1.The relationship between enhancement degree on spiral computed tomography and the vascular endothelial growth factor and microvessel density expression in colorectal carcinoma

Objective To investigate the expression of vascular endothelial growth factor (VEGF) and microvessel density (MVD) in colorectal carcinoma, and to study its relationship with the enhancement degree of the lesion on SCT scan. Methods Thirty patients with colorectal carcinoma confirmed by colonofiberscope or operation were examined by SCT, and the CT attenuations were measured. The resected tumor specimens were immunohistochemically stained with CD34 for measuring MVD and stained with VEGF. The results were compared with the enhancement degree of the lesions observed on SCT scan. Results The enhancement correlated well positively with MVD and VEGF of the colorectal carcinoma. The degree of enhancement correlated closely with the blood supply and MVD and VEGF of colorectal carcinoma (r_1=0.5905, P_1=0.0015; r_2=0.4574, P_2=0.0126). It is meaningful to prompt the research-aimed experiments in the laboratory edu-cation of undergraduate students majoring in pharmacy,which means under the guidance of tutors, students collect information,design the experiment procedure and conduct the whole experiment independently. Under this novel mode of laboratory education, students’ abilities of indepen-dent-thinking and comprehensive-experimental conduction are largely improved. Meanwhile,the sense of team-work is enhanced. To sum up,the research-aimed experiments is significantly ben-eficial to training undergraduate students for future scientific researches.
3.Effects of culture supernatant of Toxoplasma gondii on CD4~+CD25~+ T cells in mice in vitro

Objective To explore the effect of culture supernatant of Toxoplasma gondii on CD4+CD25+ T cells in mice in vitro. MethodsCD4+CD25+ T cells were separated from spleens of C57BL/6 mice and incubated in the culture superntant of Toxoplasma gondii. The apoptosis percentage of the CD4+CD25+ T cells were detected by FACScan, and the suppression of the CD4+CD25+ T regulatory cells were examined by 3H-TdR incorporation. ResultsCompared with the CD4+CD25+ T cells incubated with RPMI-1640, (36.90?0.36)% CD4+CD25+ T cells took apoptosis after incubated with the culture supernatant of Toxoplasma gondii for 10 hours,and the Annexin-v positive rate increased by (13.60?2.15)%. Compared with RPMI-1640, the culture supernatant of Toxoplasma gondii incubating the CD4+CD25+ T regulatory cells for 5,10 hours reduced their suppressive potential on the proliferation of the CD4+CD25- T cells significantly. ConclusionsSome composition of the culture supernatant of Toxoplasma gondii might cause apoptosis of CD4+CD25+ T cells and as a result, could reduce their suppression on CD4+CD25- T cells.

To prepare the RNAse-resistant virus particles containing the partial gene fragments of avian influenza virus H5N1 for use as RNA standard and control in RNA virus detection, the genes coding the coat protein and maturase of E.coli bacteriophage MS2 were amplified by PCR and then cloned into prokaryotic expression vector pET32a to construct the intermediate vector pET32a-MS2. In addition, the gene sequences coding hemagglutinin (HA), neuraminidase(NA) and M protein of the H5N1 virus were also cloned separately to the down-stream of plasmid pET32a-MS2, thus constructing the prokaryotic expression vectors pET32a-NS2-HA, pET32a-MS2-NA and pET32a-MS2-M. These recombinant plasmids were then transformed separately to E.coli BL21(DE3) with induction by IPTG. to express the virus-like particles. The virus-like particles observed under electron microscopy were identified by RT-PCR ,while their stability was confirmed by real-time RT-PCR. In this way, the virus-like particles were successively constructed and identified through PCR amplification, enzymolysis identification and sequencing analysis. These virus-like particles observed under electron microscopy appeared to be circular in shape with a diameter of about 50 nm. Their stability was proved to be rather good. From these observations, it is apparent that these virus-like particles can be used as RNA standard and quality control in the detection of avian influenza virus H5N1.

Objective To observe the changes of CD4+CD25+ regulatory T cells in the spleen of mice infected with T.gondii. Methods Twenty-eight female C57BL/6 mice were randomly divided into four groups. Three groups of mice were inoculated intraperitoneally with 104 tachyzoites in 200 ?l sterile PBS. At 2, 4 and 6 days post-infection, the spleens were removed. The expression level of Foxp3 mRNA in splenic CD4+ T cells was quantitated by real-time PCR. The percentage of CD4+CD25+ regulatory T cells in CD4+ T cells was determined by flow cytometry, and the absolute numbers of splenic CD4+CD25+ regulatory T cells and CD4+ T cells were assessed. The fourth group was injected intraperitoneally with 200 ?l sterile PBS as control. Copper is a trace element essential for the maintenance of normal physiological functions in cardiovascular system,and its transport and metabolisms are regulated by various copper proteins such as copper-based enzymes,copper chaperones and copper transporters.The disturbance of copper level or abnormal expression of copper proteins are closely associated with the development of cardiovascular diseases such as atherosclerosis,hypertension,ischemic heart disease,myocardial hypertrophy and heart failure.Thus,intervention of copper ion signaling pathways is expected to be an effective measure for treating cardiovascular diseases.Some copper complexes,such as trientine,copper-aspirinate complex and copper(Ⅱ)diethyldithiocarbamate,have been found to play a role in the prevention and treatment of cardiovascular diseases and possess potential prospects.Exploring the role of copper in maintaining normal cardiovascular status and the potential application of copper complexes in the treatment of cardiovascular diseases may lay a foundation for finding new targets for prevention and treatment of various cardiovascular diseases,and provide new ideas for clinical treatment of cardiovascular diseases.
7.18F-FDOPA PET/CT Semi-Quantitative Analysis in Improving the Diagnostic Efficacy of Parkinson's Disease

