1.A case report of childhood Pseudomonas aeruginosa sepsis

Objective To summarize the clinical features of Pseudomonas aeruginosa sepsis in children. Methods We retrospectively review the clinical data of a baby boy with Pseudomonas aeruginosa sepsis and summarize its clinical character-istics. Results A ten-month male infants with onset symptom of fever, irritability, drowsiness, cough and sputum was diagnosed as Pseudomonas aeruginosa sepsis through blood and sputum culture. The baby recovered well after anti-infection treatment. Conclusions Timely and appropriate use of sensitive antibiotics can decrease severe complications and mortality rate of Pseu-domonas aeruginosa sepsis in children.

ObjectiveToinvestigatetheroleofpsoriasininthepathogenesisofpsoriasis.MethodsImmunohistochemicaltechnique(SABC)andflowcytometrywereemployedtodetecttheexpressionofpsoriasinin14patientswithpsoriasis.ResultsTheexpressionofpsoriasinwasup-regulatedinpsoriatickeratinocytesandtheamountofitsexpressionwas23.38%?4.49%.Thepresenceofpsoriasinwaslocatedinthebasalandsuprabasallayersofpsoriaticlesions.Theepidermisinnormalcontrolsdidnotexpresspsoriasin.ConclusionTheoverexpressionofpsoriasininpsoriaticepidermissuggeststhatpsoriasinbeoneofthefactorsleadingtotheinfiltrationofinflammatorycellsinpsoriaticepidermisandberelatedtothehyperproliferationofpsoriatickera-tinocytes.
3.Expression, purification Orai2 protein and preparation, application it's polyclonal antibody

Objective:To prepare GST-Orai2 fusion protein and to prepare polyclonal antibody against Orai2 by immunizing rabbits.To further investigate the function of Orai2,a transmembrane protein,this antibody was used to identify the Orai2 conditional gene knockout mice.Methods:ORF of Orai2 was amplified by PCR and subcloned into pGEX-6p-1 vector.After transforming BL21 (DE3) competent cells,we succeeded in inducing the expression of GST-Orai2 fusion protein using IPTG.Then the GST-Orai2 protein was purified by immobilized Glutathione affinity chromatography and identified by SDS-PAGE.A New England rabbit was immunized with the prepared fusion protein in Freund's adjuvant to prepare specific antibody.Finally,the prepared antibody was identified by Western blot by checking its titer and specificity.Furthermore,we took use of the prepared Orai2 antibody to identify Orai2 conditional gene knockout mice,with comparing to the wildtype ones in the same cage.Results:The purity of purified GST-Orai2 reached to 90% and the concentration was 0.35 mg/ml by BCA kit.We could detect Orai2 protein even in dilution of 1:10,000.Also,the prepared polyclonal antibody agianst Orai2 could detect both overexpressed and endogenous Orai2 protein in mouse-brain,without crossing reaction with Orai1.As well,we found that the Orai2 protein expression was of obvious reduction in Orai2 conditional gene knockout mice,compared with the wildtype ones in the same cage.Conclusion:We successfully obtain the purified GST-Orai2 fusion protein and prepare specific and highly sensitive polyclonal antibody against Orai2.The antibody can be used to detect overexpressed and endogenous Orai2 protein inmouse-brain specifically,and to identify Orai2 conditional gene knockout mice,without any crossing reaction with Orai1.Our work contributes a lot to the future investigation of functions of Orai2.
4.A precise tooth preparation technique assisted with quantitive bur and microscope.

Although traditional tooth preparation techniques (e.g., depth-groove-guided and index-guided techniques) are designed to improve preparation precision, the results are unsatisfactory because of the lack of proper estimating tools. This study proposed a novel technique, in which relevant details for preparation of drilling holes are provided and corresponding depth is estimated using a quantitive bur under a microscope. This technique offers a viable option for precise tooth preparation.

5.The Apoptosis Effect of Docetaxel Combined with Gamma Knife on Hepatoma Cells Xenograft in Nude Mice

Objective To explore the apoptosis effect of docetaxel combined gamma knife on hepatoma cell SMMC-7721 subcutaneous xenograft in nude mice.Methods Subcutaneous xenogyaft models were constructed and were divided into two groups:control group and experimental group.The experimental group was treated with docetaxel 60ug/0.3ml once every 3 days for 6 times and gamma irradiation once every other day for 6 times (with indoor temperature of 137Cs radiation source irradiating the tumor and of fractionated schedule 5Gy with the total dose of 10Gy every time).The control group was treated with physiological saline with the same dose of 60 ug/0.3 mL.Tumor growth was observed.Tumor samples were cut 30 days after the treatment and TUNEL was used to detect the apoptosis of tumor cells.Results Tumor growth rate in experimental group significantly slowed down.Apoptotic index in experimental groups was significantly higher than that in control group (P < 0.05) Cornclusion Docetaxel combined gamma knife can inhibit the growth of hepatocellular carcinoma in nude mice.
6.Preliminary study on the mechanism of reading recovery in a pure alexia by using functional MRI

