, three times a week for 8 weeks. Patients whose warts cleared completely were followed up for one month to determine recurrence rates. Results Two hundred and thirty-one patients with anogenital warts were enrolled in this trial. One hundred and sixteen patients were randomly selected to receive 5% imiquimod cream; and the other receive placebo cream. 9.Clinical study of endoscopic underwater cold snare polypectomy treating small colorectal polyps

Objective To explore the clinical effect of underwater cold snare polypectomy(CSP)for the treatment of small colorectal polyps.Method A total of 186 patients with small colorectal polyps(5~10 mm)admitted to the hospital from July 2021 to June 2022 were selected for the study.They were randomly divided into underwater CSP group(n=93)and traditional CSP group(n=93).To compare the polyp surgery time,polyp complete resection rate,specimen retrieved rate,consumed clips,intraoperative and postoperative bleeding complications,and recurrence between the two groups.Results There were 156 polyps in underwater CSP group and 140 polyps in traditional CSP group.The complete resection rate in underwater CSP group was significantly higher than that in traditional CSP group(95.51%and 89.29%),the difference was statistically significant(P<0.05);The incidence of intraoperative bleeding in underwater CSP group was significantly lower than that in traditional CSP group(13.46%and 23.57%),the difference was statistically significant(P<0.05);The specimen retrieval rate in underwater CSP group was significantly higher than that in traditional CSP group(99.36%and 95.71%),the difference was statistically significant(P<0.05);The surgical time of single polyp in underwater CSP group was significantly shorter than that in traditional CSP group[(86.23±33.66)and(111.77±40.06)s],the difference was statistically significant(P<0.01).There were no significant differences in terms of consumed clips,postoperative bleeding incidence,and polyp recurrence rate between the two groups(P>0.05).There were no cases of perforation in either group.Conclusion Underwater CSP is a safe and effective treatment for small colorectal polyps,with a higher complete resection rate,lower intraoperative bleeding rate,and shorter surgical time.
