1.Tea Polyphenol Influences Blood Plasma Endotoxin Level in Alcohol Liver Hurted Rat Model

OBJECTIVE To investigate the influence of tea polyphenol in alcoholic liver(hurted) rat by observing its blood plasma endotoxin level,and to provide the direction of new(treatments) and means for the prevention of alcoholic liver disease.METHODS Thirty S-D rats(15 female,15 male) were divided into five groups randomly(noted as A,B,C,D,and E,with equal male and female),and fed in the cleanliness class laboratory.Excepting group E which was given saline,all other groups were given alcohol(56% Hongxing spirits) with the dosage of 7 g/kg once everyday.At the same time,rats in groups B,C and D were given tea polyphenol with the dosage of 0.05,0.125,0.25 g/kg.In the end of the fourth week,got the rats vein blood to determinae serum ALT,AST level and blood plasma endotoxin level.RESULTS The serum AST of the group A was higher than other 4 groups;the blood plasma endotoxin level in groups B,C,and D were(consequently) lower than in group A.CONCLUSIONS The tea polyphenol could reduce the blood plasma endotoxin level in alcoholic liver hurted rat.Its possible mechanism is to repress the isotopy of bowel flora,and to maintain the normal function of bowel mucous membrane.This research provides a new way for curing the alcohol liver hurt.
2.Status of Traffic Pollution in Main Crossroads of Changsha

Objective To understand the traffic pollution in main crossroads of Changsha. Methods The levels of noise, NO 2, O 3 and TSP and traffic volumn were monitored in 6 main crossroads of Changsha in situ, 3 times per day, continuously for 5 days. Results The traffic volumns were 1 203~3 715/h, the noise levels were 69~88 dB(A), the levels of NO 2, O 3 and TSP in air of 6 main crossroads were 0.092~0.238 mg/m 3, 0.238~0.303 mg/m 3 and 0.71~1.47 mg/m 3 respectively. The over standard rates were 93%~100% for O 3 with a average rate of 98%. Conclusion The traffic pollution in main crossroads of Changsha mainly induced by noise and O 3, which might be associated with the traffic volumn, the environment around the crossroads and the types of the automobile running in the crossroads.
3.Changes of plasma p-selectin and nitric oxide in patients with pregnancy induced hypertension

Objective To study the roles of P-selectin and nitric oxide (NO) on the etiology of pregnancy induced hypertension (PIH). Methods The levels of plasma P-selectin and nitrite and nitrate (NO2-/NO3-) were determined in 36 patients with PIH. The data were compared with those of normal pregnant group (n=18) and healthy non-pregnant group (n=19). The levels of plasma P-selectin were measured by means of enzyme-linked immunosorbent assay (ELISA) and levels of plasma NO2-/NO3- were measured by Griess reaction after the nitrate was reduced by a Cadmium-reduction method. Results The results showed: (1)The level of P-selectin in the normal pregnant group (48.92±15.16) μg/L was higher than that in non-pregnant group [(30.16±8.42) μg/L, P<0.001]. There was no significantly statistical difference in the levels of P-selectin between the normal pregnant group and the mild PIH group (P>0.05). The levels of P-selectin in the moderate and severe PIH group (70.11±17.65) μg/L were higher than those in the mild PIH group [(47.33±17.95) μg/L, P<0.01] and the normal pregnant group (P<0.001). (2)The concentration of NO2-/NO3- in normal pregnant group was higher than that in non-pregnant group (P<0.01). The concentration of NO2-/NO3- in PIH group were lower than was in normal pregnant group (P<0.001). The NO2-/NO3- in mild group was lower than that in normal pregnant group (P<0.001). The Plasma concentration of NO2-/NO3- in moderate and severe PIH group was lower than that of mild group (P<0.05). (3) There was a negative correlation between the plasma concentration of P-selectin and NO2-/NO3- in the PIH group. Conclusion The changes of plasma levels of P-selectin and NO are related to the occurrence and development of PIH.
4.The effects of connective tissue growth factor on bovine lens epithelial cells in vitro