Objective To observe the changes of the brain function during reading recovery by using functional MRI (fMRI),and to provide the experimental data in elucidating the mechanism on the recovery of reading and language function.Methods fMRI was performed in a native Chinese patient with pure alexia on the 45 th and 130 th day after the onset,respectively.Three kinds of Chinese characters were presented during the scan and the patient was asked to make the judgement weather he could recognize the characters or not.The brain activation maps were acquired after postprocessing,and the activated location and volume were compared between the first and second experiments.Results In both experiments,Broca area,Wernicke area,and the right extrastriate were significantly activated,while the left extrastriate around the lesion was markedly activated only in the second experiment,and the volume of activation in the right extrastriate in the second experiment was about 3 times as large as that in the first experiment.Conclusion The left extrastriate cortex is one of the key areas responsible for reading function in the brain.The recovery of reading function can be compensated in contralateral corresponding cortical area,or it can be the result of reorganization in ipsilateral peri-lesion cortex.Both mechanisms may simultaneously play important roles in reading recovery.
7.The effects of social isolation on the exploring behavior and working memory in mice

Objective To study the effect of social isolation( SI) on the exploratory behavior and working memory in mice. Methods The Kunming mice of postnatal 21 days were divided into the control group,SI 2 weeks group,SI 2 weeks gregarious group,SI 4 weeks group and SI 8 weeks group,according to randomized design with ten animals each. All isolated mice were isolated for 2, 4 and 8 weeks respectively, the gregarious group were housed under normal grouped housing enviroment after isolation until adult, the mice with the relative same age were control groups. All animals were measured for exploratory behavior and working memory by performing open field and T?maze after the treatment. Results In the open field,compared to the relative control group,the central area of the total time in the SI 4 weeks group(0.07±0.04) was less than the control (0.10±0.04) obviously. The central area percentage of total time in SI 8 weeks group (0.64±0.12) were more than the control (0.43±0.08). In the T?maze,the alteration times in SI 2 weeks group (first day (5.92±0.79),second day (6.67±1.3),third day (7.42±1.08),fourth day (8.17±1.27)) were less than the control (first day (6.80±1.14); second day (7.60± 0.84);third day (8.30±0.95);forth day (9.20±1.32)). However,the alteration times of gregarious group showed no obvious change. Both the alteration times of SI 4 weeks (8.18±1.99) in the second day and that of SI 8 weeks (8.29±3.04) in the forth day were more than the control (6.60±2.11) and (7.80±2.53) respectively.Conclu?sions Working memory of SI 2 weeks rats decrease,which can be improved by the resocialization.SI 4 weeks and 8 weeks rats show the decreasing exploring ability and increasing anxiety and work memory.
8. The visualization analysis of research hotspot of lower respiratory disease nursing SCI paper

Objective:
To grasp the research focus and development situation of the disease through the Web of Science database core journals of nearly ten years in nursing the lower respiratory disease is combing with visual analysis literature, as the nursing researchers further provided a reference for the research of this field.
Method:
Using literature metrology method and information visualization software Citespace, from 2008 to 2017 Web of Science a stronger core journals on the respiratory system disease, a total of 2557 articles were visualized analysis, the hot spot and research frontier of knowledge graph were draw.
Results:
The volume of lower respiratory disease in nursing has risen steadily in the past 10 years. The research focuses on pneumonia, asthma, mortality, self-management, risk factors for care and prevention, etc. The research frontier is mainly focused on the research of pulmonary embolism, deep vein thrombosis and other advanced diseases and children asthma. China and the United States have different research hot spots for lower respiratory disease in the same time zone, and China′s research hot spot is five years later than in the United States.
Conclusion
The research focus of lower respiratory disease in nursing field in the past ten years is mainly centered on the two centers of pneumonia and asthma. Lower respiratory disease in the field of nursing research China steadily rising trend, but compared with developed countries such as United States, England, the total number too little.So, China should draw lessons from foreign developed countries research hot spot and front, increase investment in the nursing of lower respiratory disease research.
9.The Study on the Distribution of High-quality Medical Resources in China from 2012 to 2021 Based on Agglomeration Degree and Theil Index

Objective It aims to analyze the resource distribution of Chinese gradeA teritary hospitals across six regions from 2012 to 2021,so as to further understand the changes in inter-regional health resource allocation.Methods Based on the"Thirteenth Five-Year Plan"for the Establishment of National Medical Centers and Regional Medical Centers,it utilizes indicators such as the annual growth rate,coefficient of variation,agglomeration degree,and Theil index to explore the resource allocation in Chinese gradeA teritary hospitals from 2012 to 2021.Comparative and trend analyses are also conducted.Results The regional difference of high-quality medical resources in the country has continued to narrow,especially with an increased rate of reduction since 2017.The Theil index of resource allocation in grade A teritary hospitals in various regions of the country has decreased from 0.035 0 to 0.020 5,indicating a positive trend towards fairness and a gradual increase in regional development coordination.An imbalance in the geographical distribution of resources among third-class medical institutions persists across various regions of China.Conclusion The resource allocation of grade A teritary hospitals in the western region should fully consider geographical factors and change the traditional single"human-oriented"resource allocation mode.Grade A teritary hospitals in economically developed areas still need to increase resource investment,conduct proactive population monitoring,early warning,and macro resource planning.The resource allocation of third-class medical institutions should focus on both intra-regional development and inter-regional collaboration to enhance the quality and efficiency of resource distribution.