Purpose To investigate the diagnostic value of 18F-FDOPA PET/CT imaging and semi-quantitative analysis platform for the diagnosis of Parkinson's disease(PD).Materials and Methods There were 27 healthy controls and 56 clinically diagnosed PD patients,including 33 early PD(Hoehn-Yahr class Ⅰ-Ⅱ)and 23 advanced PD(Hoehn-Yahr class Ⅲ-Ⅳ),underwent 18F-FDOPA PET imaging in Nanjing First Hospital,Nanjing Medical University were consecutively enrolled from January 2018 to December 2019.The striatal to occipital ratio(SORs)in radioactivity was calculated by HERMES BRASS platform,thereby completing the semi-quantitative analysis of the brain based on regions of interest and observing the asymmetry of the striatal subregions in early-stage PD and late-stage PD patients.Using artificial intelligence techniques to perform principal component analysis on the SORs of the striatal subregions in PD group and healthy control group,the degree of data aggregation and the distinguishability between groups were observed.Results The SORs was significantly reduced in the whole caudate,anterior,posterior putamen and striatum of advanced PD patients(t=9.02-11.72,P<0.000 1).The area under the curve was 0.952,0.973,0.995 and 0.982,respectively.Compared with the healthy control group,the loss of striatal asymmetry index(mean)in each subregion of the striatum in early PD group was caudate(7.61±5.50)%,anterior putamen(11.43±8.97)%,posterior putamen(17.17±11.63)%,and whole striatum(10.65±7.46)%,respectively.The uptake of 18F-FDOPA in the striatum of PD patients was significantly reduced,and the most obvious loss of early PD patients was contralateral posterior putamen,with a decrease of 34%.Conclusion The platform semi-quantitative analysis of 18F-FDOPA PET/CT images provides objective semi-quantitative values for early diagnosis and differential diagnosis of PD.Asymmetry in the striatum,especially in the putamen,may be an important parameter for early diagnosis of PD..

Breast cancer is a malignant tumor originating from breast epithelial tissue. Ferroptosis is a novel type of programmed cell death which differs from apoptosis and necrosis. Research found that the accumulation of lipid peroxides in cells, a crucial process of ferroptosis, can be induced by multiple mechanisms. The ferroptosis regulation is closely related to the occurrence and development of breast cancer, and it induced by drugs is a potential and valuable research direction in breast cancer.
9.Control effect of high aspherical lenticule in children and adolescents with low myopia

AIM:To explore the effect of high aspherical lenticule on controlling low myopia.METHODS: Prospective study. A total of 100 patients aged 7 to 12 years old with low myopia who visited our hospital from May 1 to 31, 2022 were collected. They were divided into two groups with 50 cases in each group according to the wishes of patients. The control group was given single vision glasses after optometry, while the study group was given high aspherical lenticule. The myopia progression(absolute value), axial length(AL)growth, transition rate to moderate myopia, and AL negative growth rate over 6 mo and 1 a were compared between the two groups.RESULTS: The myopia progression and the AL growth of study group was lower than that of the control group after 6 mo and 1 a(all P<0.001).The negative growth rate of AL after 6 mo of treatment was significantly higher than that of the control group(P<0.001). The transition rate to moderate myopia between the two groups was not statistically significant(P=0.62); while the transition rate to moderate myopia in the study group was significantly lower than that in the control group after wearing lens for 1 a(P<0.001), and there was no statistically significant difference in AL negative growth rate between the two groups(P=0.12). Compare with single vision glasses, high aspherical lenticule achieved an 88.2% control rate for low myopia progression over 6mo and a 90.0% control rate of AL growth. The control rate for low myopia to moderate myopia was 66.7%; while the control rate of myopia progression growth was 75.6% after wearing lens for 1a, the control rate of AL growth was 69.2%, and the control rate of the transition rate to moderate myopia was 88.9%.CONCLUSION: For children and adolescents aged 7 to 12 with low myopia, high aspherical lenticule was more effective than single vision glasses in controlling myopia, making it one of the optimal choices for myopia control.
10.Effect of ozone oil for prevention and treatment of sorafenib-induced hand-foot skin reactions: a randomized controlled trial.

OBJECTIVE:
To compare the effects of medical ozone oil and urea ointment for prevention and treatment of hand-foot skin reaction (HFSR) caused by sorafenib in patients with hepatocellular carcinoma (HCC).
METHODS:
A total of 99 patients diagnosed with advanced HCC according to National Comprehensive Cancer Network (NCCN) who were scheduled to receive sorafenib treatment for the first time were enrolled in this study between April, 2018 and January, 2020. The patients were randomized into medical ozone oil group (
RESULTS:
Eight patients were excluded for poor compliance or protocol violations, leaving a total of 91 patients for analysis, including 44 in medical ozone oil group and 47 in urea ointment group. Sixteen (36.4%) of patients in ozone oil group developed HFSR, a rate significantly lower than that in urea ointment group (57.4%;
CONCLUSIONS
Medical ozone oil can significantly reduce the incidence and severity of HFSR to improve the quality of life of HCC patients receiving sorafenib treatment.