Objective This study was to observe the effects of connective tissue growth factor(CTGF) on migration and transdifferentiation of bovine lens epithelial cells(BLECs). MethodsThe culture and identification of BLECs adopted the method of Hu(reference 1).The 2-3 passages of BLECs were collected and used in this experiment at the concentration of 1×10~6 cells/hole.The free-serum DMEM containing 0.1 ng/L,0.5 ng/L,1.0 ng/L of CTGF was added into medium for 24 hours in different experimental group respectively,and only equal volume of free-serum DMEM was added in control group.Expression of α-smooth muscle actin(α-SMA) mRNA and protein in the BLECs were examined by semiquantitative RT-PCR and Western blot,respectively.The transwell inserts were used to evaluate the migration ability of BLECs. ResultsThe expression of α-SMA mRNA in cultrued BLECs was gradually increased in different concentrations of CTGF groups.Compared with control group,the expression of α-SMA mRNA in experimental group was significantly enhanced (F=66.56,P<0.01).The expression of α-SMA protein followed the same pattern(F=65.43,P<0.01).The migration ability of BLECs was obviously elevated after CTGF stimulation under the light microscope.The migration rate of BLECs was considerably increased in experimental group compared with blank control group (t=51.7,P<0.01).ConclusionCTGF promotes the migration and transdifferentiation of BLECs at a dose-dependent manner in vitro.CTGF plays an important role in the formation of posterior capsule opacification.
5.Effect of tTG inhibitor on the expression of FN and Col-Ⅳ induced by TGF-β_2 in human lens epithelial cells

BackgroundOur previous research and other reports disclosed that the expression of tissue transglutaminase(tTG)in lens epithelial cells(LECs) of patients with cataract is enhanced,indicating tTG is related to formation of posterior capsule opacification(PCO).ObjectivePresent study is to observe the effect of tTG specific inhibitor monodansyl-cadaverineon(MDC) on the expression of fibronectin(FN) and collagen Ⅳ(Col-Ⅳ) induced by TGF-β_2 in human LECs.MethodsHLE-B3 was cultured in vitro in DMEM containing 10% fetal bovine serum and then were divided into 5 groups.The free-serum culture was used as normal control group.Free-serum culture containing 10μg/L TGF-β_2 was utilized as treatment group.10μg/L TGF-β_2 plused 100μmol/L,200μmol/L and 400μmol/L MDC respectively in different concentrations as MDC-treatment group.Semiquantitative RT-PCR was used to assay the expression of tTG,FN and Col-Ⅳ in HLE-B3.A(tTG/β-actin),A(FN/β-actin) and A(Col-Ⅳ/β-actin) was calculated separately as the detecting indexes.ResultstTG,FN and Col-Ⅳ were positively expressed in cultured HLE-B3.The expression levels of tTG,FN and Col-Ⅳ in HLE-B3 were remarkably increased in the group with 10μg/L TGF-β_2 compared with normal control group(t=33.95,P<0.01;t=38.24,P<0.01;t=13.48,P<0.01).The expression levels of FN and Col-Ⅳ were gradually declined in 100,200 and 400μmol/L MDC groups in comparison with TGF-β_2 treatment(P<0.01).The significant differences were also found in the expressions of FN and Col-Ⅳ in HLE-B3 among 100,200 and 400μmol/L MDC groups(P<0.01).ConclusionMDC inhibits the expression of FN and Col-Ⅳ induced by TGF-β_2 in human LECs at a concentration-dependent manner.tTG may be involved in the formation of posterior capsule opacification through up-regulating the expressions of FN and Col-Ⅳ in human LECs.
6.Detection and anslysis of infection indicators of 2 576 patients before transfusion

Objective To analyze the patients with infection before transfusion in onder to avoid misdiagnosis and blood transfusion caused by cross-infection and remind medical staff attention to operation and self-protection.Methods HBsAg,Anti-HCV,Anti-TP,Anti-HIV1/2 were detecced by 2 576 cases of pre-operative,prenatal,blood transfusions before the patient's blood ELISA assay.Results 2 576 cases of infectious markers in patients with serum positive rate was 12.15%.The positive rates of was HBsAg,Anti-HCV,Anti-TP,Anti-HIV1/2:11.06%,2.13%,1.67%,0.00%,HBsAg and Anti-HCV were positive for both the percentage of 0.54%.Conclusion The transfusion of pre-infection detection is necessary for patients with early diagnosis and treatment of infectious diseases to avoid and prevent hospital infections,medical personnel,as well as occupational infection to reduce or avoid blood transfusion caused by medical disputes.
7.Flow Cytometry Analysis of 24 Snap-frozen Tissue Specimens of Ovarian Carcinoma

Flow cytometry analysis of DNA ploidy level and S-phase fraction (SPF) was carried out in 24 snap-forzen tissue specimens of ovarian carcinoma. The relationship of DNA ploidy level and SP-F with age,clinical stage, ascites, histologic type,pathological grade,lymphocyte infiltration and psammoma bodies was analyzed.Results showed that the relationship of DNA ploidy level and SPF with ascites and pathological grade was significant. Aneuploid portion and SPF in patients with ascites or pathological grade Ⅲ or Ⅱ were higher than those in patients with no ascites or pathological grade I. The author considers that DNA ploidy level and SPF of ovarian carcinoma can serve as a relatively independent objective index which reflects tumor's biological behaviours,and will be of great auxiliary value to early diagnosis and therapy of ovarian carcinoma.
8.Observation on Decline Phase of Maintenance of Brugia malayiMeriones unguiculatus Model

Objective To observe the degeneration of Brugia malayi in Meriones unguiculatus model. Methods Microfilaria of Brugia malayi derived from Meriones unguiculatus was used to infect Anopheles sinensis . Infective stage larvae (L 3) from mosquito vector were collected and inoculated into abdomen of Meriones unguiculatus. Successive 33 generations of the parasite in the rodent model have been observed. Results Since 1974 when the animal model was established, the parasite has lasted for 33 generations, the positive rate of Meriones unguiculatus with microfilaria gradually reduced from 80% of the 28th generation to 16% of the 32nd generation and finally to 0 at the 33rd generation. Conclusion It became difficult for the larvae of Brugia malayi to develop and/or reproduce in the animal model after multiple inoculations for generations.
9.Effect of glucocorticoid inhaled on bone metabolism, bone density and adrenal function in children with asthma

ObjectiveTo explore the effect of glucocorticoid inhaled on bone metabolism, bone density and adrenal function in children with asthma.Methods60 children were randomly divided into the treatment group and control group with 30 cases in each group, and having 30 pneumonia children as another control. Children of the treatment group were treated with budesonide (200~600 μg/d) inhaled, asthma cases in the control group without budesonide inhaled. Bone metabolism, bone density and adrenal function of children in three groups were compared.ResultsAfter 18 months treatment, there were no significantly differences of bone metabolism, bone density and adrenal function among children of three groups (P>0.05).ConclusionThe treatment of glucocorticoid inhaled is safe to asthma children.
10.Treatment of old acromioclavicular dislocation by reconstruction of acromioclavicular and coracoclavicular ligaments

Objective To explore new treatment methods for old acromioclavicular dislocation. Methods Twenty-nine cases of old acromioclavicular dislocation were treated with augmenting reconstruction of acromioclavicular ligament by coracoacromial ligament and augmenting reconstruction of coracoclavicular ligament by conjoining tendon with biceps brachii muscle and coracobrachialis muscle in coracoid. Twenty-five cases were followed up with follow-up period of 3-36 months. There were 10 cases in grade Ⅱ and 15 in grade Ⅲ according to classification of Allman. Results Of 25 cases with follow-up, 16 were excellent and 9 satisfactory according to the Lazzcano standard. There were no other complications or dislocations. Conclusions Reconstruction of acromioclavicular and coracoclavicular ligaments for treatment of old acromioclavicular dislocation is an efficient method with advantage of rigid fixation and accords with micro-motion physiological function of acromioclavicular joint.
